Effects of ST36, BL21 on the Serum Gastrin Level, Endocrine Cells and Mucus of Gastric Mucosa In Rats (족삼리(足三里) 위유(胃兪) 혈(穴)이 흰쥐 혈중 Gastrin 농도, 위점막의 내분비세포 및 점액에 미치는 영향)

  • Objective : The purpose of this study is To investigate the effects of acupuncture and moxibustion at Zusanli(ST36) and Weishu(BL21) Methods : serum gastrin level by radioimmunoassay was measured at 5 days after acupuncture and moxibustion of those acupoints. Gastric endocrine cell(G cell and Histamine immunoreactive density) by avidin-biotinylated complex(ABC) technique, histological examinations(Alcian Blue-PAS Stain; Alcian blue-Periodic Acid Schiff reagent) of the gastric mucosa were also performed. Acupuncture applied to the ST36 acupoint increased gastrin level of serum, but moxibustion did not produced significant effect. All of acupuncture and moxibustion at BL21 acupoint increased gastrin level of serum significantly. In moxibustion at ST36 and BL21, the number of gastrin secreting cells in gastric mucosa, the density of immunoreactive histamin secreting cells and the density of body mucosa stained by PAS were decreased compare to acupuncture at ST36 and BL21. In acupuncture and moxibustion at BL21, the density of pylorus mucosa stained by PAS were increased compare to the groups applied to ST36. In the density of body mucosa stained by AB, moxibustion at BL21 and ST36 were increased compare to the other groups. Results : These data suggest that acupuncture and moxibustion at BL21 increased gastrin level of serum and those effects were more potent than acupuncture at ST36.

Developmental Patterns of Gal$\beta$1,3(4)GlcNAc $\alpha$2,3-Sialyltransferase (ST3Gal III) Expression in the Mouse: In Situ Hybridization Using DIG-labeled RNA Probes

  • Sialic acids are key determinants for biological processes, such as cell-cell interaction and differentiation. Sialyltransferases contribute to the diversity in carbohydrate structure through their attachment of sialic acid in various terminal positions on glycolipid and glycoprotein (N-linked and O-linked) carbohydrate groups. Gal$\beta$ 1,3(4)GlcNAc $\alpha$2,3-sialyltransferase (ST3Gal III) is involved in the biosynthesis of $sLe^{X}$ and sLe^{a}$ known as selection ligands and tumor-associated carbohydrate structures. The appearance and differential distribution of ST3Gal III mRNA during mice embryogenesis [embryonic (E) days; E9, E11, E13, E15] were investigated by in situ hybridization with digoxigenin-labeled RNA probes coupled with alkaline phosphatase detection. On E9, all tissues were positive for ST3Gal III mRNA expression whereas ST3Gal III mRNA on E11 was not detected throughout all tissues. On E13, ST3GAl III mRNA was expressed in different manner in various tissues. In this stage, ST3Gal III mRNA was positive only in the liver, pancreas and bladder. On E15, specific signal for ST3GAl III was detected in the liver, lung and forebrain. These results indicate that ST3Gal III is differently expressed at developmental stages of mice embryo, and this may be importantly related with regulation of organogenesis in mice.

  • Purpose: Severe aplastic anemia (SAA), a fatal disease, requires multiple transfusion, immunosuppressive therapy, and finally, hematopoietic stem cell transplantation (HSCT) as the definitive treatment. We hypothesized that iron overloading associated with multiple transfusions and HSCT-related complications may adversely affect cardiac function. Left ventricular (LV) function was assessed in children after HSCT for SAA. Methods: Forty-six consecutive patients with a median age of 9.8 years (range, 1.5-18 years), who received HSCT for SAA and who underwent comprehensive echocardiography before and after HSCT, were included in this study. The data of LV functional parameters obtained using conventional echocardiography, tissue Doppler imaging (TDI), and speckle-tracking echocardiography (STE) were collected from pre- and post-HSCT echocardiography. These data were compared to those of 40 age-matched normal controls. Results: In patients, the LV ejection fraction, shortening fraction, end-diastolic dimension, mitral early diastolic E velocity, TDI mitral septal E' velocity, and STE LV longitudinal systolic strain rate (SSR) decreased significantly after HSCT. Compared to normal controls, patients had significantly lower post-HSCT early diastolic E velocity and E/A ratio. On STE, patients had significantly decreased LV deformational parameters including LV longitudinal systolic strain (SS), SSR, and diastolic SR (DSR), and circumferential SS and DSR. Serum ferritin levels showed weak but significant correlations (P<0.05) with LV longitudinal SS and SSR and circumferential SS and DSR. Conclusion: Subclinical LV dysfunction is evident in patients after HSCT for SAA, and was associated with increased iron load. Serial monitoring of cardiac function is mandatory in this population.

Decontamination methods to restore the biocompatibility of contaminated titanium surfaces

  • Purpose: The reaction of cells to a titanium implant depends on the surface characteristics of the implant which are affected by decontamination. The aim of this study was to evaluate the cytocompatibility of titanium disks treated with various decontamination methods, using salivary bacterial contamination with dental pellicle formation as an in vitro model. Methods: Sand-blasted and acid-etched (SA) titanium disks were used. Three control groups (pristine SA disks [SA group]; salivary pellicle-coated SA disks [pellicle group]; and biofilm-coated, untreated SA disks [NT group]) were not subjected to any decontamination treatments. Decontamination of the biofilm-coated disks was performed by 14 methods, including ultrasonic instruments, rotating instruments, an air-powder abrasive system, a laser, and chemical agents. MG63 cells were cultured in the presence of the treated disks. Cell proliferation assays were performed on days 2 and 5 of cell culture, and cell morphology was analyzed by immunofluorescence and scanning electron microscopy (SEM). A vascular endothelial growth factor (VEGF) assay was performed on day 5 of culture. Results: The cell proliferation assay revealed that all decontaminated disks, except for the 2 groups treated using a plastic tip, showed significantly less cell proliferation than the SA group. The immunofluorescence and SEM analyses revealed that most groups showed comparable cell density, with the exception of the NT group, in which the cell density was lower and bacterial residue was observed. Furthermore, the cells grown with tetracycline-treated titanium disks showed significantly lower VEGF production than those in the SA group. Conclusions: None of the decontamination methods resulted in cytocompatibility similar to that of pristine SA titanium. However, many methods caused improvement in the biocompatibility of the titanium disks in comparison with the biofilm-coated, untreated titanium disks. This suggests that decontamination is indispensable for the treatment of peri-implantitis, even if the original biocompatibility cannot be restored.

Seeding Metastasis of Chromophobe Renal Cell Carcinoma after Robot-Assisted Laparoscopic Partial Nephrectomy

  • Chromophobe renal cell carcinoma (RCC) is an uncommon subtype of RCC having a better prognosis than clear cell RCC. Although there are several reports of seeding metastasis of RCC after biopsy, seeding metastasis of chromophobe RCC after surgical resection has seldom been reported. Here, we describe a case of multiple seeding metastases in the abdomen and pelvis 78 months after robot-assisted laparoscopic partial nephrectomy, without prior history of biopsy for chromophobe RCC in the right kidney. As magnetic resonance imaging (MRI) of the pelvic mass showed a similar appearance to the primary renal mass and displayed separate margins with the rectum and prostate gland, we were able to make a diagnosis before pathologic confirmation.

  • The electrification of the waste heat of stack is necessary to enhance the efficiency of fuel cell system. For this purpose, the hybrid SOFC/GT/ST system is suitable. The purpose of this work is to predict the performance of methane fueled SOFC/GT/ST hybrid power system and to analyze the influence of operating temperature of stack, current density of stack, and gas turbine pressure ratio. According to the analysis, it is proved that the SOFC/GT/ST hybrid system suppress the rapid decrease in efficiency and lead to the significant improvement of efficiency as compared with SOFC system.

MULTIPLE MYELOMA IN THE MANDIBLE MANIFESTED AS ORAL LESION OF PLASMACYTOMA: A CASE REPORT (악골에서 발견된 multiple myeloma의 구강내 plasmacytoma 병소: 증례보고)

  • Plasma cell neoplasms are generally categorized into four groups; multiple myeloma(MM), solitary plasmacytoma of the bone(SPB), plasma cell leukemias, and extramedullary plasmacytomas(EMP). These tumors may be further described as localized or diffuse in presentation. Localized plasma cell neoplasms are rare occurrences and include solitary plasmacytomas of the skeletal system, which account for 2-5% of all plasma cell neoplasms and extramedullary plasmacytomas of the soft tissue, which account for approximately 3% of all such neoplasms. A plasmacytoma is defined as any discrete, most likely solitary mass of neoplastic plasma cells either in the bone marrow or in various soft tissue sites. Diffuse lesions include the other two groups, multiple myeloma and plasma cell leukemia. The relationship between these processes has not yet been definitively characterized, but there appears to be a continuum in which both SPB and EMP often progress to MM. The patient was referred who had continuous deep throbbing bone pain and swelling on the left posterior gingival area of the mandible after extraction of the first and second molar. The result of intraoperative excisional biopsy of the lesion was confirmed as a plasmacytoma. And it revealed systemic multiple myeloma through the further diagnostic work-up. It is worth to report because of a rare case of multiple myeloma found in oral cavity as a form of plasmacytoma.

A Study on Serum Glucose Levels and the Pancreatic Beta-cell Protective Effect of Acupuncture on Streptozotocin-treated Rats by Subcutaneous Implantation of Osmotic Pump (Osmotic Pump모델 당뇨병 실험동물에서 침자극이 혈당조절 및 베타세포 보호작용에 미치는 영향)

  • Objective: To evaluate the effect of acupuncture on streptozotocin(STZ)-treated rats by subcutaneous implantation of osmotic pump. Methods: STZ was administered to rats at a low dose with osmotic pump to induce beta cell death and diabetes (STZ osmotic pump model), The experimental animals were divided into 4 groups: 1. The control group which was not treated in the STZ osmotic pump model 2. The sham group which was acupunctured at an arbitrary point in the STZ osmotic pump model 3. The sample A group which was acupunctured at the Chung-wan($CV_{12}$) in the STZ osmotic pump model 4. The sample B group which was acupunctured at the Chok-samni($ST_{36}$) in the STZ osmotic pump model. The effect of acupuncture in the STZ osmotic pump model was observed by measuring the serum glucose level and immunostaining of pancreatic tissue of the rats. Results : STZ injection by subcutaneous implantation of osmotic pump caused hyperglycemia by destroying the pancreatic beta cell selectively. Acupuncture at the Chung-wan acupuncture point($CV_{12}$) and Jhok-samni acupuncture point ($ST_{36}$) in the STZ osmotic pump model separately resulted in a decrease of the serum glucose level. In addition, the cyto-protective effect of the pancreatic beta-cell was detected in the STZ osmotic pump model by acupuncture. And there were few differences between the effects of acupuncture at the CV12 and $ST_{36}$. Conclusion : Acupuncture at the CV12 and ST36 had beneficial effects on Type II diabetes mellitus, and action mechanism of the effect was thought to be concerned with secretion of endogenous beta-endorphin.

Neuroprotection of Dopaminergic Neurons by Hominis Placenta Herbal Acupuncture in in vitro and in vivo Models of Parkinson's Disease Induced by MPP+/MPTP Toxicity

  • Objectives : This study was designed to investigate the neuroprotective effects of Hominis-Placenta (HP)on dopaminergic neurons. Methods : We examined the effect of invitro administration of HP against 1-methyl-4-phenylpyridinium( MPP+)-induced dopaminergic cell loss in primary mesencephalic culture and also used behavioral tests and performed analysis in the striatum and the substantia nigra of mouse brain, to confirm the effect of HP on dopaminergic neurons in an invivo 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)-induced PD mouse model. Animals were assigned to four groups: (1) Group 1(vehicle-treatedgroup), (2) Group 2(MPTPonlytreated group), (3) Group 3(MPTP+ saline-treated/$ST_{36}$ group), and (4) Group 4(MPTP+HP-treated/$ST_{36}$ group). HP at $20{\mu}L$ of 48 mg/kg dose was injected at $ST_{36}$ for 4 weeks at 2-day intervals. MPTP in saline was injected intraperitoneally each day for 5 days from the $8_{th}$ treatment of HP. We performed the pole test and rota-rod test on the first and seventh day after the last MPTP injection. To investigate the effect of HP on dopaminergic neurons, we performed analysis in the striatum and the substantia nigra of mouse brain after treatment with HP and/or MPTP. Results : Treatment with HP had no influence on cell proliferation and caused no cell toxicity in $PC_{12}$ and $HT_{22}$ cells. Our study showed that HP significantly prevented cell loss and protected neurites against MPP+ toxicity. Although the invivo treatment of HP herbal acupuncture at $ST_{36}$ showed a tendency to improve movement ability and protected dopaminergic cells and fibers in the substantia nigra and the striatum, it did not show significant changes compared with the MPTP treated group. Conclusions : These data suggest that HP could be a potential treatment strategy in neurodegenerative diseases such as Parkinson's disease.

A Study on the Effect of Herbal-acupuncture with Eucomiae Cortex Solution at Joksamni$(ST_{36})$ on Collagen-induced Arthritis (족삼리(足三里) 두충약침(杜沖藥鍼)이 Collagen-induced Arthritis에 미치는 영향)

  • Objectives : The purpose of this study is to observe the effects of Eucomiae Cortex herbal-acupuncture solution(EC-HAS) at Joksamni(ST36) on arthritis of mice induced by Collagen II. Methods : The author performed several experimental items. First, it is the cell survival rate of mice lung fibroblasts. Second, it is the incidence rate of arthritis and arthritis index of CIA. Third, it is the levels of IL-6, $TNF-{\alpha}$, $IFN-{\gamma}$, $IL-{\beta}$, IgG, IgM and anti-collagen II in serum and the level of IFN-y,$IFN-{\gamma}$/IL -4 ratio in CIA mouse spleen cell culture. Fourth, it is histological analysis of the mice joint. Fifth, it is expression ratio of $CD3e^+$ to $CD19^+$+ cell, $CD4^+$ to $CD8^+$ cell, $CD69^+/CD3e^+$/cells, $CD11a^+/CD19^+$/cells, $CD11b^+/Gr-1^+$ cells and $CD4^+/CD25^+$ cells. Results & Conclusion : 1. In the EC-HA, the incidence of arthritis and arthritis index were significantly decreased. 2. In EC-HA, the levels of IL-6, $IFN-{\gamma}$, $TNF-{\alpha}$, $IL-1{\beta}$, IgG, IgM and anti-collagen II in serum of CIA mice and the level of $IFN-{\gamma}$, IL-4, $IFN-{\gamma}$/lL-4 ratio in CIA mouse spleen cell culture were significantly decreased. 3. In the histological study, the cartilage destruction and synovial cell proliferation were decreased in the EC-HA, and the collagen fiber expressions in the EC-HA were similar with that of the Normal group. 4. In the EC-HA, the expression ratio of $CD3e^+$ to $CD19^+$ cell and $CD4^+$ to $CD8^+$ cell were similarly maintained as Normal group in lymph nodes, and $CD69^+/CD3e^+$ cells and $CD11a^+/CD19^+$ cells were decreased in lymph nodes, and $CD11b^+/Gr-1^+$ cells and $CD4^+/CD25^+$ cells were decreased in synovium. These results suggest that EC-HA at ST36 has an effect to control synovial cell proliferation and cartilage destruction in rheumatoid arthritis, and to be put to practical use in the future rheumatoid arthritis clinic.
