Cucumber Mosaic Virus 1a Protein Interacts with the Tobacco SHE1 Transcription Factor and Partitions between the Nucleus and the Tonoplast Membrane

  • The transcription factor SHE1 was identified as an interacting partner with the cucumber mosaic virus (CMV) 1a protein in the yeast two-hybrid system, by a pull-down assay, and via bimolecular fluorescent complementation. Using fluorescent-tagged proteins and confocal microscopy, the CMV 1a protein itself was found distributed predominantly between the nucleus and the tonoplast membrane, although it was also found in speckles in the cytoplasm. The SHE1 protein was localized in the nucleus, but in the presence of the CMV 1a protein was partitioned between the nucleus and the tonoplast membrane. SHE1 expression was induced by infection of tobacco with four tested viruses: CMV, tobacco mosaic virus, potato virus X and potato virus Y. Transgenic tobacco expressing the CMV 1a protein showed constitutive expression of SHE1, indicating that the CMV 1a protein may be responsible for its induction. However, previously, such plants also were shown to have less resistance to local and systemic movement of tobacco mosaic virus (TMV) expressing the green fluorescent protein, suggesting that the CMV 1a protein may act to prevent the function of the SHE1 protein. SHE1 is a member of the AP2/ERF class of transcription factors and is conserved in sequence in several Nicotiana species, although two clades of SHE1 could be discerned, including both different Nicotiana species and cultivars of tobacco, varying by the presence of particular insertions or deletions.

  • The enterprise are facing potential SHE(safety, health and environment) accident to cause casualties or a financial loss and increasing social responsibility. So, SHE management system has to be voluntarily accomplished rather than passively with regard to SHE regulation. In this study, KW_SHE(Kwangwoon Safety, Health and Environment) management model is developed which materialize self-regulation SHE management system of individual workplace by the research and analysis of many domestic enterprise. This KW-SHE management model is intend to assess work-product vs. management expenses investment and critically evaluated for studying its benefits and limitations

Historical Review of Lee Keumjeon, a Pioneer in Community Health Nursing in Korea (한국 지역사회간호의 선구자 이금전에 관한 역사적 고찰)

  • Purpose: The purpose of this study is to show the development of community health nursing in Korea in light of the life of Lee Keumjeon (1900~1990), who devoted her life to community health nursing. Methods: Primary and secondary sources were collected and analyzed. Results: Lee could get high level education up to college courses, which was very exceptional at that time in Korea. She got nursing and midwifery education in Severance Hospital (1929) and majored in public health nursing at Toronto University (1930). Then, she worked in mother-and-child health practice for more than 10 years. She helped the Korean Nurses' Association to publish Public Health Nursing (1933) and other nursing books. After the liberation of Korea, she became a governmental official in the public health nursing field and tried to establish the national public health nursing system. During the Korean War, she devoted herself to nursing education and practice at nursing schools and hospitals. After the war, she worked as president of the Korean Nurses' Association. In 1959, Lee was given the Nightingale award. Although she retired in 1960, she continued to devote herself to the development of nursing, and published her book Public Health Nursing (1967). Conclusion: Lee worked from 1920s to 1960s for the development of nursing in Korea and during the period Korean nursing showed great development to national system and professional status.

Ethanol extract separated from Sargassum horneri (Turner) abate LPS-induced inflammation in RAW 264.7 macrophages

  • Background: This study is aimed at identifying the anti-inflammatory properties of 70% ethanol extract produced from an edible brown seaweed Sargassum horneri (SJB-SHE) with industrial-scale production by Seojin Biotech Co. Ltd. S. horneri is a rich source of nutrient and abundantly growing along the shores of Jeju, South Korea. Methods: Here, we investigated the effect of SJB-SHE on LPS-activated RAW 264.7 macrophages. The cytotoxicity and NO production of SJB-SHE were evaluated using MTT and Griess assays, respectively. Additionally, protein expression and gene expression levels were quantified using ELISA, Western blots, and RT-qPCR. Results: Our results indicated that pre-treatment of RAW 264.7 macrophages with SJB-SHE significantly inhibited LPS-induced NO and $PGE_2$ production. SJB-SHE downregulated the proteins and genes expression of LPS-induced iNOS and COX2. Additionally, SJB-SHE downregulated LPS-induced production of pro-inflammatory cytokines (tumor necrosis factor-${\alpha}$, interleukin (IL)-6, and IL-$1{\beta}$). Furthermore, SJB-SHE inhibited nuclear factor kappa-B (NF-${\kappa}B$) activation and translocation to the nucleus. SJB-SHE also suppressed the phosphorylation of mitogen-activated protein kinases (ERK1/2 and JNK). Conclusions: Collectively, our results demonstrated that SJB-SHE has a potential anti-inflammatory property to use as a functional food ingredient in the future.

Lived experience of mothers who have child with cerebral palsy (뇌성마비아 어머니의 경험)

  • The purpose of the study is to identify the lived experience of mothers who have children with cerebral palsy in order to understand their agony. Moreover, the result of study was to find some nursing intervention for disabled children and their mothers. For this purpose, ten mothers who are willing to cooperate with this research were selected at random from those who have children with the cerebral palsy, currently using the municipal facilities for the handicapped with cerebral malfunction. Data collection was done from October 4, 1994 th December 31, 1994. The data were collected by asking the mothers mentioned above with some unstructured open-ended questions, recorded on the tapes with permission by the interviewee in order to prevent missing of the interviewed contents. These collected data have been substantiated and properly analyzed on the basis of phenomenological approach initiated by Colaizzi's method. The results and validity are proved to be credible by means of the individual checking of the interviewed mothers. The results of this study are as follows : 1. When the mother is first informed of the diagnosis of cerebral palsy on her child, she usually misses the crucial timing needed for proper treatment of the child's disorder because she is notified through the doctor's indifference and his apparently inactive, matter-of-fact attitude. At first she suspects the doctor's diagnosis and tries to attribute it to the unknown cause from a certain genetic problem and then she quickly wants to deny the whole situation that her child is really suffering from the cerebral palsy. The reality is too much for her to accept as it is and she would not believe her child is abnormal. Therefore, she even attempts depend on the power of God for its solution. 2. The mother, who goes thorough this kind of uncommon experiences, is totally devoted to the treatment and care of the child and completely ignores her own life and happiness. At the same time, she feels sorry for her other normal children she believes having not enough care and concern. Also, she feels sorry for the sick child when the child's brothers or sisters show special concern for the patient out of sympathy. It is sorry and not satisfied for her that the child is growing with abnormality and neighbor other around have inappropriate attitudes. Likewise, she is discontent with her husband's lack of concern about the child's treatment. She believes that the health care system in this society isn't fulfilling its due purpose. In the state of her utmost distress and anxiety, she always feels the need of competent consultants, and is angry about that her child is treated as an abnormal being, she is trying to hide the child from other people and to make him or her disappear, if possible. Although she doesn't have harmonious relation with her husband, she id happy when he shows his affection for the child and she feels relieved and thankful when the relatives don't mention about the child's condition Since the child's overall status of health is continuously in unstable conditions, requiring her all-time readiness for an emergency, she feels guilty of her child's illness toward the fEmily members as if it was her own fault to have borne such an abnormal child and she feels responsible for the child morally and financially if necessary Because her life is centered on taking care of the child, she cannot afford to enjoy her own life and happiness. She is a lonely mother, fatigued, with no proper relationship with other people around her. With this sense of guilt and responsibility as a mother of an unusual disease, she has no choice but to grieve her destiny from which she is not allowed to escape. 3. Nevertheless, the mother with the child suffering from the cerebral palsy does not easily give up the hope of getting her child cured and she believes that in the long run, though slower than hoped, her abnormal son or daughter will be eventually cured to become a normal sibling someday. This kind of hope is sustained by the mother's strong faith coming from observing the progress of other similar children getting better. Sometimes she is encouraged to have this faith by other mothers who share the same painful experiences, believing that her child will improve even more rapidly than others with the same palsy. Full of hope, she painstakingly waits for the child's healing. Moreover, she plans to have another child. she thinks that the patient child's brothers and sisters only can truly understand and look after the patients. However, when she notices that the progress of other children under the treatment does not look so hopeful, she is distressed by the thoughts that her child may never get well. Too, she is worried that the patient's brother or sister will be born as the same invalid with the cerebral disease. She is discouraged to have another baby as much as she is encouraged to. She is also troubled by the thought that in case she has another baby, she will have to be forced. to neglect the patient child, especially when she does have an extra hand or some reliable person to help her with taking care of the patient.

Living as a ChoSun-jok Aunt in Korea : Hermeneutical Phenominological Analysis of the Life Experience of ChoSun-jok Female Domestic Worker (한국에서 조선족이모로 살아가기 : 조선족 육아.가사도우미의 삶에 대한 해석학적 현상학)

  • The purpose of this study is to understand a ChoSun-jok female domesic worker's experience in a Korean home. This study adopted hermeneutic phenomenological method which was developed by van Manen. The participant for this study was a ChoSun-jok woman who had worked as a care-giver and housekeeper in Korean home for more than 2 years. She missed the time when she was raising her own daughter and son and expected the time that her family would be together. So, her present time wears away for her stable future in Korea which give an opportunity to make a good money. Also, she realized that she was a marginal existence : it meant she was neither a family member nor truly worker. She nursed the Korean children and did for the Korean family for money but wanted to be treated as a family member like a grandmother who could be honored about what she had done for the family. She felt that she was disregarded from the family when she miscommunicated with them because of the difference between cultures. She anticipated that she was appreciated for being here in the Korean home.

Effects of Siegesbckiae Herba on the Brain Ischemia (희렴 이 뇌허혈에 미치는 효과)

  • This experimental study was designed to investigate the effects of SIEGESBECKIAE HERBA extract (SHE) on the change of cerebral hemodynamics 〔regional cerebral blood flow (rCBF) and mean arterial blood pressure(MABP)〕 in normal condition and cerebral ischemic rats, and to determine the mechanism of action of SHE. This study was designed to investigate whether or not SHE inhibit lactate dehydrogenase (LDH) activity in neuronal cells and cytokines production in serum of cerebral ischemic rats. The results were as follows SHE increased rCBF significantly in a dose-dependent manner, but MABP was not changed by SHE in normal rats. The SHE-induced increase in rCBF was significantly inhibited by pretreatment with indomethacin (IDN), an inhibitor of cyclooxygenase but was increased by methylene blue (MTB), an inhibitor of guanylate cyclase. SHE inhibited lactate dehydrogenase (LDH) activity significantly in neuronal cells. rCBF was increased significantly and stably by SHE(10㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group in ischemic rats. In serum by drawing from femoral arterial blood after middle cerebral arterial occlusion(MCAO) for 1hr and reperfusion for 1hr, the sample group was decreased IL-1β production significantly compared to that of the control group. In serum by drawing from femoral arterial blood after MCAO 1hr and reperfusion 1hr, sample group decreased TNF-α production significantly compared to that of the control grolilp. In serum by drawing from femoral arterial blood after reperfusion 1hr, sample group increased TGF-β production significantly compared to that of the control group. In serum by drawing from femoral arterial blood after MCAO for 1hr and reperfusion for 1hr, IL-10 production of the sample group was similar to that of control group. These results suggested that SHE had inhibitive effect on the brain damage by inhibited LDH activity, IL-1β and TNF-α production, but accelerated TGF-β production.

A Study on Sonia Delaunay's Painting, Fashion and Fabric Design (쏘냐 들로우네의 회화와 의상$\cdot$직물디자인 세계)

  • Sonia Delaunay(1885$\~$1979) was one of great pioneers of abstract art, who looked at clothes and fabrics with a plastics eyes. In Association with her husband Robert Delaunay, they were instrumental in founding the movement of Orphism, she proceeded to mix strong and bright colors into her art and had a brilliant influence on the decoration and women's fashion of the 1920's. Having a strong sense for dramatic and decorative color derived in part from childhood remembrances of Russian folk art she initiated a total revolution in which she created her first simultaneous dress with multi-colored samples of materials. She extended the principle of color's simultaneity to the field of fashion, fabric design and applied art. She was interested in the dress for ballet and opera. Fashion designer Paul Poiret asked her to design the fabrics and she created the geometric and abstract patterns with her strong color. It seemed that her design was revolutionary and avant-garde. Always she desired not only art of seeing but also art of living. The purpose of this study is to recognize the influence she had upon the history of modern fashion and fabric design. It was remarked that her paintings served as a basis for later developments in Kinetics Art and had influences on 1980' s abstract patterns for silk dress. Finally, the concept 'simultaneity' of her art signifies endless rhythmes in space and time.

Inhibitory Effects of Siegesbeckia Herba Extracts on the Melanin Production and Tyrosinase Activity in B16F10 cells (희첨 추출물이 B16F10 세포에서 Melanin 생성과 Tyrosinase 활성에 미치는 억제효과)

  • Objective : Siegesbeckia Herba can treat various skin disease by expelling wind and removing dampness and clearing away heat and toxic material effects. This study was designed to investigate effects of Siegesbeckia Herba Extracts(SHE) on skin elasticity and whitening using B16F10 cell lines. Method : In this experiment, We observed effect of SHE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : 1. SHE treated group showed decreased cell viability rates significantly compared with non-treated group. More than SHE $250{\mu}g/ml$, $500{\mu}g/ml$ and $1,000{\mu}g/ml$ of treated groups were lower levels of melanin synthesis respectively. 2. SHE significantly showed tyrosinase inhibitory activity in vitro, SHE increased tyrosinase inhibitory activity and elastase inhibitory activity in B16F10 cells, and tyrosinase inhibitory activity in vitro. 3. Tyrosinase inhibitory activity and elastase inhibitory activity in B16F10 cells, tyrosinase inhibitory activity in vitro were not accepted statistical significance compared with non-treated group. 4. SHE treated group showed increased SOD-like activity rates significantly compared with non-treated group. More than SHE $250{\mu}g/ml$, $500{\mu}g/ml$ and $1,000{\mu}g/ml$ of treated groups were lower levels of melanin synthesis respectively. Conclusion : These results suggest that SHE can inhibit melanin synthesis and tyrosinase inhibtory activity. So, We suggest that SHE can be maintained skin whitening.

Postpneumonectomy Syndrome -A Case Report- (전폐절제술후 증후군 -1례 보고-)

  • Airway obstruction may be caused by extreme mediastinal shift and rotation after right pneumonecotmy or after left pneumonecotomy in the presence of right aortic arch.We experienced such a complication after right pneumonectomy, so called right postpneumonectomy syndrome. The patient was 28 years old female, and 4 month ago she had undergone right pneumonecotomy via video assisted thoracoscopic surgery[VATS] for endobronchial tuberculus dissemination and secondary pulmonary infection. She was treated by mediastinal repositioning which were composed of substernal fixation of pericardium and insertion of expandable prosthesis of 1000 cc capacity. She had good postoperative course and now she feels no obstructive symptoms.
