Analysis of Geosmin and 2-MIB in Water by Stir Bar Sorptive Extraction (SBSE) and GC/MS (교반막대추출법(SBSE)과 GC/MS를 이용한 수중의 Geosmin과 2-MIB의 분석)

  • Musty and earth odors caused by geosmin and 2-MIB are the major complaints from the drinking water consumers. Therefore, early detection of geosmin and 2-MIB is the key to prevent off-flavor occurrence. In this study, a rapid method using stir bar sorptive extraction (SBSE) in combination with the GC/MS was investigated to analyze geosmin and 2-MIB in water. The SBSE method, a solventless extraction technique, was optimized and then applied to the analysis of geosmin and 2-MIB in water. The SBSE technique was found to be a simple and fast procedure that allows many samples to be extracted simultaneously using very small volume (10~20 mL). In addition, the SBSE method offered high recovery and good linear regression coefficient for the geosmin and 2-MIB. The good repeatability of this method can be deduced from the low RSD (7.2~7.6%) at 10 ng/L for geosmin and 2-MIB. The limit of detection was determined 1~2 ng/L and the limit of quantitation was 3~6 ng/L. Above all, the SBSE method proved to be a very practical technique for the analysis of geosmin and 2-MIB in water.

Analysis of E,E-farnesol and squalene in makgeolli using stir bar sorptive extraction coupled with gas chromatography-mass spectrometry (SBSE-GCMS를 이용한 막걸리 중의 E,E-farnesol과 squalene분석법)

  • The aim of this study was to establish an analytical method for the determination of E,E-farnesol and squalene in makgeolli, which is a traditional type of Korean fermented rice wine. E,E-farnesol and squalene in makgeolli were extracted using stir bar sorptive extraction (SBSE) coupled with gas chromatography-mass spectrometry. SBSE was found to be an effective method for analyzing the E,E-farnesol and squalene levels in makgeolli. The linear dynamic range of the SBSE method for detecting E,E-farnesol and squalene ranged from 0.5 to 200 ng/mL with $R^2=0.9974$ for E,E-farnesol and 100 to 50000 ng/mL with $R^2=0.9982$ for squalene. The limit of detection and the limit of quantification using the SBSE method were 0.1 and 0.5 ng/mL for E,E-farnesol and 15.0 and 40.0 ng/mL for squalene, respectively. The average recoveries obtained were, quantitatively, 101-107% for E,E-farnesol and 98-103% for squalene, respectively, supporting the accuracy of the SBSE-GCMS method.

  • Simultaneous determination of crop protection agents(CPAs) in food are done with multi-residue methods, which are composed of sample clean-up, concentration, chromatographic separation and detection. Stir Bar Sorptive Extraction(SBSE) technique is used for sample preparation of various analytes in several fields. The aim of this study was to develop a sensitive and fast method based on SBSE followed by thermal desorption - gas chromatography - mass spectrometry(TD - GC/MS) to determine CPAs in tobacco sample. For the analysis of tobacco sample prior to the SBSE method, solvent extraction or ultrasound-assisted solvent extraction was performed. methanol was used as the extraction solvent. The extract was then diluted with water. Finally, the sample was subjected to SBSE. A method for fast screening of crop protection agents in tobacco using SBSE-TD - GC/MS has been developed. About 17 CPAs including organochlorine, organophosphorous and others were identified and quantified. This method showed good linearity and high sensitivity for most of the target CPAs. The method was applied to the determination of CPAs at ng/mL levels in tobacco sample. This method is simple, rapid and may be applied in detection of other components.

Analysis of Synthetic Fragrances (SFs) in Water Using Stir Bar Sorptive Extraction (SBSE) and GC-MS/MS (교반막대 추출법과 GC-MS/MS를 이용한 수중의 합성 향물질류 분석)

  • A highly sensitive analytical method based on stir bar sorptive extraction (SBSE) technique and gas chromatography/tandem mass spectrometry (GC-MS/MS) has been developed, allowing the simultaneous multi-analyte determination of 11 synthetic fragrances (SFs) in water samples. The stir bar coated with polydimethylsiloxane (PDMS) was added to 40 mL of water sample at pH 3 and stirred at 1,100 rpm for 120 min. Other SBSE parameters (salt effect and presence of organic solvent) were optimised. The method shows good linearity (coefficients > 0.990) and reproducibility (RSD < 10.9%). The extraction efficiencies were above 83% for all the compounds. The limits of detections (LOD) and limits of quantification (LOQ) were 2.1~4.1 ng/L and 6.6~12.9 ng/L, respectively. The developed method offers the ability to detect 11 SFs at ultra-low concentration levels with only 40 mL of sample volume. Matrix effects in tap water, river water, wastewater treatment plant (WWTP) final effluent water and seawater were investigated and it was shown that the method is suitable for the analysis of trace level of 11 SFs. The method developed in the present study has the advantage of being rapid, simple, high-sensitive and both user and environmentally friendly.

  • Pyroligneous liquor as a byproduct from charcoal production of bamboo sprout produced damyang-gun has the broad benefits such as improvement of soil quality, plant growth control and is mainly used for the treatment of atopic dermatitis, fungi and many other skin diseases. In this study, flavor compounds of pyroligneous liquor from bamboo sprout produced in Damyang-gun were analyzed and compared using three different methods including direct analysis (DA), headspace solid phase microextraction (HS-SPME) and stir bar sorptive extraction (SBSE). Simultaneously, the analytical conditions of GC-MS for the determination of volatile compounds were optimized. Based on volatile organic compounds detected by GC-MS, SBSE and SPME methods showed higher sensitivity than direct analysis. Major compounds of pyroligneous liquor were cresol, guaiacol, p-ethyl guaiacol and syringol. These phenolic compounds are reported as the useful chemicals with medicinal activity.

Analysis of UV Filters in Water using Stir Bar Sorptive Extraction (SBSE) and GC/MS-MS (교반막대 추출법과 GC/MS-MS를 이용한 수중의 자외선 차단제 분석)

The developed method offers the ability to detect 8 UV filters at ultra-low concentration levels with only 40 mL of sample volume. Matrix effects in tap water, river water, wastewater treatment plant (WWTP) final effluent water and seawater were investigated and it was shown that the method is suitable for the analysis of trace level of 7 UV filters except of benzophenone (BP). The method developed in the present study has the advantage of being rapid, simple, high-sensitive and both user and environmentally friendly.

Chemical Characteristics and Flavors of Bamboo-shoot Vinegar (죽순식초의 화학적 특성 및 향기분석)

  • We assessed the quality of a bamboo shoot vinegar produced in Damyang-gun in terms of parameters that included acidity, mineral contents, amino acids, and flavonol aglycones. The flavor compounds of the bamboo shoot vinegar were also analyzed using the stir bar sorptive extraction (SBSE) method. The acidity of the bamboo shoot vinegar was 4.49%, which was the lowest value among the commercial vinegars studied. The bamboo shoot vinegar had a lower concentration of Na (8.36 mg/100 g) than the other commercial vinegars. There was a relatively large amount of tyrosine and lysine in the bamboo shoot vinegar. The concentration of quercetin, a flavonol aglycone, was 3.29 mg/100 g. The results of the flavor analysis showed that hexanal, 2-furancarboxaldehyde, and benzaldehyde were high in the bamboo shoot vinegar. Oleamide, a compound that is known to induce sleep, was first found in bamboo shoot vinegar using the SBSE method.

Comparison of the Activity and Proprioception of Trunk Muscles According to Different Types of Bridge Exercises in Subjects with and without Chronic Low Back Pain

  • Purpose: The purpose of the present study was to examine joint position senses and muscle activity in subjects with and without chronic low back pain and to determine the effects of different types of bridge exercises on their trunk muscle activity. Methods: Thrity-eight subjects with chronic low back pain and thrity healthy controls participated in the experiment. Joint position senses and trunk muscle activity levels were measured during the different bridge exercise methods. Results: The joint position senses of the healthy group and chronic low back pain group showed significant differences during lumbar flexion, lumbar extension, lumbar lateral flexion, and lumbar rotation. The muscle activity levels of the transversus abdominis (TrA), internal oblique (IO), and external oblique (EO) were highest in the prone bridge exercise (PBE) group, followed by the supine bridge swiss ball exercise (SBSE) group and supine bridge exercise (SBE) group in order of precedence. The muscle activity level of the erector spinae (ES) was highest in the SBSE group, followed by the SBE and PBE groups in order of precedence. Conclusion: Overall, the results suggest that chronic low back pain is associated with declines in joint position senses and that PBEs increase trunk muscle activity more than conventional bridge exercises.

The Comparison between Sabaek-san Water and 30% EtOH Extracts for Anti-inflammatory Effects (사백산 물 추출물과 30% EtOH 추출물의 항염증 효과 비교연구)

  • Sabaek-san has been used for the treatment of inflammatory diseases derived from the cold with high fever, cough, and lung dysfunction in Korea and China. There is no study for the comparison between different solvent extracts of Sabaek-san. We made two samples, one is Sabaek-san water extract (SBSW) and the other is Sabaek-san 30% EtOH extract (SBSE). Both extracts inhibited inducible nitric oxide synthase(iNOS) protein, reduced iNOS-derived nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Also, they reduced tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-$1{\beta}$ (IL-$1{\beta}$) production. These anti-inflammatory effects caused by induction of heme oxygenase (HO)-1. HO-1 enzyme plays an important role of cellular anti-oxidant and anti-inflammatory systems. The induction of HO-1 is primarily regulated at the transcriptional level, and its induction by various inducers is related to the nuclear transcription factor-E2-related factor 2 (Nrf2). However, it is worth taking note that SBSE has more powerful anti-inflammatory effects than SBSW. In this study we suggest that different solvent extraction makes different therapeutic actions.

A Clinical Report on Chicken Pox Scars Treatment with Sae-ssack Therapy (새싹테라피를 이용한 수두 흉터의 치료 16례)

  • Objectives: The purpose of this study was to evaluate the therapeutic efficacy of Sae-ssack(sprout) therapy on the face Chicken Pox scars. Methods: Sixteen patients had scars with varicella on the face were treated with Sae-ssack therapy. Sae-ssack therapy consists of a total of two treatments, a first-stage Sae-ssack acupuncture and a second-stage texture treatment(texture acupuncture and AMTS). The evaluation of the treatment effect was made by the medical staff who did not participate in the procedure, and the PGA(Physician's Global Assessment), SBSES(The Stony Brook Scar Evalustion Score) and patient satisfaction were used as the evaluation index. Results and Conclusions: According to the above indices, the Sae-ssack therapy was evaluated to be effective in the repair of atrophic scars caused by varicella. There were no visible or consistent side effects and patient satisfaction was also very high.
