Purification and Characterization of 2,4-Dichlorophenol Oxidizing Peroxidase from Streptomyces sp. AD001

  • Streptomyces sp. AD001 is a Gram-positive soil actinomycetes secreting an uncharacterized 2,4-dichlorophenol (DCP) oxidizing enzyme, whose activity is similar to the previously known Actinomycetes lignin-peroxidase (ALiP). This extracellular peroxidase was purified from Streptomyces sp. AD001 as a single protein band on an SDS-PACE by ammonium sulfate fractionation, Q-sepharose, concanavalin A, and Bio-Gel HTP column chromatographies. The molecular mass of the purified peroxidase was determined by SDS-PAGE to be 45.2 kDa, and 49.7 kDa with MALDI-TOF-MS, respectively. The highest level of peroxidase activity was observed at pH 7.5 and $30^{\circ}C$. The amino terminal sequence of the purified peroxidase (G-E-P-E-E-G-N-V-D-G-T-L) showed no significant homologies to my known proteins, suggesting that Streptomyces sp. AD001 may secrete a novel kind of bacterial peroxidase Initial rate kinetic data of the 2,4-DCP oxidation were best modeled with a random-binding bireactant system.

  • The analogs of cyclic hydroxamic acids containing N-OH group have been proposed to play effective laccase-mediators in kraft pulp bleaching with laccase/mediator system. The existing mediators (NHA, 1-HBT, VA), the best laccase-mediators reported so far, and selected several analogs of cyclic hydroxamic acids were evaluated as a laccase-mediator for kraft pulp bleaching. It was found that NHA was the most effective mediator for the Trychophyton sp. LKY-7 laccase (TrL) in kraft pulp bleaching with TrL/mediator system, increasing about 10% ISO of brightness and decreasing about 2.8 of kappa number after alkaline-peroxide bleaching. Of the cyclic hydroxamic acidstested, the NHP.1(N-hydroxypyridone analog) was shown to enable TrL to effectively degrade lignin in kraft pulp bleaching, demonstrating approximately similar effect with that of NHA. However, the effect of substituent patterns of cyclic hydroxamic acid analogs in kraft pulp bleaching was not observed. The inhibitions of NHA and NHP.1 on TrL were not exhibited in TrL/mediator system. As a new mediator for TrL, NHP.1 was considered to be able to use in kraft pulp bleaching with TrL/mediator system.

Application of Alkaline Xylanase of Cephalosporium sp. RYM-202 in Enzymatic Treatment of Kraft Pulps (Cephalosporium sp. RYM-202가 생산하는 알카리내성 xylanase를 이용한 크라프트 펄프의 효소적 처리)

  • Enzyme-aided bleaching of softwood and hardwood kraft pulps by a xylanase preparation from an alkalophilic fungus Cephalospotium sp. RYM-202 was studied. Maximal solubilization of Pulp xylan was obtained at 5$0^{\circ}C$ in both kraft pulps. The optimum pH of the enzyme for the hydrolysis of pulp xylan was 8.0 and more than 90% of the maximal activity was detected at 9.0. The positive effects of xylanase pretreatment on bleachability of softwood and hardwood kraft pulps were observed. The kappa number of softwood and hardwood kraft pulps was decreased by 3.7 and 2.0 units, respectively. The pulp fibre integrity was not significantly affected by xylanase pretreatment when the physical properties of handsheets made from xylanase-treated pulps were compared with those of handsheets from untreated pulps. These results indicate that the alkaline xylanase of Cephalospotium sp. RYM-202 is well suitable for application in enzymatic prebleaching of softwood and hardwood kraft pulps under the alkaline conditions.

  • The ever increasing demand for energy and the shortage of resources all over the world have generated interest in recycling renewable sources such as lignocelluloses which otherwise would go to waste and cause environmental pollution. Lignin is the incrustation material for cellulose and hemicellulose, therefore, cellulose and hemicellulose are not easily degraded. Recycling lignocellulosic wastes as agricultural material are benefit to everybody and everything. In order to improve ligninase and laccase production of Phanerochaete chrysosporium, BKM-F-1767 and Ceriporiopsis subvermispora, FP 90031-SP, were compared. The ligninase activity of P. chrysosporium was maximum on day 4.5 of shaking culture at 150 rpm 2.5 cm in a back and forth cycle. The laccase activity of C. subvermispora was maximum on day 5.5 for 2% malt extract+0.1% yeast extract+0.1% Tween 20+6 mM Benzyl alcohol culture medium at stationary state.

Nutritive Evaluation of Some Browse Tree Legume Foliages Native to Semi-arid Areas in Western Tanzania

  • Browse tree legume leaves from Acacia spp (A. nilotica, A. tortilis, A. polyacantha), Dichrostachys sp, Flagea villosa, Piliostigma thonningii, Harrisonia sp were evaluated for nutritive potential (chemical compositions and degradability characteristics) compared to Gliricidia sepium. Effect of tannins anti-nutritive activity on digestibility was also assessed by polyethylene glycol (PEG) tannin bioassay. Crude protein (CP), ash, neutral detergent fiber (NDF), acid detergent fiber (ADF) and acid detergent lignin (ADL) differed (p<0.05) between legume foliages. Mean CP, ash, NDF, ADF and ADL for fodder species tested were 158, 92, 385, 145, and 100 g/kg DM, respectively. CP ranged from 115 (P. thonningii) to 205 g/kg DM (G. sepium). Acacia spp had moderate CP values (g/kg DM) of 144 (A. nilotica), to high CP in A. tortilis (188) and A. polyacantha (194) comparable to G. sepium. The forages had relatively lower fiber compositions. A. nilotica had (p<0.05) lowest NDF, ADF and ADL (182, 68 and 44) compared to P. thonningii (619, 196 and 130) g/kg DM, respectively. Except G. sepium, all fodder species had detectable high phenolic and tannin contents greater than 5% DM, an upper beneficial level in animal feeding and nutrition. Mean total phenolics (TP), total tannins (TT) and condensed tannins (CT) (or proanthocyanidins) for fodder species tested were 139, 113 and 43 mg/g DM, respectively. F. villosa had (p<0.05) lowest TP and TT of 65 and 56 mg/g DM, respectively, compared to A. nilotica (237 and 236 mg/g DM, respectively). The CT varied (p<0.05) from 6 (F. villosa) to 74 mg/g DM (Dichrostachys sp). In vitro organic matter (OM) degradability (OMD) differed (p<0.05) between fodder species. G. sepium had (p<0.05) high degradability potential compared to A. polyacantha that had (p<0.05) the lowest OMD values. Forage degradability ranked: G. sepium>A. nilotica>P. thonningi>F. villosa>Dichrostachys sp>A. tortilis>A. polyacantha. Addition of PEG resulted to (p<0.05) improvement in in vitro OM digestibility (IVD). Increase in IVD was mainly due to binding action of PEG on tannins; and represents potential nutritive values previously depressed by tannins anti-nutritive activity. Browse fodder has potential as sources of ruminal nitrogen especially for ruminants consuming low quality roughages due to high protein, lower fiber compositions and high potential digestibility. However, utilization of browse supplements in ruminants is hampered by high phenolic and tannin contents. Deactivation of tannin anti-nutritive activity, possibly by feeding tanniniferous browse with other readily available nitrogen sources to dilute tannin anti-nutritive activity could improve utilization of browse fodder supplements. Further studies are needed to assess browse fodder palatability and intake, and their effect on growth performance in ruminants.

  • This study was carried out to investigate the wood properties for development the proper uses of lesser-known tree species. The tested species are Kebambang(Pentace sp.), Bawang-bawang(Dysoxylum alliaceum Bl .), Garu buaya( Gonystylus brunescens A. Shaw)grown in Indonesia. fundamental wood properties such as anatomical, physical, mechanical and chemical properties and characteristics related to wood processing were examined. The results are summarized as follows : The woods of these species are diffuse porous, coarse textured, and interlocked grained. The heartwood of Kebambang is distinguished clearly to the sapwood, but the heartwood of other species are not distinguished clearly to the softwood. The woods of Kebambang and Bawang-bawang have the moderate weight density and the low strength properties, but Garu buaya has the high weight density and the high strength properties. The woods of these species has high shrinkage. The sawing characteristics of Kebambang are excellent but the other species are moderate. The machining characteristics by planer and drying characteristics of these species are moderate. These woods has poor characteristics in bending processing and decay durability. The wood of Bawang-bawang has high content of the extractives and lignin.

Evaluation of Soil Streptomyces spp. for the Biological Control of Fusarium Wilt Disease and Growth Promotion in Tomato and Banana

  • Fusarium oxysporum f. sp. lycopersici (Fol) and Fusarium oxysporum f. sp. cubense (Foc), are the causal agent of Fusarium wilt disease of tomato and banana, respectively, and cause significant yield losses worldwide. A cost-effective measure, such as biological control agents, was used as an alternative method to control these pathogens. Therefore, in this study, six isolates of the Streptomyces-like colony were isolated from soils and their antagonistic activity against phytopathogenic fungi and plant growth-promoting (PGP) activity were assessed. The results showed that these isolates could inhibit the mycelial growth of Fol and Foc. Among them, isolate STRM304 showed the highest percentage of mycelial growth reduction and broad-spectrum antagonistic activity against all tested fungi. In the pot experiment study, the culture filtrate of isolates STRM103 and STRM104 significantly decreased disease severity and symptoms in Fol inoculated plants. Similarly, the culture filtrate of the STRM304 isolate significantly reduced the severity of the disease and symptoms of the disease in Foc inoculated plants. The PGP activity test presents PGP activities, such as indole acetic acid production, phosphate solubilization, starch hydrolysis, lignin hydrolysis, and cellulase activity. Interestingly, the application of the culture filtrate from all isolates increased the percentage of tomato seed germination and stimulated the growth of tomato plants and banana seedlings, increasing the elongation of the shoot and the root and shoot and root weight compared to the control treatment. Therefore, the isolate STRM103 and STRM104, and STRM304 could be used as biocontrol and PGP agents for tomato and banana, respectively, in sustainable agriculture.
