The composition of biochemical components such as lipids, proteins and their amino acid components and inorganic elements in the ashes in unossified antlers from Cervus nippon Temminck var. mantchuricus grown in Korea were analyzed to obtain fundamental data for quality control. As a result, it was found that total lipids were 20.75% which was approximately similar contents with those of proteins (21.8%). Sixteen amino acids were identified from the hydrolysate of the protein fraction. Three gangliosides with very similar TLC patterns of those such as $GM_3$, $GM_1$ and $GM_{1a}$ were identified from the water soluble layer of Folch's partitions. Ash contents were revelaed to be much higher in the sponge layer (40.0%) than in the velvet layer (3.7%).

To investigate the expression patterns of proteins and growth factor signals in differentiated rabbit embryonic stem (ES) cells, ES cells with confluent stage grown of feeder layer and differentiated cells into embryoid bodies (EB) without feeder cell were applied to protein gel and Western blotting analysis. There were 66kDa and 28kDa specifically expressed in differentiated ES cell but not in undifferentiated ES cell while 25kDa protein band showed up in only undifferentiated ES cells. Also there were some difference of protein bands in several area of gel between differentiated and undifferentiated ES cells such as about 100 kDa, 50kDa and 27kDa areas, but there was no difference in band pattern of one-dimensional gel analysis between mouse ES cells and rabbit ES cells. IGF-I receptor and EGF receptor were expressed in differentiated cells and undifferentiated cells. And ICF-I and EGF were not expressed in both differentiated and undifferentiated cells. These results indicated that ES cells express their own proteins to inhibit differentiation while EB cells synthesize different proteins to differentiate, and 16F-I receptor and EGF receptor were expressed in both ES and EB cells probably for the different functions.

Cytokeratin 19 (CK19) expressed in epidermis of skin, bulge region of hair follicle, outermost layer of outer root sheath and proximal and distal to bulge. Vimentin is a fibrous protein that localized in cytoplasm of fibroblast and forms cytoskeleton to maintain shape of cell and nucleus. In this study, CK19 and vimentin in skin were confirmed with light, fluorescence and transmission electron microscope. As a result, CK19 was localized epidermis, hair follicles, outer root sheath and nucleus of Merkel's cell. However, vimentin was localized some epidermis, dermis, hypodermis and nucleus of Merkel's cell. The role of CK19 is self-renewal and homeostasis in skin. Also, hair follicle regeneration and hair growth is known to be related. It is supposed that required of structural proteins that make up cytoskeleton is increased. Thereby, expression of CK19 is increased. It is considered that vimentin localized in order to stabilize structure of cell and cytoskeleton of fibroblasts. Also, CK19 and vimentin present in nuclei of Merkel's cell, and to act as a fibrous protein that make up end of a nerve fiber present in Merkel's cell and paracrine function of Merkel's cell.
Objective: The aim of this study is to elucidate the effects of transforming growth factor-${\beta}$ (TGF-${\beta}$)1 and L-ascorbic acid on proteoglycan synthesis, and the relationship between Sox9, proteoglycan, and TGF-${\beta}1$ in intervertebral disc cells. Methods: Human intervertebral disc tissue was sequentially digested to 0.2% pronase and 0.025% collagenase in DMEM/F-12 media and extracted cells were cultured in $37^{\circ}C$, 5% $CO_2$ incubator. When intervertebral disc cells were cultured with TGF-${\beta}1$ or L-ascorbic acid, the production level of sulfated glycosaminoglycan (sGAG) was estimated by dimethyl methyleneblue (DMMB) assay. The changes of Sox9 mRNA and protein levels via TGF-${\beta}1$ were detected by RT-PCR and Western blot analysis in each. Results: The amount of sGAG was increased with the lapse of time during incubation, and sGAG content of pellet cultured cells was much larger than monolayer culture. When primary cultured intervertebral disc cells in monolayer and pellet cultures were treated by TGF-${\beta}1$ 20 ng, sGAG content of experimental group was increased significantly compared to control group in both cultures. L-Ascorbic acid of serial concentrations (50-300 ug/ml) increased sGAG content of mono layer cultured intervertebral disc cells significantly in statistics. The co-treatment of TGF-${\beta}1$ and L-ascorbic acid increased more sGAG production than respective treatment. After treating with TGF-${\beta}1$, Sox9 mRNA and protein expression rates were significantly increased in disc cells compared with the control group. Conclusion: This study suggests that TGF-${\beta}1$ would increase sulfated glycosaminoglycan (sGAG) and other proteoglycans such as versican by elevating Sox9 mRNA and protein expressions in order.
The present was undertaken to establish a model for the control of adipocytes differentiation by using antibody from egg yolk. The emulsion of membrane protein of 3T3L-1 cell membrane protein with the complete Freund's adjuvant was firstly immunized in layer. Second and third boosting were undertaken with two weeks intervals by injection of the emulsion of the same antigen with the incomplete Freund's adjuvant. After 4 week of the first immunization, eggs were collected and antibody (IgY) was purified from egg yolk. The purity of IgY was 60-98% determined by single radial immunodiffusion (SRID) methods. Titer value of the antibody showed high reactiviy for the preadipocytes membrane protein measured by ELISA. When the IgY was added in the test media containing either 2.5% porcine serum or 10% FBS(control), the differentiation of 3T3L-1 cells and Glycerol-3-phosphate dehydrogenase(GPDH) activities was significantly decreased compared to the control cells(p〈0.05). When mice were subcutaneously injected with IgY raised against membrane protein of 3T3L-1 cells for 3 weeks, adipose tissue mass around ovary was tended to be decreased in female mice compared to those of control mice. It is suggested that a potential for manipulating of lipid accumulation through decrease in 3T3L-1 cell differentiation and fat accumulation in female mice by IgY treatment.
An In vitro protein sulfation in the soluble fraction of rat brain was charaderized further by an improved method of alkaline hydrolysis and thin layer ceflulose electrophoresis TLE) The protein sulfation was carried out in a reaction system containing [35 S] 3'-phosphoadenosine-5'-phosphosulfate (PAPS), Tris-maleate buffer (pH 8), MgCI$_2$, and soluble proteins from rat brain. The sulfated proteins were precipitated by acetone and alkaline hydrolysis was performed to obtain sulfated amino acids. The hydrolysate was separated further by TLE and the separated residues were identified by fluorography. The Iluorography of one-dimensional The showed at least nine sulfated residues including tryosine-O-sulfate. The other spots were not identified yet positively. General properties of protein sulfotransferases (PST) using this method were re-examined such as effects of concentrations of PAPS, pH, incubation temperature and $Mg^2$+. These results suggest a possible occurrence of several PST corresponding to each sulfated residue in rat brain and that the sulfation can occur not only in tyrosine but also in other residues as well.

Allergic contact dermatitis is a common skin disease resulting from specific immumologic sensitization to topically applied various allergen. Purpose of this study was to investigate skin morphologic chganges by light microscopic, changes of protein band by SDS-PAGE in the skin treated on the surfactant. Furthermore, lectin histochemistry is used to know the chagnges of the terminal sugar of the glycoconjugate in the skin treated on the surfactant. The results were as follows : The 1 day of treated group was indicated the enlargement of capillary, the 3 days of treated group was showed that the cytoplasm was eosinophlic by the pyknotic of nucleus. The 6 days of treated group was observed hyperkeratinoid, increased of inflammation cells to epithelium. The 9 days of treated group was appeares that serum crust was sheded and formed a acanthosis. But dermis layer was cytoplasmic vaculation and enlargement of intercellular space. The 12 days treated group made up new epidermis layer of seven layer and of observed an decreased in quantity of inflammation cells. The mast cell of degranulated type was increased in treated surfactnat. It was secreating granules hold histamin, serotonin and heparin. Accordingly, protein band of electrophortic phase was observed a sudden changes since 3 days treated group. At observeation of the cell-surface glycoconjugates, LCA, PNA, SBA and WGA lectin positive cells but inducated lectin negative cells in Con-A. Furthermore, PNA and SBA showed stroger positive reaction as treated surfactant group.
Cage layer waste and sudex grass were ensiled in the proportions of 0:100, 30:70, 40:60 and 50:50 wet basis, respectively. The influence of ensiling cage layer waste on Salmonellae, Shigella, Proteus, and total number of colony forming units (CFU) was investigated. The nutritive value of the silages was evaluated in a digestion trial. The experiment was conducted with 24 wethers allotted to four silages. Initial samples of cage layer waste showed $0.11{\times}10^6$ CFU and salmonellae, Shigella and Proteus were present. Ensiling was effective in complete elimination of all the pathogens. Dry matter, crude protein and ash contents were increased linearly (p < .01) with the increase of cage layer waste in the silages ($358g{\cdot}kg^{-1}$ to $484g{\cdot}kg^{-1}$; $76.3g{\cdot}kg^{-1}$ to $183.2g{\cdot}kg^{-1}$ and $38.5g{\cdot}kg^{-1}$ to $169.4g{\cdot}kg^{-1}$; DM basis, respectively). Water solubles carbohydrate values for silages were 38.3, 22.5, 20.1 and $20.0g{\cdot}kg^{-1}$ DM basis, respectively. Ensiling decreased the pH values for all the silages and the decrease was higher for sudex grass ensiled alone than grass ensiled with cage layer waste. Lactic acid concentration in silages increased linearly (p < .01) with the increase of cage layer waste. The apparent digestibilities of DM, OM and CP for the animals fed sudex grass silage alone were 496.0, 516.1, $496.7g{\cdot}kg^{-1}$ DM basis respectively. However, digestibilities of all the components were higher (p < 0.01) for the animals fed silages containing cage layer waste. Among waste containing diets, the digestibilities of all the components showed quadratic affect (p < 0.01), increased for silages containing 30 and 40% cage layer waste and decreased when the level of waste increased from 40 to 50%. The results indicated that cage layer waste can be used upto 40% in ruminant diet as a source of N without any adverse effect on the health of animals. Ensiling appeared to be feasible and effective method for eliminating the pathogen present in cage layer waste.
In this experiment 5 Korean varieties and 3 U.S.A. varieties of wheat were investigated for their moisture content, ash content, crude protein content and extraction rate. The results are summarized as follows; 1. In the case of U.S.A. wheat, the ash content is directly proportional to the content of protein but the extraction rate is negatively correlated to the content of ash. 2. In the case of korean wheat, there is no proportional tendency as in the U.S.A. wheat. It might be cause by the low extraction rate where the endosperm is hardly separated from the bran layer at the same conditions of temering process. 3. The concentration of protein particles in the endospern, is centrifugal in the U.S.A. wheats, but in the Korean varieties the direction is oppositional to the U.S.A. varieties. 4. Yooksung #3, Cahngkwang, Youngkwang and Hard winter wheat are adapted to the production of all purpose flour or noodle flour; Noring #4, Jinkwang and Softwhite wheat are suitable for the process of cake flour; and Dark northern spring is just adaped to the production of bread flour.
The morphological and molecular characteristics of Pachydictyon coriaceum (Holmes) Okamura (1899) are described. Plants are collected from Korea all year round and have maximum height from August to September. The monthly variability of thallus growth is in the way with that of the seawater temperature. Two types of thallus structures, thick cortical layer tallus type and thin cortical cell layer type, are distinguished according to growing seasons. The habit of Korean plants is also classified into two thallus types, slender type and wide type, based on the length and the width of internodes, but this distinction between two types is not supported by either anatomical or molecular characteristics. P. coriaceum shares typical morphology in branching pattern and morphogenetic processes with the other species of Dictyota: 1) multi-cellular cortical and medullar layer in the partial of thallus, 2) same development of thallus from apical meristem cell, and 3) sub-lineage within Dictyota species lineage in rbcL, psaA and psbA gene sequences analyses. These characteristics lead to propose the new combination of Dictyota coriacea (Homes) I.K. Hwang, H.S. Kim et W.J. Lee, comb. nov.
