Purification and Characterization of Mouse Liver Rhodanese

  • Rhodanese from mouse liver was purified to near homogeneity by ammonium sulfate precipitation, CM-Sephadex ion exchange, hydroxyapatite and Sephacryl S-200-HR gel filtration chromatographies with a purification of 776 folds. The molecular weight was determined by Sephadex G-150 gel filtration and found to be 34.8 KDa. SOS-PAGE showed molecular weight 34 KDa and two identical subunits splitting by aging for 3 weeks at $-70^{\circ}C$ the molecular weight of which was 17 KDa. The optimal pH of enzyme activity was 9.4 and the pI value of the enzyme was 6.6. Rhodanese showed the optimal reaction temperature of $25^{\circ}C$ and near linear increasing pattern until 10 min. incubation. $K_m$ values of rhodanese for KCN and $Na_{2}S_{2}O_{3}$ as substrates were 12.5 mM and 8.3 mM, respectively. Rhodanese activity was inhibited by more than 70% at a concentration of 100 ${\mu}M$ of $Ni^{2+}$, $Zn^{2+}$, $Cd^{2+}$, $Hg^{2+}$ and $Cu^{2+}$. Other metal ions, such as $Mn^{2+}$, $Mg^{2-}$, $Ca^{2+}$, and $Fe^{2+}$ showed no effect on rhodanese activity.

Protective Activities of Fractions of Water Extract Obtained from Artemisia iwayomogi Kitamura against Oxidative Stress-induced Mutagenesity: Correlation with Their Reactive Oxygen Scavenging Activity

  • Water extracts of injinssuk (Artemisia iwayomogi Kitamura) (WE) were obtained from the dried and ground leaves and stems of injinssuk. The WE was further fractionated into crude polysaccharide (C-PS) and nonpolysaccharide fractions (N-PS). The protective activities against the tert-butyl hydro peroxide induced mutangenecity on Escherichia coli PQ37 and reactive oxygen species scavenging activity of the WE, C-PS, and N-PS were studied. The WE obtained from leaves showed a significantly higher inhibitory effect on the mutagenicity than WE from stem. The WE obtained from the leaves having higher crude polysaccharide content but lower content of total carbohydrates had significantly higher antimutagenicity than that from the leaves with lower crude polysaccharide but higher total carbohydrate contents. Further study showed that C-PS fraction showed markedly stronger antimutagenic effect than N-PS. C-PS was also more effective than N-PS for hydroxyl radical scavenging activity, but was similar to N-PS in superoxide radical scavenging activity.

  • The action mechanism of the inhibitory effect of some natural products on the DNA strand break and DNA damage was investigated in vitro and in vivo. In the E. coli chromosomal DNA strand break experiment in vitro, three mushroom water extracts were effective on the DNA strand breaking by daunorubicin. Phellinus linteus water extract inactivated daunorubicin, a DNA strand breaking agent, but did not protect DNA from daunorubicin-induced DNA strand breaking. Agaricus blazei water extract inhibited DNA strand breaking action of daunorubicin not only by daunorubicin inactivation, but also by DNA protection from daunorubicin. An inhibitory effect of Ganoderma lucidum water extract on the DNA strand break was based on the DNA protection rather than daunorubicin inactivation. In vivo mutagen assay system (SOS-chromotest), among three mushroom water extracts Phellinus linteus water extract was the most effective one on the inhibition of DNA damage by 4-NQO. The results suggest that all three mushroom water extracts inhibit daunorubicin-induced DNA damage and in vivo DNA damaging action of 4-NQO by the reaction of mutagen inactivation or DNA protection from the mutagen.

Genotypic and Environmental Effects on Flour Properties in Korean Winter Wheat

  • Flour characteristics of Korean winter wheat grown in Suwon in 1997 and 1998, and in Suwon and Deokso in 1998 were evaluated. Korean winter wheat cultivars were significantly influenced by years and locations in flour properties such as ash content, protein content, damaged starch content, starch swelling volume and power. Protein content was highly correlated with starch damage and alkaline water retention capacity. There were highly significant correlations between mixing time of mixograph and SDS sedimentation volume. Swelling properties of flour and starch were highly correlated with pasting properties of flour and starch, respectively. Compared to commercial flours for baking, Alchanmil, Gobunmil, Keumkangmil and Tapdongmil showed similar protein content, SOS sedimentation volume and mixograph mixing time. Eunpamil, Geurumil, Olgeurumil, Suwon 258, Suwon 261, Suwon 265, Suwon 275, Suwon 276, Suwon 277, Suwon 278 and Urimil had similar values to commercial noodle flours in SDS sedimentation volume. Alchanmil, Olgeurumil, Suwon 274, Suwon 275, Suwon 276 and Urimil showed higher swelling and pasting properties than the others. Chokwang, Olgeurumil, Suwon 277 and Urimil were similar to commercial cookie flours. Friabilin-absence lines showed higher protein content and starch damage than those of friabilin-presence lines. Absence lines of 1D$\times$2.2 + 1Dy12 subunit in high molecular weight glutenin subunits showed higher SDS sedimentation volume and mixing time of mixograph than those of presence lines.

버섯첨가된장의 생리활성 작용 (Physiological Activity in Doenjang Added with Various Mushrooms)

  • The study was carried out to evaluate the radical scavenging activity, and antimutagenic and anticancer effects in Korean soybean paste(doenjang) added with various mushrooms. Ganoderma lucidum doenjang(Gl-TD) showed significant inhibitory activity against the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, with an IC$\sub$50/ value of 245$\mu\textrm{g}$/$m\ell$. In contrast, the other doenjang varieties appeared to have weaker antioxidant activity. Four kinds of doenjang didn't have any antimutagenic activity against N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) by adding of 5mg/plate, but exhibited a strong inhibitory effect(55∼70%) against aflatoxin B$_1$(AFB$_1$). Especially Phellinus linteus doenjang(Pl-TD) inhibited more than 70% of the mutagenicity induced by AFB$_1$ in Salmonella typhimurium TA100. On the other hand, the mushroom doenjang varieties showed relatively weak activity toward MNNG in SOS chromotest, their inhibitory rate ranging from 23% to 33%. Methanol extracts of Gl-TD and Pl-TD inhibited by 91∼92% the growth of AGS gastric cancer cells in a concentration of 100$\mu\textrm{g}$/$m\ell$.

SOS 구조를 개선한 OSOn 및 OSOSOn 구조의 비휘발성 메모리

  • 유리 기판 상에 system on panel (SOP) 구현을 위한 비휘발성 메모리 (NVM)를 제작하였다. 기존에 사용되던 charge storage layer인 SiNx 대신에 a-Si를 사용하여 전하 저장량 증가 및 전하유지 특성 향상시켰다. 그 결과 bandgap이 작아 band edge 저장 가능하였으며, SiNx 와 마찬가지로 a-Si 내 트랩에 저장되었다. $SiO_2$/a-Si와 a-Si/SiON 계면의 결함 사이트에 전하 저장되었으며, 또한 bandgap이 작아 트랩 또는 band edge에 위치한 전하들이 높은 bandgap을 가지는 blocking 또는 tunneling layer를 통하여 빠져 나오기 어려웠다. ONOn 구조의 두께와 동일한 OSOn 박막을 사용한 구조에서는 전하 저장 특성은 뛰어나나 기억유지 특성이 나빴다. 이에 대한 향상 방안으로는 Tunneling 박막의 두께를 증가시키는 것과 OSOSOn 적층 구조 소자를 만드는 방법이 있다. Tunneling 박막의 두께를 증가시킨 소자는 기억유지 특성 향상되는 특성을 보였으며 OSOSOn 적층 구조 소자는 전하저장 및 기억유지 특성 향상을 보였다. 특히, OSOSOn 구조의 경우 2개의 터널링 barrier를 사용함으로써 전하 저장 사이트의 증가에 기여하며, 기억 유지 특성도 좋아졌다. 본 연구에서 소자는 NVM이 아닌 MIS 구조로만 제작되었다.

Purification and Characterization of Trypsins Affecting on the Autolysis of Shrimp, Penaeus japonicus

  • Two trypsins were purified from shrimp hepatopancreas through ammonium sulfate fractionation, Q-Sepharose ionic exchange, benzamidine Sepharose-6B affinity, and Sephacryl S-300 gel chromatography. Both enzymes had a single polypeptide chain with a molecular weight (M.W.) of 32 kDa by sodium dodecylsulfate polyacrylamide gel electrophoresis (SOS-PAGE), although trypsin A and B were estimated to be a molecular weight of 27.2 and 22.8 kDa, respectively, using Sephacryl S-300 gel filtration. Both trypsins had similar amino acid compositions and rich in glycine, valine, alanine, aspartic acid, and glutamic acid, but low in methionine and basic amino acids. Both enzymes were completely inactivated by soybean trypsin inhibitor (SBTI), phenylmethylsulfonyl fluoride (PMSF), tosyl-L-lysine chloromethyl ketone (TLCK), benzamidine, leupeptin, however, not affected by tosyl-L-phenylalanine chloromethyl ketone (TPCK) and pepstatin.

Antioxidative and Radical Scavenging Properties of Extracts from Geum japonicum

  • Extracts from Geum japonicum (Bammu in Korean) were analyzed for their antioxidative activities and scavenging effects on free radicals. The ethyl acetate fraction of G. japonicum methanol extract (EFGJ) showed a remarkable scavenging activity on the 1,1-diphenyl-2-picrylhydrazyl radical. EFGJ also showed excellent antioxidative activity on linoleic acid during long-period storage and on rat liver microsome peroxidation system, and good anti-peroxidation effect on lipid in Rancimat system using lard, palm oil, and perilla oil, as compared with BHT and ${\alpha}$-tocopherol. Varying effects of antioxidative activity of the EFGJ on various fatty acids were observed. The prevention of linoleic acid and linolenic acid peroxidation was superior to ${\alpha}$-tocopherol, but inferior to BHT. The methanol extract of G. japonicum did not show mutagenicity as revealed through SOS chromotest. Based on the results, the extracts of G. japonicum may be useful source as natural antioxidants.

Production of nitric oxide by murine macrophages induced by lipophosphoglycan of Leishmania major

  • Protozoan parasites of the genus Leishmania cause a number of important human diseases. One of the key determinants of parasite infectivity and survival is the surface glycoconjugate lipophosphoglycan (LPG). In addition, LPG is shown to be useful as a transmission blocking vaccine. Since culture supernatant of parasite promastigotes is a good source of LPG, we made attempts to characterize functions of the culture supernatant, and membrane LPG isolated from metacyclic promastigotes of Leishmania major. The purification scheme included anion-exchange chromatography, hydrophobic interaction chromatography and cold methanol precipitation. The purity of supernatant LPG (sLPG) and membrane LPG (mLPG) was determined by SOS-PAGE and thin layer chromatography. The effect of mLPG and sLPG on nitric oxide (NO) production by murine macrophages cell line (J77 4.1 A) was studied. Both sLPG and mLPG induced NO production in a dose dependent manner but sLPG induced significantly higher amount of NO than mLPG. Our results show that sLPG is able to promote NO production by murine macrophages.

Sejong Open cluster Survey - II. The star forming region IC 1848 (W5)

  • UBVI and $H{\alpha}$ CCD photometry of IC 1848, one of active star-forming regions in Cas OB6, is carried out as a part of Sejong Open cluster Survey (SOS) project. An OB association is an ideal laboratory for studying the triggered star formation and star formation history. Our purposes are to provide deep photometric data up to 21 mag in V and physical parameters of IC 1848. We classify 79 early-type stars and 186 pre-main sequence (PMS) stars as being the members of the cluster using photometric criteria. The IR excess emission PMS stars by Koenig et al. (2008) are also included as members of IC 1848. Total number of members is 414. We derive the interstellar reddening (=$0.659{\pm}0.058$ mag), reddening law ($R_V=4.0{\pm}0.1$), distance modulus ($V_0-M_V=12.0{\pm}0.1$ mag) using the early-type members of IC 1848. We also determine the age of the cluster ($3.5{\pm}1.5$ Myr) by placing the theoretical isochrones on the HR diagram.
