모과의 추출조건에 따른 Protocatechuic Acid 함량 변화 및 활성 평가 (Protocatechuic Acid Content and Physiological Activities of Chaenomeles sinensis Extracts Prepared with Different Methods)

  • The aim of this study was to investigate the potential of Chaenomeles sinensis as a content of protocatechuic acid and physiological activities. The analyzed the content of protocatechuic acid of Chaenomeles sinensis water extract (CSW) according to different extract temperature and time for extraction. It was analyzed as 0.14±0.00 mg/g in the extract at 100℃ for 3 h. Protocatechuic acid content increased as extraction time and temperature increased. The activity of the CSW at 100℃ for 3 h was measured. As a result of measuring DPPH, ABTS radical scavenging activity and SOD activity, antioxidant activity was found and α-glucosidase inhibition activities. Cell viability was shown for RAW 264.7 cells, and NO (nitric oxide) production was also inhibited. Finally, content of protocatechuic acid increased with increasing extraction time and extraction temperature, and exhibited antioxidant activity, inhibition of NO production, and anti-diabetic activity. It is believed that the Chaenomeles sinensis is likely to be developed as a functional material.

Curcumin-induced Growth Inhibitory Effects on HeLa Cells Altered by Antioxidant Modulators

  • Curcumin (diferuloyl methane), originated rhizomes of Curcuma longa L. has been suggested as an anti-inflammatory and anti-carcinogenic agent. In the present study, modulation of cytotoxic effects of curcumin on HeLa cells by different types of antioxidants was investigated. Cytotoxic effects of curcumin were significantly enhanced in the presence of superoxide dismutase (SOD) by decreasing $IC_{50}$ to 15.4 from $26.0\;{\mu}M$ after 24 hr incubation; the activity was not altered by catalase. The effect of curcumin was significantly less pronounced in the presence of 4 mM N-acetylcysteine (NAC). Low concentration (<1 mM) of NAC, however, increased the efficacy of curcumin. Cysteine and ${\beta}$-mercaptoethanol that have a thiol group, showed the similar biphasic patterns as NAC for modulating curcumin cytotoxicity, which was, however, constantly enhanced by ascorbic acid, a non-thiol antioxidant. In the presence of SOD, ascorbic acid, and 0.5 mM NAC, cellular levels of curcumin were significantly increased by 31-66%, whereas 4 mM NAC decreased the level. The present results indicate that thiol reducing agents showed a biphasic effect in modulating cytotoxicity of curcumin; it is likely that their thiol group is reactive with curcumin especially at high concentrations.

퉁퉁마디의 아미노산함량과 항산화 활성 (The Amino Acid Content and Antioxidant Activities of Glasswort (Salicornia herbacea L.))

  • 퉁퉁마디를 기능성 식품소재로 이용하기 위한 자료로 전남지역(해남 황산면, 신안 증도면, 영광 염산면)의 퉁퉁마디를 6월부터 10월까지 채취하여 구성아미노산, 유리아미노산, 폴리페놀함량, 전자공여능, SOD 유사활성을 조사하였다. 그 결과, 3지역 퉁퉁마디의 구성아미노산은 6월부터 9월까지는 Glutamic acid, Aspartic acid, Leucine 순으로, 10월에는 Glutamic acid, Aspartic acid, Arginine 순으로 많은 함량을 나타났다. 구성아미노산 총함량은 해남지역의 경우, $3,338.75{\sim}4,491.63\;mg$/100 g를, 신안지역의 경우, $3,086.48{\sim}3,782.89\;mg$/100 g를, 영광지역의 경우, $3,134.3{\sim}4,364.17\;mg$/100 g를 함유하였다. 유리아미노산 함량은 전체적으로 6월부터 9월까지 Proline이 가장 많았으며, 초기성숙 6월에 높았다가 성숙하면서 감소하는 경향을 보였는데, 유리아미노산 총함량은 해남지역의 경우, $472.75{\sim}1,080.98\;mg$/100 g를, 신안지역의 경우, $440.46{\sim}621.77\;mg$/100 g를, 영광지역의 경우, $387.64{\sim}669.28\;mg$/100 g를 함유하였다. 폴리페놀함량을 분석 결과, 8월까지 함량이 높았다가 퉁퉁마디가 붉어지는 10월에 가장 낮아졌으며 영광지역이 대체로 높았으며 해남지역은 $11.1{\sim}15.61\;mg/g$, 신안지역은 $12.71{\sim}21.03\;mg/g$, 영광지역은 $13.13{\sim}22.28\;mg/g$을 나타냈다. 전자공여능 분석한 결과, 신안지역이 좀더 높았고 전반적으로 8월이후 높게 나오는 경향을 보였는데 해남지역은 $53.8{\sim}70.9%$, 신안지역은 $62.7{\sim}88.8%$, 영광지역은 $63.0{\sim}72.7%$ 범위를 나타내었다. 퉁퉁마디 추출물의 SOD 유사활성은 해남지역 $28.7{\sim}41.0%$, 신안지역 $28.5{\sim}36.5%$, 영광지역 $23.5{\sim}39.6%$로 모두 7월에 가장 낮았고 10월에 가장 높은 활성을 보였다.

대금음자가미와 이(梨)추출물 배합제제가 알코올로 유발된 간손상에 미치는 영향 (Effects of combination pear extract with Daekumeumjagami medication on hepatic injury induced by alcohol in mice)

  • Objectives : The effect of pear extract with Daekumeumjagami and vitamin C medication(PDV) on alcohol metabolism and hepatic injury was assessed following hepatic injury induced by alcohol in mice. Methods : The model of alcoholic hepatic injury was established by orally administration with 3 g/kg 25% alcohol in mice. PDV was orally administrated once a day for 5 days. Mice were randomly divided into 5 groups : normal group, control group, and PDV groups (PDV-A, PDV-B and PDV-C). The activities of aspartate amino transferase (AST) and alanine amino transferase (ALT) and alcohol dehydrogenase (ADH) in serum, superoxide dismutase (SOD) and catalase in liver were determined after alcohol exposure. Results : Compared with control group, treatment with PDV-B and PDV-C significantly elevated activities of ADH. Moreover, the index of hepatic injury in serum was significantly decreased by treatment with PDV-B and PDV-C in ALT activity and PDV-C in AST activity. Additionally, enhanced catalase activities in liver was found in PDV-C treated mice after exposure to alcohol. Also, WBC in blood was significantly lower by treatment with PDV-B and PDV-C. Conclusions : This study suggests that PDV treatment could enhance alcohol metabolism, and prevent hepatic injury after alcoholic hepatic injury and that this effect is likely related to its modulation on the alcohol metabolizing and antioxidant enzymes.

LPS로 유도된 RAW 264.7 세포에 대한 레몬 머틀 잎 추출물의 항염증 효과 (Anti-inflammatory Effects of Lemon Myrtle (Backhousia citriodora) Leaf Extracts in LPS-induced RAW 264.7 Cells)

  • 본 연구에서는 레몬 머틀 잎 열수 및 에탄올 추출물을 이용하여 항산화 활성과 NO 소거능 및 RAW 264.7 세포에서의 항염증 활성에 미치는 영향을 확인하였다. 레몬 머틀 열수 및 에탄올 추출물의 총 페놀 함량은 각각 207.44와 $331.54{\mu}g\;TAE/mg$이었고, DPPH radical 소거능은 0.5 mg/ml 농도에서 각각 90.69%와 92.50%로 나타났으며, SOD 활성은 1 mg/ml 농도에서 각각 106.22%와 103.58%로 나타났다. 아질산염 소거능 분석에서는 열수와 에탄올 추출물 1 mg/ml 농도, pH 1.2에서 81.06%와 91.03%로 에탄올 추출물의 소거능이 높은 것으로 나타났다. 레몬머틀의 항염증 활성을 측정하기 위하여 대식세포에 LPS를 처리하여 세포 내 NO 생성을 유도한 후 열수 및 에탄올 추출물을 처리하여 각각의 추출물이 NO 활성에 미치는 영향을 분석한 결과 각 추출물 모두 농도 의존적으로 감소하였다. 또한 LPS에 의해 증가된 iNOS 단백질 발현 또한 농도 의존적으로 감소하였다. 이상의 결과에서와 같이 레몬 머틀 잎은 항산화 및 항염증 효과가 있는 것으로 나타났기에 천연물 소재로서의 활용도가 높을 것으로 생각된다.

지황(地黃)과 발효(醱酵) 지황(地黃)의 생리활성 비교 연구 (Comparison of Biological Activities on Rehmanniae Radix and Fermented Rehmanniae Radix)

  • Herbal medicines are medicinal products containing a single or a mixture of two or more different herbal substances or herbal preparations as active principles. Recently, much attention has been paid to developing various kinds of fermented herbal extracts, a new type of traditional herbal medicine in the field of Korean traditional medicine. The fermentation of medicinal herbs is intended to exert a favorable influence on bioestability, bioavaliablilty and pharmacological activity of herbal extract in the gastrointestinal tract as well as intensifying the nutritional and pharmacological aspects of the medicinal herbs. The purpose of this study was to investigate biological activities of fermented Rehmanniae Radix by lactic acid bacteria at $30^{\circ}C$ for 3 days in comparison with those for Rehmanniae Radix The fermented Rehmanniae Radix exhibited different chemical profile to Rehmanniae Radix generated with HPLC, indicating production of new ingredients during fermentation. Rehmanniae Radix served as good nutritional sources for the growth of lactic acid bacteria showing increased number of bacteria during fermentation. Toxic effect of the fermented Rehmanniae Radix to cells were not seen judged by the MTT assay. The fermented Rehmanniae Radix exhibited better antioxidant effect than non-fermented Rehmanniae Radix analyzed by a SOD-likely assay. Both hypoglycemic and hypotensive effects of the fermented Rehmanniae Radix were also detected and better than those for Rehmanniae Radix in showing dose-dependent inhibitory effects on alpha-glucosidase and ACE, respectively. In conclusion, fermented Rehmanniae Radix appears to have more biological activities than non-feremented Rehmanniae Radix showing not only antioxidant effect but also cardiovascular protection.

장뇌산삼이 Benzopyrene으로 유도된 간조직의 $TNF-{\alpha}$와 COX-2의 면역조직학적 분포에 미치는 영향 (Immunohistochemical Study of Wild Ginseng on Benzopyrene Induced $TNF-{\alpha}$ and COX-2 Expression in Rats)

  • Polycyclic aromatic hydrocarbon (PAH), such as benzo(a)pyrene (B(a)P), are toxic environmental contaminants known to enhance oxidative stress, production of pro-inflammatory and inflammatory cytokines. The present study was designed in order to determine whether wild ginseng (Panax ginseng C. A. Meyer) protect PAH-induced oxidative stress and inflammation. B(a)P (0.5 mg/kg, i.p.) treatment increased the distribution of immunoreactive cells for tumor necrosis factor $(TNF)-\alpha$ and cyclooxygenase (COX)-2 in peri-portal triad region and immunoreaction was shown in the cytoplasm of macrophage. Pre-treatment with wild ginseng significantly decreased immune responses in the rats treated with B(a)p. The rats given 50 mg/kg/day for 4 weeks before B(a)P treatment had 1.39-fold and 1.5-fold inhibition of $TNF-\alpha$ and COX-2 positive reaction, respectively. Wild ginseng extract alone had no effect on the distributional changes. The SOD activity as scavenger enzymes after wild ginseng administration dose-dependantly increased compared with butylated hydroxytoluene, a general radical scavenger. These data likely indicate that wild ginseng extract may act as inflammatory regulator in conjunction with inhibition of oxidant dependent metabolic activation in environmental contaminants-induced hepatic inflammation.

전처리 방법을 달리한 곰취 인절미의 제조법 표준화 연구 (A Study on Different Pre-processing of Ligularia fischeri Injeolmi to Standardize Its Processing)

  • In this study Ragwort, which had various and excellent pharmacological activity, was selected as a functional material, and intended to present an optimum condition of Ragwort Injeolmi by the blanching method, commonly used in the field. The general components, antioxidation, and dietary fiber of Ragwort were analyzed, and produceableness of Ragwort added Injeolmi was reviewed through instrumental inspection and sensory evaluation of various adding ratio of lyophilized Ragwort power and Ragwort pre-processed by blanching. It was proved that the SOD-likely active of Ragwort was 45.69%, total poly- phenol content was 12.45 mg, and total flavonoid content was 10.25 mg. The pH of Injeolmi adding Ragwort powder showed an increasing trend in the order of 0%, 1%, 3%, 5%, while that of Injeolmi adding blanching Ragwort showed increasing trend in the order of 0%, 10%, 15%, 20%, and there were significant difference (p<0.001). The soluble solid content of all sample groups showed constant value, and it was understood that no sugar was added in the process. From chromatography measurement result, a-value (redness) of 1% sample group was the lowest, and there were significant differences among sample groups (p<0.001). During 3 days of storage period, total microbial count of Injeolmi showed increasing trend while the storage period was getting longer. From the acceptability test Injeolmi adding blanching Ragwort had shown generally higher value.

세포투과 펩티드를 함유한 고분자 미셀 및 리포좀을 이용한 배나무 잎 추출물의 피부 흡수 증진 (Enhancement of Skin Permeation of Pyrus serotina var Leaf Extract Using Polymer Micelle and Liposome Containing Cell Penetrating Peptide)

  • 세포투과 펩티드를 함유한 고분자 미셀 및 리포좀을 이용한 배나무 잎 추출물의 피부 흡수 증진 및 화장품 성분으로의 응용가능성에 대해 알아보기 위해 항산화, 항균 실험 및 제형별 피부 침투 실험을 진행하였다. 총 polyphenol 함량은 배나무 잎 에탄올 추출물에서 $118.83{\pm}9.39mg/g$, 배나무 잎 열수 추출물에서 $106.89{\pm}4.45mg/g$로 확인되었다. DPPH radical 소거능 측정 결과, 500 mg/L의 농도에서 배나무 잎 에탄올 추출물이 $74.39{\pm}7.48%$의 가장 높은 라디칼 소거능을 나타냈다. SOD 유사 활성능은 1,000 mg/L의 농도에서 열수 추출물이 $91.62{\pm}0.43%$로 가장 높은 효능을 나타내었다. 이 후 실험으로부터 항산화, 주름 개선, 미백 활성이 확인되어 배나무 잎 추출물이 항산화 및 항균소재로서의 실현가능성이 높다고 판단했다. 배나무 잎 에탄올 추출물을 함유한 고분자 미셀 피부침투 실험에서는 24시간 동안의 실험 결과, 총 축적된 tannic acid의 투과량은 Formulation 2($55.45{\mu}g/cm^2$), Formulation 1($46.43{\mu}g/cm^2$), Formulation 0($34.36{\mu}g/cm^2$)의 순서로 확인되었다. 배나무 잎 열수 추출물을 함유한 리포좀 피부침투 실험에서는 24시간 동안의 실험 결과, 총 축적된 tannic acid의 투과량이 Formulation 5($75.01{\mu}g/cm^2$), Formulation 4($64.01{\mu}g/cm^2$), Formulation 3($36.60{\mu}g/cm^2$)의 순서로 확인되었다. 이 연구를 통해 배나무 잎 추출물이 가지고 있는 항산화, 주름개선의 효능에 대한 가능성을 확인하였고 고분자 미셀 및 리포좀을 이용한 배나무 잎 추출물의 피부침투 결과를 통해 향후 화장품 산업에 긍정적으로 이용될 것이라고 사료된다.
