Objectives: Radix Ginseng has been used for thousands of years to treat a wide variety of diseases. Radix ginseng has also been used as a traditional medicine for boosting Qi energy and tonifying the spleen and lungs. Traditionally, its effect could be obtained orally. Nowadays, a new method, the injection of herbal medicine, is being used. This study was performed to investigate the single-dose intravenous toxicity of water-soluble ginseng pharmacopuncture (WSGP) in Sprague-Dawley (SD) rats. Methods: All experiments were carried out at Biotoxtech, an institute authorized to perform non-clinical studies under the regulation of Good Laboratory Practice (GLP). At the age of six weeks, 40 SD rats, 20 male rats and 20 female rats, were allocated into one of 4 groups according to the dosages they would receive. The WSGP was prepared in the Korean Pharmacopuncture Institute under the regulation of Korea-Good Manufacturing Practice (K-GMP). Dosages of WSGP were 0.1, 0.5 and 1.0 mL/animal for the experimental groups, and normal saline was administered to the control group. The rat's general conditions and body weights, the results of their hematological and biochemistry tests, and their necropsy and histopathological findings were investigated to identify the toxicological effect of WSGP injected intravenously. The effect was examined for 14 days after the WSGP injection. This study was performed under the approval of the Institutional Animal Ethics Committee of Biotoxtech. Results: No deaths were found in this single-dose toxicity test on the intravenous injection of WSGP, and no significant changes in the rat's general conditions and body weights, the results on their hematological and biochemistry test, and their necropsy findings were observed during the test. The local area of the injection site showed minial change. The lethal dose was assumed to be over 1.0 mL/animal in both sexes. Conclusion: These results indicate that WSGP is safe at dosages up to 1 mL/animal.
Naringin, major citrus flavonoids, has been identified to exert antioxidative, antidiabetic, and lipid lowering effects. In this study, we examined the effect of 0.2 g/kg, 0.5 g/kg naringin supplementation for 3 times/week for 5 weeks on lipid metabolism and antithrombotic capacity in rat. Eighteen five week-old Sprague Dawley(SD) female rats, which had initial body weights of $246{\pm}9g$, were randomly divided into three groups: Control (non naringin group); Low (0.2 g/kg naringin-supplemented group); High (0.5 g/kg naringin-supplemented group). Three groups of rats were supplemented with three experimental diets for 5 weeks and we investigated antithrombotic capacity before sacrifice. Naringin did not significantly alter the body weight gain, relative organ weight. However, the level of serum triglyceride, serum free fatty acid, serum total lipid and serum glucose levels were significantly lowered compared to those of control. The high group (0.5 g/kg naringin-supplemented group) was showed significantly increased bleeding time compared to control group. These results suggest that naringin supplemental diets reduces the level of hypertension, glycosuria and fatness on the female SD rats, when orally administered below the dosage 0.5 g/kg for 5 weeks.

This study confirmed the radioprotective effects of a mixed extracts of Hottuynia, Perilla Frutescens, and Camellia Sinnensis as natural radioprotectors on the prostate, small intestine, and liver of SD rats. It is known that Hottuynia, Perilla Frutescens, and Camellia Sinnensis have antioxidant effects on the prostate, small intestine, and liver, respectively.In this study, SD rats were irradiated with 8 Gy of gamma rays to confirm the radioprotective effects of a mixed extracts of Hottuynia, Perilla Frutescens, and Camellia Sinnensis. After radiation irradiation, histological analysis of the prostate, small intestine, and liver was performed. After radiation irradiation, histological analysis of the prostate, small intestine, and liver was performed. In the case of the prostate, the HPC+IR Group had less prostate damage and better recovery due to radiation compared to the IR Group. It was confirmed that the prostate size of the HPC+IR Group was 11.48%p and 24.54%p higher than the IR Group on 1st and 7th days. In the case of the small intestine, the HPC+IR Group had less radiation-induced small intestinal damage and recovery was better than the IR Group. The length of small intestine villus in the HPC+IR Group was confirmed to be 23.73%p and 24.27%p higher than the IR Group on the 1st and 7th days. In the case of the liver, the HPC+IR Group had less liver damage due to radiation and had better recovery than the IR Group. This was confirmed through the hepatic portal vein and surrounding cells. The results of this study are considered to be used as basic data for research on natural radiation protection using mixed extracts.
KIOM-MA128 is a novel Korean herbal medicine with anti-atopic, anti-inflammatory and anti-asthmatic effects. This article presents the first pharmacokinetic study on KIOM-MA128. The purpose of this study was to characterize a pharmacokinetic characteristic of matrine, a potential marker of KIOM-MA128, in rats using population pharmacokinetic model. 1, 2 and 8 g/kg of KIOM-MA128 were administered to rats orally and plasma concentrations of matrine was determined by HPLC-MS/MS. Non-compartmental analysis (NCA) was performed using Phoenix$^{(R)}$ and pharmacokinetic model was built using NONMEM$^{(R)}$. This model was validated with internal validation which is visual predictive check (VPC) and bootstrap. The NCA result of matrine showed that $C_{max}$ was 294.24, 552.22 and 868.65 ng/ml, $AUC_{inf}$ was 1273.05, 2724.76 and $9743.25ng{\cdot}hr/ml$ and $T_{max}$ was 1, 1.3 and 2.3 hr for the doses of 1, 2, and 8 g/kg, respectively. The rat plasma concentrations were described very well with one-compartment model. Pharmacokinetic model for matrine was successfully developed and evaluated. Finally, our model is helpful to understand pharmacokinetic characteristic of KIOM-MA128.
Because moire artifacts overlap with fine lesions and frequency bands, image processing software that removes moire artifacts can lead to loss of micro lesions. In this study, microscopic lesions such as microcalcification and microfracture were randomly formed on SD (Sprague Dawley) rats and image and optimized grid images were compared and analyzed using reference image and grid suppression software. The images were read by two consecutive radiologists using a McNemar's test. Among the 73 microcalcifications, in the 13 cases after grid suppression, the image of the optimized grid shows the loss of image in 3 cases, and the image after grid suppression shows statistically significant image loss (p=0.021). In all 53 fracture lines, there were 19 cases of image loss after the grid suppression, and only one case of the optimized grid showed no image loss. Therefore, the use of grid suppression software to remove moire artifacts should be carefully considered in the diagnosis of micro lesions.
The aim of this study was to evaluate gamma-aminobutyric acid (GABA)-induced erythropoietin (EPO) and EPO-receptor expression in human Hep3B cells and Sprague Dawley (SD) rats during hypoxia. Expression levels of EPO, EPO-R mRNA, Janus kinase-2 (JAK-2), vascular endothelial growth factor (VEGF), hypoxia inducible factor-1 (HIF-1), and HIF-2 in response to GABA treatment were evaluated in cell lines. SD rats were randomly divided into 5 groups of 8 rats each, and GABA was orally administered; the groups were the normal control (NC), hypoxia-exposed (G0), as well as the GABA 1 mg/100 g body weight (BW) GABA treated group (G1), 5 mg/100 g BW GABA treated group (G5), and 10 mg/100 g BW GABA treated group (G10) with hypoxia. We analyzed EPO levels and red blood cell counts in rat blood and EPO gene expression in kidney tissue. EPO and VEGF mRNA levels in Hep3B cells exposed to hypoxia were significantly increased and further increased after GABA treatment. However, the expression of EPO-R and JAK-2 mRNAs were not affected by GABA, but hypoxia-induced HIF-1 and HIF-2 mRNA expression was inhibited by GABA. In the kidney tissue of rats exposed to hypoxia, the expression level of EPO mRNA was greatly increased, but levels in the GABA treatment groups significantly decreased. EPO levels in the serum showed the same significant trend, but the red blood cell counts were not significantly different. These findings demonstrate that HIF-1 and HIF-2 activation increase EPO expression in Hep3B cells exposed to hypoxia. However HIF decreased by GABA addition and VEGF increased significantly.
Molecular karyotyping was applied to Pneumocystis carinii (Pc) from two strains of experimental rats, Sprague Dawley(SD) and Fisher(F), in Korea. Field inversion gel electrophoresis and contour clamped homogeneous electric field electrophoresis resolved 15 chromosomal bands from the Pc. The size of the bands was estimated 270kb to 684kb from SD rats, and 273kb to 713 kb from F rats. The bands of 283 kb from SD rats and of 273 kb from F rats stained more brightly suggesting duplicated bands. Total number of chromosomes was at least 16, and total genomic size was estimated 7×106 bp. All of the bands from F rats hybridized to the probe of repeated DNA sequences of Pc and the band of 448 kb size was proved to contain rDNA sequences, but Pc. chromosome bands from SD rats showed no reactions to the probes. The 2 different karyotypes of p. carinii from 2 strains of rats were maintained consistently for 2 years.
Objective : This study was carried out to systemically investigate the effect of Joongwan(中脘; CV12) acupuncture in cerebrum and cerebellum of indomethacin-induced gastrointestinal disease in SD-rats. Method : 1. We induced gastrointestinal disease by indomethacin oral administration in SD-rats. 2. We selected Joongwan(中脘; CV12) acupuncture point that generally have been used to treat gastrointestinal disease. 3. We categorized SD-rats into three groups as followings. (1) Normal group : The group without any management (2) Control group : The group with indomethacin-induced gastrointestinal disease (3) Treated group : The group that Joongwan(中脘; CV12) acupuncture was performed after inducing gastrointestinal disease 4. We figured out the effect of acupuncture by analyzing staining degree of NADPH-diaphorase in cerebrum and cerebellum. Results : 1. Cerebrum (1) Normal group : The degree of staining was very low. (2) Control group : NADPH-diaphorase was mainly stained in cerebral cortex and the stained region was wider than Normal group. (3) Treated group : The degree and region of staining was higher and wider than the other goups. Sometimes the intensively stained regions were observed. 2. Cerebellum In both cases of Control group and Treated group, the regions in cortex were stained mainly. But, between Control group and Treated group, there was no remarkable difference. Conclusion : In case of cerebellum, there was no remarkable result. On the other hand, in case of cerebrum, there were certain differences among three groups. Through those results, we could conclude that Joongwan(中脘; CV12) acupuncture treatment was able to affect NADPH-diaphorase expression in the cerebrum of SD-rats that have gastrointestinal disease with indomethacin-inducing.

The aim of this study was to estimate time of biomineralization in developmental stages of rat lower incisors. Eruption length was measured. Four stages of incisor development were identified on histologic and microscopic computerized tomography (micro-CT) sections: (1) preodontoblast, (2) dentin matrix secretion, (3) enamel matrix secretion, and (4) enamel calcification. The overall eruption rate of the rat lower incisor was $600{\pm}70{\mu}m/day$ ($mean{\pm}SD$; n = 12). The length of the enamel secretion was $4.59{\pm}0.75mm$ in histologic section, was $3.64{\pm}0.63mm$ in radiographic section, which converts to $180.4{\pm}30.0hours$, $145{\pm}25hours$ respectively (n = 24). These findings suggested that the four biomineralizing developmental stages of the rat incisor took only several days. The significance of this animal study was to provide understanding for the rapid biomineralization process of developing rat tooth germ by analysis of tooth forming period.
This study was performed to evaluate the toxicity of polysaccharide-based Streptococcus pneumoniae vaccine in Specific Pathogen Free (SPF), Sprague-Dawley (SD) rats. S. pneumoniae vaccine was administrated subcutaneously each dose level of high (560 ${\mu}g$/rat), medium (280 ${\mu}g$/rat) and low (140 ${\mu}g$/rat) on days 0, 14, 28. The rats were observed for 2 weeks or 4 weeks after the final injection. During this test, there were no significant dose-dependent changes in body weight, water and food consumption. In urinalysis and serum chemistry, dose-related changes were not detected. In hematology, the percent of neutrophils and lymphocytes in white blood cells were changed significantly. According to the measurement of organ weight, only spleen weight was significantly increased in all groups of administration compared to the control group. In the histopathological examination, an antigen-deposit, vacuolated macrophages, infiltrated inflammatory cells and a formation of granulation tissue were observed at the site of an administration. These results are considered as an outcome by immune responses through a vaccination. Consequently, the results of this study demonstrated that S. pneumoniae vaccine has no toxicity when it was administrated subcutaneously three times in 2-week interval at a high dose of 560 ${\mu}g$/rat.
