Optimum Culture Environment of the Benthic Copepod, Tigriopus japonicus (부착성요각류 Tigriopus japonicus의 최적배양환경)

  • The harpacticoid copepod, Tigriopus japonicus is one of the most important zooplankton as a live food for the production of marine fish fry. Thus, the salinity tolerance and the optimum culture environment of this copepod in terms of salinity, temperature and light were examined. The food values of 6 kinds of phytoplankters and 2 kinds of yeast were also investigated for mass culture of this copepod. The results are as follows: After 5 day culture in the experiment of salinity tolerance, the survival rates of the gravid female at $0\%\;and\;90%o\;were\;40\%\;and\;70\%$, respectively. However, at salinity ranging from $2\%o\;to\;80\%o$, high survival rates above $85\%$ were observed. It means T. japonicus is very euryhalinous species. Temperature was more important factor than salinity for the fecundity of T. japonicus. The optimum culture conditions of this species were $24^{\circ}C,\;24\%o$, and 3,000 lux with 24 L: 0D. Under these culture conditions, the average fecundity from a gravid female per spawning was 38 nauplii, and the interval time between spawnings were 2.05 days. Phaeodactylum tircornutum seemed to be the most suitable phytoplankton as a live food for T. japonicus, and the large chlorophyta, Tetraselmis suecica showed the lowest food value among 6 phytoplankters and 2 yeasts. The food value of w-yeast was better than that of baker's yeast, and it is similar to that of phytoplankton such as Amphora sp., Chlorella ellipsoidea and Nannochloris oculata. So, the w-yeast seems to be appropriate food source for mass culture of T. japonicus.

  • The purpose of this paper is to establish an IoT-based big data platform that can check the space and form analysis in various play cultures of children. Therefore, to this end, in order to understand the healthy play culture of children, we are going to build a big data platform that allows IoT and smart devices to work together to collect data. Therefore, the goal of this study is to develop a big data platform linked to IoT first in order to collect data related to observation of children's mobile movements. Using the developed big data platform, children's play culture can be checked anywhere through observation and intuitive UI design, quick information can be automatically collected and real-time feedback, data collected through repeaters can be aggregated and analyzed, and systematic database can be utilized in the form of big data.

  • Science & Technology (S&T) is the most important thing to attain competitiveness in the 21st century. Our traditional S&T policies have been focused on hardware infra structure. But, we should consider soft infra of S&T to enhance welfare development of S&T personnel. S&T should be had a correct understanding method of the creation for S&T culture. It implies that S&T policy in the 21st century should consider non-material factors which include social, human, environment and culture. Under this background. This paper defines the concept of welfare of S&T personnel. Also this paper deals with the change of S&T policy of Korea and analyzes the characteristic of each phase. This paper argues that all actors (for example S&T personnel, corporate using S&T, public) should participate in the process of S&T innovation since the 21st century is the generation of popularization of S&T. The purpose of this paper is to suggest a few methods of welfare improvement of S&T personnel to reinforce soft infra structure of S&T.

Feeding of Bacteria by Copepod Tigriopus japonicus (Tigripus japonicus의 세균 섭이)

  • This study was carried out to investigate feeding of bacteria by Tigripus japonicus (Copepoda : Harpacticoida) under axenic culture. The ovigerous females and nauplii were grown with feed of aquatic bacteria. Growth of RT bacteria strain was suppressed by feeding of co-existing T. japonicus. T. japonicus of non-axenic culture was observed with oil bead in the egg sac. On the other hand, early nhauplius stage did not develop to the next stage without stage took bacteria as food. And the adult of T. japonicus may utilize the baxteria as nutrient source for egg development.

  • The removing copper from CCA, CCFZ treated wood were tested with Tyromyces palustris. The shacking culture, solid culture, and stationary culture methods were tested to removal of copper in CCA, CCFZ treated wood. The steam pressure method and oxalic acid method were tested as pretreatment. To investigate of copper removal mechanism, the oxalic acid that produced by T. palustris and combination with copper was examined on stationary culture that contained CCA and CCFZ treated wood chips. Oxalic acid increased copper removing rate of T. palustris as pretreatment. Stationary culture had most removal effectiveness among the culturing methods. The removal rate of copper on bioreactor was 61% on 7 days. Copper-oxalate was detected on liquid culture, which show the oxalate from liquid of culture combined with copper from treated wood. It would be related to the removal of copper from treated wood by T. palustris.

Composition of a Medium for Serum-free Culture of an Adipose-derived Stem Cell Line Established with a Simian Virus 40 T Antigen (Simian virus 40의 T항원 도입으로 수립한 지방유래줄기세포주의 효율적인 무혈청 배양법 및 무혈청 배지조성)

  • Adipose-derived stem cells (ADSCs) are considered promising tools for tissue regeneration. However, ADSCs have very poor proliferation capacity. Therefore, fetal bovine serum (FBS) is generally added to the culture media of ADSCs. As FBS contains many uncharacterized components that may affect cellular functions, methods for serum-free cultures of ADSCs have been widely investigated. In this study, to develop an efficient method for a serum-free culture of ADSC-T, we used an ADSC line established by introducing the simian virus 40 (SV40) T gene into primary ADSCs. We then investigated the effect of amino acids, vitamins, and other components on the growth of ADSC-T. When the ADSC-T cells were plated with DMEM/F12 serum-free medium, the cells did not proliferate, and the mixture of amino acids, vitamins, and B27 supplement did not increase the growth of the cells. However, when the ADSC-T cells were provided with serum-free DMEM/F12 after they had been cultured with serum-supplemented DMEM for 24 h, the cells proliferated, and the vitamins and B27 supplement increased the cell growth. Stem-Pro serum-free medium also appeared to be useful as a suspension culture for the ADSC-T cells. The ADSC-T cells secreted large amounts of proteins of around 70 kDa. Insulin-like growth factor (IGF) and fibroblast growth factor basic (FGF basic) were secreted by ADSC-T in larger amounts in the serum-free culture than in the serum-supplemented culture.

In addition, Recent construction sector integration and fusion technologies in various areas of research and planning system is being maintained by the system. Past experience of conservative management in this emerging technology-driven technology commercialization in management of technology through the convergence of various technologies and to find synergies to secure the future of construction technology because it is a need.

A study on the Turfan's costume -Centering around Kochang- (트르판의 복식에 관한 연구 -고창을 중심으로-)

  • The costume style of Turfan had changed three times which were the period of peculiar style the period of Kochang style the period of influence of T'ang's culture. Before the founding of Kochang country the costume style of Turfan was two piece style. It was a character of nomadic people's clothes. Tops were tunic and caftan. The method of adjust caftan were two types. The one is wrapping toward left so the right edge of the top covers the left. The other is wrapping toward right. Bottoms were pants and skirts. This period was characterized by the traditional style. I called in it the period of peculiar style After the founding of Kochang country style as Kochang style. In this period Silkroad was so activated that cultural exchanges between many people and materials passed through Turfan. Turfan's culture soaked up the products of civilization of chinese and western Turkestan. Chinese and western Turkestan's costume influencd under the direct control of T' ang dynasty. Such an aspects were reflected the Trufan's costume. The Trucan's costume had changed into the foreign style that was the T'ang's costume. It was the period of influence of T'ang culture.

Cellulose Utilization and Protein Productivity of Some Cellulolytic Fungal Co-cultures

  • Protein productivity by the cellulolytic fungi, Trichoderma viride(MTCC 800), Chaetomium globosum and Aspergillus terreus was compared in co-culture and mixed culture fermentations of cashewnut bran. Co-cultures were more effective in substrate saccharification, which ranged between $85{\sim}88%$ compared to the $62{\sim}67%$ saccharification shown by the monocultures. Maximum saccharification was induced by T. viride and C. globosum co-culture resulting in the highest 34% release of reducing sugars. The maximum 16.4% biomass protein and the highest protein productivity(0.58%) were shown by T. viride and A. terreus co-culture. A. terreus performed better in co-culture in the presence of T. viride rather than with C. globosum. Among the cellulolytic enzymes, FPase(Filter Paper Cellulase) activity was significantly higher in all the co-cultures and in the mixed culture than in their respective monocultures. Mixed culture fermentation involving all the three fungi was not effective in increasing the per cent saccharification or the biomass protein content over the co-cultures.

  • A mixed culture of Candida tropicalis and Trichosporon cutaneum was carried out using a n-paraffin medium. The growth of C. tropicalis was markedly enhanced by the mixed culture with T. cutaneum which did not grow on n-paraffin. C. tropicalis extracellularly excreted free fatty acids as metabolic products of n-paraffin in the culture medium. T. cutaneum appeared to assimilate these free fatty acids which were growth inhibitors for C. tropicalis, threreby enhancing the growth of C. tropicalis.
