인삼의 뿌리절편, 줄기, 엽병에 Agrobacterium을 접종하여 고생장 모상근을 선발하고자 수행하였다. 인삼 뿌리 절편에서 A. rhizogenes R1000과 A. rhizogenes $A_4$에 의하여 모상근이 유도되었으며, 인삼의 줄기 및 엽병에서도 모상근이 유도되었다. 유도된 모상근은 rolC및 virC DNA절편의 확인으로 형질전환체임을 확인하였으며, 균이 제거된 모상근을 선발하였다. 선발된 모상근 300 세포주중 성장과 형태적 특징이 뚜렷한 11 모상근을 선발하였다. 이들 모상근은 주근이 비후된 것, 주근이 가는 것, 측근의 돌기가 많은 것, 측근의 생장이 높은 것 등의 특징을 나타내었다. 이들 11 모상근중 주근이 가늘고 측근의 생장이 왕성한 형태가 모상근의 생장이 좋았으며, 주근이 비후되고 측근의 돌기가 많은 형태는 생장이 낮았다. 본 연구에서 인삼 뿌리절편으로부터 고생장 모상근인 KGHR-1, KGHR-5, KGHR-8을 선발하였다.
Burdock (Arctium sp.) is known as a nutraceutical vegetable, especially in Japanese and Korean cuisine. While burdock plants are generally harvested for their tap roots, different parts of the plant are consumed as food or used as traditional medicines. This study investigated the physicochemical properties of the leaves, stems, roots, and peeled roots of the burdock plant based on their pH, soluble solid content, titratable acidity, color values, and mineral content. The pH differed significantly among the different plant parts, with the highest value in the leaves and the lowest in the stems. However, for the soluble solid content, the leaves had the lowest, while the peeled roots had the highest. The titratable acidity of the stems was significantly lower than that of the leaves, roots, and peeled roots. As regards the color values, the lightness value was highest for the stems, while the roots showed the highest redness value, followed by the peeled roots, and the leaves had the highest yellowness value. The leaves and stems contained almost three times more potassium than the roots and peeled roots. Thus, the higher content of different minerals in the leaves and stems of the burdock plant shows that these plant parts could be used as potential sources of dietary minerals.
The purpose of this investigation is to observe tha variation of vitamin $B_{12}$, nicotinic acid and folic acid contents in relation to the growing years of Panax Ginseng roots. The contents of the vitamins were estiamted microbiologically with Lactobacillus leichmannii, Lactobacillus arabinosus and Streptococcus faecalis, respectively. It is found that the content of vitamin $B_{12}$ in Panax Ginseng roots somewhat increases according to their growing years except 3-year-old roots. It is probable that the lower content in these roots should be due to the cultivating soil. The content of nicotinic acid in all the roots is not significantly different. The result of paper chromatography using the concentrated extract of the roots suggests that there exists some other substance besides nicotinic acid and nicotinamide, of which Rf value is 0.5 in comparison with the Rf 0.75 of nicotinic acid and nicotinamide. It is thought that this substance stimulate the growth of L. arabinosus. The content of folic acid is significantly different. The content level is the highest in the 4-year-old roots and the lowest in the 6-year-old roots.
Computing square, cube and n-th roots in general, in finite fields, are important computational problems with significant applications to cryptography. One interesting approach to computational problems is by using polynomial representations. Agou, Del$\acute{e}$eglise and Nicolas proved results concerning the lower bounds for the length of polynomials representing square roots modulo a prime p. We generalize the results by considering n-th roots over finite fields for arbitrary n > 2.
Induction and culture of hairy roots by Agrobacterium rhizogenes A4 were carried out in Platycodon grandiflorum DC. After 2-4 weeks of inoculation with Agrobacterium rhizogenes hairy roots were formed at root segments in the balloon flower. Optimized growth of hairy roots was obtained in hormone-free MS medium, 6% sucrose and pH 5.8. The pattern of ginsenoside in the transformed roots was not different with that in the ordinary roots.
An ameloblastoma produces more extensive resorption of the teeth on which it encroaches than do most of the other lesions. In this study, intraoral and extraoral radiographs of 78 cases of ameloblastoma were observed and the root resorption associated with ameloblastoma was classified into four types of resorption. With these, the following conclusions were obtained. 1. The root resorption was observed in 72 cases of ameloblastoma. (92.3%) 2. In the aspect of resorptive changes of 424 roots observed, 342 roots showed smooth resorption. (80.6%) 3. The 424 roots associated with ameloblastoma revealed the following radiographic features. a) Root apex resorption in contact with the lesion appeared in 189 roots. (44.5%) b) The resorption of roots projected into the lesion appeared in 147 roots. (34.6%) c) Slight resorption of root surface in contact with the lesion was observed in 56 roots. (13.2%) d) Loss of lamina dura and periodontal space without root resorption were observed in 32 roots. (7.7%)
The development of cracking in ginseng roots was studied to elucidate the factors affecting it in the field. The cracking of 6-year-old ginseng roots harvested in late October callld be induced in 2 days after soaking in water or in watersatllrated soil. However, it callld be slowed down by approximately 10 days when the roots were kept at a low temperature(5$^{\circ}C$). Roots excavated in late May did not develop cracking tinder the conditions tested. There was no significant difference between rice-straw shade and P.E. net shade in the rate of cracked ginseng roots. Which varied with ginseng varieties in relation to shade material. The rate of cracked roots was about 40% in 6-year plants, but it was extremely high (55.6%) in rusty roots. Healing of the wound formed by cracking was generally low but varied between harvest years.
Effect of root-knot nematode (Meloidigyne sp.) 1 infection on the growth and quantity of the inorganic substances in 6 year-old ginseng (Panax ginseng C.A. Meyer) was investigated. Naturally infected 6 year-old ginseng roots were grouped into 3 such as healthy, moderate, and heavy according to the degree of the nematode infection. The roots were sampled in the middle of October, 1980 for chemical analysis. Growth of roots in terms of fresh weight, dry weight, length and diameter was greatly reduced by the nematode infection. The content of inorganic substances was different among the various parts of ginseng roots and according to gall index. Phosphorous and nitrogen contents of the healthy ginseng roots were tend to higher than those of the infected ones, whereas calcium was high in infected roots in the order of heavy, moderate, and healthy. Content of the other inorganic substances was highest in the moderately infected roots, and lowest in the heavy one.
The expression of nestin and vimentin in the spinal nerve roots of rats with experimental autoimmune encephalomyelitis (EAE) was studied to ascertain whether Schwann cells in the peripheral nerves respond to acute central nervous system autoimmune injury. Immunohistochemistry demonstrated that nestin was constitutively expressed in the dorsal roots of spinal nerves in control rats; its expression was enhanced in the spinal nerve roots of rats with EAE. Vimentin expression was weak in control rat spinal nerve roots, and it was increased in the dorsal roots of rats with EAE. It is postulated that normal animals have multipotent progenitor cells that constitutively express nestin and vimentin in the spinal nerve roots. In response to an injury of the central nervous system, these multipotent Schwann cells are activated in the spinal nerve roots through the expression of the intermediate filament proteins vimentin and nestin.
적변인삼의 원인 물질을 구명코저 인삼뿌리의 적변현상과 항산화 활성과의 상관관계를 조사하기 위하여 적변뿌리와 건전뿌리의 각 부위별 항산화 활성을 조사하였다. 적변삼의 표피와 피층조직의 superoxide dismutase(SOD) 활성(units/mg protein)은 각각 126.9와 79.6으로 정상뿌리에 비해 각각 3.5배, 2.0배 높았다. 적변뿌리의 catalase활성은 약 1.6배 높았으나, peroxidase 활성은 오히려 약간 낮았다. 적변뿌리 각 조직의 DPPH 자유라디칼 포착활성은 정상뿌리에 비해 약 2.0배 높았다. 정상뿌리의 ascorbate 함량은 조직에 따라 약간의 차이를 나타내었지만(166~240 $\mu$g/g fr. wt.), 산화형dehydroascorbate(DHA) 함량이 전체 ascorbate 함량의 약 97~79%를 차지하였다. 적변뿌리의 전체 ascorbate함량은 정상뿌리와 비슷하였지만 환원형 ascorbate의 함량은 조직에 따라 다르지만 정상뿌리에 비해 3.5~7.5배 높았다. 적변뿌리의 표피조직, 피층조직, 중심주 조직의 전체 glutathione 함량은 정상조직에 비해 각각 7.3배, 4.8배, 1.2배 높았다. 적변화에 따른 환원형 glutathione(GSH)과 산화형 glutathione(GSSG)의 함량비는 전체 glutathione 함량변화와 비슷하였다. 이상의 결과는 인삼의 적변조직은 어떤 외부스트레스에 의해 과다하게 발생되는 활성산소종을 제거하기 위하여 SOD 활성, 환원형 ascorbate 함량, 환원형 glutathione 함량이 크게 증가하는 것으로 사료된다.
