Development and evaluation of protective capacity of Salmonella Enteritidis polyphosphate kinase-deleted and temperature-sensitive mutant (Salmonella enterica serovars Enteritidis의 온도감수성 변이주 및 폴리인산키나아제 변이주의 제작과 방어효과)

  • This study was focusing on evaluating the protection of polyphosphate kinase (ppk) deleted and/or temperature-sensitive (ts) Salmonella Enteritidis (SE) as an attenuated vaccine in chickens. We constructed SEppk, SEts and SEppk::ts mutants and screened those mutants by growth capability in vitro, protection study in mice model and antibody response in chickens. Among the mutants, SEppk::ts-3 was selected because it showed higher growth capability, good protection against highly virulent SE in mice model, and good antibody response in chickens. SEppk::ts-3 also showed good protection against highly virulent SE isolate because it decreased colonization of virulent SE challenge strain in spleen, liver and cecum compared with the non-vaccinated control. The SEppk::ts-3 mutant showed cross-protection against S. Gallinarum (SG) challenge although the its cross-protection rate was a little lower than that of SG9R, a commercial vaccine against SG infection. To use for live attenuated vaccine in chickens, it should further be characterized.

Isolation, Identification and Production of Salmonella Pullorum Coloured Antigen in Bangladesh for the Rapid Whole Blood Test

  • Postmortem examination was conducted on 350 (three hundred and fifty) chickens. Related samples (Liver, heart, ovary, spleen, bone-marrow, and caecal junction) were collected. The appropriate materials from the samples were cultured into different media. A total 40(forty) isolates of salmonella pullorum and S. gallinarum were identified and preserved. Characterization of the isolates were done by cultural, morphological, biochemical, and serological tests. Salmonella pullorum antigen was prepared from the local isolate, standardized and tested. This antigen was used in the field for the detection of pullorum or fowl typhoid infection or carrier birds. The antigen consisted of suspension of Salmonella pullorum in 0.50 percent sodium chloride plus 1.5 percent sodium sulfate and inactivated with 1% formalin U.S.P. and standardized with McFarland scale iv or by pour plate method containing 800 million organisms per milliliter and stained by the addition of alcoholic crystal violet. Sterility, safety and potency were tested and found as good as other international antigens. The antigen was found to retain its quality for six months when preserved at room temperatures. The test was made by mixing one drop of the antigen with a drop of blood or a drop of serum, on a glass plate or white tile. The locally produced antigen was as good as antigens from Japan, Hungary, Holland and India. A serological study was conducted with the locally prepared antigen in different farms, and the incidence was 0-4% in government farms, 5-10% in commercial imported breeds and 0-3% in cross breed local farms respectively.

Antimicrobial Resistance Patterns of Enterococci spp. Isolated from Raw Milk Samples (원유시료에서 분리한 장구균속 세균의 항생제 내성 양상)

  • From April 2008 to January 2009, a total 458 raw milk samples were randomly collected from 15 stock raising farms located in northern area of Kyunggi province and cultured for the presence of Enterococci spp. A total 170 enterococcal isolates were recovered from the raw milk samples. Enterococcus faecalis was predominant species recovered (64.7%), followed by E. faecium (18.8%), E. avium (5.9%), E. gallinarum (5.9%) and E. durans (4.7%). Antimicrobial resistance patterns of 170 Enterococci spp. against ampicillin, erythromycin, tetracyclin, chloramphenicol, vancomycin, ciprofloxacin and streptomycin were tested. According to the result, they showed high level resistance to erythromycin and streptomycin (82,9% and 93,5% respectively), moderately resistance to ampicillin, chloramphenicol and tetracyclin (50%, 45.9% and 32%, respectively) but fortunately, vancomycin and ciprofloxacin are still effective against this species.

The Effects of the Transgenic Aspergillus oryzae Supplementation on Performance, Egg Quality and Intestinal Microflora of Layers (형질 전환 Aspergillus oryzae의 첨가가 산란계의 생산성, 계란 품질 및 장내 미생물 변화에 미치는 영향)

  • An experiment was conducted to investigate the dietary effects of a transgenic Aspergillus oryzae(AO) culture on the performance, egg quality and intestinal microflora of layers. A total of 840 Hy-line Brown layers of 39wks old were assigned to one of the following 7 dietary treatments: control(C), C+0.2% AO culture, C+0.5% AO culture, C+0.2% transgenic AO culture, C+0.5% transgenic AO culture, C+0.2% transgenic mutant AO culture, and C+0.5% transgenic mutant AO culture. The transgenic AO was made by inserting Salmonella gallinarum gene to AO. And the transgenic mutant AO was made by inserting Salmonella gallinarum gene to mutant AO which was mutated by UV irradiation. Each treatment was replicated six times with 20 birds housed in 2 bird cage. Twenty birds units were arranged according to completely randomized block design. Feeding trial lasted for 8wks under 16 hour lighting regimen. Laying performance and egg quality were significantly(P<0.05) affected by the treatments. Transgenic AO culture supplementation at the level of 0.2% significantly increased egg production, while its egg weight was significantly decreased compared to that of the control. Feed intake and feed conversion ratio(FCR) were not significantly different among the AO treatments and the control. The eggshell strength of the AO treatments was significantly higher than that of the control. Transgenic mutant AO culture supplemented at the level of 0.5% significantly increased egg yolk color. Intestinal microflora were significantly(P<0.05) affected by the treatments. The cfu of Lactobacilli spp. significantly increased and those of Salmonella species and E. coli decreased in the AO treatments. The transgenic AO and transgenic mutant AO culture were more effective than the AO culture in reducing the cfu of Salmonella species and E. coli. It is concluded that supplementation of the transgenic AO culture at the level of 0.2% could be recommended for the improvement of egg production. Supplementation of transgenic AO or transgenic mutant AO culture at 0.2% level effectively controlled intestinal Salmonella species population.

Antibiotic Resistances of Enterococcus Isolated from Salad and Sprout (샐러드와 새싹채소에서 Enterococcus의 분리와 항생제 내성특성)

  • To evaluate the antibiotic resistance of Enterococcus from salad and sprout, Enterococcus were isolated and identified from 47 salad samples and 37 sprout samples, and then their antibiotic resistances were analyzed. Ninety five Enterococcus, 41 strains from salad and 54 strains from sprout, were ultimately isolated. The frequent Enterococcus in salad and sprout were E. gallinarum, E. faecalis, E. faecium, E. hirae, and E. avium. Minimum inhibitory concentrations of the isolates for vancomycin were below $4{\mu}g/mL$, which were not high levels of resistance. All Enterococcus proved to be resistant to streptomycin and chloramphenicol. Twenty two percentage of the isolates were resistant to penicillin, however, almost the isolates were sensitive to tetracycline. Eighteen percentage of the isolates were resistant to erythromycin. All E. faecium and E. faecalis were found to be ampicillin-resistant, and seven E. faecalis and five E. faecium were resistant to rifampicin. Overall antibiotic resistances of Enterococcus isolates were relatively low and low resistance to vancomycin was similar to those evidenced by Enterococcus isolated from the other foods. Therefore, there may be no special risk from the antibiotics resistances of Enterococcus and especially vancomycin-resistant Enterococcus from the fresh-cut salads and the sprouts.

White Rice Fermented by Clostridium butyricum IDCC 9207 as an Alternative to Antibiotic having Antibacterial and Immunostimulatory Activity (항생제 대체제로서 Clostridium butyricum IDCC 9207 백미 발효물의 항균작용과 장관 면역증진효과)

  • The antagonistic activities against animal entero-pathogenic bacteria were investigated with 444 natural substances fermented by various probiotics. A white rice product fermented (FWR) by Clostridium butyricum IDCC 9207 with a high growth inhibition of Salmonella typhimurium KCTC 2054 and Escherichia coli O157:H7 was selected. Also, a FWR was shown to suppress 8 among 21 pathogenic bacteria. In a mouse model with salmonella (${\times}10^9$ CFU/mouse) infection, 5 samples (200 ${\mu}{\ell}$/mouse/day) were fed to mice (n = 25) for 18 days. A fermented white rice containing C. butyricum IDCC 9207 (FWRCb9207) among 5 samples significantly inhibited the growth of salmonella, while in the control group (PBS, tetracycline) the number of salmonella increased. And the treatment with FWRCb9207 was found to increase the secretory immunoglobulin A (sIgA) level in the feces of salmonella-infected mice. The results obtained in this study suggest that a FWRCb9207 might be utilized as a feed additive in pigs and poultry diets.

Strain Selection and Optimization of Mixed Culture Conditions for Lactobacillus pentosus K1-23 with Antibacterial Activity and Aureobasidium pullulans NRRL 58012 Producing Immune-Enhancing β-Glucan

  • Lactobacillus pentosus K1-23 was selected from among 25 lactic acid bacterial strains owing to its high inhibitory activity against several pathogenic bacteria, including Escherichia coli, Salmonella typhimurium, S. gallinarum, Staphylococcus aureus, Pseudomonas aeruginosa, Clostridium perfringens, and Listeria monocytogenes. Additionally, among 13 strains of Aureobasidium spp., A. pullulans NRRL 58012 was shown to produce the highest amount of ${\beta}$-glucan ($15.45{\pm}0.07%$) and was selected. Next, the optimal conditions for a solid-phase mixed culture with these two different microorganisms (one bacterium and one yeast) were determined. The optimal inoculum sizes for L. pentosus and A. pullulans were 1% and 5%, respectively. The appropriate inoculation time for L. pentosus K1-23 was 3 days after the inoculation of A. pullulans to initiate fermentation. The addition of 0.5% corn steep powder and 0.1% $FeSO_4$ to the basal medium resulted in the increased production of lactic acid bacterial cells and ${\beta}$-glucan. The following optimal conditions for solid-phase mixed culture were also statistically determined by using the response surface method: $37.84^{\circ}C$, pH 5.25, moisture content of 60.82%, and culture time of 6.08 days for L. pentosus; and $24.11^{\circ}C$, pH 5.65, moisture content of 60.08%, and culture time of 5.71 days for A. pullulans. Using the predicted optimal conditions, the experimental production values of L. pentosus cells and ${\beta}$-glucan were $3.15{\pm}0.10{\times}10^8CFU/g$ and $13.41{\pm}0.04%$, respectively. This mixed culture may function as a highly efficient antibiotic substitute based on the combined action of its anti-pathogenic bacterial and immune-enhancing activities.

Studies of the egg drop laying diseases from the mass zone layer (산란계 밀집지역의 산란저하성 질병에 관한 연구)

  • Newcastle disease (ND), infectious bronchitis (IB), low pathogenic avian Influenza (LPAI) and fowl typhoid (FT) have been known as egg drop laying diseases because of the serious layer damage from mass zone layer. In this study, such egg drop laying diseases were investigated. To access this study, we peformed to evaluate antibody titers in serum and isolated bacteria and virus from organs and feces on May, July and September in 2003. The distribution of ND from January to May, IB and LPAI from October to February of the next year, and FT from March to September were inspected by the question survey in 21 farms. ND revealed to be positive rates of 490 to 474 $(96.7\%)$ in May, 510 to 506 $(99.2\%)$ in July and 510 to 510 $(100\%)$ in September with hemagglutination inhibition (HI) test. The mean antibody titers were 10.2, 9.9 and 10.2, respectively. With regard to IB, 484 out of 490 samples $(98.7\%)$ in May, 508 of 510 $(99.6\%)$ in July and 509 of 510 $(99.8\%)$ in September showed positive results and the mean antibody titers were gradually increased with 8.2, 9.0 and 9.4, respectively. According to HI test of LPAI, the positive results were shown in 442 of 480 $(92.1\%)$, 394 of 494 $(79.8\%)$ and 402 of 483 $(83.2\%)$ in May, July and September, respectively The mean antibody titers were decreased with 4.6, 4.3 and 4.0. The distribution of LPAI also elicited the positive rates of 480 to 475 $(99.0\%)$ in May, 494 to 485$(98.2\%)$ in July, 483 to 472 $(97.7\%)$ in September as determined by ELISA and the mean S/P ratio were 2.319, 2.557 and 2.380, respectively. Compared ELISA results with HI test of LPAI the positive results were 480 to 422 $(92.1\%),\;475(99.0\%),\;494\;to\;394 (79.8\%),\;485 (98.2\%)\;and\;483\;to\;402(83.2\%),\;472(97.7\%)$. Therefore, the positive rate determined by ELISA was higher than that of HI test with 6.9, 18.4 and $14.5\%$, respectively. When performed RT-PCR for ND using organ and feces samples, the pathotypes were detected $5(15.6\%)\;in\;May,\; 2(5.3\%) in\;July,\;2(7.1\%)$ in September but there is no samples showing positive band for LPAI. In attempt to isolate Salmonella gallinarum, bacteria were obtained from 4 cases (12.5%) in May, 9 (23.6%) in July, 5 (17.8%) in September. Thus the highest rate for isolation revealed to be shown in July When evaluated the antimicrobial susceptibility to 18 isolated strains of 5. gallinarum, bacteria were sensitive to trimethoprim/sulfamethox$(61.1\%),\;kanamycin\;(55.5\%),\;ampicillin\;(55.5\%)$ and amoxacillin/clavulanic acid $(55.5\%)$, cephalothin $(50.0\%)$, but resistant to penicillin $(88.9\%)$, streptomycin $(88.9\%)$, erythromycin $(83_4\%)$ and tetracycline $(61.1\%)$. According to HI test of ND and LPAI using captured 164 wild Korean tree sparrows (Passer nontanus), the positive rates were $47.6\%\;and\;57.3\%$, and the mean HI titers were 5.32 and 4.02, respectively. 71 $(43.2\%)\;and\;58(35.3\%)$ in captured sparrows also showed more than 4 titers for HI test to ND and LPAI, respectively However, the attempt for isolation of viruses failed in all samples.

Evaluation of zinc oxide and copper oxide nanoparticles as potential alternatives to antibiotics for managing fowl typhoid in broilers

  • Antimicrobial resistance poses challenges to humans and animals, especially to the poultry sector in control of fowl typhoid with antibiotics, leading to increased mortality and food insecurity. Therefore, it is essential to develop more effective medications as alternatives to antibiotics. Currently, zinc oxide and copper oxide nanoparticles are of such significant interest due to their antibacterial properties. This study aimed to evaluate antimicrobial activity of zinc oxide and copper oxide nanoparticles against fowl typhoid in broilers. Ninety broiler chicks were raised under suitable management conditions. On day 10 of age, chicks were divided into six groups: control negative, control positive, T1, T2, T3, and T4. On day 19 of age, chicks in all groups except control negative were infected with Salmonella gallinarum (0.2 mL, 108 CFU/mL). After appearance of clinical signs, the treatments (Florfenicol; 50 mg/L drinking water [T1], and zinc oxide + copper oxide nanoparticles; 25 + 10 mg/kg/d [T2], 37.5 + 15 mg/kg/d [T3], and 50 + 20 mg/kg/d [T4]) were administered to chicks. Chicks were sacrificed on 26th and 30th day of age, and samples of blood and tissue were obtained. Hematological analysis with gross and histopathological examination of spleen, thymus and bursa of Fabricius was performed. Results revealed that there was no visible congestion in spleen and thymus of T3 and T4 at 11th day post infection. Antibody level against new castle's disease and lymphoproliferative response showed no significant difference in all groups. However, phagocytic response in nanoparticles treated groups exhibited a notable (p < 0.01) distinction compared to control positive. Notably, T3 demonstrated the highest level of phagocytic activity. Hematological parameters, including lymphocytes, heterophils, eosinophils, and heterophils/lymphocytes ratio in groups T2, T3, and T4, indicated significant (p < 0.01) difference compared to control positive. However, lymphocytes, heterophils, and heterophils/lymphocytes ratio in groups T2, T3, and T4 showed no significant difference when compared to T1. Nanoparticle treated groups showed decreased (p < 0.01) congestion of spleen and thymus as compared to control positive. Overall, zinc oxide and copper oxide nanoparticles have potential to serve as an alternative to florfenicol in treatment of fowl typhoid.

Formation of Biogenic Amines by Lactobacillus plantarum Isolated from Makgeolli (막걸리에서 분리한 Lactobacillus plantarum의 biogenic amine 생성능)

  • We examined biogenic amine (BA) production as well as the diversity of bacterial flora in 11 types of commercial makgeolli stored at 4 and $20^{\circ}C$. Moreover, we studied the BA-producing activity of three L. plantarum strains isolated from makgeolli. At $20^{\circ}C$, the BA content was highly increased and the denatured DNA bands were more variable in non-sterilized makgeolli compared to sterilized makgeolli. The major BAs produced in commercial makgeolli were histamine and putrescine. Histamine, tyramine, putrescine, and cadaverine were produced in excess by inoculation of the three L. plantarum isolates to makgeolli stored at $20^{\circ}C$ for 21 days. These results suggest that some L. plantarum strains in makgeolli can produce different types of BAs, depending on the extent of degradation of makgeolli.
