THE COMPARISON OF STREPTOCOCCUS MUTANS ISOLATED FROM OCCLUSAL SURFACES OF CARIES AND NON-CARIES TEETH (우식치아와 정상치아의 교합면에서 분리한 Streptococcus mutans의 비교)

  • When oral microorganisms were sampled from occlusal surfaces of caries and non-caries teeth, $3.43\times10^5$ CFU and $3.47\times10^3$ CFU of bacteria were counted on MSB agar plates, respectively. All the 20 colonies isolated from a caries surface were Streptococcus mutans but, only two of 20 colonies were identified as Streptococcus mutans by API test. S. mutans SM1 from caries tooth and S. mutans SM2 from non-caries tooth showed the same results except for $\alpha-galactosidase$ activity on sugar fermentation tests and biochemical tests. For the bacterial replication, both SM1 and SM2 were actively multiplicated at pH 5.5. And the viability of SM1 was high at 20% of sucrose, while that of SM2 was high at 5% of sucrose in the media. SM1 actively replicated at 16mM of $CaCl_2$, 160mM of KCl, and 6.4mM of $MgCl_2$, and the replication of SM2 was increased at 16mM of $CaCl_2$, 40mM of KCl, 6.4mM of $MgCl_2$. At 1mM of sodium bicarbonate and sodium phosphate, both bacteria were actively multiplicated. SM1 and SM2 were actively replicated at 1mM and 10mM of Tris, respectively. For potassium phosphate buffer, SM1 grew well proportionally to the concentration up to 100mM, while the growth of SM2 were inhibited by the increase of concentration. The 4.6 kb of gtf gene was amplified with a pair of primer, gtfB-F961 and gtfC-R5574 by polymerase chain reaction from the chromosomal DNA of SM1 and SM2. When 4.6kb bands were eluted from gel and were treated with restriction enzyme, EcoR I produced the same RFLP like 0.8kb and 3.8kb of DNA fragments for S. mutans GS-5, SM1 and SM2. By Hind III, the PCR products weren't digested for S. mutans GS-5 and SM1, but 3 fragments such as 2.4kb, 1.8kb and 400bp were examined for SM2. These results indicated the difference between gtf genes of SM1 and SM2. BamH I treatment showed 4 fragments for SM1 and SM2, while the 3 fragments for S. mutans GS-5. The PCR products were not digested by Kpn I, Sma I, Xho I and Pst I.

  • Sm-doped YBCO high $T_c$ superconductor was directionally grown by zone melt growth process in air atmosphere. Cylindrical green rods of $(Sm/Y)_{1.8}Ba_{2.4}Cu_{3.4}O_{7-x}$[(Sm/Y)1.8] oxides were fabricated by cold isostatic pressing (CIP) method using rubber mold. Based on the variation of melting temperature and growth rate, the microstructure and superconducting properties were systematically measured by using optical micrographs, TEM and SQUID magnetometer. In this study optimum melting temperature and growth rate were $1085^{\circ}C$ and 3.5 mm/hr respectively. Nonsuperconducting $(Sm/Y)_2BaCuO_5$ inclusions of (Sm/Y)1.8 superconductor were uniformly distributed within the superconducting (Sm/Y) $Ba_2Cu_3O^{7-x}$ matrix. The directionally melt-textured (Sm/Y)1.8 superconductor showed an onset $T_c$ $\geq$ 90K and sharp superconducting transition.

  • High T$\_$c/ (Sm/Y)$\_$1.8/Ba$\_$2.4/Cu$\_$3.4/O$\_$7-$\delta$/ [(Sm/Y)] superconductor, a combination of Y and Sm(50% each), was systematically investigated by the zone melt growth process. A sample prepared by this method showed well-textured microstructure, and (Sm/Y)$_2$BaCuO$\_$5/[(Sm/Y)211]inclusions were uniformly dispersed in large (Sm/Y)Ba$_2$Cu$_3$O$\_$y/ [(Sm/Y)123]matrix. The sample showed a sharp superconducting transition at 91 K. The magnetization measurements of the (Sm/Y)1.8 sample exhibited the enhanced flux pinning, compared with Y$\_$1.8/Ba$\_$2.4/Cu$\_$3.4/O$\_$7-$\delta$/(Y1.8) sample without Sm. Critical current densities of (Sm/Y) 1.8 sample was 3.5${\times}$10$^4$A/$\textrm{cm}^2$ at 1 T and 77 K.

Luminescent Characteristics and Synthesis of Sm3+-Doped CaWO4 Phosphors (CaWO4:Sm3+ 형광체의 합성과 발광특성)

  • $CaWO_4:Sm_x$(x = 0, 0.5, 1.0, 1.5, 2.0 mol%) white phosphors with different concentrations of $Sm^{3+}$ ions were synthesized using the hydrothermal method. The crystal structure, surface, and optical properties of the $CaWO_4:Sm_x$ phosphors were investigated using X-ray diffraction(XRD), field-emission scanning electron microscopy(FE-SEM), photoluminescence(PL) and photoluminescence excitation(PLE). From the XRD results, the crystal structure of the $CaWO_4:Sm$ phosphors was found to be tetragonal. The $CaWO_4:Sm$ phosphors became more cohesive with increasing $Sm^{3+}$-ion concentration. The photoluminescence excitation(PLE) peak of the phosphors, at around 250 nm, was ascribed to the transition from the 1A1 ground-state to the high-vibration level of 1T2 in the $WO{_4}^{2-}$ complex. The maximum emission spectra of the phosphors were observed when the $Sm^{3+}$ concentration was 0.5 mol%. The luminescence intensity of the $CaWO_4$ phosphors was decreased for $Sm^{3+}$ concentrations greater than 0.5 mol%.

Medicinal Plant Lemon Grass (Cymbopogon Citratus) Growth under Salinity and Sodicity

  • Salinity with sodic condition disturbs germination, retards emergence, and slow down seedling development of Lemon Grass (Cymbopogon citratus).Lemongrass is a perennial grass plant widely distributed worldwide and most especially in tropical and subtropical countries. This research experiment was designed to evaluate the influences of (4 dSm-1+ 13.5 (mmol L-1)-1/2, 5 dSm-1+ 25 (mmol L-1)1/2, 5 dSm-1+ 30 (mmol L-1)1/2, 10 dSm-1+ 25 (mmol L-1)1/2 and 10 dSm-1+ 30 (mmol L-1)1/2) on biomass produce of lemon grass against salt tolerance. The uppermost biomass yield (45.53 gpot-1) was produced by 4 dSm-1+ 13.5 (mmol L-1)1/2 treatment. The increase in the intensity of salts reduced the growth of lemon grass. Lower biomass yield (79.33 gpot-1) was gained at 10 dSm-1+ 30 (mmol L-1)1/2. 5 dSm-1+ 25 (mmol L-1)1/2 treatment performed enhanced outcome i.e. the least reduction % over control (5.87). Salinity- sodicity showed serious effect on the growth reduction from 5.87% to33.60%. This reduction gap was affected by the negative effect of salinity and sodicity on Linseed growth. Salinity- sodicity showed severe impact on the growth reduction from 5.87% to33.60%. Based on the findings, lemon Grass (Cymbopogon citratus).was capable to grow up the maximum at 4 dSm-1+ 13.5 (mmol L-1)1/2 treatment.

Feasibility Study of HDDR and Mechanical Milling Processes for Preparation of High Coercivity SmCo5 Powder

  • HDDR (hydrogenation, disproportionation, desorption, recombination) and mechanical milling processes have been applied to the $SmCo_{5}$ alloy in an attempt to produce a highly coercive powder. The $SmCo_{5}$ alloy had very high structural stability under the hydrogen atmosphere and the 1:5 phase was only partially disproportionated under up to 10 kgf/$\textrm{cm}^2$ hydrogen gas. The partially disproportionated material was recombined not into 1:5 phase after the HDDR, but rather into multi-phase mixture consisting of 1:5, 2:17, 2:7 and 1:7 phases. The $SmCo_{5}$ alloy HDDR-treated with hydrogen up to 10 kgf/$\textrm{cm}^2$ had poor coercivity. For a useful HDDR to prepare a high coercivity $SmCo_{5}$ alloy powder, much higher hydrogen pressure well exceeding 10 kgf/$\textrm{cm}^2$ would be required. The $SmCo_{5}$ alloy lump was amorphized by an intensive mechanical milling, and it was crystallised ultra-finely by a subsequent optimum annealing. The optimally annealed material had very high coercivity, and it was found that the mechanical milling followed by an annealing was an effective way of producing highly coercive $SmCo_{5}$ alloy powder.

Separation and Purification for the Determination of Samarium and its Isotopes in PWR Spent Nuclear Fuels (PWR 사용후핵연료 중 Sm 동위원소 정량을 위한 분리 및 정제)

  • A method of separation and purification of Sm for quantitation of Sm isotopes from various fission products in PWR spent nuclear fuels has been studied. Simulated solution containing inactive metal ions(Cs, Ba, Gd, Eu, Sm and Nd) in place of radioactive fission products was prepared. Sm was separated with 0.5 M $HNO_3$/80% MeOH after washing with 1 M $HNO_3$/90% MeOH on AG $1{\times}8$, anion exchange resin. Sm was purified on cation exchange resin, AG $50W{\times}8$, pretreated with 0.2 M alpha-hydroxisobutyric acid(pH 4.5-4.6) to remove Ba causing isobaric effect Sm from PWR spent fuel. As a result of mass spectrometric measurement, eluted Sm portion did not include isobars form other elements such as Gd, Eu, Pm, Nd and BaO. The contents of Sm and its isotopes($^{147}Sm$, $^{148}Sm$, $^{149}Sm$, $^{150}Sm$, $^{151}Sm$, $^{152}Sm$ and $^{154}Sm$) in spent fuel were determined by isotope dilution mass spectrometric method spiking $^{154}Sm$.

Salinity and sodicity disturbs growth of medicinal crop Guar (Cyanoposisa tetragonoloba)

  • Salinity is one of the major and increasing problems in irrigated agriculture in Pakistan. Salinity stress negatively affects the growth and yield of plants guar (Cyanoposisa tetragonoloba). This experiment was conducted to evaluate the effects of ($4dSm^{-1}+13.5(mmol\;L^{-1})^{1/2}$, $5dSm^{-1}+25(mmol \;L^{-1})^{1/2}$, $5dSm^{-1}+30(mmol\;L^{-1})^{1/2}$, $10dSm^{-1}+25(mmol\;L^{-1})^{1/2}$ and $10dSm^{-1}+30(mmol \;L^{-1})^{1/2}$) on biomass yield of guar against salinity tolerance. Maximum biomass yield ($54.50gpot^{-1}$) was produced by $4dSm^{-1}+13.5(mmol\;L^{-1})^{1/2}$ treatment. Biomass produce was reduced with the increase of the salts toxicity. Minimum biomass yield ($30.17gpot^{-1}$) was attained under $10dSm^{-1}+30(mmol \;L^{-1})^{1/2}$. $5dSm^{-1}+25(mmol\;L^{-1})^{1/2}$ treatment exhibited improved outcome i.e. the least diminution % over control (18.66). Salinity cum sodicity showed staid effect on the growth reduction from 18.66% to 44.64%. This reduction fissure was impacted by the toxic effect of salinity and sodicity on Guar growth. Salinity- sodicity behaved toxic impact on the growth reduction from 18.66% to 44.64%. Based on the findings, guar (Cyanoposisa tetragonoloba) grows better at $4dSm^{-1}+13.5(mmol \;L^{-1})^{1/2}$ treatment.

Epidemiological properties of salmonellosis of poultry farms in Kyongbuk province at recent year (최근 경북지역의 양계농장에서 발생한 salmonellosis의 역학적 특성)

  • The present study was conducted to investigate the epidemiological properties of salmonellosis of poultry farms in Kyongbuk province during the relied from November 1998 to November 1999 And antibiotic susceptibility and biochemical characteristics of 120 Salmonella cultures isolated from chicken samples were also investigated. The results obtained through this study were summarized as follows, 1. Among 667,200 chickens of 31 flocks in 17 layer farms and 9 broiler farms, 61,350 chickens of 31 flocks were died with salmonellosis. The death rate of 25 farms varied from 0.1% to 75.0%, and the average death rate was 9.2%. 2. According to etiological agents, fowl typhoid was shown the most predominant outbreak among the salmonellosis during a year, which accounted for 8R.0%(22/25) of the total case 3. The serotypes of 120 Salmonella isolates were identified as 7 strains(5.8%) of S pullorum, 10 strains(8.4%) of S typhimurium and 103 strains(85.8%) of S gallinarum. 4. Most outbreak of fowl typhoid were prevalent on the layer chicken farms(77.1%), and the summer season(45.5%) also appeared the most hazardous season during the year. 5. It seemed that the Hyline breed(70.6%) was the most susceptible among the layer chicken breeds, and followed by Isabrown(23.5%), Tetra(5.9%) in order. 6. In layers, 76.4% of fowl typhoid occurred commonly from 14 to 40 weeks including the early laying peroid, but in broiler farms, all cases was outbreak within first second weeks. 7 All the strains of S pullorum were resistant to lincomycin(Lm), penicillin(Pm), and steptomycin(Sm), but sensitive to amikacin(Ak), ampicillin(Am), cephalothin(Ce), ciprofloxacin (CiP), chloramphenicol(Cm), colistin(Co), enrofloxacin(Enr), furazolidone(Fu), gentamicin(Gm), kanamycin(Km), neomycin(Nm), polymyxin(Po), and teracycline(Tc). All the strains of S typhimurium were resistant to Lm(100%), Pm(100%), Po(90%), and Sm(90%), but were sensitive to Ak, Am, Ce, CiP, Cm, Co, Enr, Fu, Gm, Km, Nm, and Tc. 8. Minimum inhibitory concentration(MIC) of 103 strains of S gallinarum were also evaluated and their patterns were much more variable than others. All the strains of S gallinarum were sensitive to Ak, Am, Ce, Cip, Cm, Enr, Fu, Km, and Nm, but resistant to Lm(100%), and Sm(100%), 99(96.1%) to Co, 83(80.6%) to Pm, and 83(80.6%) to Po, 55(53.4%) to Gm, and 33(32.0%) to Tc. 9. The multiple drug resistance patterns of 120 Salmonella strains were CoLmPmPo Sm pattern(34.2%), CoGmLmPmPoSmTc(20.8%), CoGmLmPmPoSm(13.3%), CoGmLmSm(7.5%), LmPmSm(7.5%), LmPm(6.7%), CoLmSmTc(3.3%),, CoGmLmSmTc(1.7%), GmLmSmTc(1.7%), CoGmLmPoSm(0.1%), LmPmPo(0.1%), CoLm Sm(0.1%), and LmSm(0.1%), in order.

Soil Salinity and Salt Spray Drift Tolerance of Native Trees on the Coastal Windbreaks in the South-Sea, Korea (한국 남해안방풍림 자생수종의 내염성 및 내조성 수종 선발)

  • This study was carried out to investigate the soil salinity and salt spray drift of the indigenous windbreak trees, and its main purpose was to provide basic data for the selection of salt-tolerant trees in the saline coastal region in the South Sea of Korea. The soil salinity($EC_{1:5}$)was $0.18dSm^{-1}$, which was an average degree of the whole areas of investigation whose salinity degree ranged from $0.05dSm^{-1}$ to $0.58dSm^{-1}$. The level of soil salinity gradually decreased as it moved farther inland, except the belt I. The level of decreasing soil salinity was found to be in the following order: belt II, belt III, belt I, belt IV. The degree of soil salinity was $EC_{1:5}$ $0.22dSm_{1:5}$, $0.22dSm_{1:5}$ $0.19dSm^{-1}$ and $0.13dSm^{-1}$ respectively. The total 110 taxa, which consisted of 45 families, 74 genus, 101 species, and 9 varieties, were found to be tolerant to both soil salinity and salt spray drift. The trees that grow in the highest degree of salinity($EC_{1:5}$ $0.50dSm^{-1}$)were Parthenocissus tricuspidata(Siebold & Zucc.), Planch and Lonicera japonica Thunb. The next group of trees that grow in the high degree of salinity ranging from $EC_{1:5}$ 0.41 to $0.50dSm^{-1}$ was Cudrania tricuspidata(Carr.) Bureau ex Lavall$\acute{e}$e, Rubus parvifolius L., Zanthoxylum schinifolium(Siebold & Zucc.), Hedera rhombea(Miq.) Bean., Robinia pseudoacacia L., Quercus serrata Thunb., Callicarpa dichotoma(Lour.) K. Koch, and so on. The woody species which grew in the entire belts were Pueraria lobata(Willd.) Ohwi and Vitis flexuosa Thunb., and Vitex rotundifolia L. f. which was known to be highly tolerant to salt spray drift was found only in belt I. The woody species with high important value(IV) were Zelkova serrata(Thunb.) Makino., Celtis sinensis Pers., Koelreuteria paniculata Laxmann, Mallotusjaponicus(Thunb.) Muell. Arg., Trachelospermum asiaticum(Siebold & Zucc.) NAKAI, and Pueraria lobata(Willd.) Ohwi. These species were classified as native windbreak trees that are comparatively more tolerant to salt spray drift than other kinds.
