Study on the Retarding Mechanism and Strength Loss of Gypsum from Hydrolyzed Wheat Protein Retarder

  • In this article, the influence of a hydrolyzed wheat protein retarder on the hydration process, ion concentration in liquid phase, degree of supersaturation, and crystal morphology of plaster was investigated. Furthermore, the retarding mechanism and the strength loss of gypsum were also studied by scanning electron microscopy (SEM). The results indicate that the use of the hydrolyzed wheat protein retarder for plaster achieved a better retarding effect and lower strength loss. The combination of gypsum plaster with the retarder not only decreased the plaster's early hydration rate and prolonged its setting time efficiently, but also militated against the crystal morphology of dihydrate gypsum. For example, the crystal dimensions changed little, but the proportion of needle-shaped crystals decreased. Combination with calcium ions on the surface of dihydrate gypsum crystal nuclei may form a chemisorbed layer, reduce the surface energy of the crystal nuclei, and inhibit the growth of the crystal nuclei of dihydrate gypsum. Consequently, the hydration process of building gypsum becomes greatly extended and is slowed down significantly.

Islet function within a multilayer microcapsule and efficacy of angiogenic protein delivery in an omentum pouch graft

  • We have previously described a new multilayer alginate microcapsule system, and the goals of the present study were to assess the in vitro function of islets encapsulated in its inner layer, and the angiogenic ability of FGF-1 delivered from the external layer in an omentum pouch. Following isolation and culture, islets were encapsulated in the inner core of microspheres ($500-600{\mu}m$ in diameter) with a semi-permeable poly-L-ornithine (PLO) membrane separating two alginate layers, and both unencapsulated and encapsulated islet function was assessed by a dynamic glucose perifusion. For angiogenesis experiments, one group of microcapsules without FGF-1 (control) and another (test) containing FGF-1 with heparin encapsulated in the external layer were made. One hundred microcapsules of each group were transplanted in Lewis rats (n = 5/group) and were retrieved after 14 days for assessment of angiogenesis. Glucose perifusion of unencapsulated and encapsulated islets resulted in similar stimulation indices. The release of FGF-1 resulted in increased vascular density compared to controls. In conclusion, islets encapsulated in the core of multilayer alginate microcapsules maintain functionality and the microcapsule's external layer is effective in delivery of FGF-1 to enhance graft neovascularization in a retrievable omentum pouch.

Efficiency of Essential oil about the Skintroubles induced Surfactants - Palmarosa, Neroli essential oil - (계면활성제 유발된 피부장애의 아로마 에션셜 오일의 유효성 연구 - Palmarosa, Neroli essential oil을 중심으로 -)

  • The effectiveness of Palmarosa and Neroli essential oil on dry skin of rat induced by kitchen detergent are investigated. The experimental groups were divided the control group (C), group treated with surfactant (A1), group treated with Palmarosa (A2), group treated with Neroli (A3), group treated with Palmarosa and Neroli (A4). The protein analysis of all experimental groups was performed with SDS polyacrylamide gel electrophoresis and observation of epidermis and the alteration of mast cell were performed with photomicroscope. According to the protein analysis, the A3 group treated with Neroli essential oil was appeared the most similar with the control group. And then the A4 group treated with Palmarosa + Neroli essential oil was appeared the most similar with the control group. According to the results of morphologic view with keratin layer, the keratin layer's breakaway resulting from Palmarosa essential oil, the keratin layer's restoration resulting from Neroli essential oil was appeared. And then the structure of the epidermal layer was preserved by hyperkeratosis reaction. In photomicrosopic obersevation of mast cell to examine the inflammatory reactions, the increase in size and number of mast cell were showed in A1 group treated with surfactant compared to the control group (C). The number of mast cells definitely decreased in groups (A3, A4) which were treated with Neroli essential oil.

Proteomic Analysis of Global Changes in Protein Expression During Exposure of Gamma Radiation in Bacillus sp. HKG 112 Isolated from Saline Soil

  • A Gram-positive bacterium was isolated from the saline soils of Jangpura (U.P.), India, and showed high-level of radiation-resistant property and survived upto 12.5 kGy dose of gamma radiation. The 16S rDNA sequence of this strain was examined, identified as Bacillus sp. strain HKG 112, and was submitted to the NCBI GenBank (Accession No. GQ925432). The mechanism of radiation resistance and gene level expression were examined by proteomic analysis of whole-cell extract. Two proteins, 38 kDa and 86.5 kDa excised from SDS-PAGE, which showed more significant changes after radiation exposure, were identified by MALDI-TOF as being flagellin and S-layer protein, respectively. Twenty selected 2-DE protein spots from the crude extracts of Bacillus sp. HKG 112, excised from 2- DE, were identified by liquid chromatography mass spectrometry (LC-MS) out of which 16 spots showed significant changes after radiation exposure and might be responsible for the radiation resistance property. Our results suggest that the different responses of some genes under radiation for the expression of radiation-dependent proteins could contribute to a physiological advantage and would be a significant initial step towards a fullsystem understanding of the radiation stress protection mechanisms of bacteria in different environments.

Effects of Feeding Dried Leftover Food on Productivity of Laying Hens

  • This experiment was conducted to investigate the effects of different levels of dried leftover food (DLF) in the diet on feed utilization and egg-laying performance of hens. One hundred sixty-eight, 18 week old Tetra brown commercial layers, were assigned to 7 treatments in a completely randomized design. Each treatment has four replications per treatment with six animals per replication. All the experimental animals were fed diets for 7 weeks. The treatments included 1) control group without DLF, 2) diet with 10% DLF, 3) diet with 20% DLF, 4) diet with 30% DLF, 5) 10% higher protein level of diet with 10% DLF, 6) 20% higher protein level of diet with 20% DLF and 7) 30% higher protein level of diet with 30% DLF. Average daily feed intake (ADFI) tended to be improved with DLF feeding. ADFI of group fed diets with 20% was significantly higher than that of control (p<0.05). Feed conversions of DLFfed groups were higher than that of control. Egg production tended to be higher in groups fed diets with 10% DLF than control diet without significant differences (p>0.05). However, those of groups fed diets containing 20 and 30% DLF were lower than that of control. Supplementing protein source to DLF-containing diets improved egg production (p<0.05). Increasing level of DLF in the diet for layer decreased egg weight and egg mass compared to control without significant differences (p>0.05). Protein supplementation to DLF-containing diets increased egg mass without significant difference (p>0.05). The range of egg cholesterol concentration of DLF-fed groups was 11.94-14.10 mg/g while that of control group was 12.31 mg/g although there was no significant difference among treatments (p>0.05).

Photoprotective Effects of Silybum marianum Extract (흰무늬엉겅퀴 열매 추출물의 자외선에 대한 피부 보호 효과)

  • Ultraviolet rays (UV) cause photoaging by inducing skin photodamages such as erythema and sunburn. Silymarin is a mixture of antioxidant polyphenols extracted from Silybum marianum fruit (S. m), which is known as milk thistle. It is known to protect skin tissues from UV treatment and antioxidant effects. In this study, we aimed to identify the photoprotective effects of S. m extract, which has silymarin in the epidermis layer of the skin. We found that the extract can function as a UV filter, so it can reduce DNA damage by UV treatment. Especially, we found that, in the stratum corneum, the extract can suppress the protein carbonylarion and DNA damages caused by suberythemal dose of UV treatment which does not induce erythema in the skin. UV treatment also increased protein carbonylation levels in the stratum corneum by oxidation, but it was prevented by applying the extract. The extract can absorb UV with minimal phototoxicity. Together, our study suggests that S. m extract can be used as a photo-protective ingredient to avoid photoaging of the skin.

Effects of Corn Distiller's Dried Grains with Solubles on Production Performance, Nutrient Digestibilities and Fecal Ammonia Emission in Laying Hens (옥수수주정박이 산란계의 생산성, 영양소이용율 및 분 암모니아 방출에 미치는 효과)

  • A layer feeding trial was conducted to investigate the effects of high quality corn distiller's dried grains with solubles(DDGS) on laying performance and nutrient metabolizabilities. A total of 216 Hy-line Brown layers, 23-wk of age, were employed in a 10-wk feeding trial consisting of three dietary treatments(0%, 10%, and 20% DDGS), and six replicates per treatment. All experimental diets were prepared as iso-protein(17%) and iso-calorie(2,780 kcal/kg). At the end of the trial, the effect of DDGS on fecal ammonia emission was also estimated. The use of DDGS up to 20% in layer diets did not affect the feed intake, laying rate, egg weight, and feed conversion ratio(P>0.05). The DM metabolizability decreased significantly(P<0.05) by the DDGS supplementation(P<0.05). The NFE metabolizability decreased gradually as the level of DDGS increased(P<0.05). The fecal ammonia emission from DDGS-fed birds tended to decrease as the storage days extended. In conclusion, albeit DDGS has some unfavorable effects on DM and NFE metabolizabilities, it could be included in layer diets up to 20% level without any harmful effects on laying performance.

  • This study was designed to estimate effect of wood vinegar on meat quality in old Layer which was low production and low economic value. The old layer chickens were used to resource for income increase of layer farm and protein supply. Two hundred forty Isa Brown chickens were assigned with control(Broiler feed), Treatment 1 (Broiler feed with addition of domestic wood vinegar $0.8\%$) and Treatment 2(Broiler feed with addition of foreign wood vinegar $0.4\%$). Feed intake of control group was lower 3g than other treatments. This is the reason of result that what organic acid in wood vinegar of treatment groups affect to feed digestibility, The different of chicken production was depended on ingredient and included value of wood vinegar in domestic and foreign. Foreign wood vinegar have organic acids which that functionally act sexual Pheromone so that promote endocrine matter. This is reason that old Layer chicken increase to egg production rate. Treatment 2 group effect to thigh meat rather than breast meat and improve to value of crude fat and meat color. The Meat quality and sensory test of thigh meat of foreign wood vinegar was better appraise rather than that of domestic wood vinegar. Because foreign wood vinegar make to fine of meat tissue and to decline cooking loss and also to increase juiciness and tenderness on sensory test.

Anti-oxidant and Anti-inflammation Activity of Fractions from Aster glehni Fr. Schm. (섬쑥부쟁이(Aster glehni Fr. Schm.) 분획물의 항산화 및 항염증 활성)

  • The Plants and their extracts containing polyphenol have been shown to be associated with decreased the cause of aging and variety of disease such as reaction oxygen species (ROS) and reactive nitrogen species (RNS) in several recent studies. We conducted to investigate whether the extracts and fractionation isolated from Aster glehni Fr. Schm. has an inhibitory effect association with oxidation or inflammation. The Aster glehni Fr. Schm. 70% aq. MeOH was fractioned according to polarity with n-hexane layer, EtOAc layer, n-BuOH layer and water layer. The electron donating ability of EtOAc, n-BuOH solvent fraction from Aster glehni Fr. Schm. was about 58.0%, 46.4% at $50\;{\mu}g/mL$, respectively. The superoxide anion radical inhibitory effect of EtOAc extracts was about 64.65% at $50\;{\mu}g/mL$, and n-BuOH extracts was 35.66% at $50\;{\mu}g/mL$. EtOAc layer to the inhibition activity of hyaluronidase and lipoxygenase were inhibited about 24.37%, 29.5% at $5\;{\mu}g/mL$. In the anti-inflammation effect of EtOAc layer inhibited the generation of nitric oxide. also, these results showed that EtOAc extract inhibited 81.5% at $50\;{\mu}g/mL$ on the expressions of iNOS protein in Raw264.7 cell line.

Hierarchical Architecture of Multilayer Perceptrons for Performance Improvement (다층퍼셉트론의 계층적 구조를 통한 성능향상)

  • Based on the theoretical results that multi-layer feedforward neural networks with enough hidden nodes are universal approximators, we usually use three-layer MLP's(multi-layer perceptrons) consisted of input, hidden, and output layers for many application problems. However, this conventional three-layer architecture of MLP shows poor generalization performance in some applications, which are complex with various features in an input vector. For the performance improvement, this paper proposes a hierarchical architecture of MLP especially when each part of inputs has a special information. That is, one input vector is divided into sub-vectors and each sub-vector is presented to a separate MLP. These lower-level MLPs are connected to a higher-level MLP, which has a role to do a final decision. The proposed method is verified through the simulation of protein disorder prediction problem.
