Cytotoxic Activity of Vietnamese Herbal Medicines against A549 Cells (A549세포에 대한 베트남 약용식물의 세포독성)

  • Eighty methanol extracts prepared from Vietnamese plants were tested for their cytotoxic activity against A549 cells, a human lung carcinoma cell line. Nine extracts showed survival rates of tumor cells of less than 30% at $100\:{\mu}g/mL$ of the methanol extracts. Among them, three were less than 15%; Meliosma pinnata (VK03, 4%), Goniothalamus vietnamensis (VK05, 06, 0%), Garcinia sp. (VK50, 51, 0%) and Aglaia aphanamixis (VK63, 11%). Both leaf and root extracts of Goniothalamus vietnamensis and the leaf extract of a Garcinia species completely inhibited the growth of A549 cells at the concentration of $100\:{\mu}g/mL$ methanol extract.

3-Methylthiopropionic Acid of Rhizoctonia solani AG-3 and Its Role in the Pathogenicity of the Fungus

  • Studies were conducted to determine the role of 3-methylthioproprionic acid (MTPA) in the pathogenicity of potato stem canker, Rhizoctonia solani, and the concentrations required to inhibit growth of R. solani under laboratory and plant house-based conditions. The experiments were laid out in a completely randomized design with five treatments and five replications. The treatments were 0, 1, 2, 4, and 8 mM concentrations of MTPA. The purified toxin exhibited maximal activity at pH 2.5 and $30^{\circ}C$. MTPA at 1, 2, 4, and 8 mM levels reduced plant height, chlorophyll content, haulm fresh weight, number of stolons, canopy development, and tuber weight of potato plants, as compared to the control. MTPA significantly affected mycelial growth with 8 mM causing the highest infection. The potato seedlings treated with MTPA concentrations of 1.0-8.0 mM induced necrosis of up to 80% of root system area. Cankers were resulted from the injection of potato seedling stems with 8.0 mM MTPA. The results showed the disappearance of cell membrane, rough mitochondrial and cell walls, change of the shape of chloroplasts, and swollen endoplasmic reticulum. Seventy-six (76) hours after toxin treatment, cell contents were completely broken, cytoplasm dissolved, and more chromatin were seen in the nucleus. The results suggested that high levels of the toxin concentration caused cell membrane and cytoplasm fracture. The integrity of cellular structure was destroyed by the phytotoxin. The concentrations of the phytotoxin were significantly correlated with pathogenicity and caused damage to the cell membrane of potato stem base tissue.

THE INFLUENCE OF INSTRUMENTATION IN THE CANAL ON THE EXTENSION OF INFECTION (근관내(根管內) 기계조작(器械操作)이 감염확산(感染擴散)에 미치는 영향(影響))

  • This study was to confirm the influence of intracanal instrumentation whether the pulp infection could be extended to periapical tissue. Fifty three teeth (24 badly decayed and infected, 29 sound teeth) were employed for this experiment and grouped as follows; 1. The specimen taken from the tip of 12 infected pulp in which reamer was inserted to the canal up to apical 1/3 and cultured as long as 48 hours. After 24 hours culture 11 cases were positive and 1 ease was negative but the time of incubation elapsed as 48 hours a negative case turned to positive. 2. Broth immersed paper disc was placed for 1 minute on the tip of 12 infected teeth with a reamer inserted to the apical end and cultured as usual manner in the incubator. At 24 hour culture the growth was significant in 9 cases and after 48 hours total 12 cases were positive. 3. Reamer was inserted to apical 1/3 on 14 sterile pulp canals and specimens obtained from the root tip were cultured for 24 and 48 hours. The results on both group were negative. 4. Similar maner with No.3 except reamer tip was rest exactly at the apex revealed only 2 cases of positive at 48 hour culture. 5. The tip of 24 reamers which reached to apical 1/3 and apex of infected canal were cultured for 24 and 48 hours. At 24 hour culture the growth was evident. 6. The tip of 14 reamers which inserted to apical 1/3 of sterile canal showed negative at 24 hour culture. The 15 cases of the tip which reached to the apex of sterile canal were found negative except 3 positive cases at 48 hour culture.

The Effects of Sera from Amyotrophic Lateral Sclerosis Patients on Neuromuscular Transmission and Calcium Channels in Mice

  • Amyotrophic lateral sclerosis (ALS) is a degenerative neuromuscular disease of unknown etiology in which the upper and lower motor neurons are progressively destroyed. Recent evidences support the role of autoimmune mechanisms in the pathogenesis of ALS. This study investigated the effects of sera from ALS patients on neuromuscular transmission in phrenic nerve-hemidiaphragm preparations and on calcium currents of single isolated dorsal root ganglion (DRG) cells in mice. Mice were injected with either control sera from healthy adults or ALS sera from 18 patients with ALS of sporadic form, for three days. Miniature end plate potential (MEPP) and nerve-evoked end plate potential (EPP) were measured using intracellular recording technique and the quantal content was determined. Single isolated DRG cells were voltage-clamped with the whole-cell configuration and membrane currents were recorded. Sera from 14 of 18 ALS patients caused a significant increase in MEPP frequency in normal Ringer's solution $(4.62{\pm}0.14\;Hz)$ compared with the control $(2.18{\pm}0.15\;Hz).$ In a high $Mg^{2+}/low\;Ca^{2+}$ solution, sera from 13 of 18 ALS patients caused a significant increase in MEPP frequency, from $2.18{\pm}0.31$ Hz to $6.09{\pm}0.38$ Hz. Sera from 11 of 18 patients produced a significant increase of nerve-evoked EPP amplitude, from $0.92{\pm}0.05$ mV to $1.30{\pm}0.04$ mV, while the other seven ALS sera did not alter EPP amplitude. In the ALS group, EPP quantal content was also elevated by the sera of 14 patients (from $1.49{\pm}0.07$ to $2.35{\pm}0.07).$ MEPP frequency and amplitude in wobbler mouse were $4.03{\pm}0.53$ Hz and $1.37{\pm}0.18$ mV, respectively, which were significantly higher than those of wobbler controls (wobblers without the symptoms of wobbler). Sera from ALS patients significantly reduced HVA calcium currents of DRG cells to 42.7% at -10 mV. Furthermore, the inactivation curve shifted to more negative potentials with its half-inactivation potential changed by 6.98 mV. There were, however, significant changes neither in the reversal potential of $I_{Ca}$ nor in the I-V curve. From these results it was concluded that: 1) The serum factors of sporadic ALS patients increase neuromuscular transmission and can alter motor nerve terminal presynaptic function. This suggests that ALS serum factors may play an important role in the early stage of ALS, and 2) Calcium currents in DRG cells were reduced and rapidly inactivated by ALS sera, suggesting that in these cells, ALS serum factors may exert interaction with the calcium channel.

Humidification and Shading Affect Growth and Development of Cutting Propagated 'Maehyang' Strawberry (Fragaria × ananassa Duch.) at Propagation Stage (삽목번식 시 가습과 차광 처리에 따른 '매향' 딸기의 생육)

  • This study was conducted to examine the effect of humidification and shading during cutting propagation on growth and development of strawberry (Fragaria x ananassa Duch.) 'Maehyang' plants at a propagation stage. The runner cuttings were stuck on Nov. 23, 2017 in propagation benches set in a Venlo-type glasshouse. Four shading treatments, no shading (control, C), 55% shading with white lawn (W55), 55% black shading net (B55), or 100% black plastic film (B100) with either an intermittent fog system (H) or without fog system. The shading and fog systems were removed 2 weeks after sticking of strawberry cuttings. A nutrient solution for strawberry, which was developed by Yamazaki, was supplied once a day with electrical conductivity (EC) $1.6dS{\cdot}m^{-1}$ and pH 5.8. Growth parameters such as plant height, longest root, crown diameter, leaf chlorophyll, leaf area and fresh and dry weight were measured at 7 days and 26 days after sticking. There was no significant difference in growth of above-aerial part of strawberry. The overall growth of the strawberry roots was better grew by providing fog than that not provide fog. The root fresh weight and root dry weight after 26 days after sticking of strawberry cutting was the best in the treatment that provided fog system without shading (CH). The longest root after 26 days after sticking of strawberry cutting was the best in the treatments that provided fog system with either 55% white lawn (W55H) and 55% black shading net (B55H). These results suggest that morphogenesis of these plants were affected by humidification and shading types. In a broader perspective, these results can be used to optimize studies of other crops grown from cuttings.

Potential to mitigate ammonia emission from slurry by increasing dietary fermentable fiber through inclusion of tropical byproducts in practical diets for growing pigs

  • Objective: Research was conducted to test the effect of including fiber-rich feedstuffs in practical pig diets on nutrient digestibility, nitrogen balance and ammonia emissions from slurry. Methods: Three Vietnamese fiber sources were screened, namely cassava leaf meal (CL), cassava root residue (CR), and tofu by-product (TF). Accordingly, a control diet (Con) with 10% of dietary non-starch polysaccharides (NSP) and three test diets including one of the three fiber-rich feedstuffs to reach 15% of NSP were formulated. All formulated diets had the same level of crude protein (CP), in vitro ileal protein digestible and metabolisable energy, whereas the in vitro hindgut volatile fatty acid (VFA) production of the test diets was 12% to 20% higher than the control diet. Forty growing barrows with initial body weight at $28.6{\pm}1.93kg$ ($mean{\pm}standard$ deviation) were allocated to the four treatments. When pigs reached about 50 kg of body weight, four pigs from each treatment were used for a nitrogen balance trial and ammonia emission assessment, the remaining six pigs continued the second period of the feeding trial. Results: The TF treatment increased fecal VFA by 33% as compared with the control treatment (p = 0.07), suggesting stimulation of the hindgut fermentation. However, urinary N was not significantly reduced or shifted to fecal N, nor was slurry pH decreased. Accordingly, ammonia emissions were not mitigated. CR and CL treatments failed to enhance in vivo hindgut fermentation, as assessed by fecal VFA and purine bases. On the contrary, the reduction of CP digestibility in the CL treatment enhanced ammonia emissions from slurry. Conclusion: Dietary inclusion of cassava and tofu byproducts through an increase of dietary NSP from 10% to 15% might stimulate fecal VFA excretion but this does not guarantee a reduction in ammonia emissions from slurry, while its interaction with protein digestibility even might enhance enhanced ammonia emission.
