Study on the Antioxidative Effects and Amino Acid Contents of the Roots of Cynanchum Wilfori (백하수오(白何首烏)의 항산화활성(抗酸化活性)과 amino acid의 분포(分布)에 관한 실험적(實驗的) 연구(硏究))

  • The aim of the presents study is to investigate and compare the antioxidative effects and qualities of the cultivating root of Cynanchum Wilfori, which is increasingly used in recent days, with those of the wilding root, which has mainly been used in the past in oriental medicine for a tonic and also for prevention and treatment of various geriatric diseases including aging progress. For comparison of their antioxidative effects, the activities of the total extracts on lipid peroxidation and the activity of aldehyde oxidase(EC 1.2.3.1) as well as xanthine oxidase(EC 1.2.3.2) were investigated in vitro. In addition, their inhibitory effects on the activity of 5-lipoxygenase, which is known to induce inflammation and concerned with free radicals, were also determined in vitro. Furthermore, the amino acid contents of both roots were analyzed in order to compare their qualities. The results are as follows: 1. The wilding root inhibited significantly the activity of 5-lipoxygenase, showing five times more portent than the cultivating root. 2. Both of the wilding root and the cultivating root inhibited aldehyde oxidase activity in a dose-dependant manner. The wilding root was more effective than the other. 3. Both of the wilding root and the cultivating root dose-dependently suppressed lipid peroxidation in rat brain, kidney, and liver. 4. The anti-peroxidative effects of both roots appeared to be most strong in brain and least in liver. In particular, the cultivating root exhibited a significant inhibition on brain lipid peroxidation. 5. The cultivating root contained 15 amino acids including five essential amino acids in contrast with the less contents in the wilding root.

  • The effect of various supplies of lead singly and in combination with aluminium on growth and chlorophyll biosynthesis was investigated in 7-day-old Vigna anguluris seedlings. Expose to 50 $\mu$N Pb or more drastically reduced root elongation rate. Significant depressions in root growth was observed within 1 day and no recovery of growth was seen over the duration of treatment period. Root elongation decreased depending on the Pb concentrations. Root growth inhibition was stronger than shoot growth inhibition. The initiation of lateral roots appeared to be more sensitive to Pb than the growth of main roots. Inhibition of root and shoot elongation by Pb was lessened by combined exposure of Pb and Al, suggesting that the presence of AA reverse the inhibitory effect of Pb alone. With the histochemical sodium rhodizonate method the rate of Pb uptake was dependent on the Pb concentration and exposure time of the roots to Pb salts. Pb was first deposited on the root surface and then translocated radially in the root cap cells. During a longer Pb administration (up to 72 h) Pb penetration was nonuniform, with accumulation within the cortex or endodermis. There was drastic reduction in chlorophyll content by Pb. The Pb inhibition of chlorophyll synthesis was concentration dependent. 5-Aminolevulinic acid dehydratase (ALAD) activity exhibited distinct inhibition from control. Reduction in chlorophyll content was accompanied by proportional changes in ALAD activity. Chlorophyll content and ALAD activity were less affected by combined exposure of Pb and Al, suggesting that Al has a protective effect against the inhibiting action of Pb on photosynthetic activity.

EFFECT OF POLYPHOSPHATE IN ROOT CANAL SEALERS ON THE GROWTH OF ORAL BACTERIA (Polyphosphate가 함유된 근관충전재가 구강세균의 성장에 미치는 영향)

  • Eliminating the infecting bacteria of the root canal system and preventing reinfection must be the main objectives of all endodontic works. None of commercially available root canal sealers have the properties of desirable tissue compatibility and strong antibacterial activity. The purpose of this study is to develope an ideal root canal sealer using commercially available polyphosphate (polyP), Calgon, which is known to be antibacterial and safe. For the study. resin type AH26, zinc oxide eugenol type Tubli Seal. Ca(OH)$_2$ type Apexit as base sealers for polyP (0~3%) and para formaldehyde containing N2 as a control base were selected. Specimens (3$\times$4mm) of the sealers were prepared in a 37$^{\circ}C$ incubator for 3 and 10 days and their antibacterial activity against streptococci and black pigmented anaerobic rods was observed using an agar diffusion method. The result were as follows: 1. Among 3 day old root canal sealers. N2 as a positive control showed the strongest antibacterial effect. followed by AH26. Tubli Seal and. Apexit which barely showed antibacterial activity against the test bacteria. In contrast. 10 day old AH26 showed a greater antibacterial activity than 10 day old N2. 2. All sealer specimens showed a greater antibacterial activity against black pigmented anaerobic rods than streptococci. Three day old ones appeared to be more antibacterial than 10 day old ones except for Apexit. 3. As compared to N2, 3 day old AH26 demonstrated a similar antibacterial activity against black pig mented anaerobic rods but to a lesser extent to streptococci. Ten day old AH26 showed a greater antibacterial activity against black pigmented anaerobic rods than 10 day old N2. 4. As compared to AH26. Tubli Seal generally revealed a lower antibacterial activity but it showed a greater antibacterial activity aginst S. gordonii Challis. 5. Enhancement of antibacterial activity by polyP was more clearly observed when it was added to Ca(OH)$^{\circ}C$ based root canal sealers. Tubli Seal and N2. 6. The addition of polyP enhanced the antibacterial activity of 3 day old AH26 against S. gordonii G9B (16%) and Challis (29%), and P. gingivalis 2561 (24%) only. Moreover, polyP failed to increase antibacterial activity of 10 day old AH26 against the test strains but P. gingivalis A7A1 28(13%). 7. The addition of polyP increased the antibacterial effect of 3 day old Tubli Seal on several test bacteria including s. mutans GS 5 (50%). s. gordonii G9B (47%) and Challis (122%). and all the test strains of P. gingivalis (13~35%) except for 9 14K 1. The addition of polyP to 10 day old Tubli Seal increased antibacterial activity of the root canal sealer against most test strains. 8. 3 day old Apexit failed to show antibacterial activity. if any very little against S. mutans GS 5 and Pr. intermedia ATCC 49046. However. polyP increased its antibacterial activity by 50 and 69%, respectively. Increase of antibacterial activity of 10 day old Apexit by polyP was more clearly observed than that of 3 day old one.

  • The activity of Daucus carota L. root $\beta$-glucan synthetase II was observed to increase in the presence of polyamines such as putrescine, spermidine and spermine in vitro, whereas the activity of Daucus carota L. root $\beta$-glucan synthetase I was not affected by the polyamine. The activity $\beta$-glucan synthetase II from Daucus carota L. root protoplasts cultured on medium containing 10-6 M polyamines such as putrescine, spermidine and spermine was observed to be higher than that of the control. Daucus carota L. root protoplasts were observed to have the activities of arginine and ornithine decarboxylases and it was noted that they could produce polyamines, which might have an effect on $\beta$-glucan synthetase II activity.

Effects of Antioxidative, DPPH Radical Scavenging Activity and Antithrombogenic by the Extract of Sancho (Zanthoxylum Schinilolium) (산초(Zanthoxylum Schinifolium) 뿌리, 줄기 및 잎 추출물의 항산화, DDPH Radical 소거 작용 및 항혈전 효과)

  • Effects of root, stem and leaf extract of sancho (Zanthoxylum schinifolium) on the inhibition of lipid peroxidation in the hepatic microsome of rat, DPPH radical scavenging activity and activated partial thromboplastin times (APTT) were examined in vitro. The highest inhibition of hepatic microsomal lipid peroxidation was observed by ethyl acetate fraction than that of methylene chloride fraction of the root and stem extracts. The high inhibition of lipid peroxidation was determined in the leaf, the root and the stem in order. The DPPH radical scavenging activity of ethyl acetate fraction was higher than that of n-butanol fraction and it was similar to the root and the steam extract. It was similar to the inhibition of hepatic microsomal lipid peroxidation. The DPPH radical scavenging activity was the highest in 2.500mg/mL of ethyl acetate fraction and it was 4.4 fold higher than that of $\alpha-tocopherol$, as an antioxidant standard. The DPPH radical scavenging activity was dependent on the extract concentration in the range of 0.125-5.000 mg/mL. The throm-boplastin times were higher than that of n-butanol fraction and it was similar to the root and the steam extracts. The leaf extract showed the highest antithrombogenic effect followed by the stem and then the root extract. The activated partial thromboplastin times were ependent on the extract concentration in the range of 0.100-2.000 mg/mL. Consequently, the effects of antioxidative, DPPH radical scavenging activity and antithrombogenic of Z. schinifolium was observed due to the inhibition of lipid peroxidation and the DPPH radical scavenging activity by methylene chloride, n-butanol and ethyl acetate fraction of the leaf extract. Antioxidant and Antidiabetic Effects of Various Sections of Astragalus membranaceus (연근별 황기의 항산화 및 항당뇨 활성 평가 고찰)

  • The antioxidant and anti-diabetes effects were evaluated by the ethanol extracts from Astragalus membranaceus root classified by years using through DPPH radical scavenging activity, hydroxyl radical (${\cdot}OH$) scavenging activity, total phenolic and flavonoid contents, reducing power activity, $\alpha$-amylase and $\alpha$-glucosidase inhibitory activities. $IC_{50}$ values for DPPH radical scavenging activity of ethanol extract from 6 years old root ($749.25{\mu}g/mL$) was higher than 1 year ($1452.67{\mu}g/mL$) and 3 years old root ($1095.61{\mu}g/mL$). 6 years old root showed better effects in ${\cdot}OH$ scavenging activity ($IC_{50}$: $10.58{\mu}g/mL$), reducing power, total phenolic contents ($26.13{\pm}0.79\;Tan\;{\mu}g/mg$, $24.03{\pm}0.74\;Cat\;{\mu}g/mg$) $\alpha$-amylase ($33.33{\pm}0.55%$) and $\alpha$-glucosidase inhibitory activity ($49.71{\pm}1.01%$). On the other hand, total flavonoid compound contents were estimated much higher in 1 year old root ($44.93{\pm}1.35\;Que\;{\mu}g/mg$, $70.32{\pm}2.03\;Rut\;{\mu}g/mg$) than others. Based on these results, It was suggested that 6 years old root of A. membranaceus has a potential candidate for functional cosmetic and medicine.

  • In order to investigate the inter-relation with the growth of the rice-root and its transaminase-activity, by measuring the growth of its root and transaminase-activity supplying this root with various nitrogen Compounds($NO_3$-N, $NH_4$-N and Amino acid.). The obtained results are summarized as follows; 1. Growth of rice-root supplied with $NO_3$-N is generally increased in length and weight, compared with that of the root fertilized by $NH_4$-N. 2. The above-metnioned root with $NH_4$-N is not only decreased in its weight and length but also is apt to inhibited its growth as the nitrogen concentruration is increased, in compared with the root provided with $NO_3$-N. 3. The activity of G.O.T. and G.P.T. for the root fertilized by $NH_4$-N, the badly grown root is generally increased, while of the root supplied with $NO_3$-N is decreased. 4. The activity of G.O.T. and G.P.T. for the root provided with amino acid known as the considerable growth inhibiting compound is generally decreased, while that of the badly-grown root is increased. 5. The activity of G.O.T. and G.P.T. in the supernatant fraction of the rice-root is for the most part, high and low in the mitochondrial fraction.

Antimicrobial Activity in 2 Angelica Species Extracts (당귀속 2종 식물의 항균활성)

  • We investigated antimicrobial activity of methanol extract from Angelica gigas and A. acutiloba against three gram-positive bacteria(Bacilius cereus, B. subtilis, and Staphylococcus aureus), two gram-negative bacteria (Escherichia coli and Pseudomonas fluorescens), and one yeast (Saccharomyces cerevisiae). The antimicrobial activity was shown in only ethyl acetate fraction of shoot and root extracts from A. gigas. The activity of the fraction of shoot extract was remarkably potent against S. aureus. The ethyl acetate fraction of root extract showed great antimicrobial activity against B. subtilis and E. coli. The n-hexane fraction of shoot extract and the water fraction of root extract from A. acutiloba were not showed antimicrobial activity, in contrast, the ether fraction of shoot and root extracts showed great antimicrobial activity. S. cerevisiae was not inhibited by any fractions and species.

  • We investigated the physicochemical and antioxidant activity characteristics of Yakju with different ratios (0, 5, 10, and 15%) of dandelion (Taraxacum officinale) root powder in order to find out potential as a functional fermented rice alcohol. Yakju samples were measured to obtain total flavonoid content, total polyphenol content, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, and reducing power. Total flavonoid and polyphenol contents increased with increasing amounts of dandelion root. The DPPH radical scavenging activity increased with the addition of dandelion root. Also, the reducing power was higher in Yakju with dandelion root compared with controls such as gallic acid, butylated hydroxytoluene, and ${\alpha}$-tocopherol. It is concluded that the Yakju fermented with additional dandelion root is functionally superior to the rice alcohol without dandelion root.
