Alkaloids from Beach Spider Lily (Hymenocallis littoralis) Induce Apoptosis of HepG-2 Cells by the Fas-signaling Pathway

  • Alkaloids are the most extensively featured compounds of natural anti-tumor herbs, which have attracted much attention in pharmaceutical research. In our previous studies, a mixture of major three alkaloid components (5, 6-dihydrobicolorine, 7-deoxy-trans-dihydronarciclasine, littoraline) from Hymenocallis littoralis were extracted, analyzed and designated as AHL. In this paper, AHL extracts were added to human liver hepatocellular cells HepG-2, human gastric cancer cell SGC-7901, human breast adenocarcinoma cell MCF-7 and human umbilical vein endothelial cell EVC-304, to screen one or more AHL-sensitive tumor cell. Among these cells, HepG-2 was the most sensitive to AHL treatment, a very low dose ($0.8{\mu}g/ml$) significantly inhibiting proliferation. The non-tumor cell EVC-304, however, was not apparently affected. Effect of AHL on HepG-2 cells was then explored. We found that the AHL could cause HepG-2 cycle arrest at G2/M checkpoint, induce apoptosis, and interrupt polymerization of microtubules. In addition, expression of two cell cycle-regulated proteins, CyclinB1 and CDK1, was up-regulated upon AHL treatment. Up-regulation of the Fas, Fas ligand, Caspase-8 and Caspase-3 was observed as well, which might imply roles for the Fas/FsaL signaling pathway in the AHL-induced apoptosis of HepG-2 cells.

Acaricide susceptibilities of Field-Collected Populations of Two-spotted Spider Mite, Tetranychus urticae (Acari:Tetranychidae) from Apple Orchards (사과원에서 채집된 점박이응애(Tetranychus urticae)의 지역별 살비제 감수성)

  • Susceptibilies of tow-spotted spider mite (Tetranychus urticae Koch) collected at 6 locations were assessed to 15 acaricides. The response to acaricides were almost similar in female adults and eggs. There were considerable difference in susceptibility depending on the acaricide treated and the region from which the population was collected. The population showing resistance ratio of more than 20 with respect to certain acaricide was regarded as a resistant population to the acaricide. The resistant populations in terms of female adult were as follows: Suwon population to azocyclotin, cyhexatin, and fenbutatin-oxide; Taejon population to dicofol and fenbutatin-oxide; Chongju population to dicofoll Chinju populatin to cyhexatin, dicofol, and fenbutatin-oxide. The resistant populations in terms of egg were as follows; Suwon population to bifenthrin, clofentezine, hexythiazox, and tetradifon; Kunwi, Chongju, and Kwangju populations to bifenthrin; Taejon population to amitraz and bifenthrin; Taejon populatin to amitraz and bifenthrin; Chinju population to amitraz, bifenthrin, clofentezine, dicofol, and tetradifon. However, the female adults and eggs of all field populations were susceptible to abamectin, chlorfenson, and fenpyroximate. This tendency was also reported previously in the susceptible strain from laboratory.

Inheritance if Resistance in Dicofol-selected Strain of the Two-spotted Spider Mite, Tetranychus urticae Koch (Acarina: Tetranychidae), and its Cross Resostance (Dicofol 저항성 점박이응애의 저항성 유전과 교차저항성)

  • Dicofol susceptible two-spotted spider mite(S) which was selected by 150 times of dicofol treatment showed 27 5-fold increase in resistance as compared to S strain. Inheritance of resistance and crossresistance in selected strain($R_d$) were investigated. There were differences oi susceptibility in the dicofol concentmtion-moltaliity relationships between $F_1$ progenies (R♀, XS♂, R♂, XS♀) obtained from reciprocal cross with the S strain and $R_d$, strain. Degree of dominance of the $F_1$ progenies (R♀, XS♂, R♂, XS♀) were -0.29 and -0.72 in adult females and -0.42 and -0.93 in adult males, respectively. These results suggest that inheritance of dicofol resistance is controlled by an incomplete recessive. $R_d$ strain exhibited a high level of cross resistance to amitraz, and also showed cross resistance to pyrethrotd acaricides, acrinathrin and bifenthnn. $R_d$ strain showed non-cross reslstancz to abamectin, chlolfenson, clofentezine, cyhexatm, fenbutatin oxlde, fenpyroximate, hexythiazox, monocrotophos, propargite and tetradifon, but negatively correlated cross-resistance to azocyclotin and fenbutatin oxide belonging to tin acaricides.

Evaluation of Toxicity of Plant Extract Made by Neem and Matrine against Main Pests and Natural Enemies (멀구슬과 고삼을 원료로 한 식물추출물의 주요해충과 천적에 대한 독성평가)

  • KNI3126 which is the eco-friendly material made by plant extracts - neem and matrine - have been evaluated for efficacies against 6 main pests and toxicity on natural enemies. Insecticidal efficacies of KNI3126 against plant hopper were above 95% at 5 days after treatment (DAT), whereas pesticidal effects against the cotton aphid were above 95% at 3 DAT and slightly decreased at 5 DAT. Insecticidal efficacy against the palm thrips resulted in lower control value as 68.1% than that of chemical insecticides. KNI3126 showed more than 95% control value against diamond back moth, suggesting that it could suppress the population of pest hard to control as eco-friendly material. Mortalities against two-spotted spider mite were about 80% at 1 DAT and over 90% at 5 DAT, respectively. For evaluation of the toxicity on natural enemies, effect against predatory natural enemy was classified as moderate selective toxicity based on the criterion of International Organization of Bio-Control (IOBC), whereas against parasitic natural enemies was found to be relatively safe. Repellent effect was weak against plant hopper, but strong against two-spotted spider mite. In the toxicity test for safety, KNI3126 was classified as the lowest level at acute oral, acute dermal and fish toxicity test and did not induce the irritancy at skin irritation test and eye irritation test.

Inheritance of Tetradifon Resistance in Two-spotted Spider Mite (Acari: Tetranychidae) and Its Cross Resistance (Tetradifon에 대한 점박이응애(Tetranychus urticae Koch) 저항성의 유전양식과 교차저항성)

  • A field colony of the two-spotted spider mite, Tetranychus urticae Koch, was selected with tetradifon for 4 mousing whole-plant residual method. This strain showed 371-fold resistance to tetradifon. The log dosehatchability lines of $F_{1}$ eggs(RS cross; Td5female$\times$Smale, and SR cross; Sfemale$\times$Td5male) were closer to the line of the resistant colony than to that of the susceptible. These differences could be due to cytoplasmic inheritance or maternal effect. The estimate of dominance index (D) for the $F_{1}$ eggs of RS cross was 0.998 and that for $F_{1}$ eggs of SR cross was 0.262. This indicates that tetradifon resistance is completely dominant in RS cross and incompletely dominant in SR cross. Td5 strain exhibited high levels of resistance to clofentezine, benzoximate, and chlorfencon, and no cross resistance to fenazaquin, pyridaben, flufenoxuron, tebufenpyrad, and fenothiocarb.

Control Efficacy of Controlled Atmosphere and Temperature Treatment System Against the Hawthorn Spider Mite, Tetranychus viennensis (환경조절열처리 기술을 이용한 벚나무응애(Tetranychus viennensis) 살비 효과)

  • The hawthorn spider mite, Tetranychus viennensis, is a pest of apples and a quarantine pest from some countries that import apples from Korea. A controlled atmosphere and temperature treatment system (CATTS) was developed as an alternative disinfestation method to methyl bromide fumigation treatment, and has been applied to control various insects and other arthropod pests on fruits. We applied CATTS to disinfect T. viennensis under conditions that were previously developed to control the peach fruit moth, Carposina sasakii. First, T. viennensis was sampled from Japanese apricot, Prunus mume, and identified by its morphological characters. In addition, both cytochrome oxidase I (COI) and internal transcribed spacer (ITS) sequences supported the morphological identification. Second, the heat-tolerant developmental stage was determined in T. viennensis. When a $46^{\circ}C$ heat treatment was applied to egg, nymph, and adult stages of T. viennensis, adults were the most tolerant stage. Third, when heat temperature was used along with 1% $O_2$ and 15% $CO_2$, the mites showed a significant increase in susceptibility to the heat treatment. Finally, CATTS at $46^{\circ}C$ with 15% $CO_2$ and 1% $O_2$ for 30 min resulted in 100% mortality of all T. viennensis development stages. These results indicated that CATTS isapplicable to disinfest T. viennensis in post-harvest apples.

  • Web structure of spiders is a foraging strategy as well as an investment to get prey. In order to make a good foraging decision spiders should change its foraging strategy in the basis of sensory information provided by potential prey species. We conducted experiments to demonstrate whether a wasp spider, Argiope bruennichi modifies its web-building behavior after experiencing sensory information emitted by a cricket species, Teleogryllus emma. To know how the web structures would be modified, we quantified web characteristics after providing sensory information of the cricket and compared to the control group. Web construction decreased in the course of the experiment in both, control group(without stimulus) and experimental group(with stimuli). The results did not show remarkable differences between two groups. However, on the first day after providing sensory information of the cricket, the web-building behavior of A. bruennichi decreased only in 3.7% of the experimental group individuals against 11.8% of the control group individuals. The number of the stabilimentum constructions decreased overall in both groups. On the first day the decreasing proportion was doubled in the control group(14.7%), comparing to the experimental group(7.4%). The individuals did not lower its web height and did not extend its web area according to sensory information of the prey.

Persistence of Chlorfenapyr in Paprika Leaf and Its Residual Biological Activity to Two Spotted Spider Mite, Tetranychus urticae (파프리카 잎 중 Chlorfenapyr의 잔류량 변화와 점박이응애에 대한 잔효 생물 활성)

  • A comparative experiment was conducted to investigate the persistence of chlorfenapyr residue and its biological activity to two spotted spider mite, Tetranychus urticae, after treatment of chlorfenapyr suspension concentrate (10% SC) on paprika plants at recommended (2000 times dilution) and double dosage. Biological half-lives of chlorpfenapyr were estimated as 8 and 5 days in leaves and fruits of paprika plants, respectively. While initial deposits of chlorfenapyr residues in leaves at recommended and double dosages were 22.22 and 37.75 ${\mu}g\;g^{-1}$ at the time of application, its residue decreased to 1.56 ${\mu}g\;g^{-1}$ and 3.62 ${\mu}g\;g^{-1}$ at 29 days after treatment, respectively. Residual biological activity of chlorfenapyr SC to T. urticae at recommended dosage lasted for 7 or 15 days on the basis of 24 or 48-hrs mortality test assayed with feeding on excised leaf chlorfenapyr-treated. The control efficacy to the mite showed a good agreement with the persistence of chlorfenapyr residues in leaves.

Biochemical Characterization of an Extracellular Protease in Serratia proteamaculans Isolated from a Spider (무당거미에서 분리한 Serratia proteamaculans에서 분비되는 단백질분해효소의 생화학적 특성)

  • Serratia proteamaculans isolated from the midgut of a spider formed big halos around the bacterial colonies, indicating that the bacterial strain produces an extracellular protease. Activity staining of the extracellular pro­tein fractions using zymogram also demonstrated that the major protein with an estimated molecular mass of 52 kDa contained a high proteolytic activity. The protease was purified to near electrophoretic homogeneity from the culture supernatant after filtration and ion exchange and size exclusion chromatography. The purified enzyme had a relatively high proteolytic activity between pH 6.0 and 10.0 and at broad temperature range. The proteolytic activity of the enzyme was not inhibited by phenylmethylsulfonyl fluoride but strongly inhibited by 1, 10-phenanthroline and EDTA. The activity also was dependent on the presence of $Ca^{++}\;and\;Zn^{++}$ ions. These observations indicate that the enzyme is a metalloprotease.

Biomimetic Analysis on the Spider Silk Apparatus for Designing the Nanofiber-spinning Nozzle (나노섬유 방사노즐 설계를 위한 거미 실크 방적장치의 생체모사 분석)

  • The biomimetic approach on the cuticular spinning nozzles of the major ampullate silk glands in the golden-web spider Nephila calvata has been attempted using various visualizing techniques of light and electron microscopes to improve the design of spinning nozzle for producing synthetic nanofibers spun from electrospinning apparatus. The major ampullate spigot which has the most effective nozzle system to produce nanofibers for dragline silk with high strength and elasticity is connected via the bullet type spigot on anterior spinneret with flexible terminal segment. The excretory duct which transports the liquid silk feedstock from ampulla to spigot is divided into 3 limbs by loops back on itself to form an S-shape morphology that is bundled in connective tissue. Final diameter of the nanofibers at nozzle was dramatically reduced by gradual narrowing of duct cuticle less than 10 times comparing to its original size of funnel region. Moreover, the funnel has a characteristic cuticular organization with porous microstructure which seems to be related to water removal from feedstock of silk precursors. High magnification electron micrographs also reveal the presence of the spiral grooves on the surface of the cuticular intima near the valve which presumed to reduce friction during rapid flow of liquid silk.
