Caprylic Acid와 Cyclodextrin 복합물이 In vitro 반추위 발효성상 및 메탄 생성에 미치는 영향 (Effects of Caprylic Acid and Cyclodextrin Complex on In vitro Fermentation Characteristics and Methane Production)

  • 본 연구에서는 caprylic acid와 cyclodextrin과의 complex(CA-CD complex)의 첨가수준이 in vitro 반추위 발효 성상과 메탄 생성에 미치는 영향을 알아보고자 하였다. CA-CD complex 무첨가구를 대조구로 설정 하였으며, 처리구로는 CA 20mg 단독 첨가구(T1), CD 830mg 단독 첨가구(T2), CA-CD complex 425mg 첨가구(T3, CA 함량 10mg), CA-CD complex 850mg 첨가구(T4, CA 함량 20mg), CA-CD complex 1700 mg 첨가구(T5, CA 함량 40mg)이며, 3반복으로 4, 8, 12시간의 배양 실험을 수행하였다. 모든 시험구에서 배양 12시간까지 pH가 지속적으로 감소하였다. 특히, 배양 8시간 후부터 T2, T3, T4, T5 처리구에서 대조구보다 유의적으로 감소하였다(P<0.05). Total VFA 농도는 대조구에 비해 T2, T3, T4, T5구들이 8시간과 12시간에서 유의적(P<0.05)으로 감소하였다. Propionate 농도는 배양 8시간에서만 다른 처리구보다 T4, T5 처리구가 유의적(P<0.05)으로 높게 나타났다. Acetate:Propionate(A/P) 비율은 모든 배양시간에서 처리구간 유의적인 차이가 없었다. 암모니아 농도는 대조구에 비해 T2, T3, T4, T5 처리구에서 배양 4시간부터 대조구보다 암모니아 농도가 유의적(P<0.05)으로 감소하였다. Total gas는 T1구에서 대조구보다 유의적(P< 0.05)인 감소가 있었으나, T2, T3, T4, T5 처리구에서는 대조구에 비하여 12시간에서 유의적(P<0.05)으로 증가하였다. Total gas 발생량에 대한 메탄 비율(%)은 12시간 배양시간에서 대조구는 3.4%이었지만, 메탄 발생량이 가장 적었던 T1구는 2.8%, 메탄 발생량이 가장 많았던 T2구는 5.1%, 그리고 T3, T4, T5구는 평균 2.7%로 T1구와 통계적 차이가 없었다. 따라서 CA-CD complex를 이용하면, 반추위내 발효에 부정적 영향 없이 메탄을 감소시킬 수 있을 것으로 기대된다.

소에 의한 다양한 사일리지의 이용율 및 TDN 평가 (Estimation of Availability and TDN of Various Silages by Cattle)

  • 본 시험은 국내에서 생산되어 곤포형태로 포장된 청보리 사일리지, 호밀 사일리지, 이탈리안 라이그라스 사일리지 및 수단그라스 (수수${\times}$수단그라스 교잡종) 사일리지의 반추위 내 발효특성과 분해율 및 전장소화율과 TDN을 평가하기 위해 반추위 누관이 장착된 소 4두를 대상으로 실시되었다. 사일리지가 반추위액의 pH에 거의 영향하지는 않았으나 사일리지 급여 후 3시간에서 다른 종류의 사일리지를 섭취한 경우와 비교할 때 수단그라스 사일리지를 섭취한 소의 반추위액의 암모니아 농도가 7.28 mg/100 ml로 가장 낮았다 (p<0.049). Acetate 조성 비율은 다른 종류의 사일리지에 비해 수단그라스 사일리지 급여 1시간 (p<0.018), 3시간 (p<0.004) 및 6시간 (p<0.019) 후에 채취한 반추위액에서 높은 수준을 보였으며, propionate 조성 비율은 사일리지 급여 후 6시간까지 호밀사일리지와 청보리 사일리지를 섭취한 소에서 높은 (p<0.001~p<0.042) 반면 수단그라스 사일리지를 섭취한 소에서 가장 낮았다. Butyrate 조성 비율은 사일리지 급여 후 대부분의 반추위액 채취 시간 (1~6시간)에 걸쳐 청보리 사일리지를 섭취한 소에서 가장 낮은 (p<0.007~p<0.027) 것으로 나타났다. 반추위 내 건물의 유효분해율은 호밀 사일리지에서 59.64%로 가장 높았으며, 그 다음으로 청보리 사일리지(56.12%) 및 이탈리안 라이그라스 사일리지(55.64%)에서 높았고 수단그라스 사일리지의 유효분해율이 54.02%로 가장 낮았다 (p<0.048). 이러한 경향은 조단백질 (p<0.014) 및 유기물(p<0.039)의 유효분해율에서도 비슷한 것으로 조사되었다. 수단그라스 사일리지 내 건물(p<0.032), NDF (p<0.034) 및 유기물 (p<0.041)의 전장소화율은 다른 종류의 사일리지에 비하여 현저히 낮았으나 다른 세 종류의 사일리지 간에는 차이가 없었다. 전장소화율을 이용하여 산출된 TDN 값은 호밀 사일리지가 61.1%로 가장 높았으며 (p<0.001), 청보리 사일리지, 이탈리안 라이그라스 사일리지 및 수단그라스 사일리지의 TDN 값은 각각 57.1, 57.9 및 50.7%인 것으로 조사되었다. 이상의 결과로 미루어 보아 국내에서 생산, 이용되고 있는 사료작물 사일리지의 성분이 VFA 조성 비율 및 소화율에 영향하는 것으로 보이며, 특히 사일리지의 NDF 함량이 소화율에 기초를 둔 TDN 값에 크게 영향하는 것으로 여겨진다.

Effects of Three Feeding Systems on Production Performance, Rumen Fermentation and Rumen Digesta Particle Structure of Beef Cattle

  • The effects of three different feeding systems on beef cattle production performance, rumen fermentation, and rumen digesta particle structure were investigated by using 18 Limousin (steers) with a similar body weight ($575{\pm}10kg$) in a 80-d experiment. The animals were equally and randomly divided into three treatment groups, namely, total mixed ration group (cattle fed TMR), SI1 group (cattle fed concentrate firstly then roughage), and SI2 group (cattle fed roughage firstly then concentrate). The results showed that the average daily gain was significantly higher in cattle receiving TMR than in those receiving SI1 and SI2 (p<0.05). Consumption per kg weight gain of concentrate, silage, and combined net energy (NEmf) were significantly decreased when cattle received TMR, unlike when they received SI1 and SI2 (p<0.05), indicating that the feed efficiency of TMR was the highest. Blood urea nitrogen (BUN) was significantly decreased when cattle received TMR compared with that in cattle receiving SI1 (p<0.05), whereas there was no difference compared with that in cattle receiving SI2. Ammonia nitrogen concentration was significantly lower in cattle receiving TMR than in those receiving SI1 and SI2 (p<0.05). The rumen area of cattle that received TMR was significantly larger than that of cattle receiving SI1 (p<0.05), but there was no difference compared with that of cattle receiving SI2. Although there was no significant difference among the three feeding systems in rumen digesta particle distribution, the TMR group trended to have fewer large- and medium-sized particles and more small-sized particles than those in the SI1 and SI2 groups. In conclusion, cattle with dietary TMR showed increased weight gain and ruminal development and decreased BUN. This indicated that TMR feeding was more conducive toward improving the production performance and rumen fermentation of beef cattle.

Effects of Protein and Carbohydrate Supplementations on Fibre Digestion and Microbial Population of Sheep

  • The effects of two types of protein, soybean meal (SBM) and fish meal (FM); and two types of energy supplements, corn flour (CF) and paper pulp (PP), on intake of guinea grass (Panicum maximum), fibre digestion and microbial activities in four Merino rams with an average weight of $54.4{\pm}4.5kg$ were studied. Each animal was fitted with a ruminal cannula and a duodenal cannula at the proximal position. The animals were fed twice daily with chopped guinea grass (5 cm) ad libitum and one of the four dietary supplements: 170 g FM+268 g PP; 170 g FM+268 g CF; 200 g SBM+200 g PP or 200 g SBM+200 g CF. All the supplements were mixed with 100 g molasses. In sacco and in vivo digestibilities, digesta flow rates, fermentation and microbial population were studied in a $4{\times}4$ Latin square design with a $2{\times}2$ factorial arrangement of dietary treatments. The effects of energy or protein sources were not significant on grass intake of sheep. The potential degradabilities of NDF and ADF were not significantly affected by any of the supplements. However, the energy and protein sources had significant efects on disappearance rate of NDF and ADF. The disappearance rate of both NDF and ADF were significantly (p < 0.05) higher in animals fed PP when compared to animals fed CF. Animals fed FM also showed significantly (p < 0.03) higher disappearance rate of ADF than those fed SBM. Animals fed PP showed better digestion in the rumen and total tract. Total flow of NDF and ADF through the duodenum was not significantly affected by the various supplements. The mean rumen pH values (5.8-6.1) were not significantly different among the four different diets. The concentration of rumen ammonia was significantly (p < 0.0001) higher in animals fed SBM (235-266.4 mg N/L) supplement than in animals fed FM (174.9-179.7 mg N/L), while total VFA concentration was not significantly affected by both energy and protein supplements. Mean values of total VFA ranged from 72.5-82.3 mM. Molar proportions of acetate, propionate and butyrate were typical of a roughage type fermentation. Molar proportion of acetate was significantly (p < 0.0001) higher in sheep fed PP when compared to sheep fed CF. Animals fed FM had higher total viable bacterial counts, while animals fed CF showed higher protozoal numbers. Proportions of cellulolytic bacteria were only slightly higher in animals fed SBM or PP.

Effects of Micronization on the In situ and In vitro Digestion of Cereal Grains

  • The effects of micronization on in situ and in vitro nutrient disappearances of wheat, barley and corn were investigated in a series of experiments. In Experiment 1, chemical composition and in situ dry matter disappearance (DMD) of six varieties of wheat were determined. In addition, an in vitro study was completed using ground micronized and unmicronized wheat (var. Kansas). In Experiment 2, three varieties of wheat (Kansas, Sceptre and Laura) and in Experiment 3, three cereal grains (wheat, barley and corn) were either micronized for 1 min to attain internal kernel temperatures of 90-100$^{\circ}C$ or not (controls), and DM, protein and starch disappearances were estimated. In Experiment 2, an in vitro study was also completed using ground micronized and unmicronized wheat (var. Kansas). Wheat samples varied with respect to crude protein (10.0-21.2%), starch (61.6-73.9%), NDF (8.5-11.8%), volume weight (753-842 g/L) and kernel hardness (0.0-32.0). Rate (p = 0.003) and extent (p = 0.001) of in situ DMD differed among wheat varieties. Correlations between in situ kinetics, and chemical and physical properties of wheat varieties showed that protein content was negatively correlated with the rate of disappearance ($r^2$ = -0.77). Micronization of all grains markedly reduced (p = 0.001) the rate and extent of DM, and protein disappearances as compared to control samples. Micronization increased (p<0.05) the digestion of starch in wheat. However, release of ammonia into the incubation medium was markedly reduced (p<0.05), suggesting that micronization increased the resistance of protein to microbial digestion. Disappearances of DM, protein and starch differed (p = 0.001) among cereal grains with wheat>barley>corn. Micronization reduced the rate of DM disappearance (p = 0.011) and slowly degradable protein fractions (p = 0.03), however, increased (p = 0.004) slowly degradable starch fractions of all three cereals. Examination of in situ samples by scanning electron microscopy confirmed that microbial colonization focused on starch granules in micronized grains, and that the protein matrix exhibited resistance to microbial colonization. These results suggest that micronization may be used to increase the ruminal escape value of protein in cereal grains, but may lead to increased starch digestion if grains are finely ground.

Shrub coverage alters the rumen bacterial community of yaks (Bos grunniens) grazing in alpine meadows

  • Proliferation of shrubs at the expense of native forage in pastures has been associated with large changes in dry-matter intake and dietary components for grazing ruminants. These changes can also affect the animals' physiology and metabolism. However, little information is available concerning the effect of pastoral-shrub grazing on the rumen bacterial community. To explore rumen bacteria composition in grazing yaks and the response of rumen bacteria to increasing shrub coverage in alpine meadows, 48 yak steers were randomly assigned to four pastures with shrub coverage of 0%, 5.4%, 11.3%, and 20.1% (referred as control, low, middle, and high, respectively), and ruminal fluid was collected from four yaks from each pasture group after 85 days. Rumen fermentation products were measured and microbiota composition determined using Ion S5™ XL sequencing of the 16S rRNA gene. Principal coordinates analysis (PCoA) and similarity analysis indicated that the degree of shrub coverage correlated with altered rumen bacterial composition of yaks grazing in alpine shrub meadows. At the phyla level, the relative abundance of Firmicutes in rumen increased with increasing shrub coverage, whereas the proportions of Bacteroidetes, Cyanobacteria and Verrucomicrobia decreased. Yaks grazing in the high shrub-coverage pasture had decreased species of the genus Prevotellaceae UCG-001, Lachnospiraceae XPB1014 group, Lachnospiraceae AC2044 group, Lachnospiraceae FCS020 group and Fretibacterium, but increased species of Christensenellaceae R-7 group, Ruminococcaceae NK4A214 group, Ruminococcus 1, Ruminococcaceae UCG-002, Ruminococcaceae UCG-005 and Lachnospiraceae UCG-008. These variations can enhance the animals' utilization efficiencies of cellulose and hemicellulose from native forage. Meanwhile, yaks grazed in the high shrub-coverage pasture had increased concentrations of ammonia nitrogen (NH3-N) and branched-chain volatile fatty acids (isobutyrate and isovalerate) in rumen compared with yaks grazing in the pasture without shrubs. These results indicate that yaks grazing in a high shrub-coverage pasture may have improved dietary energy utilization and enhanced resistance to cold stress during the winter. Our findings provide evidence for the influence of shrub coverage on the rumen bacterial community of yaks grazing in alpine meadows as well as insights into the sustainable production of grazing yaks on lands with increasing shrub coverage on the Qinghai-Tibet Plateau.

Near Infrared Spectroscopy for Measuring Purine Derivatives in Urine and Estimation of Microbial Protein Synthesis in the Rumen for Sheep

  • The efficiency of the luminal fermentation process influences overall efficiency of luminal production, animal health and reproduction. Ruminant production systems have a significant impact on the global environment, as well. Animal wastes contribute to pollution of the environment as ammonia volatilized to the air and nitrate leached to ground water. Microbial protein synthesis in the rumen satisfies a large proportion of the protein requirements of animals. Quantifying the microbial synthesis is possible by using markers for lumen bacteria and protozoa such as nucleic acids, purine bases, some specific amino acids, or by isotopic $^{15}N,^{32}P,\;and\;^{35}S$ labelled feeds. All those methods require cannulated animals, they are time-consuming and some methods are very expensive as well. Many attempts have been made to find an alternative method for indirect measurement of microbial synthesis in intact animals. The present investigations aimed to assess possibilities of NIRS for prediction of purine nitrogen excretion and ruminal microbial nitrogen synthesis by NIR spectra of urine. Urine samples were collected from 12 growing sheep,6 of them male, and 6- female. The sheep were included in feeding experiment. The ration consisted of sorghum silage and protein supplements -70:30 on dry matter basis. The protein supplements were chosen to differ in protein degradability. The urine samples were collected daily in a vessel containing $60m{\ell}$ 10% sulphuric acid to reduce pH below 3 and diluted with tap water to 4 liters. Samples were stored in plastic bottles and frozen at $-20^{\circ}C$ until chemical and NIRS analysis. The urine samples were analyzed for purine derivates - allantoin, uric acid, xantine and hypoxantine content. Microbial nitrogen synthesis in the lumen was calculated according to Chen and Gomes, 1995. Transmittance urine spectra with sample thickness 1mm were obtained by NIR System 6500 spectrophotometer in the spectral range 1100-2500nm. The calibration was performed using ISI software and PLS regression, respectively. The following statistical results of NIRS calibration for prediction of purine derivatives and microbial protein synthesis were obtained.(Table Omitted). The result of estimation of purine nitrogen excretion and microbial protein synthesis by NIR spectra of urine showed accuracy, adequate for rapid evaluation of microbial protein synthesis for a large number of animals and different diets. The results indicate that the advantages of the NIRS technology can be extended into animal physiological studies. The fast and low cost NIRS analyses could be used with no significant loss of accuracy when microbial protein synthesis in the lumen and the microbial protein flow in the duodenum are to be assessed by NIRS.

지방급원 형태와 수준에 따른 연속배양장치 내 반추위 발효성상, 지방산의 수소첨가 현상 및 Conjugated Linoleic Acid 생산에 미치는 영향 연구 (Effects of Type of Oilseed and Level of Concentrate on Fermentation, Biohydrogenation of Fatty Acids and Conjugated Linoleic Acid Production in a Rumen-Simulated Continuous Culture System)

  • 본 연구는 사료 내 농후사료의 비율을 높임으로서 불포화 지방산의 반추위 내 by-pass율의 향상 가능성을 조사하고, 아마종실과 전지대두 급여 시 CLA 생산을 상호 비교하기 위하여 연속배양장치를 이용하여 수행하였다. 지방 급원에 따른 발효 성상의 차이는 보이지 않았다. 한편 사료 내 농후사료 비율이 높은 (80%) 처리구는 농후사료 비율이 낮은 (40%) 처리구와 비교하여 pH는 감소하였으나, 암모니아, 총 휘발성 지방산, acetate, butyrate 및 valerate 농도가 증가되었다. 지방급원 (전지대두 vs 아마종실)과 사료 내 농후사료 비율은 organic matter (OM), total nitrogen, neutral detergent fiber (NDF) 및 acid detergent fiber (ADF)의 소화율에 영향을 끼치지 않았다. 반면에 전지대두는 아마종실과 비교하여 trans C18:1, C18:2 n-6 및 C18:3 n-3 유출율은 증가시켰다. 지방 급원에 의한 CLA flow는 영향을 받지 않았으나 사료 내 농후사료 비율이 높을 때와 전지대두와 아마종실의 함량이 높았을 때는 증가되었다. 수소 첨가현상은 C18:1 n-9 와 C18:2 n-6에서 지방 급원에 의하여 영향을 받지 않았으나, 아마종실 처리구에서는 C18:3 n-3 과 총 C18 불포화지방산의 수소 첨가현상이 전지대두 처리구와 비교하여 높은 비율로 발생했다. 한편 사료 내 농후사료 비율이 높을 때 처리구에서 C18:2 n-6, C18:3 n-3 및 총 C18 불포화 지방산의 수소 첨가현상은 농후사료 저 처리구와 비교하여 감소되었다.

Rumen fermentation and performance of Hanwoo steers fed total mixed ration with Korean rice wine residue

  • Background: This study was conducted to evaluate the effects of adding Korean rice wine residue (RWR) in total mixed ration (TMR) on in vitro ruminal fermentation and growth performance of growing Hanwoo steers. Methods: For in vitro fermentation, the experimental treatments were Control (Con: 0 % RWR + TMR), Treatment 1 (T1: 10 % RWR + TMR), and Treatment 2 (T2: 15 % RWR + TMR). The rumen fluid was collected from three Hanwoo steers and mixed with buffer solution, after which buffered rumen fluid was transferred into serum bottles containing 2 g dry matter (DM) of TMR added with or without RWR. The samples were then incubated for 0 h, 12 h, 24 h, or 48 h at $39^{\circ}C$ and 100 rpm. For the in vivo experiment, 27 Hanwoo steers (6 months old) with an average weight of $196{\pm}8.66kg$ were subjected to a 24-week feeding trial. The animals were randomly selected and equally distributed into three groups. After which the body weight, feed intake and blood characteristics of each group were investigated. Results: The pH of the treatments decreased significantly relative to the control during the 12 h of incubation. Total gas production and ammonia nitrogen ($NH_3-N$) was not affected by RWR addition. The total volatile fatty acid (VFA) was lower after 24 h of incubation but at other incubation times, the concentration was not affected by treatments. Feed cost was 8 % and 15 % lower in T1 and T2 compared to control. Blood alcohol was not detected and a significant increase in total weight gain and average daily gain were observed in Hanwoo steers fed with RWR. Conclusion: Overall, the results of this study suggest that TMR amended with 15 % RWR can be used as an alternative feed resource for ruminants to reduce feed cost.

Ensiled or Oven-dried Green Tea By-product as Protein Feedstuffs: Effects of Tannin on Nutritive Value in Goats

  • Ensiled or oven-dried green tea by-products (GTB) were evaluated in goats for their nutritive potential as protein feedstuffs based on in vitro and in vivo digestibility. To elucidate the effects of tea tannin on in vitro digestibility, polyethylene glycol (PEG) was used as a tannin binding agent. Both ensiled and dried GTB contained 31.9 to 32.6% of crude protein (CP) on a dry matter (DM) basis. Phenolics and tannins in soybean meal and alfalfa hay were low or not detected, but they were high in both ensiled and dried GTB (7.3-10.1% DM as total extractable tannins). In vitro protein digestibility in the rumen ranked: soybean meal>alfalfa hay cube>ensiled GTB = dried GTB. The protein digestibility post-ruminally of these feedstuffs showed a similar trend to that in the rumen, but the digestibility of ensiled GTB was significantly higher than that of dried GTB. Addition of PEG improved the in vitro protein digestibility of both kinds of GTB in the rumen and post-ruminally, indicating that tannins suppressed the potential protein digestibility of GTB. The increased protein digestibility by PEG addition was not significantly different between ensiled and dried GTB in the rumen, but the percentage increment of ensiled GTB was higher than dried GTB post-ruminally. In the in vivo digestibility trial, ensiled and dried GTB were offered to goats as partial substitutes for soybean meal and alfalfa hay cubes. Offering both GTB to goats as 5-10% on a DM basis did not affect nutrient digestibility, ruminal pH, volatile fatty acids, and ammonia concentration. However, the eating time of the GTB-incorporated diet was longer than that of the basal diet. It took 1.4 and 1.6 times longer than the control diet, to eat the diet completely when GTB silage was offered at 5 and 10% levels, respectively, of the total diet. These results show that ensiled and dried GTB are useful as partial substitutes for soybean meal and alfalfa hay cubes for goats with respect to nutritive value. Because of lessened palatability, it is recommended that GTB be incorporated into the diet at 5% on a DM basis.
