Cymbopogon citratus (CC) and Perilla frutescens (PF) are known to exert various biological effects. However, their skin hydration and skin barrier effects remain unclear. This study investigated effects of their extracts on skin hydration and skin barrier and analysed the phenolic compounds. effects of these extracts on skin hydration in HaCaT cells showed that Hyaluronic acid production in cells treated with ethanol extracts was higher than that treated with water extracts for both CC and PF. HPLC was used to analyse 19 phenolic compounds in CC and PF ethanol extracts (CCE and PFE). Results revealed chlorogenic acid and p-coumaric acid in CCE and rosmarinic acid and caffeic acid in PFE. Expression levels of hyaluronan synthase 1 (HAS1), HAS2, HAS3, and aquaporin 3 (AQP3), which are related to skin moisturization, and filaggrin and loricrin, which are related to skin barrier were higher in cells treated with CCE than with PFE. CCE and PFE also increased expression of PPAR-a protein involved in skin moisturization and epidermal differentiation in a concentration-dependent manner. As major components of CCE, chlorogenic acid and p-coumaric acid increased PPAR-a protein expression. Thus, CCE and PFE could be used as functional cosmetic materials for skin hydration and skin barrier effects.

The objective of this study was to select the right species among Agastache in the family Lamiaceae as the result of growth analysis and functional substance analysis. Among 22 species of Agastache, five species (Agastache cana, Agastache foeniculum, Agastache rugosa 'Spike Blue', Agastache rugosa 'Spike Snow', Agastache rupestris) were selected by referring to United States Department of Agriculture (USDA)'s plant hardiness zone area and Korea's one based on USDAs. These plants were cultured at 24 ± 1℃ and 18 ± 1℃ (day and night temperatures, respectively) and 65 ~ 75% relative humidity in a hydroponic culture system for 4 weeks. In terms of growth analysis experiment, stem length, root length, leaf width, leaf area, leaf number, SPAD value, and fresh and dry weights of shoots and roots were measured. The results showed that A. rugosa SS and A. rugosa SB have a higher overall balance value than other species. When compared functional substance value (rosmarinic acid, tilianin, and acacetin) of five species, A. rugosa SS and A. rugosa SB are significantly higher than other species in Agastache. It seems to be directly proportional to the growth analysis results. When considering the growth and functional substance part comprehensively, A. rugosa SS and A. rugosa SB are the most optimal high-value species among Agastache.
Background: The purpose of this study was to investigate the anti-oral microbial activity and anti-inflammatory effects of rosmarinic acid (RA) in lipopolysaccharide (LPS)-stimulated MC3T3-E1 osteoblastic cells on a titanium (Ti) surface during osseointegration, and to confirm the possibility of using RA as a safe natural substance for the control of peri-implantitis (PI) in Ti-based dental implants. Methods: A disk diffusion test was conducted to confirm the antimicrobial activity of RA against oral microorganisms. In order to confirm the anti-inflammatory effects of RA, inflammatory conditions were induced with 100 ng/ml of LPS in MC3T3-E1 osteoblastic cells on the Ti surface treated with or without 14 ㎍/ml of RA. The production of nitric oxide (NO) and prostaglandin E2 (PGE2) in LPS-stimulated MC3T3-E1 osteoblastic cells on the Ti surface was confirmed using an NO assay kit and PGE2 enzyme-linked immunosorbent assay kit. Reverse transcription polymerase chain reaction and western blot analysis were performed to confirm the expression of interleukin (IL)-1β and tumor necrosis factor (TNF)-α in total RNA and protein. Results: RA showed weak antimicrobial effects against Streptococcus mutans and Escherichia coli, but no antimicrobial activity against the bacteria Aggregatibacter actinomycetemcomitans and the fungus Candida albicans. RA reduced the production of pro-inflammatory mediators, NO and PGE2, and proinflammatory cytokines, TNF-α and IL-1β, in LPS-stimulated MC3T3-E1 osteoblastic cells on the Ti surface at the protein and mRNA levels. Conclusion: RA not only has anti-oral microbial activity, but also anti-inflammatory effects in LPS-stimulated MC3T3-E1 osteoblasts on the Ti surface, therefore, it can be used as a safe functional substance derived from plants for the prevention and control of PI for successful Ti-based implants.
The objective of this study was to determine the proper night interruption of photoperiods and dark periods for accumulating bioactive compounds of Agastache rugosa without decreasing plant growth. Five-week-old seedlings were transplanted in a DFT system with white LEDs. A. rugosa was treated with night interruption time treatments of 18:1:2:3, 18:2:2:2, 18:3:2:1 (light:dark:light:dark), and 20:4 (control) for 4 weeks. There were no significant differences except for leaf length, leaf width, and the number of flowers. The content of antioxidants in the shoot of A. rugosa was high in tilianin and acacetin, and the content of rosmarinic acid (RA) was significantly higher in the underground part. The RA content per dry weight of A. rugosa was 47.92 and 51.46% higher than that of the control in 18:1:2:3 and 18:2:2:2, and tilianin and acactin per dry weight were significantly higher in 18:3:2:1. There was no significant difference in growth due to the same day light integral, but 18:2:2:2 showed high total polyphenol contents. Therefore, it is thought that the effect of increasing secondary metabolites of A. rugosa without degradation of growth can be expected through night interruption treatment in plant factory cultivation systems using artificial light.
Rosmarinic acid (RA) is a phenolic ester that protects human keratinocytes against oxidative damage induced by ultraviolet B (UVB) exposure, however, the mechanisms underlying its effects remain unclear. This study aimed to elucidate the cell signaling mechanisms that regulate the antioxidant activity of RA and confirm its cyto-protective role. To explore the signaling mechanisms, we used the human keratinocyte cell line HaCaT and SKH1 hairless mouse skin. RA enhanced glutamate-cysteine ligase catalytic subunit (GCLC) and glutathione synthetase (GSS) expression in HaCaT cells in a dose- and time-dependent manner. Moreover, RA induced nuclear factor erythroid-2-related factor 2 (NRF2) nuclear translocation and activated the signaling kinases protein kinase B (AKT) and extracellular signal-regulated kinase (ERK). Treatment with the phosphatidylinositol 3-kinase (PI3K) inhibitor LY294002, the ERK inhibitor U0126, and small interfering RNA (siRNA) gene silencing suppressed RA-enhanced GCLC, GSS, and NRF2 expression, respectively. Cell viability tests showed that RA significantly prevented UVB-induced cell viability decrease, whereas the glutathione (GSH) inhibitors buthionine sulfoximine, LY294002, and U0126 significantly reduced this effect. Moreover, RA protected against DNA damage and protein carbonylation, lipid peroxidation, and apoptosis caused by UVB-induced oxidative stress in a concentration-dependent manner in SKH1 hairless mouse skin tissues. These results suggest that RA protects against UVB-induced oxidative damage by activating AKT and ERK signaling to regulate NRF2 signaling and enhance GSH biosynthesis. Thus, RA treatment may be a promising approach to protect the skin from UVB-induced oxidative damage.
Rosmarinus officinalis extracts had a significant antimicrobial activity against Helicobacter pyori. Total phenolic contents and inhibition zone of rosemary extracts were estimated to be 25.7 mg/g and 14 mm at $200{\mu}g/mL$ of phenolic contents, respectively. The Sephadex LH-20 and MCI-gel CHP-20 column chromatographic separations for the phenolic extracts from R. officinalis leaves led to isolation of five acids, whose structures were determined as protocatechuic acid (A), coumaric acid (B), caffeic acid (C), chlorogenic acid (D), and rosmarinic acid (E), from interpretation of spectroscopic data including nagative fast atom bombardment (FAB)-mass, $^1H$-NMR, $^{13}C$-NMR, and IR. All isolated compounds were tested for antimicrobial activity against H. pyori. The purified single compound showed less antimicrobial activity against H. pylori than the mixed purified compounds, which generate A+B, A+E, C+D, C+E (each $200{\mu}g/disc$) excellent as large clear zone by synergy effect. These results indicate rosemary extracts are preventive agents against H. pyori.
Three phenolic compounds, rosmarinic acid (1), methyl rosmarinate (2), ethyl rosmarinate (3), and two flavonoids, luteolin (4), luteolin-7-O-$\beta$-D-glucuronide methyl ester (5) were isolated from the aerial part of Lycopus lucidus (Labiatae). Their structures were determined by chemical and spectral analysis. Compounds 1-5 exhibited potent antioxidative activity on the NBT superoxide scavenging assay. The $IC_{50}$ values for compounds 1-5 were 2.59, 1.42, 0.78, 2.83, and 3.05 $\mu\textrm{g}$/mL respectively. In addition, five compounds were isolated from this plant for the first time.

Perilla seed oil is considered to be a rich source of omega-3 fatty acids and phenolic compounds, which have various health benefits. To improve its oil content and nutritional quality, 29 varieties of perilla have been bred, but their nutritional qualities, including the fatty acid and phenolic compound contents, have not been investigated. Therefore, in this study, the seed metabolite profiles, including the fatty acids, of these 29 perilla varieties were analyzed. Palmitic acid, stearic acid, linoleic acid, linolenic acid, rosmarinyl glucoside, and rosmarinic acid were identified as the major metabolites of perilla. The fatty acid contents were higher in the Okdong, Deulsaem, Daesil, Joim, Yeupsil, Danjo, and Joongmo5103 varieties (in order). However, the total phenolic compounds and antioxidative activities of Okdong and Deulsaem were relatively low, whereas the fatty acid and total phenol contents of Joim, Yeupsil, and Danjo were relatively high. These results may be useful for comparing the nutritional quality of the 29 perilla varieties.
This study was performed to evaluate the antioxidant and antimicrobial activity of methanol extract from Rosmarinus officinalis L. and its fractions. The ethyl acetate fraction of rosemary had a higher antioxidant activity in both DPPH ($3.22\;{\mu}g/ml$) and ABTS ($5.05\;{\mu}g/ml$) compared to other extracts and fractions. Based on the results of the FRAP assay, the ethyl acetate fraction of rosemary showed a value of $5.9{\pm}0.3\;{\mu}M/{\mu}g$, and buthanol fraction and rosmarinic acid exhibited values of $4.8{\pm}0.2\;{\mu}M/{\mu}g$ and $5.1{\pm}0.1\;{\mu}M/{\mu}M$, respectively. Measurements of the antimicrobial activities of the extracts, fraction against gram positive, negative bacteria revealed that the methanol extract, hexane, ethyl acetate, and chloroform fraction of rosemary caused Staphylococcus aureus and Escherichia coli to form clear zones greater than 12 mm. Furthermore, the methanol extract and chloroform fraction showed high antibacterial activity, with inhibition zone exceeding 13 mm. The methanol extract and chloroform fraction of rosemary had broad antimicrobial spectrums and low MIC values. Therefore, methanol extracts of rosemary could serve as potential antibacterial agents to inhibit pathogen growth in food and hand sanitizers.
Perilla frutescens is known as the herb helps digestion, tonifies stomachache, and decreases fever in oriental medicine. And it is reported it possess the anti-pyretic effect, anti-inflammatory effect, anti-allergy effect, anti-tumor effect etc. The components isolated from this herb consist of perilla aldehyde, d-limonene, ${\alpha}-pinene$, cyanin, linoleic acid, palmitic acid, menthol, rosmarinic acid and luteolin etc. But there is no effective tools to determine the quality of this herb. In this study, we aimed to analyze the changes of liquid chromatogram pattern, one of major standardization method, to determine the quality of Perilla frutescens.
