In this research, microporous and bicontinuous poly vinylidene fluoride(PVDF) hollow fiber membranes were prepared via hybrid process of the thermally induced phase separation (TIPS) and stretching method. The mechanism of the membrane preparation is based on liquid-liquid phase separation. The final membranes have characteristic structures which have both bicontinuous and fibrillar morphology by applying the stretching method. They showed quite different structure when compared with the spherulitic or nodular structure from S/L TIPS and bicontinuous structure from L/L TIPS. At first, PVDF hollow fiber precursors were prepared via TIPS method using various kind of diluent mixtures. We used gamma-butyrolacton, dimethyl phthalate (DMP), diethyl phthalate (DEP), dibutyl phthalate (DBP) as diluents. We could make hollow fiber membranes which had porous outer surface or dense outer surface by controling the parameters such as cooling conditions, PVDF concentration and the ratio of diluent mixtures. Finally, these hollow fiber were stretched at room temperature and diluents were extracted by ethanol. Effects of the stretching ratio on the membrane morphology were investigated using scanning electron microscope (SEM), and its effects on water flux, porosity, pore size, roughness and tensile strength were examined.
Shoot proliferation was obtained from shoot tips and nodal explants of Vitex negundo L. on MS medium supplemented with either BAP or KIN (0.1-2.0 mg/L) alone or in combination with NAA (0.1 mg/L). The concentrations of cytokinins combined with NAA produced multiple shoots from shoot tips and nodal explants. The highest mean percentage (84.3$\pm$8.0) of shoot multiplication's were observed on nodal explants in the presence of BAP (1.5 mg/L) and NAA (0.1 mg/L) followed by shoot tips (65.0$\pm$5.0). The regenerated shootlets were rooted on MS basal medium IAA, IBA, NAA (0.1-1.5 mg/L). The maximum number of roots (51.0$\pm$2.6) was achieved on the medium containing IBA (1.0 mg/L) followed by other auxins (NAA, IAA). The regenerated plants were successfully transferred to a mixture of vermiculate and soil. About 95% of the plantlets survived when transferred to the field.

An experimental investigation of the machining characteristics such as cutt- ing resistance, surface roughness and tool wear in turning the test pieces of SM45C steel with both coated and uncoated carbide tool tips under various cutting conditions was conducted. Also a specially designed simple vibration damping device was experimentally evaluated for its effectiveness on machined surface roughness and a vibration test was conducted to confirm its ability to reduce the amplitude. Based on these tests finding, the following conclusions are made; 1. The cutting resistance($\textrm{p}_{1}$) increases as the depth of cut(d) increases at fixed feed rate(f) over the cutting speed(v) range of 43-226 m/min and p decreses about 18% average when V is increased for fixed d and f. At V= 226m/min, $\textrm{p}_{1}$/for A, C tips are about the same level but $\textrm{p}_{1}$ for B tip is 15% less than A, C tips. 2. The specific cutting resistance(Ks) at V=226 m/min was derived for A, B, C tips respectively and the value of Ks for B rip is about 20% less than A, C tips. 3. The surface roughness(Ra) improves significantly as the cutting speed(V) is increased and this effect was greater when V>100 m/min. On the other hand, Ra deteriorates as the feed rate(f) is increased and this trend was accelerated when f>0.3 mm/rev. With regard to the difference of Ra values among A, B, C tips, at V=226m/min, d=0.4mm, and f=0.31-0.61mm/rev, Ra values for B.C tips are about 17% less than tip A. 4. The experimental tool wear equations were derived for A, B, C tips and from these equations, the tool life($\textrm{T}_{\textrm{L}}$) baced on the I.S.O. criteria was calculated to be $\textrm{T}_{\textrm{L}}$<$\textrm{T}_{\textrm{LB}}$<$\textrm{T}_{\textrm{LC}}$ for both flank wear($\textrm{V}_{\textrm{B}}$) and boundary wear($\textrm{V}_{\textrm{N}}$). Hence, the coated tips are superior to the uncoated tip and tip C is considered to be the best. 5. The cutting resistance may be slightly reduced and the surface rounghness improved when the damper is used especially when V>100 m/min. Therefore this damping device is considered to be effective and practical. The experimental surface roughness equations were also derived. Based on the vibration test, it is established that the surface roughness improvement was the result of amplitude reduction made possible by the damper.
The effect of alachlor treatment on peroxidase synthesis in oat root tips was studied. Alachlor caused increase in the amount of soluble peroxidase in oat root tips, peroxidase activity increase as the rate of alachlor application increased, Alachlor treatment of oats with $1{\times}10^{-6}M$, peroxidase activity increased 0.20 unit higher than that of nontreatment. After 12hr, 65mM of peroxide treatment of oats inhibited 16% root growth, and 130 mM peroxide treatment caused 59% inhibition. With PAGE of peroxidase extracted from normal root tips, PAGE give 4 species($P_2$, $P_3$, $P_4$, and $P_5$ band) of peroxidase. Alachlor significantly activated isoperoxidase. Three isoperoxidase($P_1$, $P_6$, and $P_7$) are synthesized at a increased concentration of alachlor, SDS-PAGE analysis of proteins extracted from oat root tips showed that they were made up of subunits blow 100 kD polypeptide.
Sweetpotato shoot tops (leaves, tips and petioles) are known to be very useful parts as vegetables because of their high nutritive values and great biomass yield. In this study, the phenolic compound contents, antibacterial activity, mutagenic activity, and antimutagenic activity were investigated in sweetpotato tips that were 10-15cm of shoot top including stems, petioles and tender leaves after sprout of storage roots. The study was done by extracting sweetpotato tips with 80% ethanol and the ethanol fraction was re-extracted with hexane, chloroform, ethyl acetate, butanol and water. In ethyl acetate and butanol fractions, total phenolic compounds contained 95. 6mg/g extract and 69.3 mg/g extract, respectively, The antibacterial activity was measured using the paper disk method with concentrations of 1, 2, 5 and 10 mg/disk of butanol and ethyl acetate fractions against L. monocytogenes and S. Typhimurium strains. Higher doses of solvent extracts showed the higher antibacterial activities. In addition, 5, 10 and 20 mg/mL of the extracts were tested to determine the antibacterial activity in liquid culture. The sweetpotato leaf extract by ethyl acetate showed 1 log reduction compared to control after 24 hrs on Listeria monocytogenes, but 20 mg/ml of butanol extract completely inhibited the growth of the pathogen after 12 hrs. The extracts from ethyl acetate or butanol on Salmonella Typhimurium did less than 1 log reduction during cultivation compared to control. The numbers of S. Typhimirium TA98 and TA100 revertant colonies were 29-33 and 159-188 CFU/plate, respectively, indicating that solvent extracts were no mutagenic activity. The antimutagenic test was performed by adding direct mutagen 2-NF and MMS, and butanol and ethyl acetate showed antimutagenic effect. Thus, this study showed that sweetpotato tips had high phenolic contents and both antimicrobiol and antimutagenic properties. Sweetpotato tips would be good nutritive source because of their high nutrient content without any toxicity in consuming.

Organic solvent resistant hollow fiber membranes were fabricated via a thermally induced phase separation (TIPS) method using Polyketone polymer, a material with excellent resistance to organic solvents. The PEG300, DMSO2 and Glycerine called the "green solvents" were used as diluents for TIPS method. The spherulite structure was formed with DMSO2 by S-L phase separation behavior whereas the bicontinuous structures were formed with PEG300 and Glycerine, respectively. The morphology of the PK hollow fiber membranes was investigated using SEM. The pure water permeability and the durability test were conducted to understand the permeation properties of PK hollow fiber membranes. The tensile strength test was conducted for the property of mechanical strength. In this study, the fabrication of PK hollow fiber membranes with various diluents was discussed to understand the correlation between diluent and polymer in detail.
Beet curly top virus (BCTV) mutant has been constructed in vitro that contain G-to-T transversions at nucleotide 2727 within overlapping open reading frames (ORFs) L1 and L4. The mutations introduce termination codon in ORF L4 without affecting the amino acid encoded by ORF L1. When agroinoculated into Arabidopsis thaliana the mutant caused mild stunting and stem curling, but not the callus induction and hyperlasia on infected tissues of Sei-O ecotype. However, this mutant was not infectious on Col-O. Levels of single stranded DNA forms were similar in mutant and wild type BCTV infections. The DNA quantitation data showed that the DNA of BCTV-L4 mutant virus was accumulated in shoot tips, infection origin and roots with similar levels to those of wild type virus infected. Three tissues of asymptomatic ecotype Col-O also had as much as virus DNA from wild type virus infections. In both ecotypes infected with BCTV-Logan and BCTV-L4 mutant, root tissues contained more virus DNA than any other tissues by the Southern hybridization data. The results suggest that ORF L4 encodes a functional protein that is a major determinant of pathogenesis that might affect the hyperplastic response of the host to BCTV infection.
Objective of this study was to select ectomycorrhizal fungi for black spruce(Picea mariana) inoculation to overcome the growth inhibitory effects of Kalmia angustifolia. Nineteen isolates representing 11 species of ectomycorrhizal fungi were tested for their abilities to grow and form mycorrhizae with black spruce seedlings in the presence of water leachate of leaves of Kalmia. Mycelium growth of 9 isolates were inhibited by the leaf leachate. Colony diameter and biomass of the other 10 isolates were either increased or unaffected under the same conditions. Acidic pH of the culture medium(pH 3 and 4) inhibited some of the fungi, but a combination of acidic pH and the leaf leachate was more inhibitory. Thirteen isolates were able to form ectomycorrhizae with black spruce in presence of 25% leaf leachate in pure culture. Four isolates, Paxillus involutus(NF4), Cenococcum geophilum(GB12), Laccaria laccata(GB23), and E-strain(GB45) formed mycorrhizae more successfully than the others in presence of up to 50% Kalmia leaf leachate. Black spruce seedlings pre-inoculated with these fungi were grown with Kalmia leaf leachate and live Kalmia plants during a four month greenhouse experiment. Abundant mycorrhizae(77-91% of root tips) were developed on seedlings pre-inoculated with P. involutus, L. laccata and E-strain but relatively poor mycorrhization(32% of root tips) resulted with C. geophidum. Over 90% of the short root mycorrhizae were attributed to the inoculated fungi although indigenous mycorrhizae also occurred on most seedlings. Persistence of the mycorrhizae was not affected by living Kalmia plants. Over 80% of the mycorrhizae on seedlings inoculated with P. involutus, L. laccata and E-strain and 53% of the mycorrhizae on seedlings inoculated with C. geophilum were attributable to the inoculant fungi. Control seedlings formed about 45% ectomycorrhizal short roots with indigenous fungi. The L. laccata and C. geophilum inoculated seedlings exhibited enhanced mycorrhizae formation in presence of Kalmia leaf leachate. Mycorrhizae formation with inoculant fungi was 4-15% lower at pH 4 than at pH 5, with the greatest inhibition occurring for L. laccata. Seedlings inoculated with P. involutus had the greatest shoot and root growth followed by L. laccata and E-strain inoculated seedlings. The P. involutus and L. laccata inoculated seedlings were significantly taller with more shoot dry biomass than the uninoculated(control) seedlings. E-strain inoculated seedlings had significantly higher shoot dry biomass and significantly lower number of first order lateral roots compared to the control but other growth parameters such as height, root dry weight and number of short root tips were not significantly different from the control. Seedlings inoculated with C. geophilum were not significantly different from the uninoculated seedlings in any of the growth parameters except for the number of first artier lateral roots which was significantly less than the control seedlings.
A protocol has been developed for rapid large scale clonal propagation of an aromatic endangered medicinal plant, Hemidesmus indicus R. Br. with the elimination of the problems such as premature leaf fall and callus formation during caulogenesis and rhizogenesis. Multiple shoots were induced from shoot tip and nodal explants on Murashige and Skoog (MS) medium supplemented with 1 mg/L Benzylaminopurine (BAP) and 0.5 mg/L Napthaleneaceticacid (NAA). Addition of 15 mg/L adenine sulphate to the above medium checked leaf abscission completely, reduced the time required for caulogenesis and restored morphogenetic potential after several subcultures. The in vitro grown propagules were rooted in 1/2 MS medium supplemented with 2 mg/L Indolebutyric acid (IBA) +1 mg/L NAA and sucrose 0.7% (w/v). Addition of charcoal at 100 mg/L to the rooting medium quickened root initiation with a complete check on callus formation. The effect of sucrose concentration on both caulogenesis and rhizogenesis was also studied. The resultant plantlets were acclimatized and grown in fields where ninety eight percent of the rooted shoots survived and grew normally. The estimation of the secondary metabolite content in the shoots of the regenerated plant and the mother plant indicated that the concentration of the three secondary metabolites lupeol, vanillin and rutin was similar.
In order to micropropagate uniform plantlets of Philodendron wend-imbe, the shoot tips were cultured on media supplemented with 0.5-10.0mg/L BA or 0.01-1.0 mg/L thidiazuron (TDZ). The multi-bud clusters from basal part of shoots formed vigorously on media containing 5.0-10.0 mg/L BA or 0.05-0.1 mg/L TDZ. Shoot formation from the bud cluster sections (5-7mm) was achieved favorably on medium with 5.0mg/L BA and 20 g/L sucrose. Lowering of sucrose in medium to 20 g/L was effective for the inhibition of callus growth from basal part of shoots. Growth of shoots and their rooting were favorable on media containing 1.0-2.0 mg/L IBA or 0.1mg/L NAA. The rooted plantlets were acclimatizated effectively in soil mixed with perlite 1: peat moss 1 or peat moss alone.
