돼지 염색체 6번의 연관지도 및 양적형질 유전자좌위 탐색 (Linkage Map and Quantitative Trait Loci(QTL) on Pig Chromosome 6)

  • 본 연구는 돼지의 염색체 6번에 존재하는 주요 경제형질에 관여하는 양적형질 유전자좌위(Quantitative trait loci; QTL)를 밝히기 위하여 초위성체 마커를 이용하여 유전자지도를 작성하였다. 기준집단의 조성은 재래돼지 수컷 5마리와 Landrace 암컷 9마리를 자연교미하여 생산된 F$_1$의 동복 자손별로 수컷 1두를 임의적으로 선발하여 암컷 2두 이상과 전형매 교배시켜 F$_2$ 240두를 생산하였고 QTL 분석에는 F$_2$ 183두만을 이용하였다. 기준집단의 표현형 성적은 3주령체중, 등지방두께, 도축 24시간 후의 pH, 전단력, 조단백질 함량 등을 조사 분석하였다. F$_2$ 집단은 재래돼지와 Landrace의 두 종의 평균성적을 갖고 있었으며, 개체간 표현형적 변이가 매우 높아서 경제형질과 연관된 QTL을 탐색하기 적절한 기준집단이라고 평가되었다. 연관지도는 29개의 초위성체 마커와 1개의 PCR-RFLP 마커(AMPKα2)를 이용하여 작성하였으며, 연관지도상의 염색체 길이는 169.3cM이었고, 마커간 평균 간격은 6.05cM이었다. 염색체 6번에서 경제형질과 연관된 유의적인 QTL은 모두 5개가 탐색 되었다. 3주령 체중과 연관된 QTL이 5cM 위치에서 탐색되었으며, 전단력, 도축 24시간 후 pH, 등지방두께, 조단백질 함량 등의 육질관련 QTL이 서로 다른 위치에서 5% 수준의 통계적 유의성이 확인되었다.

  • Calcineurin is a calmodulin dependent protein that functions as a regulator of muscle cell growth and function. Agents capable of interacting with calcineurin could have important applications in muscle disease treatment as well as in the improvement of livestock production. Calsarcins comprise a family of muscle-specific calcineurin binding proteins which play an important role in modulating the function of calcineurin in muscle cells. Recently, we described the first two members of the calsarcin family (calsarcin-1 and calsarcin-2) in the pig. Here, we characterized the third member of the calsarcin family, calsarcin-3, which is also expressed specifically in skeletal muscle. However, unlike calsarcin-1 and calsarcin-2, the calsarcin-3 mRNA expression in skeletal muscle kept rising throughout the prenatal and postnatal development periods. In addition, radiation hybrid mapping indicated that porcine calsarcin-3 mapped to the distal end of the q arm of pig chromosome 2 (SSC2). A C/T single nucleotide polymorphism site in exon 5 was genotyped using the denaturing high performance liquid chromatography (DHPLC) method and the allele frequencies at this locus were significantly different among breeds.

Chondrogenic Differentiation of Porcine Skin-Derived Stem Cells with Different Characteristics of Spontaneous Adipocyte Formation

  • The purpose of this study is to confirm whether spontaneous adipocyte generation during chondrogenic induction culture affects the chondrogenic differentiation of porcine skin-derived stem cells (pSSCs). For this purpose, chondrogenic differentiation characteristics and specific marker gene expression were analyzed using cell lines showing different characteristics of spontaneous adipocyte formation. Of the four different lines of pSSCs, the pSSCs-IV line showed higher Oil red O (ORO) and glycosaminoglycan (GAG) extraction levels. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis revealed that the levels of adipogenic markers peroxisome proliferator-activated receptor gamma 2 ($PPAR{\gamma}2$) and adipocyte Protein 2 (aP2) mRNAs were significantly higher in pSSCs-IV than those of the other pSSC lines (P<0.05). Among three chondrogenic markers, collagen type II (Col II) and sex determining region Y-box (Sox9) mRNAs were strongly expressed in pSSCs-IV (P<0.05), but not in aggrecan (Agg), which was significantly higher in pSSCs-II (P<0.05). These results demonstrate that the spontaneous adipocyte generation during chondrogenic differentiation has a positive effect on the chondrogenesis of pSSCs. More research is needed on the correlation between adipocyte generation and cartilage formation.

Molecular Characterization, Chromosomal Localizations, Expression Profile, and Association Analysis of the Porcine PECI Gene with Carcass Traits

  • The full-length cDNA of the porcine peroxisomal ${\Delta}^3$,${\Delta}^2$-enoyl-CoA isomerase (PECI) gene encodes a monofunctional peroxisomal ${\Delta}^3$,${\Delta}^2$-enoyl-CoA isomerase. Cloning and sequencing of the porcine PECI cDNA revealed the presence of an 1185-base pair open reading frame predicted to encode a 394-amino acid protein by the 5'rapid amplification of cDNA ends (5'RACE) and EST sequences. The porcine PECI gene was expressed in seven tissues (heart, liver, spleen, lung, kidney, skeletal muscle, fat) which was revealed by reverse transcriptase-polymerase chain reaction (RT-PCR). The porcine PECI was mapped to SSC71/2 p11-13 using the somatic cell hybrid panel (SCHP) and the radiation hybrid panel (RH) (LOD score 12.84). The data showed that PECI was closely linked to marker S0383. A C/T single nucleotide polymorphism in PECI exon 10 (3'UTR) was detected as a PvuII PCR-RFLP. Association analysis in our experimental pig population showed that different genotypes of PECI gene were significantly associated with the Average Backfat thickness (ABF) (p<0.05) and Buttock backfat thickness (p<0.01).

Mapping of the Porcine Calpastatin Gene and Association Study of Its Variance with Economic Traits in Pigs

  • The objectives of this study were to confirm a location of the calpastatin (CAST) gene in chromosome 2 and to detect associations of genetic variations with economic traits in the porcine CAST gene as a candidate gene for growth and meat quality traits in pigs. Calpastatin is a specific endogenous inhibitor of calpains. The calpain protease system is ubiquitous, and is involved in numerous growth and metabolic processes. Three single nucleotide variations were identified within a 1.6 kb fragment of the porcine CAST gene and these polymorphisms were used for genetic linkage mapping. Linkage and QTL mapping were performed with the National Livestock Research Institute (NLRI) reference families using eight microsatellites and SNP makers in the CAST gene. The porcine CAST gene was mapped adjacent to the markers, SW395 and SW1695 on SSC2 with LOD scores of 15.32 and 8.50, respectively. According to the QTL mapping, a significant association was detected at 82 cM between SW395 and CAST-Hinf I for weight at the age of 30 weeks. In addition, an association study was performed with the $F_2$ animals of NLRI reference families for Hinf I, Msp I and Rsa I polymorphisms in the CAST gene. Two polymorphisms, CAST-Rsa I and CAST-Hinf I, showed significant correlation for growth traits at p<0.01 and p<0.05, respectively.

수확 전·후 1-MCP처리가 '후지' 사과의 저온저장 중 과실품질에 미치는 영향 (Effect of Preharvest and Postharvest 1-Methylcyclopropene (1-MCP) Treatments on Fruit Quality Attributes in Cold-stored 'Fuji' Apples)

  • 본 연구는 수확 전 수체살포용 1-MCP($Harvista^{TM}$)와 수확 후 훈증용 1-MCP($SmartFresh^{TM}$) 처리가 '후지' 사과의 품질과 저장성에 미치는 영향을 구명하고자 실시하였다. $Harvista^{TM}$ 처리는 수확 1, 2, 3주전 0, 95, 125, $250mg{\cdot}L^{-1}$ 농도로 각각 처리하였으며, $SmartFresh^{TM}$ 처리는 수확 1일 후 밀폐된 공간에서 $1{\mu}L{\cdot}L^{-1}$의 농도로 18시간동안 처리하였다. 과실저장은 $0{\pm}1^{\circ}C$ 조건에서 180일간 저장하였다. $Harvista^{TM}$ 처리에 따른 수확 시 과실품질을 보면 과중, 과피 적색도, 내생에틸렌 발생량, 경도, 산 및 가용성 고형물 함량은 무처리구와 차이가 없었다. 저온저장동안 '후지' 사과의 품질변화를 보면 $Harvista^{TM}$$250mg{\cdot}L^{-1}$의 농도로 2, 3주전에 처리한 과실이 무처리구에 비하여 과실 경도 및 산 함량이 더 높게 유지되었으며, 내생에틸렌 발생량도 무처리구에 비하여 5.5-10.0% 수준으로 현저히 낮았다. 그리고 저장기간 중 가용성고형물 함량은 차이를 보이지 않았다. $Harvista^{TM}$ 단독처리와 $Harvista^{TM}+SmartFresh^{TM}$ 혼용처리구는 경도, 산 함량, 내생에틸렌 발생량 모두 저장 180일 후에도 수확 시와 큰 차이를 보이지 않아 과실품질이 유지되었다. 요인간 상관분석은 $Harvista^{TM}$ 처리시 처리농도와 무관하게 내생에틸렌 발생량이 경도 또는 산 함량과 부의 상관관계를 나타냈다. 그리고 $Harvista^{TM}+SmartFresh^{TM}$ 혼용처리 시 $Harvista^{TM}$ $250mg{\cdot}L^{-1}$$SmartFresh^{TM}$ 처리가 변수간 정의 상관관계를 나타내었다. 따라서, 수확 후 $SmartFresh^{TM}$처리에 의한 과실품질유지 효과와 함께, 고 농도의 $Harvista^{TM}$처리 또한 저온저장 중 과실품질 유지에도 큰 효과를 보였다.

Analysis of Swine Leukocyte Antigen Haplotypes in Yucatan Miniature Pigs Used as Biomedical Model Animal

  • The porcine major histocompatibility complex (MHC) is called swine leukocyte antigen (SLA), which controls immune responses and transplantation reactions. The SLA is mapped on pig chromosome 7 (SSC7) near the centromere. In this study, 3 class I (SLA-1, SLA-3, and SLA-2) and 3 class II (DRB1, DQB1, and DQA) genes were used for investigation of SLA haplotypes in Yucatan miniature pigs in Korea. This pig breed is a well-known model organism for biomedical research worldwide. The current study indicated that Korean Yucatan pig population had 3 Class I haplotypes (Lr-4.0, Lr-6.0, and Lr-25.0) and 3 class II haplotypes (Lr-0.5, Lr-0.7, and Lr-0.25). The combinations of SLA class I and II haplotype together, 2 homozygous (Lr-4.5/4.5 and Lr-6.7/6.7) and 3 heterozygous (Lr-4.5/6.7, Lr-4.5/25.25, and Lr-6.7/25.25) haplotypes were identified, including previously unidentified new heterozygous haplotypes (Lr-4.5/4.7). In addition, a new SLA allele typing method using Agilent 2100 bioanalyzer was developed that permitted more rapid identification of SLA haplotypes. These results will facilitate the breeding of SLA homozygous Yucatan pigs and will expedite the possible use of these pigs for the biomedical research, especially xenotransplantation research.

Effects of Suspension Culture on Proliferation and Undifferentiation of Spermatogonial Stem Cells Derived from Porcine Neonatal Testis

  • Despite many researches related with in-vitro culture of porcine spematogonial stem cells (SSCs), adherent culture system widely used has shown a limitation in the maintenance of porcine SSC self-renewal. Therefore, in order to overcome this obstacle, suspension culture, which is known to have numerous advantage over adherent culture, was applied to the culture of porcine SSCs. Porcine SSCs retrieved from neonatal testes were suspension-cultured for 5 days or 20 days, and characteristics of suspension-cultured porcine SSCs including proliferation, alkaline phosphatase (AP) activity, and self-renewal-specific gene expression were investigated and compared with those of adherent-cultured porcine SSCs. As the results, the suspension-cultured porcine SSCs showed entirely non-proliferative and significantly higher rate of AP-positive cells and expression of self-renewal-specific genes than the adherent-cultured porcine SSCs. In addition, long-term culture of porcine SSCs in suspension condition induced significant decrease in the yield of AP staining-positive cells on post-day 10 of culture. These results showed that suspension culture was inappropriate to culture porcine SSCs, because the culture of porcine SSCs in suspension condition didn't stimulate proliferation and maintain AP activity of porcine SSCs, regardless of culture periods.

Chloroplast genome of white wild chrysanthemum, Dendranthema sp. K247003, as genetic barcode

  • Dendranthema boreale and D. indicum are easily distinguished from other Korean Dendranthema spp. by having yellow flowers. We have found a putative new taxon of Dendranthema having white flowers, except for sharing most characters with Dendranthema boreale. The chloroplast (cp) genome of the putative new taxon of Dendranthema, Dendranthema sp. K247003, registered in National Agro-Biodiversity Center (ABC), was completely characterized as a genetic barcode. The cp-genome of Dendranthema sp. K247003 was 151,175-bp in size: LSC was 82,886-bp, IR 24,971-bp, SSC 18,347-bp. The cp-genome of Dendranthema sp. K247003 contains 113 genes and 21 introns consisted of 79 protein coding genes, 4 RNA genes, and 30 tRNA genes, with 20 group II introns and one group I intron. Some of the genes and there introns were duplicated in IR. The cp-DNA of Dendranthema sp. K247003 is distinguished from that of D. boreale IT121002 by 67 SNPs in genic regions of 24 protein coding genes and by a 9-bp INDEL in ycf1. Further cp-DNA study will give us better information on genetic markers of Dendranthema species.

양생조건에 따른 SSG공법의 일축압축강도 평가 (Evaluation on Uniaxial Compression Strength of SSG Method with Curing Condition)

  • 좁은 면적을 단기간에 개량시킬 수 있는 장점이 있는 그라우팅 공법은 지반의 특성과 지하수위 등에 따라 주입 효과의 차이가 있다. 특히 투수성이 큰 지반에서는 시간 경과에 따라서 용탈현상이 발생하여 강도저하 및 차수 성능이 저하되어 품질에 문제가 생길 수 있다. 최근 이와 같은 문제점을 해결하기 위해 초기강도 확보 및 용탈 방지와 동시에 균열 자기치유 특성을 가지는 SSG가 개발되었다. 이 연구에서는 두 가지 양생조건에 대해 SSG의 일축압축강도 시험을 수행하여 양생기간에 따른 일축압축 강도를 파악하였다. 또한 SSG와 기존의 LW 및 SGR의 일축압축강도와 비교하였다. 연구 결과, SSG가 상온 양생 및 저온 양생 모두에서 강도가 크게 측정되었으며, 초기 강도도 상대적으로 매우 우수한 것으로 나타났다.
