17β-estradiol mediated effects on pluripotency transcription factors and differentiation capacity in mesenchymal stem cells derived porcine from newborns as steroid hormones non-functional donors

  • The estrogen-mediated effect of mesenchymal stem cells (MSCs) is a highly critical factor for the clinical application of MSCs. However, the present study is conducted on MSCs derived from adult donors, which have different physiological status with steroid hormonal changes. Therefore, we explores the important role of $17{\beta}$-estradiol (E2) in MSCs derived from female and male newborn piglets (NF- and NM-pBMSCs), which are non-sexually matured donors with steroid hormones. The results revealed that in vitro treatment of MSCs with E2 improved cell proliferation, but the rates varied according to the gender of the newborn donors. Following in vitro treatment of newborn MSCs with E2, mRNA levels of Oct3/4 and Sox2 increased in both genders of MSCs and they may be correlated with both estrogen receptor ${\alpha}$ ($ER{\alpha}$) and $ER{\beta}$ in NF-pBMSCs, but NM-pBMSCs were only correlated with $ER{\alpha}$. Moreover, E2-treated NF-pBMSCs decreased in ${\beta}$-galactosidase activity but no influence on NM-pBMSCs. In E2-mediated differentiation capacity, E2 induced an increase in the osteogenic and chondrogenic abilities of both pBMSCs, but adipogenic ability may increased only in NF-pBMSCs. These results demonstrate that E2 could affect both genders of newborn donor-derived MSCs, but the regulatory role of E2 varies depending on gender-dependent characteristics even though the original newborn donors had not been affected by functional steroid hormones.

Concentration-dependent in vitro Anti-osteoarthritis Effects of Mixed Formula - Pomegranate Concentrate Powder: Eucommiae Cortex: Achyranthis Radix 5:4:1 (g/g) on the Primary Cultured Rat Articular Chondrocytes

  • The objective of present study is to evaluate concentration-dependent in vitro anti-osteoarthritic (OA) effects of synergic mixed formula consisted of dried pomegranate juice concentrate powder, Eucommiae Cortex aqueous extract and Achyranthis Radix aqueous extract 5:4:1 (g/g) mixture on the primary cultured rat articular chondrocytes. First, any cytotoxic effect of mixture was observed using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium Bromide) assay. Next, cyto-protective effect of test substances was evaluated by using the recombinant human interleukin $(rhIL)-1{\alpha}$ induced chondrocytes. In addition, anti-inflammatory effects were also observed on the lipopolysaccaride (LPS) treated chondrocytes through prostaglandin $E_2(PGE_2)$ productions and 5-lipoxygenase (LPO) activities, and inhibitory effects on matrix metalloproteinase (MMP)-2 and MMP-9 activities were observed on $rhIL-1{\alpha}$ treated chondrocytes with their extracellular matrix (ECM) related mRNA expressions. No obvious cytotoxic effects of mixture were demonstrated. Inflammatory damages of chondrocytes and related ECM degradations induced by treatment of LPS or $rhIL-1{\alpha}$ were significantly and concentration-dependently inhibited by pretreatment of mixture from a concentration level of 0.001 mg/ml to 1 mg/ml. In addition, mixture showed $IC_{50}$ for $rhIL-1{\alpha}-induced$ MMP-2 and MMP-9 activities as 44.01 and $162.47{\mu}g/ml$, and also showed $EC_{50}$ for $rhIL-1{\alpha}-induced$ inhibition of collagen type II, SOX9 and aggrecan mRNA expression as 8.61, 10.79 and $4.47{\mu}g/ml$, respectively. It is observed that mixture showed concentration-dependent anti-inflammatory and cytoprotective ECM preserved effects on the primary cultured rat articular chondrocytes without cytotoxicity.

Estimation of SO2 emissions in large point sources at Dangjin City using airborne measurements (항공관측 결과를 활용한 당진시 대형사업장에서의 황산화물 배출량 평가)

  • Based on the airborne measurement results over a coal fired power plant and steel work in Dangjin city, SO2 emission amounts of each site are estimated (top-down emission). Airborne measurements were carried out on May-June and October-November 2019. The estimated SO2 emission in 2019 for the power plant was 1502.1 kg/hr and that for the steel work was 2850.5 kg/hr, higher as much as a factor of 2.5 and 2.0, respectively, than the emission amounts provided by both facilities (bottom-up emission). The outcomes strongly illustrates that well designed airborne observations can serve a quantitative diagnostic tool for bottom-up emission estimates. Further research direction to improve the reliability of the top-down emission estimates is suggested.

  • The physical properties of DME(Dimethyl Ether) are very similar to LPG and well-mixed. As cetane number of DME is similar to diesel fuel that can replace diesel fuel and alternative energy. DME is a clean energy source that can be manufactured from various raw materials such as natural gas, CBM(Coal Bed Methane) and biomass. DME has no carbon-carbon bond in its molecular structure and its combustion essentially generates no soot as well as no SOx. The development of DME process in KOGAS have 4 section. First, syngas section can be manufactured various syngas ratio. This completes the tri-reforming process for the synthesis gas ratio of approximately 4.0 to 1.0 range can be adjusted. Second, $CO_2$ is removed from the $CO_2$ removal section of about 92~99%, so the maximum concentration of $CO_2$ entering the DME synthesis reactor should not exceed 8%. Third, in the DME synthesis section, if the temperature of DME reactor increases, the activity of DME catalyst increased. but for the long-term activity is desirable to maintain the proper temperature. Finally, the purity of DME in the DME purification section is over 99.6%.

Combustion characteristics of two imported Indonesia coals as a pulverized fuel of thermal power plants (인도네시아산 발전용 수입 석탄 2종의 연소특성 비교 평가)

  • Combustion reactivity and thermal behavior of two imported coals used as a pulverized fuel of commercially thermal power plant were investigated by thermogravimetric analysis (TGA) and large scale test furnace of 200 kg/hr. TGA results showed that combustion efficiency of high moisture coal has lower than reference coal due to the slow combustion completion rate although it has the low ignition temperature, and activation energies of high moisture coal with 79 kJ/mol for overall combustion was higher than reference coal of 53 kJ/mol. Test furnace results ascertained that flame of black band of high moisture coal during the combustion in boiler broke out compared to reference coal and then it becomes to unburned carbon due to the less reactivity and combustion rate. But, Blending combustion of high moisture coal with design coal of high sulfur are available because sulfur content of high moisture coal was too low to generate the low SOx content in flue gas from boiler during the combustion. The ash analysis results show that it was not expected to be associated with slagging and fouling in pulverized coal fired systems due to the low alkali metal content of $Na_2O$ and $K_2O$ compared to bituminous coal.

Effects of Microbial Phytase Supplementation on Growth Performance and Nutrient Excretion in Growing Pigs (육성돈에 있어 성장 및 영양소 배설에 대한 미생물성 Phytase의 첨가 효과)

  • For Exp, 1, eighty [(DurocYorkshire)Landrace) pigs(38.63$\pm$0.68kg average initial BW) were used to evaluate the effects microbial phytase supplementation on the growth performance and nutrient digestibility in growing pigs. There were ten pigs per pen and four pens per treatment. Treatments were 1) CON(com-SBM based diet), 2) Low-P(low-P diet+500 FTU/kg microbial phytase). For overall period, ADG, ADFI and gain/feed were significantly improved(P<0.05) by supplementation of Low-P diet. Also, pigs fed Low-P diet increased N, crude ash, Ca and P digestibilities compared to pigs fed CON diet(P<0.05). For Exp. 2, sox [(DurocYorkshire) Landrace) pigs(32.7kg average initial BW) were used to evaluate the effects microbial phytase supplementation on the nutrient excretion in growing pigs. Experimental treatments were as described for Exp. 1. pigs fed Low-P diet decreased N(8.70 vs 6.43g/d), crude ash(38.13 vs. 25.33 g/d), Ca(6.03 vs. 3.93g/d) and P(4.53 vs. 2.77g/d) excretion compared to pigs fed CON diet(P<0.05). In conclusion, microbial phytase supplementation in low P diets improved growth and nutrient digestibility.

Comparison of Growth Characteristics and Quality of Korean Ginseng (Panax ginseng C.A. Meyer) by Different Shade Materials (해가림자재 종류별 고려인삼의 생육과 품질특성 비교)

  • This research investigated growth of ginseng regard to sunshade materials and micro metrological phenomena under the sunshade material to product high quality ginseng. Followings are results of investigation of active ingredient from raw ginseng producted under the sunshade materials. The highest temperature under the sunshade material from June to August was measured from a Three-layered blue and one-layered black polyethylene net (TBOBFN) followed by a aluminium-coated polyethylene sheet (ACPS) and blue polyethylene sheet (BPS). The highest light penetration of $381.7{\mu}mol/s/m^2$ was obtained from the BPS at August, also the temperature was highest at the ginseng field. Growth of above ground part of 2~3 year old ginseng under the ACPS was the highest, followed by the ACPS and the BPS. In case of 4 year old ginseng, the ACPS and the BPS was the same, followed by the TBOBPN. Root length and diameter of 2~3 year old ginseng showed difference among treatments, but 4 year old ginseng was not showed difference. Meanwhile, the highest amount of root of 4 year old ginseng of $896g/m^2$ was obtained from ACPS. The total amount of ginsenocide of 4 year old ginseng under the ACPS was highest, followed by the BPS and the TBOBPN. Chromaticity of ginseng root under the TBOBPN was highest, followed by the ACPS and the BPS.

Protective Effect of Ginsenoside Rb1 on Hydrogen Peroxide-induced Oxidative Stress in Rat Articular Chondrocytes

  • The abnormal maturation and ossification of articular chondrocytes play a central role in the pathogenesis of osteoarthritis (OA). Inhibiting the enzymatic degradation of the extracellular matrix and maintaining the cellular phenotype are two of the major goals of interest in managing OA. Ginseng is frequently taken orally, as a crude substance, as a traditional medicine in Asian countries. Ginsenoside $Rb_1$, a major component of ginseng that contains an aglycone with a dammarane skeleton, has been reported to exhibit various biological activities, including anti-inflammatory and anti-tumor effects. However, a chondroprotective effect of ginsenoside $Rb_1$ related to OA has not yet been reported. The purpose of this study was to demonstrate the chondroprotective effect of ginsenoside $Rb_1$ on the regulation of pro-inflammatory factors and chondrogenic genes. Cultured rat articular chondrocytes were treated with 100 ${\mu}M$ ginsenoside $Rb_1$ and/or 500 ${\mu}M$ hydrogen peroxide ($H_2O_2$) and assessed for viability, reactive oxygen species production, nitric oxide (NO) release, and chondrogenic gene expression. Ginsenoside $Rb_1$ treatment resulted in reductions in the levels of pro-inflammatory cytokine and NO in $H_2O_2$-treated chondrocytes. The expression levels of chondrogenic genes, such as type II collagen and SOX9, were increased in the presence of ginsenoside $Rb_1$, whereas the expression levels of inflammatory genes related to chondrocytes, such as MMP1 and MMP13, were reduced by approximately 50%. These results suggest that ginsenoside $Rb_1$ has potential for use as a therapeutic agent in OA patients.

Effects of HPL-04 on Degenerative Osteoarthritis (퇴행성 골관절염에 대한 HPL-04의 효과)

  • HanPoong Leading (HPL)-04 were prepared with different oriental medicines (balk of Kalopanax pictus balk, Chaenomelis Fructus, Angelica gigas root, Zingiber officinale, Raphanus sativus Linne and Saururus chinensis Baill.) to investigate the protective effects of HPL-04 on cartilage degradation in knee osteoarthritis (OA). Rat articular chondrocytes incubated with rhIL-$1{\alpha}$ markedly increased matrix metalloproteinase (MMP)-2 and 9 activities, decreased cell viability and reduced chondrogenic gene expression. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, MMP-2 and 9 activities and real time RT-PCR indicated that HPL-04 counteracted these harmful effects in dose-dependent manner. In addition, for experimental OA in vivo, monosodium iodoacetate (MIA, 0.5 mg/50 ${\mu}L$) was injected into knee joints of rats and administered HPL-04 to rats for 4 consecutive weeks after MIA treatment. The experimental data showed that treatment with HPL-04 significantly prevented of MMP-2 and 9 activities in articular cartilage. Histopathological and micro-CT evaluations of the knee joints also revealed that HPL-04 effectively ameliorated MIA-induced degenerative OA. In conclusion, HPL-04 has potential applicability for the prevention and treatment of degenerative OA.

Maxillary sinus floor elevation using autogenous skin-derived mesenchymal stem cells in miniature pigs (미니돼지에서 자가 피부유래 간엽성 줄기세포를 이용한 상악동저 거상술)

  • Introduction: In our previous studies, we isolated porcine skin-derived mesenchymal stem cells (pSDMSCs) from the ears of adult miniature pigs and evaluated the pluripotency of these pSDMSCs based on expressions of transcription factors, such as Oct-4, Sox-2, and Nanog. Moreover, the characteristic of mesenchymal stem cells was revealed by the expression of various mesenchymal stem cell markers, including CD29, CD44, CD90, and vimentin. The aim of this study was to evaluate in vivo osteogenesis after maxillary sinus lift procedures with autogenous pSDMSCs and scaffold. Materials and Methods: The autogenous pSDMSCs were isolated from the 4 miniature pigs, and cultured to 3rd passage with same methods of our previous studies. After cell membranes were labeled using a PKH26, $1{\times}10^{7}$ cells/$100{\mu}L$ of autogenous pSDMSCs were grafted into the maxillary sinus with a demineralized bone matrix (DBM) and fibrin glue scaffold. In the contralateral control side, only a scaffold was grafted, without SDMSCs. After two animals each were euthanized at 2 and 4 weeks after grafting, the in vivo osteogenesis was evaluated with histolomorphometric and osteocalcin immunohistochemical studies. Results: In vivo PKH26 expression was detected in all specimens at 2 and 4 weeks after grafting. Trabecular bone formation and osteocalcin expression were more pronounced around the grafted materials in the autogenous pSDMSCs-grafted group compared to the control group. Newly generated bone was observed growing from the periphery to the center of the grafted material. Conclusion: The results of the present study suggest that autogenous skin-derived mesenchymal stem cells grafting with a DBM and fibrin glue scaffold can be a predictable method in the maxillary sinus floor elevation technique for implant surgery.
