배추무사마병의 뿌리혹 형성에 미치는 묘령, 접종원 농도 및 접종방법의 영향 (Effects of Plant Age Inoculum Concentration and Inoculation Method on Root Gall Development of Clubroot Disease of Chinese Cabbage Caused by Planmodiophora brassicae)

  • Effect of inoculum concentration inoculation method and plant age on development of clubroot disease of Chinese cabbage seedling were examined in growth chambers. Root galls were developed at the concentration of 105 resting spore or above per ml of incoulum and as the inoculum concentration became higher rate of development of root galls was faster. In the plants with root gall development fresh weight of above ground parts was reduced to 30-44% of that of healthy plants but root weight increased by 4-10 times. Growth of diseased plants was greatly reduced as compared to healthy plants. Planting in the diseased soil as a inoculation method was most effective for disease development showing uniform infections but time of initial root gall development was delayed by root soaking inoculation. Some plants inoculated by soil drenching method did not develop root galls. However root gall enlargement after its initial formation did not differ greatly among inoculation methods. Nine-day-old seedlings showed poor development of root gall but 16-days-old seedlings was found to be most adequate for inoculation for gall development.

  • Development of root galls of clubroot disease on Chinese cabbage seedlings was first observed 17days after inoculation of Plasmodiophora brassicae at $25^{\circ}C$ 4-11days earlier than at 5, 20, 3$0^{\circ}C$ and 35$^{\circ}C$. Subsequent enlargement of root galls was also fastest at $25^{\circ}C$ and 2$0^{\circ}C$ but delayed at 15$^{\circ}C$ and 3$0^{\circ}C$ or above. Chinese cabbage seedlings with root gall formation showed reduction in number of leaves above ground fresh weight and amount of root hairs but increase in root weight, Root galls development was highest at soil moisture level of 80% of maximum soil moisture capacity than at 60% and 100%. Optimum soil pH for root gall development was pH 6 although root galls were formed at a range of pH 5 to 8. Period of light illumination also affected root gall development with the greatest gall development at 12hr/12hr in light/dark period and the least at 8hr/16hr. Site of root gall formation and gall shape did not differ greatly among treatments of temperature soil moisture pH and light experiments.

  • The successful restoration program requires a comprehensive understanding of variables influencing seedling efficiency. Below-ground is hypothesized to have a major impact on seedling performance of species when planted in agriculture, and degraded areas with different types of mulching. This study investigated on Sg. Terla Forest Reserve in Cameron Highlands Pahang, Malaysia. In this study randomized complete block design (RCBD) was used. The excavation method was applied to study the root system development, above, and below ground biomass distributions under different types of mulching: coconut mulching (CM), oil palm mulching (OM), plastic mulching (PM) and control (CK). The root diameter, main root length, lateral root length, root coiling, and root direction toward to sun were recorded. The results in this study indicate that mulching had significant effect on root diameter, main root length, and root distributions among treatments while for lateral root length, root: shoot ratio, dry biomass distributions, and above and below ground biomass did not showed significant effect among treatments. The highest values for root diameter, lateral root length, main root length, root distributions, dry biomass distributions and above and below ground biomass were showed in CM treatments. However 75% of root coiling was observed in seedlings between treatments.

  • The study was carried out to clarify reasonable production of healthy seedling, optimal Pot depth, and appropriate transplanting time, which can be deduced from understanding of seedling quality. Seedling quality results from growth of root and shoot, morphology and distribution of root system under influence of Pot depth during seedling growing period. Stem height, shoot dry weight, leaf area and leaf number were increased in proportion to depth of pots. Growth of shoot and root during seedling growing period showed the most dramatic development between 20th and 25th day after temporary planting. Root number increased as pot depth decrease and total root length and dry weight increased as pot depth increase. In 5cm pot, relative multiplication rate was higher and mean extension rate was lower than other depth of Pot. The limitation of pot volume in which rhizosphere was located enhance the development of roots of second and third order. At 20th days after temporary Planting root distribution was relatively uniform in length and development of adventitious root on stem base was poor as Pot depth decreased.

  • Approximately 80% of rice field in Africa conducts in rainfed (Nishimaki 2017). The rice is damaged by water stress because fields like rainfed lowland repeat drying and humidity of soil because of impossible water control. Then water stress is one of the major limiting factors for decreasing rice yield. So, in initial growth stage, quick and efficient root development is useful way to avoid drought stress by getting water from deeper soil layer with roots elongation as the hypothesis. Daniel et al (2016) reported that NERICA1 and NERICA4 show different patterns of root plasticity for drought stress. NERICA1 has greater development of lateral root in shallow soil layer, while NERICA4 has greater development in deep root elongation to underground. This study was aimed to evaluate the effect of root development in initial growth stage on growing NERICA1 and NERICA4 under different soil moisture condition in rainfed lowland rice field. They were grown in same water condition until 35 days after sowing (35DAS), and after that each varieties were separated in dry and wet condition. The rice plants were grown until 60DAS. The results of soil moisture, the root extension angle, shoot dry weight and bleeding ratio showed that NERICA4 can mitigate dry stress from surface soil compered to NERICA1.

  • In order to evaluate the relationship of age with the developmental stage on mandibular second molars in female, the author examined the radiographs in standard films taken by intraoral paralleling technic and analysed the development of 620 roots of 310 females ranged from 10 to 18 years. The development was divided into 7 stages : Crown complete (Cr.C.), Root lengthed 1/4(R.1/4), Root length 1/2 (R.1/2), Rooth length 3/4 (R.3/4), Rooth length complete (R.C), Apex 1.2 closed (A. 1/2), Apical closure complete (A.C) The obtained results were as follows : 1. The formation of roots in fulllength of mandibular second molars was completed at 14.21-year-age in mesial root and at 14.96-year-age in distal root. 2. The formation of apical foramen of mandibular second molars was completed at 17.11-year-age in mesial root and at 17.53-year-age in distal root. 3. The mesial root of mandibular second molar was generally developed earlier than distal root (p<0.01) 4. Coefficients of correlation and regression equations between for age(Y) and the developmental stage of mandibular second molar(X) were "Y=1.53X+6.54, r=0.91 (p<0.01)" in mesial root and "Y=1.52X+7.11, r=0.92(p<0.01)" in distal root respectively.l root respectively.

Different Structural Modifications Associated with Development of Ginseng Root Rot Caused by Cylindrocarpon destructans

  • Root rot caused by Cylindrocarpon destructans is one of the most important diseases of ginseng (Panax ginseng C. A. Meyer). Two types of symptoms found in ginseng root rot are black root rot and rusty root (rusty spots), in which disease severities are high and low, respectively. Symptom development and related histopathological changes were examined in an inoculation test on 2-year-old ginseng roots using virulent (Cy9801) and avirulent (Cy0001) isolates of C. destructans under different temperature conditions (13, 18, 23, and $28^{\circ}C$). Black root rot was only induced by Cy9801 in the lower temperature range (13, 18, and $23^{\circ}C$) and not at the higher temperature ($28^{\circ}C$). No black root rot, but only rusty spot symptoms, were induced by Cy0001 at all temperatures tested except $13^{\circ}C$, at which no symptoms occurred on over half of inoculation sites, suggesting disease development was influenced by pathogen virulence and temperature. Wound periderms were formed in all root tissues with rust spot symptoms at $28^{\circ}C$ caused by Cy9801 and at 18, 23, and $28^{\circ}C$ temperatures caused by Cy0001. No wound periderm was formed at $13^{\circ}C$ by either Cy9801 or Cy0001. Light microscopy revealed that the wound periderm was formed by initial cell divisions in cell wall formation and/or additional cell wall layering in parenchyma cells without obvious nuclear division, followed by layering of the divided cells adjacent to the inoculation sites, blocking the spread of the rot. These results suggest that disease development declined at lower temperatures and by the formation of a wound periderm at higher temperatures, and that ginseng rusty root may develop under conditions unfavorable for further disease development of C. destructans.

Defoliation Effects on Root and Rhizome Development of Kura Clover

  • There is limited information on the relationship between defoliation and root and rhizome development of kura clover (Trifolium ambiguum M. Bieb.). To determine the effects of defoliation severity on root and rhizome growth of young kura clover plant (seedling about 8 wk old), this research was conducted in 2002 (Experiment 1) and 2003 (Experiment 2) in a glasshouse at the University of Wisconsin-Madison. Four kura clover entries were used in this experiment: two were started from seed materials (ARS-2678 and 'Rhizo') and two were clones from mature, field-grown Rhizo kura clover plant. Three defoliation frequencies (2-, 4- and 6-wk intervals) and two defoliation intensities (complete and partial defoliation) were imposed on each of the four kura clover entries. Root, rhizome, and leaf dry matter (DM) generally increased with less frequent defoliation, however, the increase in rhizome DM was not significant between 4- or 6-wk defoliation periods. The root and leaf DM under complete defoliation (CD) were significantly lower than under partial defoliation (PD). In Exp. 1, rhizome DM was not significantly different between CD and PD; it was significantly lower under CD in Exp. 2. ARS-2678 showed excellent root development characteristics, however, its rhizome DM was significantly lower than Rhizo clones. The rhizome development from Rhizo clones was greater than that from seed materials. If maximum root and rhizome growth are expected from young kura clover plant, the intensity and frequency of defoliation should be minimized or defoliation should be avoided.

The Danger-Associated Peptide PEP1 Directs Cellular Reprogramming in the Arabidopsis Root Vascular System

  • When perceiving microbe-associated molecular patterns (MAMPs) or plant-derived damage-associated molecular patterns (DAMPs), plants alter their root growth and development by displaying a reduction in the root length and the formation of root hairs and lateral roots. The exogenous application of a MAMP peptide, flg22, was shown to affect root growth by suppressing meristem activity. In addition to MAMPs, the DAMP peptide PEP1 suppresses root growth while also promoting root hair formation. However, the question of whether and how these elicitor peptides affect the development of the vascular system in the root has not been explored. The cellular receptors of PEP1, PEPR1 and PEPR2 are highly expressed in the root vascular system, while the receptors of flg22 (FLS2) and elf18 (EFR) are not. Consistent with the expression patterns of PEP1 receptors, we found that exogenously applied PEP1 has a strong impact on the division of stele cells, leading to a reduction of these cells. We also observed the alteration in the number and organization of cells that differentiate into xylem vessels. These PEP1-mediated developmental changes appear to be linked to the blockage of symplastic connections triggered by PEP1. PEP1 dramatically disrupts the symplastic movement of free green fluorescence protein (GFP) from phloem sieve elements to neighboring cells in the root meristem, leading to the deposition of a high level of callose between cells. Taken together, our first survey of PEP1-mediated vascular tissue development provides new insights into the PEP1 function as a regulator of cellular reprogramming in the Arabidopsis root vascular system.

Screening of Selected Korean Sweetpotato (Ipomoea batatas) Varieties for Fusarium Storage Root Rot (Fusarium solani) Resistance

  • A common post-harvest disease of sweetpotato tuber is root rot caused by Fusarium solani in Korea as well as the other countries. Storage root rot disease was monitored earlier on sweetpotato (Ipomoea batatas) in storehouses of different locations in Korea. In the present study, an isolate SPL16124 was choosen and collected from Sweetpotato Research Lab., Bioenergy Crop Research Institute, NICS, Muan, Korea, and confirmed the identification as Fusarium solani by conidial and molecular phylogenetic analysis (internal transcribed spacer ITS and translation elongation factor EF 1-α gene sequences). The isolate was cultured on potato dextrose agar, and conidiation was induced. The fungus was screened for Fusarium root rot on tuber of 14 different varieties. Among the tested variety, Yenjami, Singeonmi, Daeyumi, and Sinjami showed resistant to root rot disease. Additionally, the pathogen was tested for pathogenicity on stalks of these varieties. No symptom was observed on the stalk, and it was confirmed that the disease is tissue specific.
