AtERF11 is a positive regulator for disease resistance against a bacterial pathogen, Pseudomonas syringae, in Arabidopsis thaliana (애기장대 AtERF11 유전자에 의한 Pseudomonas syringae에 대한 병 저항성 유도)

  • AvrRpt2 protein triggers hypersensitive response (HR) and strong disease resistance when it is translocated from a bacterial pathogen Pseudomonas sp. to host plant cells containing a cognate RPS2 resistance protein through Type III Secretion System (TTSS). However, AvrRpt2 protein can function as the effector that suppresses a basal defense and enhances the disease symptom when functional RPS2 resistance protein is absent in the infected plant cells. Using Affymetrix Arabidopsis DNA chip, we found that many genes were specifically regulated by AvrRpt2 protein in the rps2 Arabidopsis mutant. Here, we showed that expression of AtERF11 that is known as a member of B1a subcluster of AP2/ERF transcription factor family was down regulated specifically by AvrRpt2. To determine its function in plant resistance, we also generated the Arabidopsis thaliana transgenic plants constitutively overexpressing AtERF11 under CaMV 355 promoter, which conferred an enhanced resistance against a bacterial pathogen, Pseudomonas syringae pv. tomato DC3000. Thus, these results collectively suggest that AtERF11 plays a role as a positive regulator for disease resistance against biotrophic bacterial pathogen in plant.

Two-Stage Biological Hydrogen Production by Rhodopseudomonas palustris P4 (Rhodopseudomonas palustris P4에 의한 이 단계(Two-stage) 생물학적 수소생산)

  • The integrated or the two-stage (dark anaerobic and photosynthetic) fermentation processes were compared for the hydrogen production using purple non-sulfur photosynthetic bacteria, Rhodopseudomonas palustris P4. Cell growth, pH changes and organic acids and bacteriochlorophyll contents were monitored during the processes. Culture broth of Rps. palustris P4 exhibited dark-red during the photosynthetic culture condition, while yellow under the anaerobic condition without light. Rps. palustris P4 grown at the photosynthetic condition evolved 0.38 and 1.33 ml $H_2$/mg-dcw during the dark and the light fermentation, respectively, which were totally 1.71 ml $H_2$/mg-dcw at the two-stage fermentation. The rate of hydrogen production using Rps. palustris P4 grown under the dark anaerobic condition was 2.76 ml $H_2$/mg-dcw which consisted of 0.46 and 2.30 ml $H_2$/mg-dcw from the dark and the photosynthetic fermentation processes, respectively. Rps. palustris P4 grown under dark anaerobic conditions produced $H_2$ 1.6 times higher than that of grown under the photosynthetic condition. However, total fermentation period of the former was 1.5 times slower than that of the latter, because the induced time of hydrogen production during the photosynthetic fermentation was 96 and 24 hours when the seed culture was the dark anaerobic and photosynthetic, respectively. The integrated fermentation process by Rps. palustris P4 produced 0.52 ml $H_2$/mg-dcw(1.01 mol $H_2$/mol glucose), which was 20% of the two-stage fermentation.

Molecular Phylogenetic Study of Anemone pendulisepala (Ranunculaceae) (태백바람꽃(Anemone pendulisepala, Ranunculaceae)의 분자계통학적 검토)

  • Anemone pendulisepala, recently described as a new species, is distributed in sympatry with A. reflexa, A. amurensis, and A. raddeana at Mt. taebeark and Mt. Baekdu. Anemone pendulisepala was previously proposed to be a hybrid species between A. reflexa and A. amurensis becaue it displavs overlapping features with them in involucre shape, petiole length, sepal apex and xylem shape, To verify the taxonomic status and to examine the hybridity of A. pendulisepala, sequences of ITS region of nuclear ribosomal DNA and the psba-trnH, rps16 and trnLF region of cpDNA from 36 accessions of 5 taxa including outgroup were analyzed. In maximum parsimony tree based on ITS sequences, A. pendulisepala had the same sequences of A. reflexa and was clustered with monophyletic A. amurensis, and then A. raddeana. Anemone pendulisepala was distinguished from the other taxa by having four base insertion in rps16 region, two species-specific bases and insertion in trnLF region. In the phylogenetic trees of combined cpDNA, A. pendulisepala showed monophyly with the bootstrap 100%. Anemone pendulisepala exhibited no polymorphism and shared no sequences with putative parental or related taxa examined in this study. Molecular data suggest that A. pendulisepala should be a distinct species, and no evidence of the hybridization was detectcd.

Estimating the Investment Value of Fuel Cell Power Plant Under Dual Price Uncertainties Based on Real Options Methodology (이중 가격 불확실성하에서 실물옵션 모형기반 연료전지 발전소 경제적 가치 분석)

  • Hydrogen energy is emerging as an important means of carbon neutrality in the various sectors including power, transportation, storage, and industrial processes. Fuel cell power plants are the fastest spreading in the hydrogen ecosystem and are one of the key power sources among means of implementing carbon neutrality in 2050. However, high volatility in system marginal price (SMP) and renewable energy certificate (REC) prices, which affect the profits of fuel cell power plants, delay the investment timing and deployment. This study applied the real option methodology to analyze how the dual uncertainties in both SMP and REC prices affect the investment trigger price level in the irreversible investment decision of fuel cell power plants. The analysis is summarized into the following three. First, under the current Renewable Portfolio Standard (RPS), dual price uncertainties passed on to plant owners has significantly increased the investment trigger price relative to one under the deterministic price case. Second, reducing the volatility of REC price by half of the current level caused a significant drop in investment trigger prices and its investment trigger price is similar to one caused by offering one additional REC multiplier. Third, investment trigger price based on gray hydrogen and green hydrogen were analyzed along with the existing byproduct hydrogen-based fuel cells, and in the case of gray hydrogen, economic feasibility were narrowed significantly with green hydrogen when carbon costs were applied. The results of this study suggest that the current RPS system works as an obstacle to the deployment of fuel cell power plants, and policy that provides more stable revenue to plants is needed to build a more cost-effective and stable hydrogen ecosystem.

Efficacy of alginate microsphere oral vaccine against Miamiensis avidus (Ciliophora: Scuticociliatida) in olive flounder (Paralichthys olivaceus) (알지네이트 코팅 Miamiensis avidus (Ciliophora: Scuticociliatida) 경구백신의 넙치(Paralichthys olivaceus)에 대한 효능평가)

  • The efficacy of the alginate microsphere (Alginate MS) oral vaccine against Miamiensis avidus in olive flounder (Paralichthys olivaceus) was confirmed through challenge infections by both immersion and injection routes. In trial 1, the formalin-inactivated M. avidus coated with alginate, designated as 'IMa+Alginate MS' group, and the IMa group were administered with vaccines mixed with feed, with a total antigen dose of 3.75 × 106 cells/fish. When challenged with immersion infection at five weeks post vaccination, the relative percent survival (RPS) in the IMa+Alginate MS group was 50% (immersed in 50% seawater) and 37.5% (immersed in 100% seawater). The group that received only IMa showed a low survival rate. In trial 2, the antigen was fed mixed with feed at a total dose of 2.38 × 106 cells/fish for 5 days. Two weeks after oral vaccination, fish were intraperitoneally injected for infection. The RPS in the IMa+Alginate MS group was 30.8%, while the IMa-only group showed no vaccine efficacy. At five weeks post vaccination, when subjected to challenge infection by immersion in 50% seawater, the IMa+Alginate MS group recorded a RPS of 42.9%, whereas the IMa group had a RPS of 14.3%. The results of this study indicate that coating M. avidus antigen with alginate can provide higher protection in olive flounder compared to administering the antigen alone.

Feature Extraction Based on Speech Attractors in the Reconstructed Phase Space for Automatic Speech Recognition Systems

  • In this paper, a feature extraction (FE) method is proposed that is comparable to the traditional FE methods used in automatic speech recognition systems. Unlike the conventional spectral-based FE methods, the proposed method evaluates the similarities between an embedded speech signal and a set of predefined speech attractor models in the reconstructed phase space (RPS) domain. In the first step, a set of Gaussian mixture models is trained to represent the speech attractors in the RPS. Next, for a new input speech frame, a posterior-probability-based feature vector is evaluated, which represents the similarity between the embedded frame and the learned speech attractors. We conduct experiments for a speech recognition task utilizing a toolkit based on hidden Markov models, over FARSDAT, a well-known Persian speech corpus. Through the proposed FE method, we gain 3.11% absolute phoneme error rate improvement in comparison to the baseline system, which exploits the mel-frequency cepstral coefficient FE method.
