Effect of Methanol Extracts of Red Colored Rices on Antioxidant Activity and Growth Inhibitory Activities of Cancer Cells (적미 추출물과 분획물의 항산화 활성 및 암세포 성장억제효과)

  • The inhibitory effects of the water and methanol extracts of Jakwangdo and Honghyangmi on the rat microsome lipid peroxidation and growth of four human cancer cells such as HepG2 (liver cancer), SNU-1 (stomach cancer) MCF-7 (breast cancer) and SNU-C4 (colon cancer) were examined. The methanol extracts of red colored rices showed the antioxidant activity and growth inhibitory effects of cancer cells. However, water extracts did not show the activities. Inhibitory activities of methanol extracts of Jakwangdo and Honghyangmi against lipid peroxidation of rat microsome was 80% and 68%, respectively, at the concentration of 1 mg/assay. Jakwangdo methanol extracts showed the highest growth inhibitory activity in MCF-7 cells among the cancer cells tested. The methanol extracts of red colored rices were further fractionated with hexane, chloroform, ethyl acetate and butanol. Both chloroform and hexane fractions showed strong growth inhibitory activity in HepG2 and MCF-7 cells.

Losses in Yield and Quality of Forage Legumes During Field Curing in Spring (봄철 포장건조 콩과목초의 수량 및 품질 손실)

  • No comprehensive study of yield and quality losses of forage legumes harvested and cured in spring has been conducted in Korea, therefore, this experiment was carried out to gain information on yield and quality, losses of alfalfa(Medicgo sativa L.), crimson clover (Trifolium incarnatum L.), and red clover(Trifolium pratense L.) during field curing in spring(mid-May). Alfalfa was highest in dry matter content of 15% at harvest, while crimson clover and red clover had 12.4 and 11.5%, respectively. Species differences for the dry matter content were maintained and consistent trends were observed during the field curing. Alfalfa took seven days to reach dry matter content over 80%, while both crimson clover and red clover took eight days. Crimson clover and alfalfa tended to show a higher leaf-stem ratio than red clover based on dry matter, but red clover showed a higher leaf-stem ratio than alfalfa and crimson clover based on crude protein content. Losses in dry matter of alfalfa from leaf shattering were higher than crimson clover and red clover, and similar trends were detected losses in crude protein for three different legumes. Fresh legume forages averaged higher in clude protein(CP), total digestible nutrients(TDN), and relative feed value(RFV), while dry legume forages averaged lower in acid detergent fiber(ADF) and neutral detergent fiber(NDF). Results of the experiment indicate that hay curing in spring affects crude protein concentration more than ADF and NDF, and this is due, in part, to leaf shattering caused by field operations.

Deep Learning-Based Algorithm for the Detection and Characterization of MRI Safety of Cardiac Implantable Electronic Devices on Chest Radiographs

  • Objective: With the recent development of various MRI-conditional cardiac implantable electronic devices (CIEDs), the accurate identification and characterization of CIEDs have become critical when performing MRI in patients with CIEDs. We aimed to develop and evaluate a deep learning-based algorithm (DLA) that performs the detection and characterization of parameters, including MRI safety, of CIEDs on chest radiograph (CR) in a single step and compare its performance with other related algorithms that were recently developed. Materials and Methods: We developed a DLA (X-ray CIED identification [XCID]) using 9912 CRs of 958 patients with 968 CIEDs comprising 26 model groups from 4 manufacturers obtained between 2014 and 2019 from one hospital. The performance of XCID was tested with an external dataset consisting of 2122 CRs obtained from a different hospital and compared with the performance of two other related algorithms recently reported, including PacemakerID (PID) and Pacemaker identification with neural networks (PPMnn). Results: The overall accuracies of XCID for the manufacturer classification, model group identification, and MRI safety characterization using the internal test dataset were 99.7% (992/995), 97.2% (967/995), and 98.9% (984/995), respectively. These were 95.8% (2033/2122), 85.4% (1813/2122), and 92.2% (1956/2122), respectively, with the external test dataset. In the comparative study, the accuracy for the manufacturer classification was 95.0% (152/160) for XCID and 91.3% for PPMnn (146/160), which was significantly higher than that for PID (80.0%,128/160; p < 0.001 for both). XCID demonstrated a higher accuracy (88.1%; 141/160) than PPMnn (80.0%; 128/160) in identifying model groups (p < 0.001). Conclusion: The remarkable and consistent performance of XCID suggests its applicability for detection, manufacturer and model identification, as well as MRI safety characterization of CIED on CRs. Further studies are warranted to guarantee the safe use of XCID in clinical practice.

Cytotoxic activities of various fractions extracted from some pharmaceutical insect relatives

  • This research was performed to screen the cytotoxic activities of some pharmaceutical insect relatives. Cytotoxic activities of total extract and fractions of hexane, ethyl acetate, methanol, water and boiling water were extracted from four pharmaceutical insect relatives: the Chinese gall, the cicada slough, the hornet nest and the batryticated silkworm. These extracts were investigated against the cancer cell lines of L1210, P388 and SNU-1 in vitro tests. Results showed that, ED, , against the cancer cell lines of L1210, P388 and SNU-1 were 0.55, 0.50, and $0.83{\mu}g/ml$ in the ethyl acetate fraction from the Chinese gall; 1.07, 2.19, and $2.24{\mu}g/ml$in the ethyl acetate fraction, 1.51, 1.26, and $1.45{\mu}g/ml$ in the water fraction and 1.48, 2.29, and $1.29{\mu}g/ml$in the boiling water fraction from the cicada slough; 3.31, 2.00, and $6.61\mug/ml$ in the water fraction from the hornet nest and 13.80, 19.95, and $31.62{\mu}g/ml$in the hexane fraction and 33.88, 21.88, and $25.12{\mu}g/ml$in the ethyl acetate fraction from the batryticated silkworm, respectively. All of the fractions mentioned above showed high cytotoxic activities and could be suggested for further studies in vivo tests.

Hay Preparation Technology for Sorghum×Sudangrass Hybrid Using a Stationary Far-Infrared Dryer (정치식 원적외선 건조기를 이용한 수수×수단그라스 교잡종의 건초 조제 기술 연구)

  • This experiment was conducted to confirm the possibility of preparing Sorghum×sudangrass hybrid artificial hay using far-infrared rays in Korea. The machine used in this experiment is a drying device based on far-infrared rays, and is designed to control temperature, air flow rate, far-infrared radiation amount, and air flow speed. The Sorghum×sudangrass hybrids harvested in late September were wilted in the field for one day, and a drying test was performed on them. Conditions for drying were performed by selecting a total of 7 conditions, and each condition induced a change in radiation amount in a single condition (42%) and two steps (4 treatments) and three steps (2 treatments). The speed of the air flow in the device was fixed at 60 m/s, and the run time was changed to 30, 60, and 90 minutes. The average dry matter (DM) content was 82.84%. The DM content was 59.94 and 76.91%, respectively, in drying conditions 1 and 3, which were not suitable for hay. In terms of drying rate, it was significantly higher than 80% in the 5, 6 and 7 treatment, and power consumption was slightly high with an average of 5.7 kw/h. As for the feed value according to each drying condition, the crude protein (CP) content increased as the drying time increased, and there was no significant difference between treatments in ADF, NDF, IVDMD and TDN content. In terms of RFV, treatment 1, which is a single condition, was significantly lower than the complex condition. Through the above results, it was determined that the drying conditions 4 and 5 were the most advantageous when considering the drying speed, power consumption, and quality.

Antitumor Activity of Kp, a Protein-polysaccharide from the Mycelial Culture of Phellinus linteus (Phellinus linteus 균사 배양물로부터 분리한 단백다당체 Kp의 항암활성)

  • A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation, Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 mg/kg. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation, Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 mg/kg. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of $200\;{\mu}g/ml$. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host's immune system.

Revelation of Antitumor Effect in Combination with 5-Fluorocytosine and Extracellular Cytosine Deaminase (5-Fluorocytosine과 세포외 Cytosine Deaminase의 병용투여에 의한 항암효과의 발현)

  • This study was carried out particularly focusing on he antitumor effect in combination with 5-fluorocytosine(5-FC), antifungal agent, and extracellular cytosine deaminase from Chromobacterium violaceum YK 391 against U-937, K-562 and SNU-C4 cells. While the addition of 10$\mu\textrm{g}$/100 ${\mu}\ell$ of anticancer agent, 5-fluorouracil(5-FU), to U-937, K-562 and SNU-C4 caused the decrease of proliferation 90%, 75% and 93% respectively, the addition of 20 $\mu\textrm{g}$/100 ${\mu}\ell$ of the extracellular cytosine deaminase and 10 $\mu\textrm{g}$/100 ${\mu}\ell$ of antifungal agent 5-FC caused the decrease of proliferation 80%, 70% and 90%, respectively. These results, therefore, reveal that this enzyme has the similar clinical effect for considering of adjuvant antitumor effect. From the above results, the treatment of 5-FC and the cytosine deaminase was very effective and showed the possibility to remove side effects which easily occur by the treatment of 5-FU only. Anti-tumor activities of Naesosungma-tang (내소승마탕(內消升麻湯)의 항암효과(抗癌效果) 연구(硏究))

  • This study was carried out to evaluate antioxidative, cytotoxic and chemosensitizing effects for anti-tumor activities of Naesosungma-tang (NST). The results were as follows ; 1. The oxidative effects were measured by DPPH radical scavenging activity. NST water extract was showed more effective than ethanol extract and also various solvent fractions from NST showed effective in the following order : butanol fraction > ethyl acetate fraction > hexane fraction > aqueous fraction. 2. NST water extract was showed cytotoxic effects on the human cancer cells Calu-6 cell and SNU-601 cell, but not on MCF-7 cell. 3. The viability of cells was measured in 4 kinds of solvent fractions from water extract of NST by MTT assay. The results were as followings : 1) On the NIH3T3 cell, $IC_{50}$ value was $200{\sim}300\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 2) On the Caco-2 cell, effective only in the butanol fraction. 3) On the Calu-6 cell, most effective in hexane fraction($IC_{50}:80\;{\mu}g/m{\ell}$) and effective in ethyl acetate and butanol fraction($IC_{50}:100{\sim}120\;{\mu}g/m{\ell}$). 4) On the MCF-7 cell, $IC_{50}$ value was similar in three fractions except aqueous fraction. 5) On the SNU-601 cell, $IC_{50}$ value was within $100\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 6) On the HCT-1l6 cell, $IC_{50}$ value was $150{\sim}200\;{\mu}g/m{\ell}$ in three fractions except aqueous fraction. 7) On the AML-2/WT cell, not effective in four kinds of solvent fractions. 4. In combined effects of NST and vincristine on AML-2/D100 cell, water and ethanol extracts of NST and hexane fraction of water extracts of NST more effectively inhibited proliferation of AML-2/D100 cell. These results Suggest that NST has antioxidative and cytotoxic effects against Caco-2, Calu-6, MCF-7, SNU-601, HCT116 and NIH3T3 cell especially Calu-6 and SNU-601 cell, and also chemosensitizing effects against AML-2/D100 cell in combined vincristine.

Comparative study of Seed Productivity of Spring Sown Italian Ryegrass(Lolium multiflorum Lam.) Depending on Seeding Distance in Gangwon Highland (강원 고지대에서 봄 파종 이탈리안 라이그라스(Lolium multiflorum Lam.)의 파종 간격에 따른 종자 생산성 비교 연구)

  • This experiment was conducted to study on the growth characteristics and seed productivity of Italian ryegrass (Lolium multiflorum Lam., IRG) planted in the Spring in Gangwon Highland according to the seeding distance (20, 30 and 40 cm). The field was located in highland around 600 m above sea level. The experimental design was randomized block design with three replication and the tested IRG variety was 'Greencall' developed by National Institute of Animal Science (NIAS). IRG was sown on March 26, 2020, and harvested on July 2. The plant height was the shortest at 80.5 cm in the 40 cm seeding distance plot (P<0.05), and there was no significant difference between the 20 and 30 cm seeding distance. The number of spike per square meter (㎡) was significantly higher in the 20 cm seeding distance plot than that of 40 cm (937 vs. 571). The dry matter (DM) content of seed and straw after harvesting was 49.70 and 33.36 % on average, and there was no significant difference between treatments (P>005). However, there was a significant difference in the fresh and DM yield of seeds and straw (P<0.05). DM yield of seeds was significantly higher in 20 cm distance than that of 40 cm, and the yield of straw was the same trend. On the other hand, there was no significant difference in DM yield between 20 cm and 30 cm and also in the feed value of straw after seed harvesting among seeding distance. The average CP, ADF, NDF, and TDN contents were 6.91, 36.76, 61.75 and 59.86%, respectively, and the RFV value was 91. Considering the above results, the production of Italian ryegrass seeds sown in the spring in the highlands of the Gangwon is lower than that of autumn sowing, but it is judged that it needs to be reviewed in case it is unavoidable. In the future, there should be an economic analysis and the development of technology that can increase production.

Antitumoral, Antioxidant and Antimicrobial Activities of Solvent Ftactions from Grifola umbllatus (저령추출물의 항암, 항산화 및 항균효과)

  • Grifola umbellatus was extracted using methanol, and the extract was further fractionated by water and ethyl acetate. Assay of each fraction with MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium-bromide] revealed significant cytotoxicity effect of the methanol extract of Grifola umbellatus against human gastric cancer cell but not normal human lymphocytes. The methanol extract showed the highest antioxidant activity as well. Antimicrobial activity of Grifola umbellants against Helicobacter pylori was higher in method extract than in other fractions. Grifola umbellatus had a significant inhibitory effect on Helicobacter pylori reducing both its growth and urease activity. These results show that the methanol extract of Grifola umbellatus possesses therapeutic potential on gastric diseases.
