Old growth and protected-trees are worth history and culturally in Korea. This research carried out to investigate habitat characterization and the vegetative propagation methods of Salix chaenomeloides(100-600 years old trees) in Gyeongsangnamdo Province. It has been preserved in good and grow naturally in area surrounding village(4 trees), riverside(1 tree) and field(1 tree). Cutting from old growth and protected trees was conducted using 10 kinds of bedsoils. It was survived above 93% in all bedsoils. Growth of cuttings is a distinct difference according to bedsoils and tree ages. Growth of cutting was high on CBS, followed mixed bedsoils(VPMP) including vermiculite, peatmoss and perlite. Root collar diameter was difference depend on tree ages and bedsoils. All trees are produced 1 to 4 branch. Root growth of cutting was 20 cm on CBS and VPMP, which is the best shoot growing on cutting. Bedsoil with physico-chemical characteristics of high moisturizing ability, high air permeability, high EC value and slightly acid may be suitable for efficiently cutting propagation of old growth and protected S. chaenomeloides. CBS and VPMP bedsoil are favorable for propagating this species by cuttings.
This study was investigated the methods of vegetative propagation for new Lespedeza varieties. Experimental matterials were used 6 Lespedeza species; L. maximowiczii, L. cyrtobotrya, L. angustifolioides, L. bicolor, L. maximowiczii var. tomentella, and L. japonica var. intermedia. In April a year branches were cut 15-20cm length and 3-4mm thickness and planted in sand bed (depth 150cm) to be sterilized by the 0.1% solution of Uspulun. And then the cutting beds were irrigated and shaded. The results are as follows. 1. The rooting rates of the cuttings could be found so variable among varieties from 69% of L. maximowiczii var. tomentella to 50.3% of L. japonica var. intermedia. 2. The growth performance showed statistically significant difference at 1% level among varieties. L. maximowiczii was the best, while L. japonica var. intermedia showed the wrost growth performance of them. 3. The average number of roots with more than 5 cm length per stock was revealed statistically significant difference at the 1% level among varieties from the greatest number of 6.4 of L. maximowiczii var. tomentella to the smallest number of 2.3 of L. japonica var. intermedia. 4. Total average length of the roots per stock was statistically significant so different at 1% level from the longest 279.8cm of L. maximowiczii var. tomentella to the shortest 41.1cm of L. japonica var. intermedia. 5. The average ndules with more than 1 mm in diameter per stock was statistically significant so different at 1% level among varieties from the greatest number of 34.4 of L. maximowcizii to the smallest numqer of 4.6 of L. japonica var. intermedia.

This research was carried out to establish the large quantity propagation system for Kalopanax pictus using root cutting. The results are as follows. As there was no significant difference in the promotion of adventitious shoot development from the root cuttings of Kalopanax pictus among kinds (IBA, NAA and Kinetin) and concentrations (0 to 2000 mg/L) of growth regulators, no application is recommended, which showed more than 90% of shoot development. Horizontal and vertical placement of rooting showed more than 93% of shoot occurrence, but the latter showed more desirable growth traits in the number of shoot development, number of leaf, petiole length, and leaf length and leaf width Polarity was observed and thus normal vertical placement of root cutting is preferred. However, root cuttings placed upside down showed the shoot development at the distal end, which indicates further sophisticated test is necessary to investigate this phenomenon.

Veronica rotunda var. subintegra (Nakai) T. Yamaz. is perennial herb belonging to the family Scrophulariaceae, native to Korea. Plant growth regulators, cutting mediums, cutting position, and leaf size were examined to investigate the requirements for cutting propagation of V. rotunda var. subintegra. This study was conducted over a period of 6 weeks from March to May 2017 in the Division of Plant Resources, Korea National Arboretum. For all treatments, survival and rooting percentage was 100%. In PGRs experiment, the number of roots and shoot length treated with 100 mg/L IBA treatment was significantly higher than that of NAA treatment. The shooting percentage was decreased with an increase of the PGRs concentration. In growing substrates experiment, the number of roots on commercial soil (61.2) was the highest, followed commercial + kanuma soil (45.7), and the lowest was kanuma soil (41.8). The shoot length treated with commercial soil (8.9 cm) was significantly greater than that of kanuma soil (4.1 cm). In cutting positions experiment, the number of roots treated with apical (56.6) was significantly higher than that of basal (38.8). In leaf-cut experiment, the number of roots and of shoot length treated with full leaf was significantly higher than that of half leaf. As a result, we recommend treating cuttings with 100 mg/L IBA treatment, commercial soil, apical position, and full leaf for successful cutting propagation of V. rotunda var. subintegra.
Present studies were carried out to understand the way of propagation of Osmanthus which has much value as ornamental trees in southern parts of Korea. The results of these experiments were summarized as follows: 1. In order to increase the survival of Osmanthus, scions of Osmanthus was grafted on the stocks of Ligustrum obtusifolium S. et Z. The survival of the grafts were 63% for Osmanthus fragrans var. aurantiacus Makino and 60% for Osmanthus asiaticus Nakai. 2. The growth of the grafts was initiated from 28th day after the grafting and the height growth resumed three times, and the growth percentage was 63% for Osmanthus fragrans var. aurantiacus and 59% for Osmanthus asiaticus. 3. Cutting medium mixed with perlite and vermiculite (1:1) gave better results than either use of them separately, and the rooting percentage was 93% for Osmanthus fragrans var. aurantiacus and 90% for Osmanthus asiaticus. 4. Rooting percentage of cuttings was lower in hard-wood cutting than that of in green-wood cutting. 5. Grafts showed fairly good height growth in July and September, and ceased growth in October. 6. Correlation between height growth and diameter of root collar of grafts in case of Osmanthus fiagrans var. aurantiacus was highly significant and showed linear correlation ($r=0.70^{**}$)
Genetic variance and heritability for height growth of Populus alba${\times}$ P. glandulosa were estimated. Remarkable different between P. alba${\times}$ alba(Italy), female and P. alba naturalized in Korea was observed in the genetic variance and therefore genetic character of the P. alba${\times}$ alba(Italy) was considered better than that of P. alba. The heritability (83%) for the female was considerably high. On the other hand, P. glandulosa, pollen tree. showed narrow genetic variance and also very heritability(17%). Two(A and B) of three pollen trees showed the same trend in the genetric variance. However, the other(c) was somewhat different from A and B. Similarly, rooting ability of cuttings differed between female trees, but did not between male trees. Naturalized Pl alba showed relatively higher rooting ability than P. alba${\times}$ alba(Italy). In the pollen trees, the ability was similar in two male trees. However, the other male tree(c) was slightly poorer than the other two.

Hardwood cutting propagation and early growth characteristics were investigated in order to develop the method of cutting propagation and to find out growth characteristics in the low altitude for in situ and ex situ conservation of Empetrum nigrum var. japonicum K. Koch, which is typical arctic alpine plants on Mt. Halla. The growth of roots and shoots was different depending on hormone concentrations or soil conditions. The survival rate, rooting rate, root growth, number of root and shoot growth increased with treatment of 100 mg/l or 500 mg/l NAA. Consequently, optimum condition of hardwood cutting was at treatment with 100 mg/l or 500 mg/l NAA. When plantlets from hardwood cuttings were exposed to the field condition, after 7 months survival rate was 73.3% without shading while $91.1{\sim}94.4%$ at shading conditions. In the green house, however, survival rate of plantlets were $95.6{\sim}97.8%$ without shading. The growth of plantlets was different depending on sites and shading conditions. Particularly, the best growth was obtained when the plantlets were grown in shading conditions. It indicates that relative humidity and light intensity are correlated with the growth in the low altitude area.
Iris dichotoma Pall. is an important endangered plant belonging to the family Iridaceae. A method was developed for the rapid micropropagation of I. dichotoma through plant regeneration from leaf, rhizome, and root explant-derived calli. Leaf, rhizome, and root segments were cultured on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxy acetic acid (2,4-D; $0-3.0mg{\cdot}L^{-1}$) for callus induction. Callus production was highest at $1.0mg{\cdot}L^{-1}$ 2,4-D, where 73.8% and 45.5% of cultured rhizome and root cuttings, respectively, produced calli. The viable calli were maintained at an induced concentration of 2,4-D ($3.0mg{\cdot}L^{-1}$). They were then transferred to MS medium supplemented with various concentrations of 2,4-D ($0-3.0mg{\cdot}L^{-1}$) in combination with 6-benzyladenine (BA: 0, 1.0 and $3.0mg{\cdot}L^{-1}$) for adventitious shoot regeneration. The addition of a low concentration of 2,4-D into BA-containing medium significantly increased the frequency of shoot regeneration in leaf, rhizome, and root-derived calli. The highest number of adventitious shoots (26.4 per callus) formed at $0.5mg{\cdot}L^{-1}$ 2,4-D and 1.0 mg/l BA. For rooting of the shoots, half- strength MS medium supplemented with different concentrations of indole 3-butyric acid (IBA) $0-3.0mg{\cdot}L^{-1}$ was tested. The optimal results were observed using half-strength MS medium supplemented with $1.0mg{\cdot}L^{-1}$ IBA, on which 98% of the regenerated shoots developed roots with an average of 3.5 roots per shoot within 45 days. The plantlets raised in vitro were acclimatized and transferred to soil with 95% success. This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.
An efficient method for the rapid micropropagation of Camptotheca acuminata from axillary buds was established by application of various plant growth regulators. Among various cytokinins, $0.5mg\;L^{-1}$ BA showed the best performance on shoot multiplication, number average multiple shoots up to 10.8. The propagated shoot cuttings in vitro were elongated on NN basal medium without plant growth regulators. The secondary multiple shoots were induced at the site of initially induced buds. Rooting was induced directly near the base of the shoot on half-strength NN medium containing $0.5mg\;L^{-1}$ of IBA, whereas high concentration of $1.0mgL^{-1}$ IBA could induce callus at the base of the shoot. The camptothecin content, anticancer compound of the micropropagated plants was contained in various tissues. Camptothecin contents were 1.8 and $2.5mg\;g^{-1}$ dry weight in stems from propagated in vitro and mother plant, respectively. This result may be used to develop strategies for large-scale propagation of elite C. acuminata trees.

To select the section with shoot formation ability, the calli and shoot formation from three sections (first leaf including cotyledonary node, hypocotyl and cotyledon explants) of 5-days-seedlings of soybean were induced on MS medium supplemented with 1.0 mg/L BAP, 3% sucrose, and 0.3% gelrite for one month. The first leaf section exhibited the highest shoot formation rate (51%), followed the hypocotyl section (10%) and the cotyledon section (0%). The shoot formation rates and shoot number of the four excised sections (whole first leaf, a half of the first leaf, a third of the first leaf and only node) of the first leaf were also investigated on the same medium. A half of the first leaf explant and the third of the first leaf explant had higher shoot formation rates (76-80%) and numbers (3-4 / explants) than those in other two explants. Effects of six cytokinins (kinetin, zeatin, BAP, 2iP, PBA, and TDZ) on shoot formation were determined, using the half of the first leaf explants. Zeatin (1.0 mg/L) exhibited the highest in shoot formation rate (94%) and numbers (8 / explant). In addition, the combined effects of three cytokinins (zeatin, BAP, and TDZ; 0.5, 1.0, 2.0 mg/L, respectively) and an auxin (IAA; 0.0, 0.5, 1.0, 2.0 mg/L) were determined. The combination (1:1, v/v) of zeatin (1.0 mg/L) and IAA (1.0 mg/L) exhibited the highest in shoot formation rate (96%) and numbers (16 / explant), twice more than zeatin (1.0 mg/L) alone. The shoot cuttings were transferred and cultivated on the rooting media supplemented with only auxin, IBA at various concentrations. The highest root formation (8 / shoot) was achieved on the medium supplemented with 1.5 mg/L. After 4 weeks of cultivation, the plantlets with an extensive root system were transplanted in pots with a soil mixture of vermiculite and fine sand. Transferred to field, about 75% of the plantlets survived.
