The Effects of Gilgyunghaedok-tang on Antitumor and Antimetastatic Activity (길경해독탕이 항암 및 항전이 효과에 미치는 영향)

  • Background and Objective : In order to investigate the effects of Gilgyunglwedok-tang (GRT) on antitumor activity and antimetastatic activity, studies were done experimentally. Materials and Methods : Experimental studies were perfonned for the cytotoxic effect on BALB/c mouse lung fibroblast cells, the proliferating effect of splenic lymphocyte, the expression of CD3e/CD4, CD3e/CD8, and B220 in peripheral blood mononuclear cells (PBMCs), the cytotoxic effect on A549, SK-OV-3, SK-MEL-2, MCF-7 cells, the inhibitory effect on the activity of DNA topoisomerase I, the T/C% in ICR mice bearing S-180, the inhibitory effect of Cell adhesive of A549 Cells and SK-OY-3 Cells to complex extracellular matrix, the inhibitory effect on lung colonies, the change of lung tissue, the antiangiogenic activity, and the effect on MMP-2 and MMP-9 gene expression in the RT1080 cell line. Results and Conclusion : The results were obtained as follows : 1. In the cytotoxic effect on BALB/C mouse lung fibroblast Cell, GHT didn't show the significant cytotoxic effect on BALB/C mouse lung fibroblast cell compared to the control group. 2. In thymidine uptake assay, GHT showed the significant proliferating effect of splenic lymphocyte in proportion to the concentration. 3. In the expression of CD3e/CD4, CD3e/CD8, and B220 in peripheral blood mononuclea cells (PBMCs) of mice, GRT had no significant change to the normal group in CD4. However, GRT showed an increase to the normal group in CD8 and GHT in the only $1\mu\textrm{g}/ml$ category showed an increase to the normal group in B220. 4. In the cytotoxic effect of GRT on A549, SK-OY-3, SK-MEL-2 and MCF-7 cells, there was no significant cytotoxic effect compared to the control group. 5. In the inhibitory effect on the activity of DNA topoisomerase I, GHT in the $10\mu\textrm{g}/ml$ category showed the inhibitory effect on the activity of DNA topoisomerase I in proportion to the concentration. 6. In the T/C% in ICRmice bearing S-180, GHTtreated group showed 123.7% of T/C% compared to the control group. 7. In the inhibitory effect of cell adhesive of A549 Cells and SK-OV-3 Cells to complex extracellular matrix, GRT in the only $100\mu\textrm{g}/ml$ category showed the significant inhibitory effect compared to the control group. 8. In the inhibitory effect on lung colonies, GHT showed the significant inhibitory effect on lung colonies compared to the control group. 9. In the change of lung tissue, GHT showed a significant decrease of lung cancer growth, interalveolar fibrosis and hyaline material compared to the control group. In the development of lymphocyte around lung cancer cells and lung parenchymal, GHT showed the significant inducement efficacy compared to the control group. 10. In CAM assay, the antiangiogenic activity of GHT showed 30%. 11. In the effect on MMP-2 and MMP-9 gene expression in the RT1080 cell line, GHT had no significant inhibitory effect on MMP-2 and MMP-9 gene expression compared to the control group. According to the above results, it could be suggested that GHT has an antitumor activity and antimetastatic activity.

Antioxidative and Neuroprotective Effects of Extract and Fractions from Adenophora triphylla (잔대 추출물과 이들 분획물들의 항산화 및 뇌신경세포 보호 효과)

  • The 70% ethanol extract from Adenophora triphylla showed a strong antioxidant effect and reduced cytotoxicity of $H_2O_2$ in SK-N-SH cells. The chloroform fraction from A. triphylla extract (AT-CH) among the six fractions showed strong DPPH radical and intracellular reactive oxygen species (ROS) scavenger effects and the highest protective effect against $H_2O_2$-induced SK-N-SH cell death. Bioactivity compounds were purified from AT-CH, and the chemical structures of the compounds were determined as ${\beta}$-sitosterol and daucosterol on the basis of $^1H-NMR$, $^{13}C-NMR$, and EI mass spectra. ${\beta}$-Sitosterol and daucosterol also had protective effects against oxidative stress in SK-N-SH cells. Phospho-p38 MAPK levels were elevated by $H_2O_2$ but were inhibited by treatment with AT-CH and phytosterols (${\beta}$-sitosterol and daucosterol) isolated from AT-CH. These results suggest that AT-CH has brain neuroprotective effects against oxidative stress ($H_2O_2$) by inhibiting activation of p38 pathways and scavenging intracellular ROS.

Anticarcinogenic Effects of Allium tuberosum on Human Cancer Cells (부추 추출 성분의 항발암 효과 연구)

  • The anticarcinogenic effects of various food components have received much attention in recent years. However mechanism of anticarcinogens in food materials on cancer cells have rarely been investigated. This study was performed to investigate the effects on the cytotoxicity and quinone reductase (QR) activity of Allium tuberusum (AT) on the human cancer cells. The six partition layers which are methanol (ATM), hexane (ATMH), ethylether (ATMEE), ethylacetate (ATMEA), butaonl (ATMB) and aqueous (ATMA) of Allium tuberusum were screened for their cytotoxic effects on HepG2, MCF-7, HeLa and SK-N-MC cells by the MTT assay. Among the six partition layers, ATMEE had the strongest cytotoxic effect at concentration of $150\;{\mu}g/mL$ which resulted over 95% on HepG2, HeLa, MCF-7 and SK-N-MC cell lines. The ATMEA also showed significant cytotoxic effect on HepG2 and SK-N-MC cell lines. The ATMB showed the highest induction activity of QR on HepG2 cells among the other partition layers. QR activity of HepG2 cells, grown in the presence of ATMB at the concentration of $50\;{\mu}g/mL$, was increased by 3.9 times, compared to the control value of 1.0. Based on these results, the ATMEE and ATMB may have potentially anticarcinogenic and chemopreventive activities.

Anti-wrinkle effects of extracts and solvent fractions from Nymphoides peltata on CCD-986sk (CCD-986sk 세포내에서의 노랑어리연꽃 추출물 및 분획물의 주름개선 효과)

  • The purpose of this study was to investigate the antiaging effect in the fibroblasts by using the extracts and fractions of Nymphoides peltata for the development of new functional cosmetic materials. The anti-aging effects of the extracts and fractions of N. peltata were measured using CCD-986sk cells. Ethyl aceate (EA) fraction inhibited UVB-induced reactive oxygen species expression by more than 50%, and n-butyl alcohol (BuOH) fraction increased expression of pro-collagen mRNA by more than 50% at $25{\mu}g/mL$. In addition, EA and BuOH fractions inhibited MMP-1 protein and mRNA expression in a concentration-dependent manner, BuOH fraction inhibited MMP-1 mRNA expression by more than 40% at $10{\mu}g/mL$. Especially, the high efficacy of EA and BuOH of the N. peltata was confirmed. Therefore, the EA and BuOH of N. peltata are expected to be useful as anti-aging materials in cosmetics field.

Induction of Caspase-3 Dependent Apoptosis in Human Ovarian Cancer SK-OV-3 Cells by Genistein

  • The present study was designed to determine how the phytochemical genistein activates caspase-3 to cause cell cycle arrest and apoptosis. When human ovarian cancer SK-OV-3 cells were treated with $200\;{\mu}M$ genistein for 24 hr, cell growth decreased significantly (p<0.05). Conversely, genistein treatment significantly increased cytotoxicity (measured as lactate dehydrogenase release) under the same conditions (p<0.05). To elucidate the mechanism behind the induction of apoptosis by genistein, we studied the cell cycle and caspase-3 activation. When cells were treated with genistein, the population of cells in sub-G1 phase increased by 44.2% compared to untreated cells. Genistein caused decrease in precursor caspase-3, increase in cleaved caspase-3 and a significant increase in caspase-3 activity (p<0.05). Therefore, genistein may induce apoptosis via caspase-3 activation. However, high-dose genistein treatment must be viewed with caution because of its potential cytotoxicity.

The Effects of Artemisia capillaries Herbal Acupuncture on Ethanol-induced Apoptosis in Neuroblastoma Cell Line (인진 약침액이 신경아세포주에서 에탄올에 의해 유발된 아폽토시스에 미치는 영향)

  • 목적:인진 약침액이 SK-N-MC 신경아세포주에서 에탄올에 의해 유발된 아폽토시스에 미치는 영향을 조사하였다. 방법 : SK-N-MC cell line에서의 아폽토시스 변화를 관찰하기 위해서 MTT assay, DAPI staining 및 flow cytometric analysis 방법을 이용하였다. 결과: MTT assay를 이용하여 분석한 결과 농도에 따른 세포 독성의 효과가 에탄올 투여로부터 관찰되었다. 또한 인진 약침액으로 전처치하고 에탄올을 처치하였을 때 세포 독성이 크게 감소되었다. DAPI staining에서 인진 약침액 투여군은 에탄올 투여군에 비해서 fragmentation이 억제되었다. Flow cytometry를 통하여 인진 약침액 투여군은 에탄올 투여군에 비하여 세포주기 중 sub $G_1$ 분획의 증가가 억제되었다. 결론 : SK-N-MC 신경아세포주에서 에탄올에 의해서 유발된 아폽토시스는 전형적인 세포사별 형태를 나타내었다. 또한 인진 약침액은 에탄올에 의해서 유발된 아폽토시스에서 세포보호 효과가 있음이 확인되었다.

Effect of Scutellariae Radix Water Extract on Hydrogen Peroxide Production in RAW 264.7 Mouse Macrophages (황금(黃芩) 물추출물이 마우스 대식세포의 hydrogen peroxide 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate effects of Scutellariae Radix Water Extract on hydrogen peroxide production in RAW 264.7 mouse macrophages. Methods : Scutellariae Radix produced from South Korea (SK) and Scutellariae Radix produced from China (SC) were extracted by hot water. Effects of SK and SC on hydrogen peroxide production in RAW 264.7 were measured by dihydrorhodamine 123 assay after 2, 4, 20, 24, 28, 44, and 48 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL. Results : SK significantly increase hydrogen peroxide production in RAW 264.7 cells for 2, 4, 20, 24, 28, 44, and 48 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL (P < 0.05). SC also significantly increase hydrogen peroxide production in RAW 264.7 cells for 4, 20, 24, 28, and 48 h incubation at the concentrations of 10, 25, 50, and 100 ug/mL (P < 0.05). For 2 h incubation, SC significantly increase hydrogen peroxide production in RAW 264.7 cells at the concentrations of 10, 25, and 100 ug/mL (P < 0.05). For 44 h incubation, SC significantly increase hydrogen peroxide production in RAW 264.7 cells at the concentrations of 10, 25, and 50 ug/mL (P < 0.05). Conclusions : These results suggest that Scutellariae Radix has the immune - enhancing property related with its increasement of hydrogen peroxide production in macrophages.

The Combined Effects of Ginkgo Biloba Extracts and Aspirin on Viability of SK-N-MC, Neuroblastoma Cell Line in Hypoxia and Reperfusion Condition

  • Objective: The purpose of this study is to investigate the combined effects of ginkgo biloba extract, ginkgolide A and B and aspirin on SK-N-MC, human neuroblastoma cell viability and mRNA expression of growth associated protein43 (GAP43), Microtubule-associated protein 2 (MAP2), B-cell lymphoma2 (Bcl2) and protein53 (p53) gene in hypoxia and reperfusion condition. Methods: SK-N-MC cells were cultured with Dulbecco's Modified Eagle's Medium (DMEM) media in $37^{\circ}C$, 5% $CO_2$ incubator. The cells were cultured for 8 hours in non-glucose media and hypoxic condition and for 12 hours in normal media and $O_2$ concentration. Cell survival rate was measured with Cell Counting Kit-8 (CCK-8) reagent assay. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to estimate mRNA levels of GAP43, MAP2, Bcl2, and p53 genes. Results: The ginkgolide A and B increased viable cell number decreased in hypoxic and reperfused condition. The co-treatment of ginkgolide B with aspirin also increased the number of viable cells, however, there was no additive effect. Although there was no increase of mRNA expression of GAP43, MAP2, and Bcl2 in SK-N-MC cells with individual treatment of ginkgolide A, B or aspirin in hypoxic and reperfused condition, the co-treatment of ginkgolide A or B with aspirin significantly increased GAP43 and Bcl2 mRNA levels. In MAP2, only the co-treatment of ginkgolide A and aspirin showed increasing effect. The mRNA expression of p53 had no change in all treating conditions. Conclusion: This study suggests that the combined treatments of Ginkgo biloba extracts and aspirin increase the regeneration of neuroblastoma cells injured by hypoxia and reperfusion.
