Orthostatic Hypotension and Cognitive Function in Parkinson's Disease (파킨슨병에서 기립저혈압과 인지 기능의 관계)

  • Background: Although orthostatic hypotension (OH) and cognitive impairment (CI) are common non- motor symptoms of Parkinson's disease (PD), the relationship between OH and CI remains to be clarified. This study was aimed to investigate the relationship between OH and CI in PD. Methods: We recruited 192 patients who were diagnosed as PD based on the UK Brain Bank diagnostic criteria. The Hoehn & Yahr stages were ranged I to III and patients underwent extensive clinical evaluation, including brain magnetic resonance imaging (MRI) for cerebral white matter hyperintensity (WMH), tilt table test, the Korean version of Montreal Cognitive Assessments and the Korean version of Mini-Mental Status Examination in one month from the first clinic visit. The participants were divided into two groups according to the presence of OH (OH+ vs. OH-) and cognitive function (cognitive normal, CN vs. CI), respectively. Results: Significant relationship between OH and cognitive function (p=0.04) was found in our patients. The patients with OH+ had higher risk of CI by 2.6 times than that of OH- patients. Maximum heart rate change during tilt table test was correlated with cognitive function and white matter changes, whereas blood pressure change during tilt table test showed no correlation with those parameters. Conclusions: There was significant relationship between OH and CI in PD. Therefore, PD patients with either symptom may need periodic evaluation and proper management for OH and cognitive functions.

Ozone Oxidation of PAHs in the Presence of Soil (I): Ozonation of Soil Slurry Phase (Ozone에 의한 PAHs 오염토양 복원 연구(I): 토양슬러리상 오존 산화)

  • A mechanism of ozonation of simulated soil slurry contaminated by phenanthrene and benzo[a]pyrene has been studied under various conditions. The effects of soil media(BS, S, GB), radical scavenger, ozone input ratio(0.17~0.73mg/min), bicarbonate ion, and humic acid were investigated, BS showed the highest removal efficiency in media tested. The generation of OH-radical via the catalytic decomposition of ozone on active sites of the natural sand was confirmed by OH-radical scavenger experiments. The enhanced removal efficiency by OH-radical was indirectly quantified to be about 22%. As initial concentration of humic acid(as sodium salt) was increased, pseudo first-order rate constant ($k_o$) of phenanthrene was decreased from $1.37{\times}10^{-2}s^{-1}$ to $0.53{\times}10^{-2}s^{-1}$. The amount of ozone required to oxidize 80% of the initial mass of phenanthrene(10mg/kg) and benzo[a]pyrene(10mg/kg) was 67.2mg/kg-soil and 48.0mg/kg-soil, respectively.

OH MASERS TOWARDS THE W49A STAR-FORMING REGION WITH MERLIN AND e-MERLN OBSERVATIONS

  • We present preliminary results from OH ground state phase referenced observations carried out with the Multi Element Radio Linked Interferometer Network (MERLIN) and e-MERLIN towards the massive star forming region W49A. There are three active SFRs within this complex: W49 North (W49 N), W49 South (W49 S) and W49 South West (W49 SW). The first epoch of observations was obtained in 2005 with MERLIN while the second epoch was obtained in 2013 with the e-MERLIN upgraded system. In this paper, we present 1665 and 1720 MHz maser emission towards W49 S and W49 SW. Overall, both epochs show good agreement with the previous observations of Argon et al. (2000) carried out with the Very Large Array (VLA). The better sensitivity and wider velocity coverage of the MERLIN/e-MERLIN observations allowed us to discover a new 1720 MHz OH maser site in W49 S.

대기압 플라즈마로 폐 암세포(H460)와 폐 정상세포(L132) 처리시, OH radical density에 따른 Cell 변화 측정

  • 대기압 플라즈마와 생체용액과의 상호작용은 Bio-medical 분야에서 주목 받고 있다. 대기압 플라즈마는 전자온도가 고온 플라즈마 보다 상대적으로 낮기 때문에 생체에 적용하기가 적합하다. 따라서 플라즈마가 세포에 미치는 영향을 관측하기 위해서 대기압 플라즈마를 이용하여 생체용액과의 반응을 살펴보고자 한다. Ar gas를 이용하여 플라즈마를 발생시켜 생체용액 표면을 처리하고 OES (Optical Emission Spectroscopy)을 이용해 방출 선을 조사했다. Ar 기체를 이용한 대기압 플라즈마를 사용하여 다른종류의 용액내의 OH Radical Density를 측정하였다. 용액으로는 DI (deionized) water 와 PBS (1x phosphate buffered saline)를 사용하였다. Ar gas를 200 sccm ($cm^3/min$) 으로 흐르게 하였을 때, DI water의 OH Radical Density 는 $4.33{\times}10^{16}cm^{-3}$ 으로 측정되었으며, 자외선 흡수분광법으로 측정한 완충용액인 PBS의 OH Radical Density 측정값은 $1.87{\times}10^{16}cm^{-3}$ 이다. 이런 특성을 기반으로, PBS 용액내의 H460 (Lung Cancer Cell) 와 L132 (Lung Normal Cell)을 깊이와 시간에 따라 대기압 플라즈마로 처리하여 cell의 변화를 보았다. 실험 각각의 조건은 깊이를 2 mm, 4 mm, 6 mm이며 시간은 10 sec, 30 sec, 60 sec 로 설정하였다. 표면으로부터의 깊이가 2 mm, 4 mm, 6 mm 일때 의 OH Radical Density는 각각 $1.87{\times}10^{16}cm^{-3}$, $0.5{\times}10^{16}cm^{-3}$, 0으로써 용액이 깊어질수록 OH Radical Density가 감소함을 볼 수 있다. OH radical density가 높은 2 mm 에서, 처리한 시간이 길어질수록 Cell 은 영향을 많이 받음을 관찰 할 수 있었다. H460 이 L132 보다 플라즈마에 영향을 많이 받음을 확인하였다. 특성변화를 알아보기 위하여 raman spectroscopy, flow cytometry, electron spin resonance로 측정한다.

Effect of deep eutectic solvent (DES) on the extraction of asiaticoside and madecassoside from Centella asiatica (병풀(Centella asiatica)로부터의 asiaticoside와 madecassoside의 추출효율에 미치는 DES의 영향)

  • Centella asiatica (C. asiatica) extracts, including asiaticoside and madecassoside, are used in ointments to treat the wound and atopic dermatitis due to their antibacterial and skin-regenerating effects in Asia. Therefore, research on the cultivation and extraction efficiency of C. asiatica is being actively conducted to increase commercialization efficiency. In this study, various deep eutectic solvents (DESs) were prepared and used as the extraction solvents according to the mole ratio between the hydrogen bond acceptor (HBA) and hydrogen bond donor (HBD). And then, the extraction yields in distilled water (DW) and methanol (MeOH), commonly used extraction solvents for C. asiatica, were compared and analyzed by HPLC in the optimized operating condition. As a result, a mixture of DW and DES at a ratio of 3:7 showed about 1.4 times higher extraction efficiency than MeOH only. Conversely, the extraction efficiency in a mixture of MeOH and DES at a ratio of 3:7 was about 6 % lower than that in MeOH only.

  • CdS and $CdS:Co^{2+}$ single crystals were grown by CTR method using iodine as transport material. The grown single crystals have defect chalcopyrite structure with direct band gap. The optical energy band gap was decreased according to add of Co-impurity. We can observed the Co-impurity optical absorption peaks assigned to the $Co^{2+}$ ion sited at the $T_d$ symmetry lattice and we consider that they were attributed to the electron transitions between energy levels of ions.

Effects of the Butanol Extact of Head of Panax Ginseng on Type II Collagen-induced Arthritis in DBA/1J Mice

  • In order to evaluate the improvement effects of head of Panax ginseng on chronic arthritis, we have investigated the activity of butanol fraction (BuOH fraction) in vitro and in vivo system. BuOH fraction showed significant inhibition on the elastase activity. Anti-arthritic activity of BuOH fraction was also examined on type II collagen-induced arthritis in DBA/1J mice. Mice were immunized with injection of type II collagen emulsified in Freund's complete adjuvant, followed by a booster injection 21 days later. BuOH fraction(BHPG) was administered at an oral dose of 500mg/kg for 2 weeks from the 1st day boost. The hind paw edema was significantly decreased in the group of treatment with BuOH fraction compared to control. In collagen-induced DBA/1J mice, BuOH fraction did not affected the collagen antibody titer but significantly inhibited the tumor necrosis factoralpha(TNF-${\alpha}$) activity. These results were confirmed with histological evaluation of joint tissues. This study may raise the possibility that the usage of BuOH fraction of head of Panax ginseng as alternative medicine for the relief and prevention of rheumatoid arthritis symptoms.

Antibacterial Activity of Medicinal Plant Extracts to S. aureus KCCM12256 and V. parahaemolyticus KCCM11965 (한약재 추출액의 S. aureus KCCM12256과 V. parahaemolyticus KCCM11965에 대한 항균 활성)

  • This study was conducted to investigate the antibacterial activities of medicinal plant extracts. The results were as follows. Crude water and methanolic extracts of Sanguisorba officinalis Linne, Coptis chinensis Makino, and Portulaca oleracea Linne, among 31 kinds of medicinal plants, had strong antibacterial activities against S. aureus and V. parahaemolyticus, with the methanolic extract more effective than the water extract. The methnolic and water extracts of S. officinalis Linne had strong antibacterial activities against S. aureus and V. parahaemolyticus. Antibacterial activity against S. aureus and V. parahaemolyticus was observed with extracts of S. officinalis Linne at dilutions of 5 and 10%, respectively. The EtOAc, BuOH, and $H_2O$ fractions of the extract of S. officinalis Linne had antibacterial activities against S. aureus and V. parahaemolyticus. Specifically, the fractions had antibacterial activity against S. aureus at dilutions of 5, 2, and 1%, respectively. Regarding V. parahaemolyticus, the EtOAc, $H_2O$, and BuOH fractions had antibacterial activities at dilutions of 5, 2, and 1%, respectively.

Antigenicity Changes of Ovomucoid and Ovalbumin in Chicken Egg White by NaOH, Heat and Protease Tratments (NaOH, 열, 및 효소 처리에 의한 계란 난백 중 ovomucoid와 ovalbumin의 항원성 변화)

  • Antigenicities of ovomucoid (OM) and ovalbumin (OA) in chicken egg white (EW) before and after NaOH, heat, and pretense treatments were examined by competitive indirect enzyme-linked immunosorbent assay (ciELISA), using rabbit anti-OM and-OA antibodies, Enzymatic hydrolysis of EW did not effectively reduce antigenicity of OM, whereas that of OA was decreased to 1/5,000-1/100,000 by treatment of plant-derived or microbial pretenses. Heat treatment below $100^{\circ}C$ for 30min did not decrease antigenicity of OM, whereas that of OA in heated EW increased maximally to 100 times, Antigenicity of OM in EW effectively decreased by NaOH treatment, disappearing at over 1% NaOH, whereas that of OA increased. Additional heat treatment of NaOH-treated EW at $70^{\circ}C$ for 15min slightly reduced antigenicities of OM and OA.

Cyclofarnesane sesquiterpene glucoside from the whole plant of Loranthus tanakae and its cytotoxicity ('꼬리겨우살이'(Loranthus tanakae) 전초로부터 cyclofarnesane sesquiterpene glucoside의 분리 동정 및 세포독성)

  • The whole plants of Loranthus tanakae were repeatedly extracted with 80% aqueous MeOH and the concentrate was partitioned into ethyl acetate (EtOAc), n-butyl alcohol (n-BuOH) and $H_2O$ fractions. The repeated silica gel ($SiO_2$), octadecyl $SiO_2$ (ODS), and Sephadex LH-20 column chromatographies for the n-BuOH fraction led to isolation of a cyclofarnesane sesquiterpene glucoside. The chemical structure including stereo structure was determined to be a (1'R,3'S,5'R,8'S,2E,4E)-dihydrophaseic acid $3^{\prime}-O-{\beta}-{\text\tiny{D}}$-glucopyranoside on the basis of spectroscopic analyses. This compound was isolated for the first time from L. tanakae in this study. Compound 1 showed a moderate dose-dependent cytotoxicity against human Caucasian gastric adenocarcinoma cells and human hepatocyte cari-noma cells cell lines at higher than $50{\mu}g/mL$.
