Cucumber fruit mottle mosaic tobamovirus (CFMMV) causes severe mosaic symptoms with yellow mottling on leaves and fruits, and occasionally severe wilting of cucumber plants. No genetic source of resistance against this virus has been identified. The genes coding for the coat protein or the putative 54-kDa replicase were cloned into binary vectors under control of the SVBV promoter. Agrobacterium-mediated transformation was peformed on cotyledon explants of a parthenocarpic cucumber cultivar with superior competence for transformation. R1 seedlings were evaluated for resistance to CFMMV infection by lack of symptom expression, back inoculation on an alternative host and ELISA. From a total of 14 replicase-containing R1 lines, 8 exhibited immunity, while only 3 resistant lines were found among a total of 9 CP-containing lines. Line 144 homozygous for the 54-kDa replicase was selected for further resistance analysis. Line 144 was immune to CFMMV infection by mechanical and graft inoculation, or by root infection following planting in CFMMV-contaminated soil. Additionally, line 144 showed delay of symptom appearance following infection by other cucurbit-infecting tobamoviruses. Infection of line 144 plants with various potyviruses and cucumber mosaic cucumovirus did not break the resistance to CFMMV. The mechanism of resistance of line 144 appears to be RNA-mediated, however the means is apparently different from the gene silencing phenomenon. Homozygote line 144 cucumber as rootstock demonstrated for the first time protection of a non-transformed scion from soil inoculation with a soil borne pathogen, CFMMV.
This study sought to control the root-knot nematode (RKN) Meloidogyne incognita using benign organo-chemicals. Second-stage juveniles (J2) of RKN were exposed to dilutions (1.0%, 0.5%, 0.2%, and 0.1%) of acetic acid (AA), lactic acid (LA), and their mixtures (MX). The nematode bodies were disrupted severely and moderately by vacuolations in 0.5% of MX and single organic acids, respectively, suggesting toxicity of MX may be higher than AA and LA. The mortality of J2 was 100% at all concentrations of AA and MX and only at 1.0% and 0.5% of LA, which lowered slightly at 0.2% and greatly at 0.1% of LA. This suggests the nematicidal activity of MX may be mostly derived from AA together with supplementary LA toxicity. MX was applied to chili pepper plants inoculated with about 1,000 J2, for which root-knot gall formations and plant growths were examined 4 weeks after inoculation. The root gall formation was completely inhibited by 0.5% MX and standard and double concentrations of fosthiazate; and inhibited 92.9% and 57.1% by 0.2% and 0.1% MX, respectively. Shoot height, shoot weight, and root weight were not significantly ($P{\leq}0.05$) different among all treatments and the untreated and non-inoculated controls. All of these results suggest that the mixture of the organic acids may have a potential to be developed as an eco-friendly nematode control agent that needs to be supported by the more nematode control experiments in fields.
Carrot (Dacus carota var. sativus) is one of the top-ten most economically important vegetable crops produced worldwide, and the root-knot nematodes, Meloidogyne spp., are one of the most important pests in the carrot. In Korea, M. hapla and M. incognita are presumed to be the major root-knot nematodes distributing mostly in open carrot fields and greenhouses, respectively. In our study, currently-developed and commercial carrot cultivars and the parental lines were examined for their pathological responses to M. incognita and M. hapla 7 weeks after inoculation with about 1,000 second-stage juveniles (J2) of the nematodes. All the carrot cultivars and lines showed susceptible responses to both nematodes with the gall index (GI) of 2.4-4.4, which were always higher on the carrot plants infected with M. incognita than M. hapla. Gall sizes were remarkably larger with more serious reduction of the root growths in the plants infected with M. incognita than M. hapla, suggesting the carrot lines examined in our study were more susceptible to the former than the latter. In the infection sites of the root tissues, giant cells were more extensively formed, occupying larger stellar regions with the prominent destruction of adjacent xylem vessels by M. incognita than M. hapla. All of these results suggest M. incognita affect more seriously on the carrot plants that are grown in greenhouses, compared to M. hapla that has a major distribution in open carrot fields, which would be used for determining cropping systems based on target nematode species, their damage and pathological characteristics.
Black root rot (BRR) caused by Lasiodiplodia theobromae is an alarming disease of mulberry that causes tremendous economic losses to sericulture farmers in India and China. Successful control of this disease can be attained by screening germplasm and identifying resistant sources. Seventy four diseased root samples were collected from farmer's fields belonging to four major mulberry growing states of South India. Based on morpho-cultural and scanning electron microscopy studies, 57 fungal isolates were characterized and identified as L. theobromae. Phylogenetic analysis of concatenated internal transcribed spacer and β-tubulin sequences revealed variation of the representative 20 isolates of L. theobromae. Following the root dip method of inoculation, pathogenicity studies on susceptible mulberry genotypes (Victory-1 and Thailand male) recognized the virulent isolate MRR-142. Accordingly, MRR-142 isolate was used to evaluate resistance on a set of 45 diverse mulberry accessions. In the repeated experiments, the mulberry accession ME-0168 which is an Indonesian origin belonging to Morus latifolia was found to be highly resistant consistently against BRR. Eight accessions (G2, ME-0006, ME-0011, ME-0093, MI-0006, MI-0291, MI-0489, and MI-0501) were found to be resistant. These promising resistant resources may be exploited in mulberry breeding for developing BRR resistant varieties and to develop mapping populations which successively helps in the identification of molecular markers associated with BRR.
새싹인삼의 뿌리썩음병 억제제로서 과산화수소의 활용 가능성을 구명하고자 항진균 활성효과와 묘삼 소독효과를 연구한 결과, 과산화수소는 300 mg/l의 농도에서 1×107 conidia/ml 농도의 Cylindrocarpon destructans 균총을 80% 이상 억제시켜 항진균 활성을 나타내었고 인위적으로 병원균에 감염시킨 묘삼의 유병율을 20% 이하로 억제시켜 소독효과가 매우 높았다. 결과적으로 과산화수소 희석수 300 mg/l를 14일에 걸쳐 분무처리하였을 때, 인위적으로 병원균에 감염된 새싹인삼의 뿌리썩음병 발생수율이 72.1%로 무처리의 발생수율 87.2%에 비해 다소 억제되었다. 그러나, 과산화수소의 주기적인 살포가 새싹인삼의 생육 부진을 다소 유발하였다. 추가 연구로서 뿌리썩음병의 방제에 효과적이고 새싹인삼에 안전한 과산화수소의 처리방법 개발이 요구되었다.
황칠나무는 두릅나무과에 속하는 난대성 상록활엽수로 우리나라 제주도, 보길도, 거문도, 거제도, 완도, 해남 등에 분포한다. 2021년 6월 전라남도 나주에서 황칠나무 유묘의 지제부가 갈변하고 잘록해지는 뿌리썩음병을 국내 최초로 발견하였다. 황칠나무 뿌리썩음병균을 분리 및 동정하기 위해, 유묘의 병든 조직에서 3개의 균주를 분리하였고, PDA 배지에서 형태적, 균학적 특성 조사한 결과 Fusarium oxysporum으로 동정되었다. Internal transcribed spacer (ITS) 및 translation elongation factor 1-α (EF1-α) 염기서열을 통한 분자계통학적 분석을 수행하여 한국의 황칠나무 뿌리썩음병균은 F. oxysporum임을 확인하였다. 병원성 테스트를 위해 유묘의 뿌리를 분생포자 현탁액에 침지하여 정식하였고, 접종 20일 후 처리구에서 뿌리썩음 및 지제부의 갈변 증상을 확인하였다. 본 연구는 한국의 황칠나무 유묘에서 뿌리썩음병균 F. oxysporum의 첫 보고이다.
본 연구는 내생균근균 접종이 목본식물의 인공 산성우에 대한 저항성에 미치는 영향을 구명하는데 그 목적이 있다. 아까시나무(Robinia pseudoacacia) 종자를 실험실 내에서 파종한 후, 멸균된 버미큘라이트, 모래, 밭토양을 1:1:1로 혼합한 토양을 담은 1l pot에 건강한 묘목을 이식하였다. 각 pot에 묘목을 이식할 때 분쇄된 뿌리혹과 균근균을 접종하였는데, 뿌리혹은 자연 상태에서 자란 아까시나무림에서 채취한 것을, 내생균근균은 상업용으로 판매하는 Glomus intraradices를 수입하여 사용하였다. pH 2.6, 3.6, 4.6, 5.6 4종류의 인공 산성우는 황산과 질산을 3:1로 혼합하여 만들었다. 각 pot에는 질소와 인을 배제시킨 양료용액과 일정한 pH 수준의 산성우를 각각 180 mm씩 매주 1회씩 50일간 처리하였으며 포트는 온실에 놓아두었다. 실험 종료시 묘목을 굴취하여 엽록소, 무기양료의 함량과 조직의 순광함성량, 건중량, 뿌리의 균근감염율 등을 측정하였다. 균근감염율은 pH 2.6에서 현저히 낮은 수준을 나타냈는데, 이는 극도로 낮은 pH에서 G. intraradices의 활력이 억제됨을 의미한다. 수고생장, 건중량, 뿌리혹 형성, 엽록소 함량, 그리고 순광합성 등은 pH 3.6을 제외한 모든 pH 수준에서 균근감염에 의해 증가하였다. pH 5.6과 4.6에서는 균근균 접종이 뿌리혹 형성을 각각 85%, 45% 증가시켰으나, pH 3.6괴 2.6에서는 균근균 접종이 뿌리혹의 형성을 거의 촉진하지 않았다. 순광합성 량은 모든 pH 수준에서 균근감염에 의해 증가하였다. 조직체 내의 인 함량도 균근감염에 의해 평균 43% 증가하였으며 pH 2.6을 제외하고는 통계적으로 유의하였다. 결론적으로 아까시나무에 대한 균근균 접종은 광합성과 인 함량, 뿌리혹 형성을 증가시킴으로써 생장을 촉진하고, 산성우에 대한 저항성을 높여준다고 할 수 있다.
오이와 방울토마토에 Glomus sp.를 종자파종과 고형배지경에 정식시 각각 접종하여 생장과 수량반응 및 균근 감염 양상 등을 비교 검토하였다. 오이와 방울토마토의 초장, 엽수, 엽면적 및 건물중 등의 전반적인 생장은 무접종 < 정식시 균근균 접종 < 파종시 균근균 접종의 순으로 양호하였으며, 과실수량반응은 종자파종시에 균근균을 접종할 경우 과실의 생체중이 가장 높게 나타났고, 과실의 당도도 증가하였다. 균근감염은 오이와 방울토마토 모두 주로 균사에 의하여 이루어 졌고. vesicle와 arbuscule에 의한 감염은 극히 적었다. 뿌리체내 균근균 감염율은 종자파종시 균근균 접종원 처리의 경우가 정식시에 균근균 접종원을 처리한 경우보다 높았다. 균근균 접종에 따른 과실 수확을 보면 오이의 경우 주당 과실수가 증가하였고, 토마토의 경우 주당 과실의 수확 중량이 증가하였다.
우리나라에서 처음 발생되었던 Kresek에 과한 발생의 균형 및 재현에 관한 시험결과는 다음과 같았다. 흰빛잎마름병균에 의한 Kresek 현상은 주로 남부방인 전라남도 화순, 무안, 강진, 영암, 곡성, 광산, 경상남도 진양군에서 볼 수 있었다. Kresek 발생지의 병원균의 균형은 I, II, IV, V군에 의한 발생을 나타냈다. 3. Kresek이 가장 심하였던 화순군에서는 21균주중 17균주는 IV군, 2균주는 II군, 2균주는 V군이었으며 광산군의 5균주는 IV군dp 속하는 균주였다. 4. 화순군에서는 II, IV, V군이 혼재하고 있음을 볼 수 있었다. 5. Kresek 재현에 관한 시험에서는 접종 5일만에 발병을 볼 수 있었으며 뿌리 절단법에서 발병율이 현저히 높았고, 분무접종, 침접종에서도 발병을 볼 수 있었다. 6. 병원균농도가 높을수록 발병이 많았다.
농업유용미생물을 이용한 원예 및 채소작물의 건전 유묘 생산을 위하여 본 연구에서는 growth pouch 실험을 통하여 6가지 식물생육촉진 미생물을 접종하여 작물의 초기 뿌리 생장에 미치는 영향을 살펴 보았다. 본 실험에 사용한 6가지 균주는 Methylobacterium oryzae CBMB20, Methylobacterium phyllosphaerae CBMB27, Methylobacterium suomiense CBMB120, Methylobacterium strains CBMB12, CBMB15와 CBMB17이었다. 대조구와 비교했을 CBMB12, CBMB17, 및 CBMB20접종은 상추 초기 뿌리 생육에 유의성있는 효과를 보였고, CBMB102접종은 배추 초기 뿌리 생육에 유의성있는 효과를, CBMB27접종은 토마토의 초기 뿌리 생육에 유의성있는 효과를 보였다. 또한 Methylobacterium suomiense CBMB120접종은 오이, 토마토, 배추, 그리고 Methylobacterium strain CBMB12접종은 고추의 뿌리 초기 생장을 크게 촉진시켰음을 알 수 있었다. 위의 결과를 통하여 실험한 균주들을 각각의 작물 육묘 포트에 처리할 때에도 유묘의 생장속도를 촉진 시킬수 있음을 예상할 수 있다.
