Production of IL-6 and IL-8 in Human Fibroblasts Stimulated with Mycoplasma Lysates and Bacterial Toxins (세균독소와 Mycoplasma 항원으로 자극시킨 사람 섬유아세포의 Interleukin-6와 Interleukin-8 생성의 변화)

  • Cytokines are hormone-like proteins which mediate and regulate inflammatory and immune responses. The purpose of this study was to investigate the effect of lipopolysaccharide (LPS), Staphylococcus enterotoxin B (SEB), and Mycoplasma lysates on regulation of IL-6 and IL-8 production by human nasal fibroblasts. Primary cultured cells were incubated with LPS ($1.0\;{\mu}g/ml$) from E.coli, SEB ($1.0\;{\mu}g/ml$) from S.aureus, or Mycoplasma lysates (M.pneumoniae, Mp; M. fermentans, Mf; M. hominis, Mh, each $1.0\;{\mu}g/ml$). The culture supernatants were collected at 2, 6, and 24 hr and assessed for IL-6 and IL-8 production by enzyme-linked immunosorbent assay. The production of IL-6 in the culture supernatant was downregulated by LPS, SEB, or Mycoplasma lysates. But IL-6 was upregulated by mixed exposure with Mp+LPS (2 hr), Mp+LPS+SEB (24 hr), Mf+LPS (24 hr), Mf+LPS+SEB (2 hr), Mh+LPS (24 hr), Mh+SEB (24 hr), or Mh+LPS+SEB (24 hr). The production of IL-8 in the culture supernatant was similar to that of IL-6 by same stimulants. But IL-8 was upregulated by mixed exposure with Mf+LPS+SEB (2 hr), Mh+LPS (24 hr), Mh+ SEB (24 hr), or Mh+LPS+SEB (24 hr). These studies show that costimulation of LPS or SEB with Mycoplasma whole cell lysates upregulates the production of IL-6 and IL-8.

In-Vitro, Anti-Bacterial Activities of Aqueous Extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo Against Gram Positive and Gram Negative Bacteria

  • Objective: Evaluations of the in-vitro anti-bacterial activities of aqueous extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and Shilajita mumiyo against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) and gram-negative bacteria (Escherichia coli, klebsiella pneumoniae, Proteus mirabilis, Pseudomonas aeruginosa) are reasonable since these ethnomedicinal plants have been used in Persian folk medicine for treating skin diseases, venereal diseases, respiratory problems and nervous disorders for ages. Methods: The well diffusion method (KB testing) with a concentration of $250{\mu}g/disc$ was used for evaluating the minimal inhibitory concentrations (MIC). Maximum synergistic effects of different combinations of components were also observed. Results: A particular combination of Acacia catechu (L.F.) Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo extracts possesses an outstanding anti-bacterial activity. It's inhibiting effect on microorganisms is significant when compared to the control group (P<0.05). Staphylococcus aureus was the most sensitive microorganism. The highest anti-bacterial activity against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) or gram-negative bacteria (Escherichia coli, Klebsiella pneumonia, Proteus mirabilis, and Pseudomonas aeruginosa) was exerted by formula number 2 (table 1). Conclusion: The results reveal the presence of anti-bacterial activities of Acacia catechu, Castanea sativa husk, Ephedra sp. and Mumiyo against gram-positive and gram-negative bacteria. Synergistic effects in a combined formula, especially in formula number 2 (ASLAN$^{(R)}$) can lead to potential sources of new antiseptic agents for treatment of acute or chronic skin ulcers. These results considering the significant anti-bacterial effect of the present formulation, support ethnopharmacological uses against diarrheal and venereal diseases and demonstrate use of these plants to treat infectious diseases.

Descending Necrotizing Mediastinitis Secondary to Peritonsillar Abscess -A Case Report- (편도주위농양에 합병된 하행 괴사성 종격동염 -치험 1례-)

  • Descending necrotizing mediastinitis(DNM) is a rare complication of the oropharyngeal and cervical infection. Descending necrotizing mediastinitis requires an early and aggressive surgical approach to reduce the high morbidity and mortality associated with this disease. A 39-year-old man complained of odynophagia, neck swelling, and disturbance of swallowing with dyspnea. CT scans of the neck suggested a peritonsillar abscess and retropharyngeal and peripharyngeal abscess. He underwent cervical drainage. He remained febrile and complained of severe both pain in both shoulders. On postoperative day 5, a follow-up CT scan confirmed a mediastinal abscess. Reexploration of the neck and right thoracotomy for debridement and drainage of the mediastinal abscess were performed.. A large amount of pus was drained from the anterior and posterior mediastinum and its necrotic tissue was debrided. The patient's condition and radiologic findings gradually improved. Cultures of the drain fluid revealed Klebsiella pneumoniae. He was discharged on the 85th hospital day. In our experience, both transcervical drainage and aggressive mediastinal exploration via thoracotomy can lead to an improvement in the survival of the patient with descending necrotizing mediastinitis. CT scanning is useful for early diagnosis of mediastinitis and for follow up.

  • 최근 환경과 건강에 대한 관심이 증대되면서, 천연 섬유를 소재로 사용한 최종 제품 역시 고급화 및 고기능화되고 있는 추세이며 섬유항균가공분야에 있어서도 천연 추출물을 이용한 항균가공이 주목을 받고 있다. 현재 항균가공에 사용되고 있는 항균제는 일반적으로 합성항균제를 사용하고 있고 이는 거의 대부분 자극성 화합물로써 인체에 잠재적 유해요인이 될 수 있고 제조 및 가공과정에서 환경오염을 유발시키는 문제점도 발생되고 있다. 반면 천연추출물을 사용할 경우 합성물질에 비해 포름알데히드 검출 등의 인체 안정성의 불안이 적어 건강차원에서 그 인식이 점차 증대되고 있는 추세이다. 천연항균물질에는 chitosan이나 식물에서 추출된 천연항균물질 등이 있는데 특히 식물에서 추출된 천연항균물질은 피톤치드(phytoncide)라고 표현되고 있으며 이는 수목들이 해충이나 미생물 등으로부터 자기방어를 위해 공기 중으로 발산하는 방향성의 항생물질을 뜻하는 말이다. 본 연구에서는 식물 중에서 피톤치드 정유의 함량이 많다고 알려진 측백나무과의 편백나무 정유를 이용하였으며 편백나무 정유가 식물성 천연 오일형태이므로 가공 처리 시 물과 계면이 발생되기 때문에 HLB(Hydrophilic Lipophlic Balance) 조건에 따른 최적 유화조건을 선정하였으며 편백나무 정유를 이용한 면직물의 항균 기능 부여를 위한 시험으로 편백나무 정유 처리 농도에 따른 항균성을 평가해 보고자 하였다. 실험 결과 편백나무 정유는 HLB 15이상 계면활성제 사용 시 물에 용해성이 좋고 안정된 에멀젼 상태를 보였으며 제조된 편백나무 정유 가공액 5%이상 처리 시 Staphylococcus aureus과 Klebsiella pneumoniae의 99.9% 정균감소율을 나타내었다.

In Vitro Screening for Antimicrobial Activity of Chitosans and Chitooligosaccharides, Aiming at Potential Uses in Functional Textiles

  • Antimicrobial finishing of textiles has been found to be an economical way to prevent (or treat) skin disorders. Hence, this research effort was aimed at elucidating the relationship between the molecular weight (MW) of chitosan and its antimicrobial activity upon six dermal reference microorganisms, as well as the influence of the interactions with cotton fabrics on said activity. Using 3 chitosans with different MWs, as well as two chitooligosaccharide (COS) mixtures, a relevant antimicrobial effect was observed by 24 h for the six microorganisms tested; it was apparent that the antimicrobial effect is strongly dependent on the type of target microorganism and on the MW of chitosan - being higher for lower MW in the case of E. coli, K. pneumoniae, and P. aeruginosa, and the reverse in the case of both Gram-positive bacteria. Furthermore, a strong antifungal effect was detectable upon C. albicans, resembling the action over Gram-positive bacteria. Interactions with cotton fabric resulted in a loss of COS activity when compared with cultured media, relative to the effect over Gram-negative bacteria. However, no significant differences for the efficacy of all the 5 compounds were observed by 4 h. The three chitosans possessed a higher antimicrobial activity when impregnated onto the fabric, and presented a similar effect on both Gram-positive bacteria and yeast, in either matrix. Pseudomonas aeruginosa showed to be the most resistant microorganism to all five compounds.

A Case of Miliary Tuberculosis Misdiagnosed as Pneumonia and ARDS Due to the Transient Improvement after Intravenous Injection of Levofloxacin (폐렴, 급성호흡곤란증후군으로 오인되어 Levofloxacin 투여 후 호전되어 진단이 지연된 속립성 결핵 1예)

  • Miliary tuberculosis is quite a rare but serious cause of acute respiratory distress syndrome (ARDS). Therefore, the early detection of military tuberculosis as the underlying cause of ARDS is very important for the prognosis and survival of the patient. We report a case of military tuberculosis mimicking ARDS. A female patient was admitted due to repeated fever and dyspnea. The initial chest CT scan showed diffuse ground glass opacity, without a miliary pattern. The case was considered to be ARDS caused by pneumonia. She showed improvement after being treated with levofloxacin. However, she was re-admitted with fever seven days after discharge. The follow up chest CT scan showed micronodules in both lungs. An open lung biopsy confirmed the diagnosis of military tuberculosis.

Natural Dyeing Absorption Properties of Chitosan and Nano Silver Composite Non-Woven Fabrics -Focus on Chrysanthemum Indicum Linn- (키토산/나노실버 복합섬유 혼방 부직포의 천연염색 염착특성 -감국을 중심으로-)

  • This study examines the dyeability, light fastness, washing fastness, and the antibacterial activity of chitosan and nano silver composite non-woven fabrics dyed with an extracted solution from Chrysanthemum Indicum Linn. The results show that an increase in the chitosan and nano silver percentage resulted in an increase in the $a^*$ values and $b^*$ values; however, the $L^*$ values decreased in the undyed condition. ${\Delta}E$ values of chitosan and nano silver composite non-woven fabrics were higher than cotton 100% non-woven fabrics in the dyed condition with an extracted solution from Chrysanthemum Indicum Linn, and mordant treatments influenced the chrominance change. In the dyed condition with an extracted solution from Chrysanthemum Indicum Linn, an increase in the percentage of chitosan and nano silver resulted in an increase of the K/S values. The dyeability of chitosan and nano silver composite non-woven fabrics increased by mordant treatments. The light fastness and washing fastness of the mordanted non-woven fabrics were better than the non-mordanted. For the antibacterial activity, the bacterial reduction rate of chitosan and nano silver composite non-woven fabrics was 99.9% to Staphylococcus aureus and Klebsiella pneumoniae.

The Dyeability of Silk Fabrics with Sweet Potato Stem·Leaf Extract (고구마 줄기·잎 추출액을 이용한 견직물의 염색성)

  • The purpose of this study was to investigate the dyeability of silk fabrics with sweet potato stem leaf extract. To obtain the optimal dyeing conditions it was examined at various dyeing conditions(temperature, pH, time and bath ratio). The dyeability and the depths of shade which were evaluated in terms of K/S and CIELAB color difference values of the dyed and mordanted fabrics were also investigated. After dyeing, various color fastness(wash fastness, dry cleaning fastness, light fastness, rub fastness, and perspiration fastness) was measured and reviewed for UV protective, deodorant and antimicrobial functionality. The optimun output, as a result, was shown at 100% of dye concentration, $90^{\circ}C$ of dyeing temperature and 80 minutes of dyeing time while in terms of dye uptake depending on the kind of mordants and mordanting, it was found that among four mordants of $Alk(SO_4)_2$, $CuSO_4$, $SnCl_2$, and $FeSO_4$, post-mordanting with $SnCl_2$ showed the best results. Color fastness to dry cleaning, washing and rubbing was found strong at grade 4-5 and the grade to perspiration was as good as 3 while to light fastness was good at 4 as well. In aspects of functional properties, it showed excellent results of 98.3% UV protection rate and 88% deodorization rate. Antibacterial activity was 99.9% against staphylococcus aureus and 73.3% against klebisella pneumoniae. In conclusion, we validated that the dyestuffs from the disused sweet potato stem leaf extract would be useful as a natural dye material using the optimized conditions and dyeability for silk dyeing.

In Vitro and in Vivo Antibacterial Activities of a New Parenteral Cephalosporin, LB10522 (주사제용 세파로스포린계 항생제 LB10522의 in vitro 및 in vivo 항균력)

  • The in vitro antibacterial activities of LB10522, a new catechol-substituted cephalosporin, were compared with those of cefpirome, ceftazidime, ceftriaxone, and cefoperaz one against clinical isolates and laboratory standard anaerobes. LB10522 had broad spectrum antibacterial activities against both gram-positive and gram-negative microorganisms. It was most active against gram-positve bacteria among the reference cephalosporins tested. Against gram-negative strains such as the family Enterobacteriaceae, LB10522 showed an activity comparable to that of cefpirome. But LB10522 was more potent than ceftazidime, ceftriaxone and cefoperazone. In particular, Pseudomonas aeruginosa was highly susceptible to LB10522, which was 32-fold and 64-fold more active than ceftazidime and cefpirome, respectively. Against anaerobic strains, the activity of LB10522 was similar to those of reference compounds. LB10522 exhibited potent therapeutic activities against experimental local infections in mice. The therapeutic effect of LB10522 against urinary tract infection (UTI) caused by P. aeruginosa 1912E in mice was superior to that of cefpirome. Against experimental respiratory tract infection (RTI) caused by K. pneumoniae DT-S in mice, LB10522 was as effective as cefpirome. The in vivo efficacy of LB10522 was correlated well with its in vitro activity.

Antimicrobial Effects of a Bacteriocin Mixture from Lactic Acid Bacteria against Foodborne Pathogens (복합 박테리오신의 항균활성 및 축산식품 저장성 증진 효과)

  • The purpose of this study was to evaluate inhibitory activity of a bacteriocin mixture from lactic acid bacteria(LAB) against foodborne pathogens. Each bacteriocin solutions were prepared by growing nine strains of bacteriocin producers in MRS broth for 18~24 h followed by centrifugation(8000$\times$g, 20 min, 4$^{\circ}C$). Bacteriocins were purified from ammonium sulfate precipitation and were resuspended in 50 mM phosphate buffer(pH 7.0). Nine bacteriocins were mixed together and then allowed to freeze at -2$0^{\circ}C$. The mixture of nine bacteriocins showed enhanced inhibitory activity compared to each of bacteriocins and inhibited the Gram negative pathogens including Escherichia coli 0157:H7, Klebsiella pneumoniae, Pseudomonas chlororaphis and Shigella sonnei. The mixture of bacteriocin solutions was significantly lower than controls when a freeze-dried bacteriocin mixture was added to frank sausage, Mozzarella cheese and pork loin. With addition of bacteriocin mixture, total mesophilic bacteria in pork loin were constant over storage period, whereas total mesophilic bacteria in Mozzarella cheese and frank sausang slightly increased. Total viable cells of control group increased during storage without bacteriocin treatment. Volatile base nitrogen content of pork loin during storage also increased significantly without bacteriocin treatment. The bacteriocin mixture was capable of inhibiting pathogenic and spoilage microorganisms and extending the shelf-life of cheese and meat products during storage.
