The Effects of Gilgyunghaedok-tang on Antitumor and Antimetastatic Activity (길경해독탕이 항암 및 항전이 효과에 미치는 영향)

  • Background and Objective : In order to investigate the effects of Gilgyunglwedok-tang (GRT) on antitumor activity and antimetastatic activity, studies were done experimentally. Materials and Methods : Experimental studies were perfonned for the cytotoxic effect on BALB/c mouse lung fibroblast cells, the proliferating effect of splenic lymphocyte, the expression of CD3e/CD4, CD3e/CD8, and B220 in peripheral blood mononuclear cells (PBMCs), the cytotoxic effect on A549, SK-OV-3, SK-MEL-2, MCF-7 cells, the inhibitory effect on the activity of DNA topoisomerase I, the T/C% in ICR mice bearing S-180, the inhibitory effect of Cell adhesive of A549 Cells and SK-OY-3 Cells to complex extracellular matrix, the inhibitory effect on lung colonies, the change of lung tissue, the antiangiogenic activity, and the effect on MMP-2 and MMP-9 gene expression in the RT1080 cell line. Results and Conclusion : The results were obtained as follows : 1. In the cytotoxic effect on BALB/C mouse lung fibroblast Cell, GHT didn't show the significant cytotoxic effect on BALB/C mouse lung fibroblast cell compared to the control group. 2. In thymidine uptake assay, GHT showed the significant proliferating effect of splenic lymphocyte in proportion to the concentration. 3. In the expression of CD3e/CD4, CD3e/CD8, and B220 in peripheral blood mononuclea cells (PBMCs) of mice, GRT had no significant change to the normal group in CD4. However, GRT showed an increase to the normal group in CD8 and GHT in the only $1\mu\textrm{g}/ml$ category showed an increase to the normal group in B220. 4. In the cytotoxic effect of GRT on A549, SK-OY-3, SK-MEL-2 and MCF-7 cells, there was no significant cytotoxic effect compared to the control group. 5. In the inhibitory effect on the activity of DNA topoisomerase I, GHT in the $10\mu\textrm{g}/ml$ category showed the inhibitory effect on the activity of DNA topoisomerase I in proportion to the concentration. 6. In the T/C% in ICRmice bearing S-180, GHTtreated group showed 123.7% of T/C% compared to the control group. 7. In the inhibitory effect of cell adhesive of A549 Cells and SK-OV-3 Cells to complex extracellular matrix, GRT in the only $100\mu\textrm{g}/ml$ category showed the significant inhibitory effect compared to the control group. 8. In the inhibitory effect on lung colonies, GHT showed the significant inhibitory effect on lung colonies compared to the control group. 9. In the change of lung tissue, GHT showed a significant decrease of lung cancer growth, interalveolar fibrosis and hyaline material compared to the control group. In the development of lymphocyte around lung cancer cells and lung parenchymal, GHT showed the significant inducement efficacy compared to the control group. 10. In CAM assay, the antiangiogenic activity of GHT showed 30%. 11. In the effect on MMP-2 and MMP-9 gene expression in the RT1080 cell line, GHT had no significant inhibitory effect on MMP-2 and MMP-9 gene expression compared to the control group. According to the above results, it could be suggested that GHT has an antitumor activity and antimetastatic activity.

  • Two new sulfated saponins designated as certonardosides P$_{2}$ and I$_{3}$ (1 and 2) were isolated from the brine shrimp active fraction of the MeOH extract of the starfish Certonardoa semiregularis. The structures were determined on the basis of spectral analysis. Compounds 1 and 2 were tested for cytotoxicity against five human tumor cell lines (A549, SK-OV-3, SK-MEL-2, XF498, and HCT15), and compound 1 displayed significant cytotoxicity against the SK-MEL-2 skin cancer cell.

  • A minor cytotoxic compound was isolated by bioassay-guided fractionation from Asiasari Radix and identified as aristolactam III(1) on the basis of spectral data and chemical evidence. This is the first report on the isolation of compound 1 from Asiasarum genus. Compound 1 exhibited a significant cytotoxic activity against the three kinds of human cancer cell lines (A 549, SK-MEL-2 and SK-OV-3).

Study on Antitumor Activity of Samiyeongeontanggamibang(SYTG) (사미연견탕가미방(四味軟堅湯加味方)이 항암(抗癌) 및 항전이(抗轉移) 효과(效果)에 미치는 영향)

  • To evaluate the antitumor and antimetastatic effects of Samiyeongeontanggamibang(SYTG), We have examined whether SYTG can inhibit the growth of several tumor cell lines, tumor cell adhesion, experimental tumor metastasis and increase survival rate of tumor-bearing mice by inhibition of DNA topoisomrase activity. The results were obtained as follows: 1. SYTG extracts revealed an efficient cytotoxicity against A549, SK-OV-3, B16-F10, and SK-MEL-2 cell lines. 2. SYTG extracts inhibited DNA topo-isomerase I from calf thymus. 3. The T/C% in S-180 bearing ICR mice treated with SYTG was 115.8% 4. SYTG extracts significantly inhibited adhesion of A549 cell to complex extracellular matrix. 5. In pulmonary colonization assay, SYTG suppressed lung metastases in tumor cell-injected mice. 6. In CAM assay, SYTG extracts inhibited angiogenesis at $15{\mu}g/egg$ concentration as compared with control. These results suggested that SYTG extracts might be a potent inhibitor of cancer.

Cytotoxic Activities of 6-Arylamino-7-halo-5,8-quinolinediones against Human Tumor Cell Lines

  • 6-Arylamino-7-halo-5,8-quinolinediones (4a-4k, 5a-5b) were tested for in vitro cytotoxicity against human solid tumor cell lines such as A 549 (non-small cell lung). SK-OV-3 (ovarian), SK-MEL-2 (melanoma), HCT-15 (colon) and XF 498 (CNS) by SRB assay. The arylamino-7-chloro-5,8-quinolinediones 4 were also evaluated for cyclin-dependent kinase (CDK2 and CDK4) inhibitory effect. Among them, the 5,8-quinolinediones 4a and 5a with 7-(4-fluorophenyl) amino group were found to be potent cytotoxic against HCT 15, SKOV-3 and XF 498, and the compounds 4f and 4i showed inhibitory activities for the CDK4.

Analysis of Cytotoxic Constituent of Berberis koreana Palibin (매자나무 세포독성성분 분석)

  • Methanol extracts of five Berberidaceae species were examined against tissue factor inhibitory and tumour cell growth inhibitory activity. Methanol extracts of Berberis koreana Palibin showed a strong cytotoxicity activity against SK-MEL-2 (Melanoma) tumour cell lines with more than 90% in $25{\mu}g/m\ell$ and against A549 (Lung carcinoma), SK-OV-3 (Ovarian cancer), XF498 (CNS cancer) and HCTl5 (Colon cancer), other Berberidaceae species except B. koreana species have no effect on the tumour cells. Biologically active compound, therefore, was isolated through the activity guided fractionation and purification. The structure was confirmed by NMR. FT-IR and MS to 2-(3,4-dihydroxybenzyl)-ethyl alcohol. It showed cytotoxicity activity against SNU-C4 tumour cell lines with 50.7% in $50{\mu}g/m\ell$. Methanol extracts of 5 Berberidacae species have no effect on the tissue factor inhibitory activity.

  • Seventy three hydrolysable tannins and related compounds were isolated from seven Euphorbia plants. Among them, 28 compounds including nine gallotannins, eleven ellagitannins and eight related compounds were selected according to the structural similarity. Cytotoxicity of them on the human tumor cell lines including A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT-15 were evaluated by the SRB method in vitro. 3,4,6-Tri-O-galloyl-D-glucose was shown to exhibit most potent cytotoxic effect($4.4{\mu}g/ml).

  • In order to study the cytotoxic properties of sesquitepenes, dehydrocostus lactone (DL) and costunolide from Saussurea lappa, cytotoxicity was measured by SRB method using various human cancer cell lines. Dehydrocostus lactone(DL) and costunolide exhibited significant cytotoxicity against A-549, SK-OV-3, SK-MEL-2, XF-498 and HCT 15 cells. The U937 human leukemia cells treated with DL showed several apoptotic evidences like chromosome condensation and formation of apoptotic bodies. From the results of FACS analysis, early apoptosis was observed by phosphatidylserine externalization detected by annexin V-FITC. Furethermore, these studies determined hypodiploid contents and effects on the cell phase distribution of DL-treated U937 cells. After exposure of U937 cells to $30\mu\textrm{M}$ DL effectively led to G2/M modified cell cycle distribution within 24hr. These observations suggest that DL can be used efficiently for the cancer treatment.

  • Repeated column chromatographic separation of the $CH_{2}Cl_{2}$ extract of Artemisia stolonofera (Asteraceae) led to the isolation of a triterpene (I), a sesquiterpene (II), two aromatic compounds (III and IV) and a benzoquinone (V). Their structures were determined by spectroscopic means to be simiarenol (I), (1S,7S)-1 $\beta$-hydroxygermacra-4(15),5, 10(14)-triene (II), 3'-methoxy-4'-hydroxy-trans-cinnamaldehyde (III), vanillin(IV) and 2,6-dimethoxy-1,4-benzoquinone (V), respectively. Among these products, compound V showed significant cytotoxicity against five human tumor cell lines in vitro, A549 (non small cell lung adenocarcinoma), SK-OV-3 (ovarian), SK-MEL-2 (skin melanoma), XF498 (CNS) and HCT15 (colon) with ED_{50}$ values ranging from 1.33~4.22${\mu}g/ml$.
