TIGGE/S2S 기반 중장기 토양수분 예측 및 검증 (Verification of Mid-/Long-term Forecasted Soil Moisture Dynamics Using TIGGE/S2S)

  • Developing reliable soil moisture prediction techniques at agricultural regions is a pivotal issue for sustaining stable crop productions. In this study, a physically-based SWAP(Soil-Water-Atmosphere-Plant) model was suggested to estimate soil moisture dynamics at the study sites. ROSETTA was also integrated to derive the soil hydraulic properties(${\alpha}$, n, ${\Theta}_r$, ${\Theta}_s$, $K_s$) as the input variables to SWAP based on the soil information(Sand, Silt and Clay-SSC, %). In order to predict the soil moisture dynamics in future, the mid-term TIGGIE(THORPEX Interactive Grand Global Ensemble) and long-term S2S(Subseasonal to Seasonal) weather forecasts were used, respectively. Our proposed approach was tested at the six study sites of RDA(Rural Development Administration). The estimated soil moisture values based on the SWAP model matched the measured data with the statistics of Root Mean Square Error(RMSE: 0.034~0.069) and Temporal Correlation Coefficient(TCC: 0.735~0.869) for validation. When we predicted the mid-/long-term soil moisture values using the TIGGE(0~15 days)/S2S(16~46 days) weather forecasts, the soil moisture estimates showed less variations during the TIGGE period while uncertainties were increased for the S2S period. Although uncertainties were relatively increased based on the increased leading time of S2S compared to those of TIGGE, these results supported the potential use of TIGGE/S2S forecasts in evaluating agricultural drought. Our proposed approach can be useful for efficient water resources management plans in hydrology, agriculture, etc.

Response of two-way reinforced concrete voided slabs enhanced by steel fibers and GFRP sheets under monotonic loading

  • Various efforts have been made to reduce the weight of concrete slabs while preserving their flexural strength. This will result in reducing deflection and allows the utilization of longer spans. The top zone of the slab requires concrete to create the compression block for flexural strength, and the tension zone needs concrete to join with reinforcing for flexural strength. Also, the top and bottom slab faces must be linked to transmit stresses. Voided slab systems were and are still used to make long-span slab buildings lighter. Eight slab specimens of (1000*1000 (1000*1000 mm2) were cast and tested as two-way simply supported slabs in this research. The tested specimens consist of one solid slab and seven voided slabs with the following variables (type of slab solid and voided), thickness of slab (100 and 125 mm), presence of steel fibers (0% and 1%), and the number of GFRP layers). The voids in slabs were made using high-density polystyrene of dimensions (200*200*50 mm) with a central hole of dimensions (50*50*50 mm) at the ineffective concrete zones to give a reduction in weight by (34% to 38%). The slabs were tested as simply supported slabs under partial uniform loading. The results of specimens subjected to monotonic loading show that the combined strengthening by steel fibers and GFRP sheets of the concrete specimen (V-125-2GF-1%) shows the least deflection, deflection (4.6 mm), good ultimate loading capacity (192 MPa), large stiffness at cracking and at ultimate (57 and 41.74) respectively, more ductility (1.44), and high energy absorption (1344.83 kN.mm); so it's the best specimen that can be used as a voided slab under this type of loading.

Perspective of breaking stagnation of soybean yield under monsoon climate

  • Soybean yield has been low and unstable in Japan and other areas in East Asia, despite long history of cultivation. This is contrasting with consistent increase of yield in North and South America. This presentation tries to describe perspective of breaking stagnation of soybean yield in East Asia, considering the factors of the different yields between regions. Large amount of rainfall with occasional dry-spell in the summer is a nature of monsoon climate and as frequently stated excess water is the factor of low and unstable soybean yield. For example, there exists a great deal of field-to-field variation in yield of 'Tanbaguro' soybean, which is reputed for high market value and thus cultivated intensively and this results in low average yield. According to our field survey, a major portion of yield variation occurs in early growth period. Soybean production on drained paddy fields is also vulnerable to drought stress after flowering. An analysis at the above study site demonstrated a substantial field-to-field variation of canopy transpiration activity in the mid-summer, but the variation of pod-set was not as large as that of early growth. As frequently mentioned by the contest winners of good practice farming, avoidance of excess water problem in the early growth period is of greatest importance. A series of technological development took place in Japan in crop management for stable crop establishment and growth, that includes seed-bed preparation with ridge and/or chisel ploughing, adjustment of seed moisture content, seed treatment with mancozeb+metalaxyl and the water table control system, FOEAS. A unique success is seen in the tidal swamp area in South Sumatra with the Saturated Soil Culture (SSC), which is for managing acidity problem of pyrite soils. In 2016, an average yield of $2.4tha^{-1}$ was recorded for a 450 ha area with SSC (Ghulamahdi 2017, personal communication). This is a sort of raised bed culture and thus the moisture condition is kept markedly stable during growth period. For genetic control, too, many attempts are on-going for better emergence and plant growth after emergence under excess water. There seems to exist two aspects of excess water resistance, one related to phytophthora resistance and the other with better growth under excess water. The improvement for the latter is particularly challenging and genomic approach is expected to be effectively utilized. The crop model simulation would estimate/evaluate the impact of environmental and genetic factors. But comprehensive crop models for soybean are mainly for cultivations on upland fields and crop response to excess water is not fully accounted for. A soybean model for production on drained paddy fields under monsoon climate is demanded to coordinate technological development under changing climate. We recently recognized that the yield potential of recent US cultivars is greater than that of Japanese cultivars and this also may be responsible for different yield trends. Cultivar comparisons proved that higher yields are associated with greater biomass production specifically during early seed filling, in which high and well sustained activity of leaf gas exchange is related. In fact, the leaf stomatal conductance is considered to have been improved during last a couple of decades in the USA through selections for high yield in several crop species. It is suspected that priority to product quality of soybean as food crop, especially large seed size in Japan, did not allow efficient improvement of productivity. We also recently found a substantial variation of yielding performance under an environment of Indonesia among divergent cultivars from tropical and temperate regions through in a part biomass productivity. Gas exchange activity again seems to be involved. Unlike in North America where transpiration adjustment is considered necessary to avoid terminal drought, under the monsoon climate with wet summer plants with higher activity of gas exchange than current level might be advantageous. In order to explore higher or better-adjusted canopy function, the methodological development is demanded for canopy-level evaluation of transpiration activity. The stagnation of soybean yield would be broken through controlling variable water environment and breeding efforts to improve the quality-oriented cultivars for stable and high yield.

한국에 있어 우주법의 주요내용, 논평과 장래의 과제 (The Main Contents, Comment and Future Task for the Space Laws in Korea)

  • 한국은 우주 탐사 프로그램에 대한 포부와 함께 급속하게 확장내지 발전하고 있다. 정부는 항공우주산업에 우선권을 주고 있으며, 더 좋은 기반위에, I987년의 항공우주산업개발촉진법, 2005년의 우주개발진흥법과 2007년의 우주손해배상법을 제정하여 시행 해오고 있다. 필자는 위성의 발사허가, 우주물체의 등록, 위성정보의 이용, 우주비행사의 구조, 손해배상, 지상 제3자에 대한 책임보험, 우주개발지원계획 등이 포함된 전기 세 가지 우주관계법의 입법경위, 주요내용 및 논평을 하고자 한다. 더욱이 필자는 한국의 우주산업을 효율적으로 개발하기 위하여 일본의 우주항공연구개발기구(JAXA), 영국의 국립우주센터(BNSC), 국립우주연구센터(CNES), 독일의 항공우주센터(DLR), 스웨덴의 우주공사(SSC), 중국의 항천과공집단공사(CASIC) 등과 유사한 한국의 새로운 우주항공개발공사 (KADA: 가칭)의 창설할 수 있는 입법이 필요로 하며 또한 정부기구로서 캐나다의 우주청(CSA), 러시아의 우주청(RSA), 이탈리아의 우주청(ISA), 이스라엘의 우주청(ISA), 인도의 우주성(IDS), 미국의 항공우주청(NASA)과 중국의 국가항천국(CNSA)등과 유사한 한국의 새로운 우주청(KSA: 가칭)을 창설할 수 있는 입법조치가 필요하다. 만약 한국 정부가 장차 정부 조직으로 한국의 우주청을 설립시키려면 정부조직법의 개정이 필요하다. 동양적인 사고와 창의력에 근거하여 아시아 여러 나라들 간에 우주산업을 발전시키기 위하여 연구협력을 증진시킬 수 있는 "아시아우주기구(ASA)" 의 설립이 필요하다.

돼지 FABP3 Promoter 부위 내 신규 돌연변이 탐색과 근내지방도와의 연관성 분석 (Detection of Novel Mutations in the FABP3 Promoter Region and Association Analysis with Intramuscular Fat Content in Pigs)

  • Intramuscular fat content(lMF) is considered as one of major economic traits in the pig breeding and industry. In general, high IMF results in better meat quality. Several approaches to detect quantitative trait 10ci( QTL) for IMF indicated a strong possibility of the existence of a QTL related to IMF between the microsatellite marker SW71 and SW1881 on SSC6q. Porcine FABP3 has been considered as a candidate gene affecting IMF due to its physiological roles and position on the pig genome. Two novel mutations, g.-114T> C and g.-158T>G were detected by duplicate sequencing of the porcine FABP3 promoter region. These two mutations were identified as absolute linkage disequilibrium. The g.-158T> G mutation was used for investigating relationships with growth and fat deposition traits. The GG genotype of the g.-158T> G polymorphism showed highly negative effects(P< 0.01) on body weights at 3 and 12 weeks of age, and a positive effect(P< 0.05) on IMF. However, backfat thickness(BF) and carcass fat(CF) content were not significantly associated with the genotype. The result indicates that the novel mutations, identified in this study, could be utilized as possible genetic markers to improve IMF, independent with BF.

돼지 등심 내 카프르산 함량과 단일염기다형 마커 rs81437607 유전자형의 상관 (Association of the Genotypes of Single Nucleotide Polymorphism Marker rs81437607 with Capric Acid Contents in longissimus dorsi Muscle in Pigs)

  • This study tested the association between genotypes of the single nucleotide polymorphism (SNP) marker, rs81437607 and capric acid (FA_C10_0) compositions in longissimus dorsi muscle in pigs. Eighteen fatty acid (FA) compositions were measured in a total of 974 $F_2$ animals among 1,106 $F_2$ progeny produced between Landrace and Jeju Black Pig (JBP). Among FA compositions tested, we identified a cluster of highly significant SNPs for capric acid compositions on 58 Mb position of Sus scrofa chromosome 12 (SSC12) using genome-wide association study (GWAS) with $F_2$ genotypes from SNP panel analysis. GWAS results showed that the rs81437607 was the highest trait-related SNP marker with capric acid levels. Three genotypes (C/C, C/T and T/T) of rs81437607 marker were found in $F_2$ population by further pyrosequencing. Association analysis results showed the significant differences between rs81437607 genotypes and capric acid compositions (P<0.05). The $F_2$ pigs harboring rs81437607 C/C ($0.119{\pm}0.002%$) and C/T ($0.116{\pm}0.002%$) genotypes showed additively higher levels of capric acid content than those of T/T homozygotes ($0.109{\pm}0.002%$) ($P=1.30{\times}10^{-12}$). These results suggested that the genetic variations of rs81437607 may be helpful to find causative variants and assist as molecular genetic markers for improving the capric acid contents in longissimus dorsi muscle in pigs.

돼지 웅성 생식선 줄기세포의 체외배양기법 개발 (Development of In Vitro Culture System for Male Germline Stem Cells in Porcine)

  • Spermatogonial stem cells(SSCs) only are responsible for the generation of progeny and for the transmission of genetic information to the next generation in male. Other in vitro studies have cultured SSCs for proliferation, differentiation, and genetic modification in mouse and rat. Currently, information regarding in vitro culture of porcine Germline Stem Cell(GSC) such as gonocyte or SSC is limited and is in need of further studies. Therefore, in this study, we report development of a successful culture system for gonocytes of neonatal porcine testes. Testis cells were extracted from $10{\sim}14$-day-old pigs. These cells were harvested using enzymatic digestion, and the harvested cells were purified with combination of percoll, laminin, and gelatin selection techniques. The most effective culture system of porcine gonocytes was established through trial experiments which made a comparison between different feeder cells, medium, serum concentrations, temperatures, and $O_2$ tensions. Taken together, the optimal condition was established using C166 or Mouse Embryonic Fibroblast(MEF) feeder cell, Rat Serum Free Medium(RSFM), 0% serum concentration, $37^{\circ}C$ temperature, and $O_2$ 20% tension. Although we discovered the optimal culture condition for proliferation of porcine gonocytes, the gonocyte colonies ceased to expand after one month. These results suggest inadequate acquirement of ingredients essential for long term culture of porcine GSCs. Consequently, further study should be conducted to establish a successful long-term culture system for porcine GSCs by introducing various growth factors or nutrients.

수확전 수체살포용 1-MCP 처리가 '감홍' 사과의 저온저장 중 과실품질에 미치는 영향 (Effect of Preharvest Sprayable 1-Methylcyclopropene (1-MCP) Treatment on Fruit Quality Attributes in Cold Stored 'Gamhong' Apples)

  • 본 연구는 수확전 수체살포용 1-MCP(Harvista) 처리가 국내 육성품종인 '감홍' 사과의 품질과 저장성에 미치는 영향을 구명하고자 실시하였다. 수체살포용 1-MCP 처리 후 수확시에 과실의 산 함량은 1-MCP 수확 10일전 처리가 무처리에 비하여 높았으나 다른 과실품질 요인들은 차이가 없었다. 그리고 저장동안 '감홍' 사과의 품질변화를 보면 저장기간이 길어질수록 1-MCP 처리 과실들의 경도와 산 함량이 무처리 과실보다 높게 유지되었으나 가용성고형물 함량은 차이가 없었다. 내생 에틸렌 발생량은 수확시에는 처리간 차이가 없이 낮은 에틸렌발생을 보였으나 무처리 과실들은 저장 180일에는 $58.3{\mu}L{\cdot}L^{-1}$로 급격히 증가한 반면 1-MCP 처리 과실들은 $18.24{\sim}25.03{\mu}L{\cdot}L^{-1}$로 현저히 낮은 발생량을 보였다. 따라서 '감홍' 사과에 수확전 수체살포용 1-MCP 처리가 저장기간동안 과실의 품질을 유지하는데 효과적이었다.

Identification and Screening of Gene(s) Related to Susceptibility to Enterotoxigenic Escherichia coli F4ab/ac in Piglets

  • In 2004, Jorgensen and coworkers proposed the MUC4 gene as a candidate gene of enterotoxigenic Escherichia coli (ETEC) F4ab/ac receptor in piglets and a mutation of $G{\rightarrow}C$ in intron 7 of MUC4 was identified to be associated with the ETEC F4ab/ac adhesion phenotypes. In this study, we used 310 piglets of three breeds (Landrace, Large White and Chinese Songliao Black) to analyze the relationship between this mutation and the F4ab/ac adhesion phenotype. The results show that the genotypes at this site and the ETEC F4ab/ac adhesion phenotypes were not completely consistent, although they are very strongly associated. Among the individuals with genotype CC, which was identified as a resistant genotype to F4ab/ac adhesion, only 72.1% (124/172) were non-adhesive to ETEC F4ab and 77.9% (134/172) were non-adhesive to ETEC F4ac infections. This suggests that this mutation may not be the causative mutation for ETEC F4ab/ac adhesion, rather, the actual causative mutation may be in another gene closely linked to MUC4, or at aother site within the MUC4 gene. Our results also suggest that the receptors of F4ab and F4ac may be determined by two different but closely linked loci. In order to screen other genes related to F4ab/ac adhesion in piglets, the mRNA profiles from six full sib piglets, of which three were adhesive to ETEC F4ab/ac and three non-adhesive, were analyzed by suppression subtractive hybridization (SSH). One up-regulated gene, Ep-CAM, was selected for further analysis based on its role in the intestinal epithelial cells adhesion. Using real-time RT-PCR, we found that the Ep-CAM gene was significantly up-regulated in the piglets adhesive to F4ab/ac. It was mapped to SSC3q11-q14 by radiation hybrid mapping.

CA 저장에 의한 청매실의 저온장해 발생 억제 및 품질 특성 유지 효과 (Suppression of Chilling Injury and Maintenance of Quality Characteristics in Prunus Mume Fruits Stored under Controlled Atmosphere)

  • 실용적인 CA 처리 방법을 이용하여 매실 '남고'의 저장성 향상 및 저온장해과 발생 억제에 관한 기초 자료를 얻고자 본 연구를 수행하였다. 매실 '남고'를 1, 5, $9^{\circ}C$에 저장하면서 최적 CA 조성으로4% $O_2$ +75% $CO_2$ 의 농도를 선정하여 사용하였다. 매실의 상온 저장시에는 황변현상과 조직의 연화가 빠르게 진행되어 수확 후 3~4일 이내에 상품성이 없어지는 문제점이 있다. 이산화탄소와 에틸렌 생성량은 CA 저장구에서 유의적으로 줄었다. 또한, Hue 값은 CA 저장으로 감소폭이 줄었으며, 특히 $9^{\circ}C$ 대조구에서 낮은 값을 나타내었고, 1, $5^{\circ}C$에서 저장 30일까지 녹색을 유지하였다. 중량감소율, 당도, 산도의 경우 대조구에 비해 CA 저장구에서 유의적으로 유지되었으며, 전해질 유출 정도도 CA 저 장구에서 낮은 값을 나타내었다. CA 처리를 한 매실의 경우 모든 저장 온도구에서 저온 저장 30일까지 과피갈변, surface pitring 등 저온장해과가 발생하지 않았고, 부패율 역시 CA 저장구에서 유의적으로 낮게 나타났다. 저장 30일경에 $5^{\circ}C$$9^{\circ}C$ 대조구에서 황변현상, 생리장해과, 부패과가 발생되었으나, CA 저장 시험구는 효과적으로 억제되었다. 따라서 매실 '남고'를 CA 저장 시$1^{\circ}C$가 가장 적합한 것으로 선정되었고, 매실 저장 시 주요한 문제점인 황변현상, 연화, 저온장해과, 부패율이 효과적으로 억제되었다.
