The present study was conducted to examine the fermentation characteristics and effective degradability (ED) in the rumen, and whole tract digestibility of whole crop silage based TMR in comparison with conventional separate feeding of concentrate and roughage. Three ruminally fistulated non-lactating Holstein cattle were used in a 3 $\times$ 3 Latin square design. The cattle were fed 8kg of whole crop barley silage based TMR (BS-TMR) or 8kg (DM basis) of whole crop rye silage based TMR (RS-TMR) twice (08:00 and 18:00) daily in an equal amount. The cattle were also fed concentrate (5.6kg) and rice straw (1.4kg) seperately (DM basis, Control) twice daily in an equal amount. The both silages were included in TMR at 20% level (as fed basis). pH in the rumen fluid was not influenced by the diets but was slightly higher from TMR than from control. No difference was found in ammonia-N concentration between diets. Total VFA concentration was relatively increased in the cattle fed RS-TMR to the other diets up to 6h post feeding. The proportion of acetate was increased in the TMR feeding at right before feeding (0 h, p<0.005) and 9 h (p<0.048) post feeding compared with control. Propionate proportion was increased (p<0.046) in both TMRs while butyrate proportion was increased (p<0.029) at 1h post feeding compared to other diets. Effective degradability (ED) of DM and CP of RS-TMR was relatively increased to other diets, and EDNDF of both TMRs was higher than that of control diet due to the increased parameters b (p<0.039) and c (p<0.006) in TMR treatments. Whole track digestibility of most components in the TMRs was slightly increased compared to that in control diet, and RS-TMR had a tendency to be increased whole track digestibility except for NDF compared to BS-TMR. Based on the results observed from the present study, nutrient availability of whole crop silage based TMR looked slightly better than conventional separate feeding of concentrate and rice straw, mainly due to the improved stabilization of fermentation in the rumen and increased NDF digestibility of whole crop silage in TMR.
This study was conducted to estimate the in vitro fermentation characteristics and in situ degradabilities of total mixed rations fermented by the synbiotic co-cultures composed of various anaerobic microorganisms in the rumen of cow. Seventy two TMR bags (4 treatments $\times$ 6 fermentation days $\times$ 3 replications) were manufactured for in vitro and in situ experiments. The experiment was composed of four treatments including the control, the mould and bacteria synbiotics (T1), the mould and yeast synbiotics (T2) and the bacteria and yeast synbiotics (T3). Each treatment had six fermentation days (1, 3, 5, 7, 14, 21 day) with three replications. Two rumen cannulated Holstein cows (550 ㎏ of mean body wt) were used for in situ trial, and a total of 96 nylon bags were retrieved from the rumen according to eight fermentation times (1, 3, 6, 9, 18, 24, 48 and 72 hr). The mean fermentation temperatures of TMRs by supplementation of anaerobic micoorganism co-cultures ranged from $22.97^{\circ}C$ to $26.07^{\circ}C$, and tended to increase steadily during the entire period. pH values of the F-TMRs ranged from 4.39 to 4.98 and tended to decrease with the extension of the fermentation period, and decreased by supplementation of synbiotics (p<0.05). The ammonia concentrations of F-TMRs were not affected by addition of synbiotic co-cultures during the early fermentation period (within 7 days), but was lowest (p<0.05) in T3 during the late fermentation periods (after 14 days). Lactic acid concentration of F-TMR was lowest in T3 at 1 day of fermentation, but was not different from treatments in the other fermentation days. Microbial growth rates of F-TMR reached a peak at 7 days of fermentation, and afterward tended to decrease. In in situ experiment, the DM disappearance rates were higher in T1 than the control during early fermentation times (within 3 hours), but was vice versa at 48 hours of fermentation (p<0.05). There was no significant difference in effective DM degradability among treatments. NDF and ADF disappearance rates in situ were similar to those of DM. From the above results, the supplementation of synbiotics, particularly the mould and bacteria synbiotics, resulted in improving the pH and concentration of lactic acid of F-TMR as parameters of fermentation compare to the control, and also had higher in situ disappearance rates of DM, NDF and ADF than the control at early fermentation time. However, effective DM degradability was not affected by supplementation of synbiotics.

A study was conducted with the objective of evaluating the nutritive value of some browse species from Kenya. The species evaluated included: Bauhinia alba, Bauhinia variegata, Bridelia micrantha, Calliandra calothyrsus, Carisa edulis, Cratylia argentea, Gliricidia sepium, Lantana camara, Maerua angolensis, Sesbania micrantha and S. sesban. The browses were evaluated by their chemical composition including phenolics, in vitro gas production and tannin activity (tannin bioassay). All the species had high crude protein content (149-268 g/kg DM) and low NDF content (239-549 g/kg DM). The feeds had varying contents of total extractable tannins (TET) ranging from low (3-22 mg/g DM), moderate (42-58 mg/g DM) and high (77-152 mg/g DM). Calliandra calothyrsus had the highest tannin content. Significant (p<0.05) variation in gas production was recorded among the species. Sesbania micrantha had the highest (p<0.05) potential gas production while Gliricidia sepium had the highest (p<0.05) rate of gas production. Use of polyethylene glycol (PEG 6000), to assess the adverse affect of tannins, indicated that tannins in browse species with high tannin content had inhibitory effects on rumen microbial fermentation as indicated by the gas production. Estimated organic matter digestibility and metabolizable energy also increased with PEG addition. The results of this study indicate that such Kenyan browse species have the potential to be used as feed supplements for ruminant animals.
This study was conducted to evaluate the effect of the thickness of corn flakes produced by pressurized steam chamber (PSC) on rumen fermentation characteristics and nutrient degradability in Hanwoo and Holstein cows. Corn flakes were treated by PSC, in three groups based on corn flake thickness: < 2.5 mm (T1), 2.5-3.0 mm (T2), and > 3.0 mm (T3). Corn flake thickness significantly influenced pH (p < 0.01) and propionate concentration (p < 0.05) and slightly but not significantly influenced acetate, butyrate, and total-volatile fatty acids (T-VFA) concentrations. The dry matter (DM) degradability increased significantly with a reduction in corn flake thickness (p < 0.01), being significantly greater in T1 and T2 than T3 groups (p < 0.01) and similar between T1 and T2 groups throughout whole incubation time. Also, starch degradability was the lowest in T3 groups than others (p < 0.01). Thus, the present results showed that considering the production efficiency and economic feasibility, the optimal thickness of corn flakes produced in a PSC is recommended to be 2.5-3.0 mm.
The study was conducted to evaluate and compare the effects of feed additives on ruminal fermentation, nutrient digestibility and performance of lambs fed diets containing 60% wet brewers grains (WBG). In Experiment 1, two simultaneous trials were conducted. Fifty intact ($20.2{\pm}0.8kg\;BW$) lambs were used in a feedlot trial and 10 (rumen cannulated; $32{\pm}1kg\;BW$) in a digestion trial. The pH, volatile fatty acids (VFA) and ammonia-N in lambs were also estimated. Lambs were randomly assigned to one of five diets: i) without additives (Con), ii) with 1% bicarbonate (Bic), iii) with 1% bentonite (Ben), iv) with 33 mg/kg monensin (Mon) and v) with 200 mg/kg fibrolityc enzymes (Enz). In Experiment 2, 120 RambouilletPelibuey intact male lambs ($19.5{\pm}1.5kg\;BW$) were used in a feedlot trial and randomly assigned to four diets: i) without additives (control), ii) with 1% Bic, iii) with 33 mg/kg Mon and iv) with 1% Bic and 33 mg/kg Mon. In Experiment 1, lambs fed diets containing Bic or Mon had significantly higher final weight, DMI, ADG than other lambs. However, apparent DM, OM, CP, NDF and ADF digestibilities and ruminal individual VFA content were similar (p>0.05) among treatments. Conversely, treatmentcollection period interaction was significant for ruminal pH and NH3. In Experiment 2, lambs fed diets containing a Bic and Mon combination had significantly higher final weight, DMI and ADG. It is concluded lambs fed Bic or Mon or Bic and Mon combination had better performance characteristics than lambs on Ben or Enz.
The effect of pH on the fermentation characteristics and the formation of cis-9, trans-11 conjugated linoleic acid (CLA) and trans-11 octadecenoic acid by mixed ruminal bacteria was examined in vitro when incubated with linseed or rapeseed. Concentrate (1%, w/v) with ground linseed (0.6%, w/v) or rapeseed (0.5%, w/v) was added to 600 ml mixed solution of strained rumen fluid with artificial saliva (1:1, v/v), and was incubated anaerobically for 12 h at $39^{\circ}C$. The pH of culture solution was maintained at level close to 4.5, 5.3, 6.1 and 6.9 with 30% $H_2SO_4$ or 30% NaOH solution. pH increment resulted in increases of ammonia and total volatile fatty acid (VFA) concentration in culture solutions containing both oilseeds. Fermentation did not proceeded at pH 4.5. Molar proportion of acetate decreased but that of propionate increased as pH increased when incubated with oilseeds. While the hydrogenating process was very slow at the pH range of 4.5 to 5.3, rapid hydrogenation was found from the culture solutions of pH 6.1 and 6.9 when incubated with linseed or rapeseed. As pH in culture solution of linseed or rapeseed increases proportions of oleic acid (cis-9 $C_{18:1}$) and trans-11 octadecenoic acid increased but those of linoleic acid and linolenic acid decreased. The CLA proportion increased with pH in culture solution containing rapeseed but CLA was mostly not detected from the incubation of linseed.
An in vitro study was conducted to determine the effect of nitrate-nitrogen used as a sole dietary nitrogen source on ruminal fermentation characteristics and microbial nitrogen (MN) synthesis. Three treatment diets were formulated with different nitrogen sources to contain 13% CP and termed i) nitrate-N diet (NND), ii) urea-N diet (UND), used as negative control, and iii) tryptone-N diet (TND), used as positive control. The results of 24-h incubations showed that nitrate-N disappeared to background concentrations and was not detectable in microbial cells. The NND treatment decreased net $CH_4$ production, but also decreased net $CO_2$ production and increased net $H_2$ production. Total VFA concentration was lower (p<0.05) for NND than TND. Suppression of $CO_2$ production and total VFA concentration may be linked to increased concentration of $H_2$. The MN synthesis was greater (p<0.001) for NND than UND or TND (5.74 vs. 3.31 or 3.34 mg/40 ml, respectively). Nitrate addition diminished methane production as expected, but also increased MN synthesis.
An in vitro study was conducted to examine the effect of addition level of concentrate on fermentation characteristics and long-chain unsaturated fatty acids composition, especially conjugated linoleic acid (CLA) and trans-octadecenoic acid (t-FA) by mixed ruminal bacteria when incubated with linseed or rapeseed. Four levels (0.83, 1.25, 1.67 and 2.08%, w/v) of concentrate and ground oilseeds (linseed or rapeseed; 0.83%, w/v) were added to mixed solution of strained rumen fluid with artificial saliva (1:1, v/v) in the glass jar with a glass lid equipped with stirrer, and was incubated anaerobically for 24 h at $39^{\circ}C$. Addition level of concentrate slightly reflect on pH and ammonia concentration of the culture solution at the various incubation times when incubated with both linseed and rapeseed. Total VFA concentration slightly increased with incubation times and concentrate levels for incubations with oilseeds. While CLA composition had a clearly increasing trend with incubation time when incubated with linseed, percent CLA was relatively stable when incubated with rapeseed. Percent CLA, however, had a clearly decreasing trend with concentrate level throughout incubation times with significances at 3 h incubations when incubated with linseed (p<0.038) and rapeseed (p<0.0009). The differences in compositions of t-FA were relatively small among concentrate levels for both incubations with linseed and rapeseed. The ratios of t-FA to CLA were lower for linseed with increased proportion of CLA than for rapeseed.
The aim of this study was to evaluate the effect of incorporating wet sorghum distiller's grains (WSDG) as part of their diet on the lactating performance of dairy cows. Twenty-seven Holstein milking cows were selected, all in the early lactating stage, with an average weight of 550 kg, and producing an average of 30 kg of milk daily. The cows were divided into three groups according to milk yield and lactation and were fed different total mixed rations. The diets were formulated according to NRC (1989) recommendations in three rations to (1) control diet, (2) 15% WSDG diet and (3) 30% WSDG diet. The three different diets were all formulated as iso-nitrogen and iso-energetic diets. After one week adaptation period, the experimental feeding was conducted for 8 weeks. Three ruminal cannulated cows were also examined in order to investigate ruminal fermentation of the three total mixed rations. The results showed that the milk yield, as corrected to the 4.0% fat standard, had no significant difference among the control, 15% WSDG and 30% WSDG treatment groups (p>0.05). The daily dry matter intake of the control group was higher than the other groups (p<0.05). with respect to milk composition, milk fat, milk protein and total solids, there was no significant difference among the treatment groups (p>0.05). The energy efficiency of the 30% WSDG group were significantly higher than the other treatment groups (p<0.05). Ruminal pH value showed no difference among the treatment groups (p<0.05). Ammonia-nitrogen concentration in the control group was higher than the other treatment groups (p<0.05). The concentration of total ruminal volatile fatty acid was similar in all three dietary groups.
This study was conducted to determine effects of salinomycin (SL) on feeding-induced changes in glucose kinetics and blood VFA concentrations in sheep fed a high-roughage diet. Four sheep were fed the diet with or without 20 mg/kg diet of SL once daily for 21 d. Glucose entry and utilization rates were determined during the prefeeding and 3 h postfeeding periods, using a [$^{13}C_6$]glucose dilution method and non-steady state equations. Ruminal characteristics and concentrations of blood VFA, plasma glucose and insulin were also measured during the same periods. A feeding-induced increase in ruminal total VFA concentration tended to be inhibited (p<0.10) with SL, although ruminal pH was unaffected (p>0.10) with SL or by feeding. Salinomycin decreased (p<0.05) acetate proportion and increased (p<0.05) propionate proportion in the rumen, but did not modify these changes in response to feeding (p>0.10). A feeding-induced increase in blood acetate concentration was attenuated (p<0.05) with SL. Salinomycin tended to increase (p<0.10) blood propionate concentration without modifying its response patterns to feeding (p>0.10). Plasma concentrations of glucose or insulin were unaffected (p>0.10) with SL. Salinomycin tended to enhance (p<0.10) glucose entry and utilization rates. Feeding also enhanced (p<0.01) both rates, whereas their interactive effect was not detected (p>0.10). We conclude that SL possibly enhances whole body glucose entry and utilization with an increase in blood propionate concentration in sheep given a high-roughage diet, although SL does not appear to affect their responses to feeding.
