소루쟁이 뿌리 추출물(Rumex crispus L.)의 라디칼 소거능과 항균력 평가 및 화장품 적용 가능성에 관한 연구 (A Study on Antioxidant and Antibacterial Efficacy Evaluation and Cosmetic Application of Rumex crispus L. Root Extract)

  • 본 연구는 소루쟁이 뿌리 추출물의 유효성 실험을 위해 항산화 활성 및 항균 효능을 확인하고 제형 안정성을 확인하였다. 항산화 활성으로는 DPPH radical scavenging, FRAP activity, ABTS+ radical scavenging, SOD-like activity를 진행하였으며 항균 활성 평가는 Staphylococcus epidermidis, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Candida albicans 균주에 대해 생육저해환과 최소저해농도를 평가하였다. 또한 소루쟁이 뿌리 추출물을 함유한 스킨을 21일 동안 pH, 온도, 일광에 대한 경시 변화를 확인하였다. 항산화 평가 결과 0.0625-1mg/mL 농도에서 농도 의존적으로 활성이 증가하는 것을 확인할 수 있었다. 생육저해환의 경우 100mg/mL 농도에서 각 균의 생육저해환이 10.45±0.34, 9.77±0.59, 9.92±0.22, 10.08±0.12로 대조군인 Methyl paraben에 비해 우수한 항균력을 확인할 수 있었고 최소저해농도의 경우 100mg/mL 농도에서 S. aureus, E. coli에 대한 항균력을 확인하였다. 스킨의 pH 농도가 4.0, 6.0, 7.0에서 흡광도의 변화가 미미하였고, 4℃, 25℃, 40℃에서 온도가 높아질수록 변색되는 것을 확인하였다. 또한 스킨을 일광과 실온에서 보관했을 때 일광에서 변색이 일어난 것을 보아 소루쟁이 뿌리 추출물을 함유한 화장품은 차광하여 저온 보관하면 변색을 방지 할 수 있을 것으로 추측된다. 본 연구 결과를 종합하였을 때 소루쟁이 뿌리 추출물은 항산화, 항균 활성을 기대할 수 있는 화장품 원료로 이용 가치가 높을 것으로 사료된다.

전도율측정법(傳導率測定法)(AHI Mastitis Detector)을 이용(利用)한 유방염진단(乳房炎診斷)의 야외적용시험(野外適用試驗) (Efficacy of the AHI Mastitis Detector for the Diagnosis of Bovine Mastitis)

  • A total of 136 dairy cows were subjected to test for bovine mastitis by AHI mastitis detector, microbiological test (MT) and California mastitis test (CMT). The results obtained were summarlized as follows: MT indicated that the most important causative microorganisms isolated from mastitic milk were Staphylococcus aureus (59.3%), Staphylococcus epidermidis (16.4%), Streptococcus agalactiae (12.4%) and Streoticoccus uberis (4.4%). In Holstein breed, the critical threshold of electrical conductivity values of normal and mastitic milk were found to be less than $6,900{\mu}S$ and above, $7,700{\mu}S$, respectively. Although there was goad agreement (92.0%) between AHI mastitis detector test and CMT for the diagnosis of bovine mastitis, the diagnostic efficiency of AHI mastitis detector (80.0%) was higher than that of CMT (74.0%) when compared with microbiological findings. In addition, handiness and objectivity of AHI mastitis detector for the detection of mastitic milk suggested that this could effectively be used for the diagnosis of both clinical and subclinical bovine mastitis in field.

제조방법에 따른 강하주의 품질 특성 (Quality Properties of Gangha-ju Liquor According to the Preparation Method)

  • This studies were performed to develop a Korean traditional folk liquor namely Gangha-ju has been prepared at Bosung district in Korea, and manufacturing conditions and anti-oxidation activity and anti-microbial activity of Gangha-ju were investigated. Ethyl-alcohol 20% and 30% Gangha-ju were brewed with glutinous rice wine, distilled liquor and 6 herbs of ginger, cinnamon, etc. Chemical and physical properties of 30% Gangha-ju were acidity 0.22, pH 4.31, amino acidity 3.26, transmittance 59 and conductivity $911\;{\mu}s/m$, and 20% Gangha-ju were 0.43, 4.20, 6.26, 62 and $924\;{\mu}s/m$. Volatile flavor compounds of ethyl alcohol, acetic acid, butanol, n-amyl alcohol, iso-pentyl alcohol, ethyl acetate, butyl acetate, acetaldehyde and furfural were detected, and main aroma compounds of Gangha-ju were isopentyl alcohol and ethyl acetate. Anti-oxidation activity by DPPH method was evaluated 31.32%, and nitrite scavenging effect was 31.79%. Anti-microbial activity against several microorganisms was pronounced strong activity over a wide range of test organisms, and Leuconostoc mesenteroids and Salmonella Ttyphimurium, Staphylococcus epidermidis were found to be more sensitive to Gangha-ju than Escherichia coli and Aspergillus flavus.

사간(射干) 에탄올추출물이 피부생리활성에 미치는 영향 (Effect of Ethanol Extract of Belamcandae Rhizoma on Dermal Bioactive Properties)

  • Objective : This experimental study was preformed to investigate the effect of ethanol extract of Belamcandae Rhizoma (EBR) on dermal bioactive properties related to cosmeceuticals such as radical scavenging activity and depigmentation. Methods : The free radical scavenging activity of EBR was assayed in cell free systems using 1,1-diphenyl-2-picrylhydrazyl (DPPH). Antibacterial potency was measured by the size of inhibition zone with change of volume. Cell viability was measured by MTT assay and tyrosinase activity was assessed using L-DOPA. Results : EBR showed slightly radical scavenging activity and protected against UVB-induced cell cytotoxicity. Futhermore, EBR showed antibacterial activities on S. epidermidis and S. aureus. Although the proliferation of B16/F10 cells was decreased by EBR, necrosis was not revealed. EBR augmented tyrosinase activity and dendrite outgrowth in B16/F10 cells. Conclusions : These results suggest that EBR has antioxidative & antibacterial activities, and stimulate melanogenesis.

Distribution of Coagulase-Negative Staphylococci and Antibiotic Resistance

  • Coagulase-negative staphylococci (CoNS) are a typical group of microorganisms, and the recent advances in laboratory technology and medicine has dramatically modified their significance in medical practice. CoNS, which were previously classified as normal bacterial flora, have recently been reported to be associated with serious infectious diseases, such as surgical wound infection or periprosthetic joint infection. Representative CoNS include Staphylococcus epidermidis, S. haemolyticus, and S. saprophyticus, which are known to cause serious problems in biomaterial-based and prosthetic device infections, as well as to cause simple urinary tract infections in sexually active women. Over the last decade, the clinical isolation rate of CoNS has been increasing, and antibiotic resistance has also been occurring. This review aimed to investigate the incidence of CoNS infection and to use the results as basic data for the management of CoNS, with a focus on the isolation rate and antibiotic resistance in clinical surgery.

Isolation and Identification of Staphylococcus sp. from Korean Fermented Fish Products

  • In order to find out if staphylococci occur in significant numbers in Korean fermented fish products, a total of 40 different fermented fish products were collected from different markets in Korea and analyzed for their physico-chemical and microbiological states. The pH, salt concentration and water activity of the products were measured and the total viable cell count and the number of Staphylococcus grown on mannitol salt agar were determined. The identification of the strains of Staphylococcus were made by API Staph Strip and MIS identification kits, and the physiological properties of the identified strains were further characterized by different conventional methods. The pH, salt content and water activity of fermented fish samples varied widely from 4.8 to 7.1, 7.4-28.7$%$ and 0.77-0.84, repectively, depending on the type of product. The total viable cell count varied from $10^4-10^9$ cfu/ml, and most of the samples had $10^5-10^6$ cfu/ml No correlation was found between the viable cell count and the pH, NaCl concentration and water activity of the samples. Among the 35 colonies identified as Staphylococcus strains by the identification kits, S. xylosus was the most frequently occurring strain marking 17, and S. warneri was 8, S. epidermidis 4 and S. cohnii 2. S. hominis, S. saprophyticus, S. haemolyticus and S. aureus were also identified once each. In some samples (K-3, P-6, K-8, G-5 and G-10), 2-3 different species of Staphylococcus were found. Considering the region of sampling, among the 10 samples from Kunsan 5 were identified as S. warneri, while in the other regions S. xylosus was predominant. Although the physiological characteristics of the identified strains were generally consistent with those in Bergey's Manual, some discrepances were also observed. All the strains were highly salt tolerant, growing in the media containing over 18$%$ NaCl. All the strains except S. aureus (G-11) showed negative in hemolysis activity, plasma coagulation and DNase tests. All the strains including S. aureus (G-11) showed negative in enterotoxin test.

소아 암 환자에서 발생한 균혈증 : 원인균과 항생제 감수성 (Bacteremia in Pediatric Cancer Patients : Causative Organisms and Antibiotic Sensitivities)

  • 목 적 : 면역 저하된 소아 암 환자에서 균혈증은 조기에 적절한 치료를 하지 못하면 이환율과 사망률이 높아 질 수 있는 질환으로서 호발 원인균의 발생 빈도 및 항생제에 대한 감수성은 지역적 시기적으로 변화하는 경향이 있으므로 이를 조사하여 경험적 항생제 선택에 도움을 주고자 하였다. 방 법 : 1995년 9월부터 2003년 8월까지 영남대학교 의과대학부속병원 소아과에 입원한 소아 암 환자를 대상으로 병력지와 혈액배양검사 결과를 조사하여 균혈증이 확진된 39명, 58례를 대상으로 원인 균의 종류 및 항생제 감수성을 조사 분석하였다. 결 과 : 소아 암 환자에서 균혈증의 발생빈도는 균혈증이 의심되어 혈액배양을 시행한 1,022례 중 58례로 5.7%였다. 균혈증을 일으킨 가장 흔한 원인 균은 그람양성균으로 총 37례(63.8%) 였고 그람음성균은 총 21례(36.2%)였다. 그람양성균의 경우 Staphylococcus epidermidis (22례, 37.9%), Staphylococcus aureus (10례, 17.3%), Streptococcus (5례, 8.6%) 순이었고, 그람음성균의 경우 Escherichia coli (7례, 12%), Enterobacter (4례, 6.9%), Klebsiella (4례, 6.9%), Serratia (2례, 3.5%), Acinetobacter (2례, 3.5%), Proteus (1례, 1.7%), Morganella morganii (1례, 1.7%) 순으로 나타났다. 항생제 감수성 검사에서는 그람양성균 37례 중 6례(16%)에서만 penicillin 감수성을 나타내었고, 15례(40%)에서 oxacillin 감수성을 나타내었다. Staphylococcus aureus는 1례를 제외한 모든 예에서 vancomycin에, Staphylococcus epidermidis는 1례를 제외한 모든 예에서 teicoplanin에 감수성을 나타내었다. 그람음성균의 경우, 21례 중 2례(10%)만이 cefotaxime에, 4례(19%)에서만 ceftazidime에 감수성을 나타내었다. Aminoglycoside에는 21례 중 6례(29%) 에서만 감수성을 나타내었으며, 21례 모두에서 imipenem에 감수성을, 2000년 이후부터 감수성 검사를 시행한 meropenem에도 모든 예에서 감수성을 나타내었다. 결 론: 균혈증이 의심되는 소아 암 환자에서 그람양성균에 의한 균혈증이 증가하는 점과 그람양성균과 그람음성균 모두 항생제 다제내성 빈도가 증가하고 있음을 유념하여 적절한 항생제를 선택해야 할 것이다.

젖염소 분방 유즙에서 분리한 세균 및 항균제 감수성 조사 (Prevalence of isolated microorganisms and antimicrobial susceptibility from half milk in dairy goats)

  • Samples of milk were collected from 425 halves of 216 dairy goats in Chonnam province over a period of January through August 2003. Bacterial isolation was carried out on those samples, and their antimicrobial susceptibility was tested. Bacteria were isolated from 166 milk samples (39.1%), either singly (74.7%) or in combination (25.3%). Of the 220 isolates, Staphylococcus spp. was the most prevalent (82.6%), followed by Streptococcus spp. (2.7%), Corynebacterium spp. (1.8%), Enterococcus spp. (1.8%), and Pasteurella spp. (1.8%). Of the 11 species identified from the 182 isolates of Staphylococcus spp., the most frequent species identified were S. epidermidis (28.6%) followed by S. chromogens (14.8%), S. haemolyticus (12.6%), S. aureus (12.1%), S. capitis (8.2%), S. lentus (8.2%), S. hyicus (4.4%), S. simulans (4.4%), S. caprae (2.8%), S. hominis (2.8%) and S. warneri (1.1%). Antimicrobial sensitivity test revealed that most isolates were highly susceptible to 11 antimicrobial agents (96.4 ~ 80.9%), while most isolates were resistant to penicillin.

준임상형(準臨床型) 유방염(乳房炎) 및 집합유(集合乳)의 Staphylococcus 속균(屬菌)에 대하여 (Studies on Staphylococci from Subclinical Mastitic and Bulk Milk Samples)

  • This paper deals with the incidence of bovine mastitis for 743 quarters and distribution of Staphylococci for the quarter and 70 bulk milk samples in the northern area of Gyeongbuk during the period from January to December 1984. Isolated Staphylococci were examined for species, subgroups, antibiotic resistance and penicillinase production. The results obtained were summarized as follows : A total of 25(73.5%) of 34 herds, 102(54.3%) of 188 cows and 208(30.3%) of 743 quarters were found to be infected with subclinical mastitis. A total of 83(83.1%) of 102 cows, 94(45.2%) of 208 mastitic quarters and 55(78.6%) of 70 bulk milk samples were isolated Staphylococci. Three hundred and eighteen strains of Staphylococci were classified into 11 species. Of these speoies, S. aureus from mastitis and S. sciuri from bulk milk were found most frequently, followed by S. epidermidis, S. simulans, S. cohnii, S. haemolyticus, S. xylosus, S. hyicus subsp. chromogenes, S. saprophyticus, S. warneri, S hyicus subsp. hyicus. Subgroups of catalase-positive and negative cocci were belonged most frequently to subgroup I, and subgroups III and III b, respectively. The method of Pelzer of al(97.8%) was more classified than that of Baird-Parker (68.5%). One hundred and sixty one strains(50.6%) of 318 Staphylococci isolates were resistance to one or more antibiotics such as ampicillin, chloramphenicol, gentamicin, kanamycin, streptomycin, and tetracycline. Isolates from subclinical mastitis were more resistant to antibiotics than its from bulk milk. Of the 318 Staphylococci Isolates, 128(40.3%) gave positive reaction for the penicillinase test, all of ampicillin resistance strains produced this emzyme.

사람 모발에 부착되어 있는 세균의 동정 및 항생제 감수성 (Identification of Bacterial Strains Adhered to Human Scalp Hair and Antimicrobial Susceptibility)

  • 사람의 모발에 부착되어 있는 세균을 분리 동정한 후 이 세균들의 항생제 감수성을 확인하였다. 중환자실에 입원해 있는 환자와 건강인의 모발에서 39개의 세균을 분리한 결과, Staphylococcus epidermidis가 19주로 가장 많았으며 그 다음으로 S. aureus가 14주, S. waneri가 5주, S. pasteuri가 1주 순으로 나타났다. Amikacin, ampicillin, bacitracin, carbenicilline, cefazolin , cefoperazone , chloramphenicol, erythromycin, gentamicin, methicillin, nalidixic acid, neomycin, oxacillin, penicillin, streptomycin, tetracycline, vancomycin에 대한 내성을 디스크 확산법으로 확인하였다. 항생제에 내성을 나타내는 세균들은 모두 입원환자의 모발로부터 분리된 것들이었다. 사람의 모발에 부착된 세균을 제거하기 위해 시중에 유통되고 있는 모발 세정제와 계면활성제인 SDS를 처리하였으나 제거효과가 없었다. 이러한 결과는 사람의 모발에 부착되어 있는 세균들이 병원 등에서 교차 감염원이 될 수 있는 가능성을 보여주며, 모발에 부착되어 있는 세균 소독의 중요성을 확인시켜주었다.
