Inhibition Effect of Phenolic Compounds from Ultra-fine Ground Chrysanthemum indicum L. on Xanthine Oxidase (초미세 분쇄한 감국으로부터 추출된 phenolic 화합물의 xanthine oxidase 저해 효과)

  • In this study, the extracted phenolic compounds from 98 species of oriental herbal medicine were examined for biological activities to be used as functional resources. In particular, the anti-gout effect by xanthine oxidase (XOase) inhibition was determined using water and ethanol as extraction solvents because of their non-toxicity in the human body. The extracts of Chrysanthemum indicum L. (83.45%), Cuscuta chinensis (60.22%), Asiasarum sieboldi F. Maekawa (51.66%), Acorus gramineus (67.8%), Aconitum pseudo-laeve var. erectum (75.23%), Thuja orientalis (47.27%), Polygonum aviculare (53.98%), Carthami semen (63.99%), and Syzygium aromaticum (40.22%) showed relatively high XOase inhibitory activity. Chrysanthemum indicum L. was selected for its high XOase inhibitory activity. The biological compounds in Chrysanthemum indicum L. were identified to contain phenolics included in extracts of solids. Ultra-fine grind technology showed a higher extraction yield than normal grind and fine grind technology. Ethanol extracts showed relatively higher XOase inhibitory activity than water extracts. XOase inhibitory activity increased in a dependent manner as phenolic concentration increased. Therefore, ultra-fine grind technology was confirmed for use in increasing the extraction yield of XOase inhibitory compounds from Chrysanthemum indicum L.. Extracts from Chrysanthemum indicum L. are expected to be a useful functional resource for the prevention or treatment of gout.

Effect of Soy Milk and Sugar Addition to Jeungpyun on Physicochemical Property of Jeungpyun Batters and Textural Property of Jeungpyun (증편 제조시 콩물과 설탕의 첨가가 반죽의 이화학적 성질 및 저장 중 증편의 품질에 미치는 영향)

  • The study was attempted to investigate physicochemical and biological changes that would occur during preparation of Jeungpyun (rice cake prepared with rice wine). Furthermore, the effect of soy milk and suga. addition to Jeungpyun batters on textural changes of Jeungpyun during storage was studied in relation to physicochemical properties of Jeungpyun batters. 1. As fermentation continued, PH of Jeungpyun batters droped from 6.01 to 4.36. The addition of soy milk and 10% sugar to Jeungpyun batters resulted in significantly lower PH during fermentation. The volume of the batters with soy milk were significantly larger than those without soy milk. Jeungpyun batters with soy milk showed dramatic increase in viscosity after 2 hours of fermentation and the viscosity of other groups increased after 3 and half hours of fermentation. The reducing sugar contents of Jeungpyun batters containing 20% sugar increased rapidly during first fermentation and then decreased. The reducing sugar contents of Jeungpyun batters containing 10% sugar increased gradually during first fermentation and then rapidly increased after addition of 10% sugar during 3rd fermentation. 2. Sensory evaluation results demonstrated that hardness of Jeungpyuns increased and tenderness, springiness, moistureness, overall acceptability decreased during storage of 4 days at 4$^{\circ}C$. QTS data showed that hardness, gumminess, chewiness of all the groups increased and adhesiveness decreased during storage. Both of sensory evaluation and QTS data demonstrated that addition of soy milk and separate addition of sugar at first and furing 3rd fermentation period induced slower changes in textural properties in Jeungpyuns during storage.

Leaching of Organophosphorus and Carbamate Pesticides in Soil Column and Prediction of Their Mobility Using the Convective Mobility Test Model in Soils (유기인계 및 카바메이트계 농약의 토주용탈과 대류이동성 모형에 의한 이동성 예측)

  • This study was conducted to investigate the downward mobility of pesticides using soil columns and to compare the experimental results with values predicted from Convective mobility test model. Nine pesticides such as metolcarb, molinate, fanobucarb, isazofos, diazinon, fenitrothion, dimepiperate, parathion and chlorpyrifos-methyl were used for leaching test in soil column for four soils; Jungdong (upland soil), Gangseo (paddy soil), Yesan (forest soil), and Sineom(upland, volcanic ash-derived soil) series. The peak concentrations leached from 10 cm-columns of three soils except Sineom series ranged 6.5 to 12.6 mg/L for metolcarb, 2.6 to 5.0 mg/L for molinate, 4.5 to 7.8 mg/L for fenobucarb, 0.39 to 1.36 mg/L for dimepiperate, 1.1 to 4.6 mg/L for isazofos, 0.01 to 0.14 mg/L for diazinon, lower than 0.01 to 0.70 mg/L for fenitrothion and lower than 0.01 to 0.44 mg/L for parathion. But chlorpyrifos-methyl was not leached from any soil columns. Elution volumes to reach the peak of metolcarb, molinate, fenobucarb, isazofos, diazinon, and dimepiperate in the leachate ranged 1.1 to 2.1 pore volume (PV), 1.6 to 3.3 PV, 1.6 to 3.3 PV, 2.1 to 4.4 PV, 6 to 15 PV, and 8 to 21 PV, respectively. On the same water flux conditions, convection times estimated by Convective mobility test model were coincided with results from soil column test in most of the soil-pesticide combinations applied. Based on convection times estimated by the model at standard conditions (water flux 1 cm/day), metolcarb was classified as most mobile, molinate, fenobucarb and isazofos as mobile or most mobile, dimepiperate as moderately mobile or mobile, diazinon as mobile, fenitrothion and parathion as slightly mobile or mobile and chlorpyrifos-methyl as immobile or slightly mobile.

Evaluation of Usefulness of SPECT-CT at the Examination of Digestive System Leakage Patients (소화기계 Leakage 환자 검사 시 SPECT-CT의 유용성 평가)

  • Purpose When examining patients with digestive system Leakage, it is not easy to distinguish between bile leakage and bleeding only with Planar images. I would like to evaluate the utility of leakage of bile, confirmation of gastrointestinal bleeding and location discrimination using SPECT-CT. Materials and Methods SPECT-CT was performed according to the request of the reading room after acquiring Planar image for a total of 13 patients, including 8 patients suspected of bile leakage and 5 patients suspected of gastrointestinal bleeding, among patients visiting this specification went. We used Symbia T16 from SIEMENS and Discovery 670 from GE. Planar and SPECT-CT images were evaluated with a score of 1 to 10 by 4 experienced nuclear medicine doctors. Using the sensitivity and the specificity, the evaluation of the inspection by the ROC curve was carried out. The final diagnosis was confirmed by follow-up observation as a result of reoperative surgery. Results The sensitivity, specificity and accuracy of SPECT-CT were 91.7%, 100% and 94.2%, respectively. Planar images were 83.3%, 68.8% and 78.8%, respectively. Planar images showed lower diagnostic accuracy compared to SPECT-CT (78.8%, 94.2%, p<0.05). Moreover, the reliability of the diagnosis of SPECT-CT by ROC curve analysis showed a more useful result than the Planar image (p<0.05). Conclusion SPECT-CT had high sensitivity and specificity for diagnosis of biliary leakage and gastrointestinal bleeding and location discrimination. When SPECT-CT is additionally performed together with Planar images, it may be considered to improve bile leakage and diagnosis of gastrointestinal bleeding patients and position discrimination evaluation. There is usefulness depending on the patient's age and position, but consideration of additional CT exposure may be done sufficiently.

The Relation of HRV, PSQI and IQ with Symptoms of Insomnia Patients (수면장애와 동반된 임상증상에 따른 심박변이도와 수면 관련 설문간의 상관성 연구)

  • Objective: This study was to investigate the relation of what insomnia patients' symptoms with Heart Rate Variability(HRV), Pittsburgh Sleep Quality Index (PSQI) and Insomnia Questionaire(IQ). Methods: For this study, we carried out HRV, PSQI and IQ of 43 patients with insomnia who have come to Donguei oriental hospital of Donguei university from September 2007 to May 2008. We studied the relationship HRV with PSQI and IQ. For every symptom, this study classified the patients as none-symptom, symptom groups and analyzed each group's HRV, PSQI and IQ. Result : 1. The rate of xerostomia in female group is significantly higher than male group. 2. P1 (hypnagogic time), P3(awakening time), LF has negative correlation with age. The score of PI (hypnagogic time), P3(awakening time), P4(time of recoverable sleep) in the group whose age is more than 50 are higher than the group whose age is lesser than 50. 3. The frequency of using hypnotics(P6) in the group whose duration of onset are over 1 year was significantly higher than the score in the group whose duration of onset were below 1 year. 4. The accompanied symptoms in the group who started sleep disorder after stressor are higher than the number in the group whose duration of onset were poor-defined. 5. The drinking group have lower rate of anorexia, higher score of P1 (hypnagogic time) and TP than non-drinking group. 6. The score of P-total(general quality of sleep) have negative correlation with NNSO, HF, TP and positive correlation with LF/HF. 7. The score of P1(hypnagogic time) have negative correlation with LF. 8. The score of P5a(frequency of delayed hypnagogue) have positive correlation with LF/HF. 9. The score of P5e(frequency of cough or snoring) have negative correlation with RMSSD. 10. The score of I1a(degree of onset insomnia), I1b(degree of maintenance insomnia) have negative correlation with NN50, pNN50, HF. 11. The score of I1c(degree of termination insomnia) have negative correlation with RMSSD, NN50, pNN50, HF, TP and positive correlation with MHR, LF/HF. 12.. The score of P5i(frequency of night pain), LF/HF in the group with headache are higher than the group without headache. 13. The score of P5a(frequency of delayed hypnagogue), P5c(frequency of awakening for urine of stool) in the group with xerostomia are higher than the group without xerostomia. 14. The score of P9(degree of unrecoverable sleep) and I5a(degree of onset insomnia) in the group with chest discomfort are lower than the group without chest discomfort. 15. The score of P7(frequency of drowsiness for daytime) in the group with palpitation are lower than the group without palpitation. 16. The score of P5c(frequency of awakening for urine of stool), P5f(frequency of burning sensation), P5h(frequency of nightmare), I1c(degree of termination insomnia) in the group with burning sensation of upper trunk are higher than the group without burning sensation. 17. The score of NN50, pNN50, LF, TP in the group with anorexia are lower than the group without anorexia. 18. The score of P-total(general quality of sleep), P2(duration for sleep start) in the group with constipation are higher than the group without constipation. 19. The score of P4(recoverable sleep time) in the group with depression are higher than the group without depression. 20. In the group with anxiety, the score of P1(time of hypnagogue), P5a(frequency of onset insomnia) are lower and the score of I1b(degree of maintenance insomnia), I4(objective degree of impairment by insomnia) are higher than the group without anxiety. 21. The score of NN50, pNN50, HF, TP in the group with inertia are lower than the group without inertia. 22. The accompanied symptoms haver posirive correlation with P5f(frequency of burning sensation), P5h(frequency of nightmare), P5i(frequency of night pain) and LF/HF. Conclusion: This study provides insights into the complicated relations of the pattern of sleep disorder with many symptoms such as headache, chest discomfort, depression and any others. And this study showed that autonomic nervous system have important function in the regulation of sleep.

Prevalence Report of Transovarian Transmitted Diseases in the Breeder Chickens, Korea (국내 종계에서 난계대 전염병 감염 실태 보고)

  • A survey of transovarially transmitted diseases, including salmonellosis [(pullorum disease; PD)/(fowl typhoid; FT)], mycoplasmosis, avian infectious anemia (CIA), and fowl adenovirus infection was conducted in the breeder chickens from August to December in 2009. The numbers of flocks sampled out were: Grand Parents Stock (GPS), 45; Parents Stock (PS) 1,018, Baeksemi breeder (BSB) 54. The seroprevalence of salmonellosis (PD/FT) were 0% (GPS), 3.2% (PS), and 3% (BSB), respectively. A total of 983 chicken farms were affected with FT outbreaks between 2000 and 2008. The incidence of FT in commercial broilers, Baeksemi, commercial layers, native chickens, and broiler breeders was 44.3%, 26.2%, 15.7%, 12.6% and 1.08%, respectively. Of the affected broilers, over 90% birds were under 2 weeks of age, indicating it was possible that they were infected with S. gallinarum via vertical transmission. The sero-positive flocks against Mycoplasma gallisepticum (MG) were 71.1% (GPS), 88.7% (PS), 88.7% (BSB), while the rates of positive flocks against Mycoplasma synoviae (MS) were 86.0% (GPS), 77.0% (PS), and 98.0% (BSB). In GP and parent farms, the detection rates on specific genes of CIA virus were 19/45 (42.2%), and 169/1039 (18.0%), respectively, whereas the seroprevalence of CIA were 86.0% in GPS and 93.7% in PS flocks. In addition, positive flocks of fowl adenoviruses were 4.4% (GPS), 2.7% (PS) and 9.35% (BSB), respectively. As the results, avian mycoplasmosis and CIA have been more prevailing in chicken breeder than avian salmonellosis and fowl adenovirus infection in Korea.

Effectiveness of Fentanyl Transdermal Patch (Fentanyl-TTS, $Durogegic^{(R)}$) for Radiotherapy Induced Pain and Cancer Pain: Multi-center Trial (방사선치료로 인한 통증 및 암성통증에 대한 듀로제식의 효과: 다기관연구)

  • $\underline{Purpose}$: To evaluate the effectiveness and safety of fentanyl-TTS in the management of radiotherapy induced acute pain and cancer pain treated with radiotherapy. $\underline{Materials\;and\;Methods}$: Our study was open labelled prospective phase IV multi-center study. the study population included patients with more 4 numeric rating scale(NRS) score pain although managed with other analgesics or more than 6 NRS score pain without analgesics. Patients divided into two groups; patients with radiotherapy induced pain (Group A) and patients with cancer pain treated with radiotherapy (Group B). All patients received 25 ug/hr of fentanyl transdermal patch. Primary end point was pain relief; second end points were change in patient quality of life, a degree of satisfaction for patients and clinician, side effects. $\underline{Results}$: Between March 2005 and June 2005, 312 patients from 26 participating institutes were registered, but 249 patients completed this study. Total number of patients in each group was 185 in Group A, 64 in Group B. Mean age was 60 years and male to female ratio was 76:24. Severe pain NRS score at 2 weeks after the application of fentanyl was decreased from 7.03 to 4.01, p=0.003. There was a significant improvement in insomnia, social functioning, and quality of life. A degree of satisfaction for patients and clinician was very high. The most common reasons of patients' satisfactions was good pain control. Ninety six patients reported side effect. Nausea was the most common side effect. There was no serious side effect. $\underline{Conclusion}$: Fentanyl-TTS was effective in both relieving pain with good tolerability and improving the quality of life for patients with radiotherapy induced acute pain and cancer pain treated with radiotherapy. The satisfaction of the patients and doctors was good. There was no major side effect.

Ionic currents elicited by the hypotonic solution in hamster eggs (저장성 용액에 노출된 햄스터 난자에 관찰되는 이온전류의 변화)

  • Cell volume regulatory mechanisms are usually disclosed by exposure of cell to anisotonic media. If a cell is suddenly exposed to hypotonic media, it swells initially like an osmometer but within minutes regains its original cell volume. This behavior has been labelled as regulatory cell volume decrease(RVD). RVD is believed to result from the loss of permeable ions through the membrane. In this study, we examined hypotonically induced changes in the membrance currents involved in RVD by using whole cell voltage clamp technique in the unfertilized hamster egg. At -40mV of the holding potential, the stationary current was maintained in the hamster egg exposed to isotonic solution composed of, mainly, 115mM NaCl and 40mM mannitol. Hypotonic solution was prepared by removing mannitol. Therefore, the concentrations of $Na^+$ and $Cl^-$ in this hypotonic media were the same as those in the isotonic solution. Following 30 to 60 sec after applying the hypotonic media to the egg, the inward current was evoked. This inward current was eliminated by $100{\mu}M$ 4-acetamido-4'-isothiocyanostil-bene-2,2'-disulfonic acid(SITS), an anion channel blocker, leaving the small outward current component. Further addition of 2mM $Ba^{2+}$, a broad $K^+$ channel blocker, completely abolished the small outward current left even in the presence of SITS during hypotonic stress. These results suggest that $K^+$ and $Cl^-$ move out of cells, resulting in RVD. To test the involvement of $Na^+$ in RVD, 20mM Na-isethionate was substituted for mannitol in isotonic media(135mM $Na^+$) and Na-isethionate (20mM) was freed the hypotonic solution. Only $Cl^-$ concentration in both isotonic and hypotonic media was kept constant at 115mM, whereas concentration of $Na^+$ was lowered in hypotonic solution to 115mM from 135mM in isotonic solution. Hypotonic medium induced the outward current in the egg equilibrated isotonically. This current was reduced by $100{\mu}M$ SITS but was augmented by 2 mM $Ba^{2+}$. In terms of RVD, these results imply that $Cl^-$ efflux is coupled with $K^+$, maybe for electroneutrality during hypotonic stress and/or with $Na^+$ via unknown transport mechanism(s). From the overall results, the hypotonic stress facilitates the movement of $Cl^-$ and $K^+$ out of the hamster egg to regain cellular volume with electroneutrality. If there exist a difference in $[Na^+]_0$ between isotonic and hypotonic solution, another transport mechanism concerned with $Na^+$ may, at least partly, participate in regulatory volume decrease.

Butachlor and Silicate Fertilizer Interaction to Rice (Butachlor와 규산질비료(珪酸質肥料)의 수도(水稻)에 대(對)한 상호작용(相互作用))

  • Pot and laboratory tests were undertaken to investigate the influence of silicate fertilization on butachlor phytotoxicity to rice. Growth of rice seedlings at 150 ppm of $SiO_2$ was stimulated, while adverse effect was observed over 300 ppm of $SiO_2$ and growth reduction was enhanced with combination of butachlor and $SiO_2$ Rice growth in pot trial at 150kg/10a of silicate fertilization was not influenced by recommended amounts of butachlor and nitrofen, however, the growth of Seokwang byeo at 300kg/10a of silicate was markedly retarded by butachlor in the initial stage of growth. Growth reduction of Seokwang byeo caused by combined application of silicate and butachlor was recovered 50 days after herbicide application. Growth reduction from butachlor was not influenced by pH level and also degradation behaviors of butachlor in submerged soil was not altered by silicate fertilization. Adsorbed amount of butachlor on rice root was increased with addition of $SiO_2$ and its amount in Seokwang byeo was higher than that of Jinju byeo. Butachlor absorption by Seokwang byeo was accelerated by 150 ppm of $SiO_2$ applied simultaneously, but those effect was not encountered in Jinju byeo. Butachlor absorption of rice seedlings was also increased by 150 ppm of $K_2O$, while CaO hindered the absorption and $Na_2O$ had no effect on the absorption. Residual level of butachlor in Seokwang byeo treated with combined solution of butachlor and $SiO_2$ was continued higher than that with butachlor alone during 10 days after transplantation to culture solution.

Characteristics of Chungkookjang Prepared by Bacillus amyloliquefaciens with Different Soybeans and Fermentation Temperatures (Bacillus amyloliquefaciens를 이용하여 콩 종류와 발효온도를 달리하여 제조한 청국장의 특성)

  • This study was carried out to investigate the characteristics of Chungkookjang depending on different soybeans and fermentation temperatures using Bacillus amyloliquefaciens C2 isolated from homemade Chungkookjang. The highest protease activity was 854 U/g in yellow soybean Chungkookjang and 847 U/g in black soybean Chungkookjang at $35^{\circ}C$. The highest amylase activity was 3.87 U/g at $40^{\circ}C$ in yellow soybean Chungkookjang and 4.96 U/g at $45^{\circ}C$ in black soybean Chungkookjang. The highest reducing sugar content was 16.11 mg/g at $40^{\circ}C$ in yellow soybean Chungkookjang and 19.08 mg/g at $45^{\circ}C$ in black soybean Chungkookjang. The highest amino type nitrogen content was 420 mg%/g in yellow soybean Chungkookjang and 194 mg%/g in black soybean Chungkookjang at $40^{\circ}C$. The highest pH was 7.92 at $40^{\circ}C$ in yellow soybean Chungkookjang and 7.59 at $45^{\circ}C$ in black soybean Chungkookjang. The highest number of viable cell was 9.3 log CFU/g at $40^{\circ}C$ in yellow soybean Chungkookjang and at $35^{\circ}C$ in black soybean Chungkookjang. On the other hand, the lowest ammonia type nitrogen content was 225 mg%/g at $45^{\circ}C$ in yellow soybean Chungkookjang and 80 mg%/g at $40^{\circ}C$ in black soybean Chungkookjang. Yellow soybean Chungkookjang showed high protease activity, pH and amino type nitrogen, whereas black soybean Chungkookjang showed high amylase activity and reducing sugar.
