Attachment of Pasteuria penetrans Endospores to Meloidogyne spp. Juveniles Affected by Temperatures and the Nematode species (선충기생세균(Pasteuria penetrans) 내생포자의 뿌리혹선충(Meloidogyne spp.) 유충 부착에 대한 온도와 선충종의 영향)

  • A greenhouse soil infested with an obligate nematode parasitic bacterium, Pasteuria penetrans, was used to test the effect of temperatures on the endospore attachment to root-knot nematode, Meloidogyne arenaria, juveniles (J2). Freshly hatched J2s were inoculated to the soil in petri dish and incubated under different temperatures of $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, and $35^{\circ}C$ for 7 days. The endospore attachment rates were 100% in all the temperatures, while the number of endospores attached per J2 was highest in $25^{\circ}C$ with 28.3 endospores/J2 followed by 20.2, 18.6, and 13.6 in $30^{\circ}C$, $20^{\circ}C$, and $35^{\circ}C$, respectively. When the soil was pre-treated under different temperatures before the J2 inoculation, the endospore attachment rates significantly decreased from 60% in room temperature to $25^{\circ}C$, 31.7, 8.3, 5.0, and 0% after the soil incubation in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$ for 10 days, respectively. The endospore numbers attached per J2 were 3.5, 4.3, 1, 1, and 0 when the soil was pre-treated in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$, respectively, which were lower than 5.3/J2 of room temperature treated soil. The P. penetrans isolate in the soil showed nematode species-specific endospore attachment characteristics with 100% attachment rate only on M. arenaria J2s while the rates were 0% on M. hapla and M. incognita J2s.

Effects of Ectomycorrhizae and Soil Condition on Growth of Pinus rigida Seedlings (외생균근(外生菌根) 및 토양조건(土壤條件)이 리기다소나무 균목생장(菌木生長)에 미치는 영향(影響))

  • The experiment was conducted to test the effects of ectomycorrhizal inoculation and soil moisture regime on growth and nutrient uptake of Pinus rigida seedlings. Two-year-old seedlings inoculated with either Pisolithus tinctorius or Thelephora terrestris were used for this experiment. There were two moisture treatments and four phosphate treatment levels with three replications. Height and root collar diameter growth and dry weight increase were measured and nitrogen, phosphorous, and potassium in the tissue were analyzed three months after the initiation of phosphate and moisture regime treatment. Results are as follows; 1) Height growth of seedlings was not affected in all treatments. (mycorrhizae, fertility, moisture). 2) High moisture regime increased root collar diameter growth 2.7 times as large as low moisture regime. 3) In low moisture regime, net production increased 26% in seedlings inoculated with Pisolithus tinctorius and increased 72% control seedlings. 4) In high moisture regime, net production of inoculated seedlings with Pisolithus tinctorius increased four times as much as that of control seedlings but increased $P_2O_5$ in fertility level did not affect net production. 5) In uptake of nitrogen, phosphate, and potassium, Pisolithus tinctorius - inoculated seedlings increased uptake by 1.9, 1.8, and 1.5 times, respectively, as much as control seedlings. 6) Increase in $P_2O_5$ fertility level enhanced the uptake of $P_2O_5$ in Pisolithus tinctorius - inoculated seedlings, but not affected the uptake of $P_2O_5$ in control seedlings. 7) Increase in $P_2O_5$ fertility levels enhanced the uptake of $K_2O$ in Pisolirhus rinctorius - inoculated seedlings.

Biocontrol of Damping-Off(Rhizoctonia solani) in Cucumber by Trichoderma asperellum T-5 (Trichoderma asperellum T-5를 이용한 오이 모잘록병(Rhizoctonia solani)의 생물학적 제어)

  • A fungal strain of Trichoderma having strong chitinolytic activity was isolated from field soil enriched with crabshell for several years. Based on 5.8S rRNA, partial 18S, 28S rRNA genes, ITS1, ITS2 sequence analysis and morphological characteristics, the fungus was identified as Trichoderma asperellum and named as Trichoderma asperellum T-5 (TaT-5). The fungus released lytic enzymes such as chitinase and ${\beta}$-1, 3-glucanse, and produced six antifungal substances in chitin broth medium. To demonstrate the protective effect of TaT-5 against damping-off in cucumber plant caused by Rhizoctonia solani, TaT-5 culture broth (TA), chitin medium (CM) and distilled water (DW) were applied to each pot at 10 days after sowing, respectively. Then, the homogenized hyphae of R. solani were infected to each pot at 1 week after TaT-5 inoculation. During experimental period, fresh weight of shoot and root in cucumber plant more increased at TA treatment compared to other treatments. PR-proteins (${\beta}$-1, 3-glucanase and chitinase) activities in cucumber leaves markedly increased at CM and DW treatments, but the activity slightly increased and then decreased at TA treatment at 3 days after infection of R. solani. The activity of PR-proteins activities in cucumber roots at all treatments decreased with time where the degree of decrement was more alleviated at TA treatment than CM and DW. These results suggest that the lytic enzymes (chitinase and ${\beta}$-1, 3-glucanse) and antifungal substances produced by TaT-5 can reduce the pathogenic attack by R. solani in cucumber plants.

  • The purposes of this study were to evaluate the effects of acid rain on tree growth and on the mycorrhizal formation and the effects of mycorrhizae on the host tolerance to acid rain. Simulated acid rain was applied for five months to Pinus $rigida{\times}taeda$ seedlings in pots inoculated with Pisolithus tinctorius (Pt) and Suillus luteus (Sl). Mycelial inocula of Pt and Sl were either mixed with entire pot soil (Mix) or casted as a band (Band) after soil sterilization. Three pH levels of acid rain (pH 3.0, 4.5 and 6.4 adjusted by 3 : 1 mixture of sulfuric and nitric acids) were tested. Pt-Mix was most effective in growth stimulation and resulted in 45-90% increase in tree height in sandy loam. Pt-Band was less effective than Pt-Mix in growth stimulation and mycorrhizal formation. Simulated acid rain at pH 4.5 stimulated height growth by 10-55%, while acid rain at pH 3.0 did not significantly affect the height growth. The top/root ratio was increased by pH 4.5 treatment, while pH 3.0 treatment reduced it. Mycorrhizal infection rate was not affected by acid rain. Pt inoculation reduced acid-induced leaf injury by 28-58% in both pH 3.0 and 4.5 compared with un-inoculated plants. Sl was also effective in growth enhancement, but was less effective than Pt in both mycorrhizal infection and reducing leaf injury.

A study of the effects of PDGF-BB on the characteristics of bone stromal and periodontal ligament cells (혈소판유래성장인자-BB가 골간질세포와 치주인대세포의 성상에 미치는 영향)

  • The main goal of periodontal therapy is to restore the lost periodontal tissue and establish the attachment appratus. Current acceptable therapeutic techniques are included : removal of diseased soft tissue, demineralization of exposed root surface, using the barrier membrane for preventing the downgrowth of gingival epithelial cell, insertion of graft materials as a scaffolding action, and biological mediators for promoting the cell activity. The latest concept one among them has been studied which based on the knowledge of cellular biology of destructed tissue. Platelet-derived growth factor(PDGF) is one of the polypeptide growth factor which have been reported as a biological mediator to regulate activities of wound healing progress including cell proliferation, migration, and metabolism. The purposes of this study is to evaluate the influences of the PDGF as biological mediator to periodontal ligament and bone marrow cell. Both right and left maxillary first molar were extracted from rat which had treated with 0.4% ${\beta}-Aminopropionitril$ for 5 days, and feeded until designed date to sacrifice under anesthesisa. Periodontal ligament were removed from the extracted socket of the rat, and cultured with Dulbecco's Modified Essential Medium(DMEM) contained with 10% Fetal Bovine Serum, 100U/ml penicillin, $100{\mu}g/ml$ streptomycin, $0.5{\mu}g/ml$ amphotericin-B. Bone marrow cell were culture from bone marrow suspension with which washed out from femur with same medium. The study was performed to evaluate the effect of PDGF to periodontal ligament and bone cell, cell proliferation rate, total protein synthesis, and alkaline phosphatase activity of rat periodontal ligament(PDL) cell and bone stromal(RBS) cell in vitro. The effects of growth factors on both cells were measured at 3, 5th day after cell culture with (control group) or without growth factors(experimental group). The results were as follows: 1. The tendency of cell proliferation under the influence of PDGF showed more rapid proliferation pattern than control at 3 and 5 days after inoculation. 2. The activity of Alkaline phosphatase revealed 14, 80% increased respectively at 3, 5 days culture than control group. Measurements of ALPase levels indicated that PDL cells had significantly higher activity when compared with that of co-culture groups and GF only(P<0.05). And, ALPase activity in 10 days was higher than that of 7 days(P<0.05). 3. The tendency of formation of the mineralized nodule were observed dose-depend pattern of PDL cells. There was statistically significant difference among group l(PDL 100%), 2(PDL 70%:GF 30%), and 3(PDL 50%:GF 50%)(P<0.01). But, there was no difference among group 3, 4(PDL 30%:GF 70%), and 5(GF 100%). 4. Also, the number of nodule was greater in co-culture of PDL 70% and GF 30% than in culture of PDL 70%(P<0.05). From the above results, it is assumed that the PDGF on PDL cells and RMB cell culture. GF stimulates the cell growth, which is not that of PDL cells but GF. And, the activity of ALPase depends on the ratio of PDL cells, and ALPase may relate to the initial phase of nodule formation. Also, it is thought that the calcified nodule formation principally depends on PDL cells, is inhibited by GF, and affected by cell density. In conclusion, platelet-derived growth factor can promote rapid osteogenesis during early stage of periodontal tissue regeneration.

Isolation of Photosynthetic Bacterium, Rhodopseudomonas palustris JK-1 and Researches on IAA and Carotenoid Production (광합성세균 Rhodopseudomonas palustis 분리 및 IAA와 Carotenoid 생성에 관한 연구)

  • The JK-1 isolate which was the best producer of indole-3-acetic acid and carotenoid among the 388 strains isolated from 28 wetlands in Jeju, was identified to be Rhodopseudomonas palustirs belongs to a typical group of non sulfur purple bacteria based on 16S sRNA sequencing. This study investigated the effect of different cultural conditions of pH, temperature, agitation, light and aeration on growth, IAA and carotenoid production of photosynthetic bacterium JK-1 for optimization of IAA and carotenoid production. It was found that growth, IAA, carotenoid, and bacteriochlorophyll production with light (3,000~3,500 Lux) and agitation (100 rpm) showed better results than those with dark/static or dark/agitation (100 rpm) in anaerobic conditions. The optimal pH, temperature and agitation speed for cell growth were 7, $30^{\circ}C$, 150 rpm, for IAA production were 9, $30^{\circ}C$, 150rpm and for carotenoid production were 6, $25^{\circ}C$, 50 rpm, cultured for 72 h under anaerobic light, respectively. The growth and IAA production were high in aerobic culture compared with anaerocic culture, whereas carotenoid and bacteriochlorophyll content were decreased extremely in aerobic condition (0.5~1 vvm). Subsequently, the optimal culture conditions for JK-1 were selected with pH 7, $30^{\circ}C$ and 100 rpm under anaerobic light and the effect on plant growth was tested by pot assay. Inoculation of JK-1 with 3% (v/v) level caused increase in shoot and root dry weigh that varied from 20%~58% to 40%~28% in young radish in camparison to uninoculated treatment at 50 days of growth. The study suggests that the JK-1 isolate may serve as efficient biofertilizer inoculants to promote plant growth.

Isolation and Evaluation of an Antiviral Producing Serratia spp. Strain Gsm01 against Cucumber mosaic virus in Korea (한국에서 CMV에 항바이러스 효과를 나타내는 Serratia spp. Gsm01 균주의 분리 동정 및 효과 검정)

  • An Antiviral producing bacterial strain was isolated from ginseng root environment in Hongcheon, Kangwon province of Republic of Korea. Identification of this bacterial strain was performed by physiological and biochemical tests along with 16S rRNA analyses. The results revealed that the bacterium was closer to genus Serratia, which was named as Gsm01. The strain was grown in Mannitol-Glutamate-Yeast (MGY) broth for 48 h. The culture was centrifuged and the filtrate obtained was tested for its ability to control Cucumber mosaic virus strain Y (CMV-Y) in greenhouse and field experiments. In the green house experiments, CF was evaluated for its ability to protect local host, Chenopodium amaranticolor and systemic host of CMV, Nicotiana tabacum cv. Xanthi-nc. It was found that, CF treatment reduced viral infection by 98% in local host; C. amaranticolor. The N. tabacum cv. Xanthi-nc plants treated with CF did not show visible viral symptoms 15 days post inoculation (dpi) and remained symptomless throughout the periods of the study. To evaluate effectiveness of CF under field conditions, experiment was carried out in a polyvinyl house. It was observed that, 52% plants were protected from viral diseases compared to non-treated plants, increasing the crop yield. This is the first report showing antiviral activity of a Serratia spp. against CMV.

Efficacy of Three Antiviral Agents and Resistant Cultivars on Tomato Yellow Leaf Curl Virus in Tomato (토마토황화잎말림바이러스병에 대한 저항성 품종과 항바이러스 활성 물질 3종의 효과 검증)

  • Recently, several in vitro studies have reported antiviral activity of agents of systemic acquired resistance against plant virus infection, but the approach has not been applied in a wide range of agricultural fields. The objective of this study was to evaluate the inhibitory effect of the exogenous application of salicylic acid (SA), chitosan (CH), or eugenol (EG) in tomato yellow leaf curl virus (TYLCV) infection of greenhouse-grown tomato plants. In vitro, the initial time of symptom was observed in TYLCV-infected plants (VP) of the resistant cultivar 'Superdotaerang' at 12 days post inoculation (dpi) after application of antiviral agents. At 32 dpi, the disease rate of TYLCV in the CHT+VP (0.1% chitosan and virus infected control) treated plants was 87.5%, lower than that of the other treatment. However, the virus content in the CHT+VP treated plants was higher than those of the other treatments, and SA, EG, and CH did not show significant effect on plant height or shoot and root fresh weight. Our results from summer-cultivated greenhouse-grown tomatoes show that none of the tested agents had an inhibitory activity on viral infection or yield of tomato 'Dotaerangsola'cultivar. In contrast, all treated 'TY Giants' cultivars that possessed TYLCV resistance genes Ty-1 and Ty-3a did not show typical symptoms and the virus content was remarkably lower than those in the TYLCV treated plants in 'Superdotaerang'. The results of this research indicated that the planting of resistant tomato cultivars was effective method instead of using SA, EG, and CH (known as resistance-inducing factors for control) of TYLCV in the field.
