Characterization of rock bream (Oplegnathus fasciatus) fin cells and its susceptibility to different genotypes of megalocytiviruses

  • Genus Megalocytivirus cause red sea bream iridoviral disease (RSIVD) and scale drop disease (SDD). Based on the phylogeny of the major capsid protein (MCP) and adenosine triphosphatase (ATPase) genes, megalocytiviruses except for SDD virus (SDDV) could be three different genotypes, red sea bream iridovirus (RSIV), infectious spleen and kidney necrosis (ISKNV), and turbot reddish body iridovirus (TRBIV). In this study, primary cells derived from the caudal fin of rock bream (Oplegnathus fasciatus) grew at 25℃ in Leibovitz's medium supplemented with 10% (v/v) fetal bovine serum and primocin (100 ㎍/mL). Rock bream fin (RBF) cells exhibited susceptibility to infections by different genotypes of megalocytiviruses (RSIV, ISKNV and TRBIV) with the appearance of cytopathic effects with an increase in the viral genome copy number. Furthermore, compared to grunt fin (GF) cells, even though 10 times lower number of RSIV genome copies were inoculated in RBF cells, viral genome copy number produced on RBF cells were 44 times higher than that of GF cells at 7 d post-inoculation. As the isolated RBF cells are sensitive to different genotypes of megalocytiviruses (RSIV, ISKNV and TRBIV), they can be used for future studies regarding in vitro viral infection and subsequent diagnosis.

Transcriptional profiles of rock bream iridovirus (RBIV) using microarray approaches

  • Rock bream iridovirus (RBIV) causes high mortality and economic losses in the rock bream (Oplegnathus fasciatus) aquaculture industry in Korea. Viral open reading frames (ORFs) expression profiling at different RBIV infection stages was investigated using microarray approaches. Rock bream were exposed to the virus and held for 7 days at 23 ℃ before the water temperature was reduced to 17 ℃. Herein, 28% mortality was observed from 24 to 35 days post infection (dpi), after which no mortality was observed until 70 dpi (end of the experiment). A total of 27 ORFs were significantly up- or down-regulated after RBIV infection. In RBIV-infected rock bream, four viral genes were expressed after 2 dpi. Most RBIV ORFs (26 genes, 96.2%) were significantly elevated between 7 and 20 dpi. Among them, 12 ORF (44.4%) transcripts reached their peak expression intensity at 15 dpi, and 14 ORFs (51.8%) were at peak expression intensity at 20 dpi. Expression levels began to decrease after 25 dpi, and 92.6% of ORFs (25 genes) were expressed below 1-fold at 70 dpi. From the microarray data, in addition to the viral infection, viral gene expression profiles were categorized into three infection stages, namely, early (2 dpi), middle (7 to 20 dpi), and recovery (25 and 70 dpi). RBIV ORFs 009R, 023R, 032L, 049L, and 056L were remarkably expressed during RBIV infection. Furthermore, six ORFs (001L, 013R, 052L, 053L, 058L, and 061L) were significantly expressed only at 20 dpi. To verify the cDNA microarray data, we performed quantitative real-time PCR, and the results were similar to that of the microarray. Our results provide novel observations on broader RBIV gene expression at different stages of infection and the development of control strategies against RBIV infection.

Effects of long double-stranded RNAs on the resistance of rock bream Oplegnathus fasciatus fingerling against rock bream iridovirus (RBIV) challenge

  • To determine whether rock bream Oplegnathus fasciatus can be protected from rock bream iridovirus (RBIV) infection by intramuscular injection of long double-stranded RNAs (dsRNAs), we compared protective effect of virus-specific dsRNAs corresponding to major capsid protein (MCP), ORF 084, ORF 086 genes, and virus non-specific green fluorescent protein (GFP) gene. Furthermore, to determine whether the non-specific type I interferon (IFN) response was associated with protective effect, we estimated the activation of type I IFN response in fish using expression level of IFN inducible Mx gene as a marker. As a result, mortality of fish injected with dsRNAs and challenged with RBIV was delayed for a few days when comparing with PBS injected control group. However, virus-specific dsRNA injected groups exhibited no significant differences in survival period when compared to the GFP dsRNA injected group. Semi-quantitative analysis indicated that the degree of antiviral response via type I IFN response is supposedly equal among dsRNA injected fish. These results suggest that type I IFN response rather than sequence-specific RNA interference might involve in the lengthened survival period of fish injected with virus-specific dsRNAs.

Long Double-stranded RNA Induces Sequence-specific RNA Interference and Type I Interferon Responses in Rock Bream (Oplegnathus fasciatus)

  • To determine whether long double-stranded RNA (dsRNA) induces RNA interference and type I interferon (IFN) responses in fish, long dsRNAs encoding enhanced green fluorescent protein (EGFP), GFPuv, and polyinosinic-polycytidylic acid sequences were co-injected with an EGFP expressing plasmid, into rock bream (Oplegnathus fasciatus). We investigated the EGFP mRNA and protein levels, and the transcriptional responses of dsRNA-dependent protein kinase and Mx1 genes. Long dsRNAs were strong inducers of a type I IFN response in rock bream, resulting in nonspecific suppression of exogenous gene expression. Furthermore, sequence-specific knockdown of exogenous gene expression at the mRNA level was detected at an early phase (24 h). These results suggested that long dsRNA may inhibit exogenous gene expression through an early mRNA interference response and a later type I IFN response in fish.

Behavioral analysis of rock bream Oplegnathus fasciatus reveals a strong attraction potential for sea urchin extracts

  • Monitoring fish movement is important to understand how physiology adapts to environmental change. To explore the applicability of a video tracking system for determining if chemical cues attract or repel aquatic animals, the movement patterns of rock bream, Oplegnathus fasciatus, were analyzed upon exposure to various materials, including extracts of sea urchin, rock worm, bait worm, krill, barley kernel, and commercial fish feed. Pellets were prepared by mixing freeze-dried tissues with a cellulose and corn flour mixture. Behavioral analysis was carried out with five fish that had been acclimated in the adaptation zone of a Y-shaped tank. Preference toward chemical cues was quantified by assessing the frequency rock bream were observed in a discrete zone around the test material located at the end of each arm and the duration each fish stayed in each zone. The analysis of fish movement upon exposure to commercial feed and barley kernel at each end, respectively, indicated a clear preference toward the feed relative to the barley kernel. Movement responses were further tested with pellets containing extracts of sea urchin, one of the species collected on a large scale, and other materials including krill and worms. A stronger preference toward sea urchin (100%) was observed based on the duration of stay in the test zone, compared to krill (90.1 ± 44.2%), bait worm (81.1 ± 39.1%), rock worm (73.7 ± 28.9%), and barley (63.9 ± 25.9%), under the conditions tested. A detailed comparison of rock bream movements toward each material revealed significant differences in frequency and duration, respectively, between pairs of test materials including krill (74 ± 29.8 and 375.6 ± 118.9) vs. rock worm (41.5 ± 18.7 and 160.2 ± 42.6), krill (86.3 ± 22.9 and 477.1 ± 84) vs. bait worm (36.2 ± 5.5 and 166.1 ± 50.7), and rock worm (45.9 ± 26.2 and 213.7 ± 100.1) vs. bait worm (34.6 ± 21.7 and 159.5 ± 98.5). Rock bream exhibited preference for the test materials in the following order: commercial fish feed > sea urchin > krill > rock worm > bait worm > barley. The results suggest a higher potency of sea urchin extract as a rock bream fishing bait compared to the other materials that are used as commercial bait.

The immune-adjuvant effect and safety of recombinant CC chemokine 1 (rRbCC1) in rock bream, Oplegnathus fasciatus

  • Adjuvants are immune enhancers that are often used in vaccination to augment the immune response of a vaccine, thereby enhancing the protective immunity against the targeted disease. In the present study, we used the recombinant protein, such as rRbCC1, this protein was produced from rock bream CC chemokine 1. To verify the adjuvant effects of this recombinant protein, the immune responses of rock bream to Streptococcus iniae (S. iniae) FKC vaccination, which alone or in combination with recombinant protein was analyzed and then also performed experimental challenge with live S. iniae. The result of serum agglutination titres was showed relatively low levels however, the efficacy of FKC vaccine still conferred protection against S. iniae. Moreover, the adverse effects result showed that no statistically significant difference was revealed between high concentration injected and non-injected fish groups, generally. The relative percent survival (RPS) of FKC + recombinant vaccination group was significantly higher than that of vaccinated group with FKC alone. After experimental challenge to the rock bream by injection with live bacteria (S. iniae), the FKC + rRbCC1 vaccination group was showed 87.0% RPS, however, the RPS of FKC alone vaccination was 68.2%. The results indicated that the recombinant protein as an adjuvant had a clear synergism to injection vaccine of rock bream.

Induction of antiviral mechanisms by interferon-related genes in rock bream (Oplegnathus fasciatus) infected with rock bream iridovirus (RBIV)

  • We evaluated the transcriptional response of interferon (IFN)-related genes in rock bream iridovirus (RBIV)-infected rock bream under high-, low-, or no-mortality conditions induced by different stocking water temperatures. Under the high susceptibility condition (group A, water temperature 26℃, 100% mortality), only the Mx gene was expressed early, with prolonged expression, and with heavy viral loads of approximately 106~107 major capsid protein gene copies/μL from 4 to 10 days post infection (dpi). However, IRF1, IRF3, IRF8, STAT1, ISG15, PKR, Viperin, GVIN1, IFI44, and ISG56 were activated at later time points (8 dpi) and then quickly decreased (10 dpi). For the low susceptibility condition, the water temperature was set at 23℃ for 7 days (group B) and then reduced to 17℃. Group B exhibited a 28% mortality rate, in which persistent and effective antiviral responses were observed for long periods of time. In particular, at 20 and 22 dpi, when virus replication was peaked at approximately 107/μL, the expressions of most of the IFN-related genes (IRF1, IRF3, IRF8, Mx, STAT1, ISG15, PKR, Viperin, GVIN1, IFI44, and ISG56) were significantly higher in group B than in the control group. Moreover, prolonged and higher levels of IRF3 (at least 30 dpi), IRF8 (at least 30 dpi), ISG15 (at least 30 dpi), PKR (at least 28 dpi), Viperin (at least 30 dpi), and IFI44 (at least 30 dpi) were also observed in the recovery stage of infection. Under the no-susceptibility condition at 17℃ (0% mortality), significantly elevated levels of IRF3, Mx, ISG15, and PKR were observed mostly until 20 dpi. The findings indicate that RBIV infection can induce an efficient IFN-mediated antiviral immune response in low- and no-susceptibility conditions. The findings could be valuable for effective control of viral pathogens in fish.

돌돔, Oplegnathus fasciatus의 Cyclooxygenase-2 유전자의 cloning 및 발현분석 (Cloning and Expression of the Cyclooxygenase-2 gene in the Rock bream, Oplegnathusfasciatus)

  • Megalocytivirus는 우리나라를 포함한 아시아 각국의 양식현장에서 고위험성 병원체이다. 그럼에도 불구하고 어류의 면역체계와 megalocytivirus의 상호관계에 관한 연구는 아직 부족한 실정이다. 다양한 연구에서 cyclooxygenase isoform중 COX-1 유전자는 constitutive 하게 발현되며, COX-2 enzyme은 면역반응에 중요한 역할을 한다고 알려져 있다. 또한 COX-2 유전자는 LPS (lipopolysaccharide) 또는 병원체의 감염과 같은 염증반응 시 그 발현이 증가한다고 알려져있다. 본 연구는 다른 어종의 COX-2 유전자를 바탕으로 제작된 degenerated primer와 5'- 그리고 3'-end RACE-PCR을 이용하여 돌돔에서 COX-2 유전자의 전체 염기서열을 밝혔으며, 그 결과 rbCOX-2 (rock bream COX-2)유전자 cDNA의 전체 길이는 2655 bp 였으며, 609개의 아미노산으로 구성되어있었다. rbCOX-2 유전자의 genomic organization은 9개의 intron과 10개의 exon으로 구성되어 있었다. 또한 본 연구에서는 돌돔에 megalocytivirus의 인위감염 시 COX-2 유전자의 발현을 조사하였다. LPS 접종 시 rbCOX-2 유전자는 접종 1일 후 대조구와 비교하여 13.10배 증가하여 최고 발현을 보였으나, megalocytivirus 접종 시 대조구와의 비교에서 유의적인 발현을 확인할 수 없었다. 돌돔에서 COX-2 유전자의 염기서열의 분석과 발현 분석은 바이러스 감염 시 어류의 방어기작을 이해하는데 도움이 될 것이며, 바이러스 백신개발 및 치료제 개발의 기초자료로 활용될 것이다.

Induced Morphological Changes in Larval Rock Bream, Oplegnathus fasciatus, under Starvation

  • Morphological changes in the reared rock bream, Oplegnathus fasciatus, from hatching to six days after hatching were examined during the early growth stage under starvation. All the larvae died within five days when feeding was delayed for three days after hatching. These results imply that initial larval food should be supplied within two days of hatching. Changes in the pectoral angle and the ratios of eye height to head height, gut height to standard length, and gut height to myotome height in the rock bream are alternative indicators for the identification of starving fish. These indicators might prove useful in evaluating the successful transition from endogenous to exogenous feeding in this species.

  • 남해안산 방어와 돌돔에 대한 수은, 구리 및 카드뮴의 중금속독성 실험을 1978년 7월 18일$\~$27일간 정수식으로 실험실에서 실시하였다. 실험결과 24시간 반수치사농도(24hr-LC50)는 방어에 있어서는 수은이 0.10mg/l, 카드뮴 0.82mg/l, 구리 1.03 mg/l였고, 돌돔에 있어서는 수은 1.61mg/l, 구리 1.73mg/l를 나타냄으로서 중금속별 독성 순위는 방어는 수은, 카드뮴 구리이고 돌돔은 수은-구리-카드뮴 순이며 도종류 공히 수은 독성이 가장 강함을 구명하였다.
