Inhibitory Effect of Rosa davurica Pall. on LPS-mediated Nitric Oxide Productionvia NF-κB signaling (NF-κB signaling을 통한 Rosa davurica Pall.의 NO 생성 저해 효과)

  • This study was designed to determine the activities of Rosa davurica Pall. leaf extract and their regulatory mechanisms in macrophage inflammation. Anti-inflammatory potential of Rosa davurica Pall. leaf extract was evaluated by measuring the nitric oxide (NO) release and inducible nitric oxide synthase (iNOS) synthesis in lipopolysaccharide (LPS)-treated macrophage Raw 264.7 cells. Rosa davurica Pall. leaf extract potently inhibited LPS-induced NO release in a dose dependent manner. However, cell viability decreased to about 50% at high dose of 500 ㎍/ml, resulting in cytotoxicity. LPS-induced iNOS protein expression was also reduced significantly after treatment with Rosa davurica Pall. leaf extract. Furthermore, extract of Rosa davurica Pall. attenuated LPS-mediated phosphorylation of IκB and nuclear factor (NF-κB). Suppression of NF-κB signaling by treatment with PDTC, an NF-κB specific inhibitor, accelerated the inhibition of NO production and iNOS protein expression. These results suggest that Rosa davurica Pall. exhibits the anti-inflammatory potential in LPS-induced macrophage inflammation, partly through inhibition of NO production by down-regulation of NF-κB signaling.

Anti-oxidant and Inhibitory Activity on NO Production of Extract and its Fractions from Rosa davurica Pall. Leaves (생열귀나무 잎 추출물과 분획물의 항산화 및 NO생성 억제 활성)

  • In the present study, we investigated biological activities of Rosa davurica Pall. leaves in order to evaluate the possibility as a natural biomaterial. The 80% ethanol extract and its subsequent fractions of Rosa davurica Pall. leaves were prepared using several solvents with different polarities. The extraction yield was 33.4, 36.6, 25.2, 18.7, 5.8 and 5.8% in ethanol extract, aqueous, butanol, ethyl acetate, n-hexane, chloroform fractions, respectively. The ethyl acetate fraction (661.38 mg/g), butanol fraction (396.68 mg/g) and 80% ethanolic extract (239.54 mg/g) has higher total polyphenol contents than other fractions. The antioxidant activity was detected in ethanolic extract, ethyl acetate and butanol fractions. The ethyl acetate fraction showed the highest levels of DPPH radical scavenging activity ($IC_{50}$, $4.77{\mu}g/m{\ell}$). Moreover, the ethyl acetate fraction significantly inhibited production of NO in LPS-stimulated macrophage RAW 264.7 cells without cytotoxicity. These results indicate that 80% ethanol extract and its fractions of Rosa davurica Pall. leaf, especially ethyl acetate fraction, have the properties of anti-oxidant and anti-inflammation, suggesting leaf of Rosa davurica Pall. may be a candidate for natural and functional materials.

Hepatoprotective Activities of Rosa davurica Root Extract in Rats Intoxicated with Bromobenzene (브로모벤젠으로 유도된 간독성 흰쥐에서 생열귀나무 뿌리의 간보호활성)

  • To investigate hepatoprotective activities of the root extract of Rosa davurica, the activities of hepatic enzymes, aminopyrine N-demethylase, aniline hydroxylase, glutathione S-transferase and epoxide hydrolase in rats intoxicated with bromobenzene were studied. Pretreatment with the methanol extract from the roots of Rosa davurica did not show any significant effects on the increases of the activities of aminopyrine N-demethylase and aniline hydroxylase, enzymes forming toxic epoxide by bromobenzene. There was no change in glutathione S-transferase activity by Rosa davurica. However, the activity of epoxide hydrolase, and epoxide-removing enzyme, was increased 33% by the administration of 500 mg/kg of the methanol extract. From the results, the protection of Rosa davurica against bromobenzene-induced hepatotoxicity is thought to be via enhancing the activity of epoxide hydrolase, an enzyme removing toxic epoxide rather than through epoxide-producing system.

  • Anti-human immunodeficiency virus (HIV) type I protease (PR) and anti-lipid peroxidation effects on Rosa davurica were investigated. Of the various parts tested from R. davurica, the water extracts of stem and leaves inhibited the HIV-1 PR activity by more than 45% at a concentration of $100\;{mu}g/mL$. Hyperoside from the percarp of title plant showed 25% inhibition on HIV-1 PR at $200\;{mu}M$. The methanol extract of the root of R. davurica reduced the level of lipid peroxides induced by bromobenzene in vivo.

  • This study was carried out to investigate the antioxidative activities of Rosa davurica Pall for the purpose of development of novel antioxidant from natural products. Antioxidant activities of four different parts of Rosa davurica Pall such as fruit, leaf, stem and root were examined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract from the root of Rosa davurica Pall showed the highest antioxidative activity among 16 samples tested. And, we also tested radical scavenging effects of 5 different extract compartments(Hexane, $CHCl_3$, EtOAc, BuOH and $H_2O$ fraction). EtOAc and BuOH fractions from the root of Rosa davurica Pall exhibited antioxidative activities higher to those of natural, ${\alpha}-tocopherol$ or synthetic antioxidants, BHT. The antioxidative substance of EtOAc fraction from the root of Rosa davurica Pall was successively purified with silica gel adsorption column chromatography and Sephadex LH-20 column chromatography. The purified active substance was isolated as crystal and identified as (+)-catechin by $^{l}H-NMR$ and $^{13}C-NMR$. This compound exhibited DPPH radical scavenging activity with the $IC_50$ value of $1.7\;{\mu}g/ml$. In the analysis of catechin content, the leaf extracts contained the highest catechin, and fruit extracts contained the lowest catechin. Considering antioxidative activity on DPPH assay, the extracts of Rosa davurica Pall showed a possibility to be used as a new material for natural antioxidant and functional food.

  • A triterpenoid glucoside and five phenolic compounds have been isolated from the roots and leaves of Rosa davurica. They were elucidated as rosamultin and gallic acid from the roots, and methyl gallate, gallic acid, protocatechuic acid, quercetin and hyperoside from the leaves of this plant, respectively.

  • The methanolic extract of Rosa davurica Pall. roots exhibited strong antioxidant activity in a 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging assay and was found to be a dose-dependent inhibitor of non-enzymatic formation of advanced glycation end products (AGEs), which are relevant to diabetes complications. HPLC-diode array detector (DAD) analysis of the R. davurica Pall. root extract led to the identification of four compounds: hydrocaffeic acid, catechin, epicatechin, and ellagic acid. Catechin was present in the largest amount and exhibited high antiglycation activity. A CYP3A4 assay was used to investigate potential interactions between drugs and the extract, and results suggest that the R. davurica Pall. root extract had moderate potential for interfering with drug metabolism. The R. davurica Pall. extract did not display anti-inflammatory activity on the level of that for tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in a lipopolysaccharide (LPS)-stimulated macrophage assay; however, the extract did exhibit low to moderate immunostimulatory activity in a pro-inflammatory macrophage assay. Therefore, we conclude that R. davurica Pall. root is a promising anti-AGE agent with low to moderate risks of associated inflammation or drug interaction.

Effect of split achene pericarp, Giberellin, and germination temperature on the germination of Rosa davurica Pall (생열귀나무 종자 발아에 있어서 개갑처리, 지배렐린, 발아온도의 효과)

  • This study was conducted to investigate the suitable conditions for seed germination of Rosa davurica PALL. In the characteristics of the fruit and seed of Rosa davurica, fruit length and width was 1.3cm and 0.9cm , respectively, and seed number was eighty-nine. Artificial and low temperature storage at 4$^{\circ}C$ increased the rate of split achene pericarp until 46.6% and storage at 15$^{\circ}C$ incubator decreased the rate of split achene pericarp (10.5%). The rate of seed germination of split achene pericarp in control at 15$^{\circ}C$ was 90% and of non-split achene in GA$_3$100ppm at 15$^{\circ}C$ was 36.8%. Average germination day of split achene pericarp seeds in GA$_3$150ppm at 2$0^{\circ}C$ was 4.2 days and non-split achene pericarp in GA$_3$100ppm at 2$0^{\circ}C$ was 7.3 days. Seed germination was not different between various concentrations of GA$_3$ treatments in split achene pericarp seeds but the rate of germination was more reduced in high concentration of 200ppm. Only the treatments of GA$_3$ was increased germination rate at 4$^{\circ}C$ and immature seed of Rosa davurica.

  • This study was conducted to determine the effect of cutting type, growth regulators and propagation media on the rooting and root growth of Roea davurica $P_{ALL}$. Three type of cutting, hardwood, half-softwood, softwood cutting and root cutting of Rosa davurica $P_{ALL}$, were used to study the rooting ability. There was no rooting in hardwood cutting while root cutting was appeared 100% of callus formation and rooting. The optimum conditions of softwood cutting for rooting were IAA 100ppm and rooton-F at vermiculite+perlite. The rate of rooting in treatment of rooton-F ranged from 10 to 60%, but such a good effect was not appeared in other growth regulators, IAA, NAA and IBA.

  • The objective of this study was carried out to investigate nutritional characteristics and biological activities effects of Korean leaf of Rosa davurica Pall. in vitro. They were extracted with methanol and then further fractionated to n-hexane, chloroform, ethyl-acetate, n-butanol and water from methanol extracts. Methods of the antigenotoxic used in this experiment were UVA/UVB absorption property and DPPH radical scavenge. The proximate compositions of leaves of Rosa dauvrica Pall were 67.5% of crude Moisture, 0.7% of crude fat, 6.8% of crude protein, 6.1% of crude ash, and 20.8% of crude fiber. The major minerals were K (1637.2 mg%), Ca (219.5 mg%), P (182.1 mg%), and Mg (135.1 mg%). Most of the fractions of methanol extract which leaves of Rosa dauvrica Pall. have strong absorbency at UVB region (308 nm) and UV A region (350nm). These fractions have a good absorbency property as synthetic filter and could be served as substitutes for synthetic UV sunscreen agents. All fractions (n-hexane, ethyl-acetate, n-butanol and water) from methanol extracts except chloroform fraction exhibited DPPH radical scavenging activity with IC$\_$50/ of 35.3, 6.0, 14.0, and 18.0 $\mu\textrm{g}$/mL.
