  • The study was conducted to determine the optimal glucose, superoxide dimutase(SOD) and catalase levels during the in vitro culture of porcine oocytes matured and fertilized in vitro for morulae and blastocyst development. Oocytes were cultured for 0~8 days in TCM-199 medium supplemented with 20% FCS, different glucose, SOD and catalase levels. The results are summairzed as follows ; 1. The in vitro developmental rates of porcine oocytes cultured in TCM-199 medium containing 0.1, 0.3, 0.5, 1.0, 3.0 mM glucose levels 0~3 and 0~8 days after insemination were 22.8, 24.2, 21.9, 20.0, 12.1 and 21.9, 26.7, 25.0, 22.6, 16.7%, respectively. 2. The in vitro developmental rates of porcine oocytes cultured in TCM-199 medium containing 100, 200, 300, 500 $\mu\textrm{g}$/ml SOD levels 0~3 and 0~8 days after insemination were 16.7~23.3 and 16.7~25.0%, respectively. High levels of SOD(500 $\mu\textrm{g}$/ml) significantly reduced the rates of molurae and blastocysts stage(P<0.05). 3. The in vitro developmental rates porcine oocytes cultured in TCM-199 medium containing 100, 200, 300, 500 $\mu\textrm{g}$/ml catalase levels 0~3 and 0~8 days after insemination were 18.8~26.7 and 19.4~28.1%, respectively, and there was significant differences on the development to the molurae and blastocysts stage among the cumulus cells and glucose levels.

  • Reactive oxygen species are produced under diabetic conditions and possibly cause various forms of tissue damage in patients with diabetes mellitus. The aim of this study was to examine the effects of high glucose on the alloxan-induced beta cell injury. The insulinoma (RINm5F) cells were clutured either with high glucose (22.2 mM) or normoglucose (5.6 mM) in RPMI 1460 media for 3 days. The SOD activities were determined by spectrophotometric assay and nitroblue tetrazolium (NBT) stain. The effects of high glucose on the cytotoxicity of alloxan were also investigated in RINm5F cells and the cells viability were determined by 3-(4,5-dimethylthiazolyl)-2,5-diphenyltetrazolium bromide (MTT) methods. Results showed that the CuZn-SOD activity was decreased but Mn-SOD activity was increased significantly in RINm5F cells cultured with high glucose (22.2 mM) media. The cytotoxicity of alloxan was increased by high glucose compared with normoglucose in RINm5F cells. Diethyl-dithiocarbarmate (DDC), as inhibitor of CuZn-SOC, also potentiate the alloxan-induced cytotoxocity in RINm5F cells. These results suggest that, in RINm5F cells, short term culture with high glucose media decreases Cu-Zn-SOD activity and the decreased activity of CuZn-SOD many one of the causative factors of beta-cell injury induced by high glucose.

  • For the study of method for promoting the germination of Korean sod seeds harvseted newly some physical and chemical treatments were tested in this experiment. The summarized results of these tests are as follows: 1. Storage of wet seed in low temperature of 0~$2^{\circ}C$ during 2~30 days was seemed the most effective treatment for promoting the germination of sod seed. 2. The treatment that Soaking and mixing up the seeds in cone sulphuric acid about one and half minutes and then washing off in the water promoted fairly the germination of sod seeds. 3. Better germination of sod seeds was found in the light than in the dark or soil. 4. Cutting the top of seed and crushing of seed coat with sand seemed to promote slightly the germination of sod seeds. 5. No promoting effect for germination of sod seeds was found with treatments of Gibberellin, $NH_4$ $NO_3$ and NAA.

  • The experiment was conducted to find the changes of early growth and chemical components such as chlorophyll content, superoxide dismutase(SOD) activity, free proline content on the different manganese chloride concentration(2,500, 3,500 and 4,500ppm) in rice seedling. Root growth was decreased in highest concentration, 4,500 ppm of Mn compared with the control and germination rate was also decreased 43% at 4,500 ppm of Mn. Chlorophyll content was decreased at Mn 4,500ppm with 1.16mg. Free proline content at 3 day after germination in Mn 4,500ppm was highest relative to the other manganese chloride concentrations. SOD activity was gradually increased as manganese chloride concentration was increased. As a result, it was suggested that an increment of free proline and SOD activity results from the higher manganese chloride concentration.

  • Cells express several antioxidant enzymes to scavenge reactive oxygen species (ROS) responsible for oxidative damages and various human diseases. Therefore, antioxidant enzymes are considered biomedicine candidates. Among them, extracellular superoxide dismutase (SOD3) had showed prominent efficacy against asthma and inflammation. Despite its advantages as a biomedicine, the difficulty in obtaining large quantity of active recombinant human SOD3 (rhSOD3) has limited its clinical applications. We found that a significant fraction of over-expressed rhSOD3 was composed of the inactive apo-enzyme and its potency against inflammation depended on the rate of metal incorporation. Also, purified rhSOD3 was unstable and lost its activity very quickly. Here, we suggest an ideal preparative method to express, purify, and store highly active rhSOD3. The enzymatic activity of rhSOD3 was maximized by incorporating metal ions into rhSOD3 after purification. Also, albumin or polyethylene glycol prevented rapid inactivation or degradation of rhSOD3 during preparative procedures and long-term storage.

  • This study aims to demonstrate the effect of SOD (superoxide dismutase), one of the antioxidant enzymes, on the ultrastructural changes in the parietal cells caused by the administration of cisplatin in the rat. A total of 60 healthy Sprague-Dawley rats weighing about 200 gm were used as experimental animals. Cisplatin (6 mg/kg) was administered intraperitoneally to rats pretreated with 15,000 unit/kg of SOD or rats without the pretreatment. The experimental animals were sacrificed at 6 hours, 12 hours, 24 hours and 3 days after the administration of cisplatin. The results were as follows: 1. SOD alone did not affect the ultrastructural changes in the gastric parietal cells in the rat. 2. Irregular shaped mitochondria, mitochondria with dim cristae, dilated cristae, ruptured outer membrane, electron lucent matrix and degenerative mitochondria were seen in cisplatin treated rat. Whorled membranous body, many lysosomes and large vacuole were observed in the gastric parietal cells in cisplatin treated rat. 3. Mitochondria with dilated cristae and electron lucent matrix and irregular shaped mitochondria were observed in the gastric parietal cells of the cisplatin treated rat with pretreatment of SOD. These results suggest that SOD attenuates the toxic effect of the cisplatin in the gastric parietal cells of the rat.

  • Leaves of two-week old seedlings of tomato (Lycopersicon esculentum) were treated with various concentrations (0∼100 M) of $CdCl_2$ for up to 9 days and subsequent growth of seedlings, symptoms of oxidative stress and isozyme activities of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and peroxidase (POX) were investigated. Compared with the non-treated control, Cd exposure decreased biomass but increased Cd accumulation, hydrogen peroxide production and lipid peroxidation as malondialdehyde (MDA) formation in leaves and roots. Further studies on the developmental changes of isozyme activities showed that Fe-SOD, Cu/Zn-SOD and one of three APX isozymes decreased and CAT and one of four POX isozymes increased in leaves, whereas Fe-SOD, one of three POX isozymes and two of four APX isozymes decreased and CAT increased in roots, showing different expression of isozymes in leaves and roots with Cd exposure level and time. Based on our results, we suggest that the reduction of seedling growth by Cd exposure is the oxidative stress resulting from the over production of $H_2O_2$ and the insufficient activities of antioxidant enzymes particularly involved in the scavenging of $H_2O_2$. Further, the decreased activities of SOD and APX isozymes of chloroplast origin, the increased activities of CAT and POX and high $H_2O_2$ contents with Cd exposure might indicate that Cd-induced oxidative stress starts outside chloroplast.

  • This study was conducted to investigate how to enhance resistance to oxidative stress in petunia progeny obtained by a crossing between transgenic plants, MnSOD (SOD2) ($T_4$) and NDPK2 ($T_2$), to develop transgenic petunia much more resistant to environmental stress. At the treatment of MV 200 ${\mu}M$, the progeny was significantly less damaged than its parental plants (SOD2- or NDPK2-transgenic lines) as well as wild type plants, implying its resistance to oxidative stress was enhanced compare to that of SOD2- or NDPK2- transgenic plants. In an expression of 11 quantitative traits, the progeny remained similar to control plants, although it infrequently displayed slightly longer or wider than either parental or wild type plants. In the expression of 6 qualitative traits, there was no significant difference between parental or non-transgenic control plants.

  • The first line of antioxidant defense against reactive oxygen species includes the enzymatic activity of the superoxide dismutase (SOD) that catalyzes the disproportionation of superoxide to hydrogen peroxide and water. The SOD mainly removes highly toxic $O_2$$^{[-10]}$ and also prevents $O_2$$^{[-10]}$ mediated reduction of iron and subsequent OH$^{[-10]}$ generation. Along with an interest in SOD as a first line of defense against damage mediated by the superoxide anion, the SOD1 enzyme has been subjected to investigation in the molecular and cellular level. • The heterologous expression of sodN gene from Streptomyces seoulensis in Streptomyces lividans together with the gel filtration and sedimentation equilibrim data indicated that the quaternary structure of NiSOD is homohexamer, which is novel among SODs, not the previously reported homotetramer. The EPR spectrum of $^{61}$ Ni (I = 3/2) substituted NiSOD showed a clear resolved hyperfine structure at g=2.016, unambiguously identifying that the EPR signal from NiSOD is due to Ni.(omitted)
