Effects of 5-azacytidine, a DNA methylation inhibitor, on embryogenic callus formation and shoot regeneration from rice mature seeds (벼 성숙종자로부터 배상체 캘러스 형성 및 식물체 재분화에 DNA methylation 억제제인 5-azacytidine의 영향)

  • The modification of DNA and histone plays an important role for gene expression in plant development. The objective of this research is to observe the effects of methylation on the gene expression during dedifferentiation from rice mature seeds to callus and differentiation from callus to shoots. The embryogenic callus with ability to shoot regeneration was not induced on the N6A medium supplemented with 5-azacytidine and abnormal callus with brown color was formed. When the normal rice callus was placed on the regeneration MSRA medium supplemented with 5-azacytidine, the shoot regeneration was inhibited. The results showed that 5-azacytidine, DNA demethylating agent, had negative effects on normal embryogenic callus formation and shoot regeneration. This suggested that DNA methylation of some genes was required for normal cell dedifferentiation and differentiation in tissue culture. The microarray and $GeneFishig^{TM}$ DEG screening were used to observe the gene transcript profile in callus induction and regeneration on N6A (N6 medium + 5-azaC) and MSRA (MS regeneration medium + 5-azaC). Subsets of genes were up-regulated or down-regulated in response to 5-azaC treatments. The genes related with epigenetic regulation, electron transport, nucleic acid metabolism and response to stress were up and down regulated. The different expression of some genes (germin like protein etc.) during callus induction and shoot regeneration was confirmed using RT-PCR and northern blot analysis.

Purification and Characterization of β-N-Acetylhexosaminidase from Rice Seeds

  • N-Acetyl-$\beta$-D-hexosaminidase ($\beta$-HexNAc'ase) (EC 3.2.1.52) was purified from rice seeds (Oryza sative L. var. Dongjin) using ammonium sulfate (80%) precipitation, Sephadex G-150, CM-Sephadex, and DEAE-Sephadex chromatography, sequentially. The activities were separated into 7 fractions($F_1-F_7$) by CM-Sephadex chromatography. Among them, F6 was further purified to homogeneity with a 13.0% yield and 123.3 purification-fold. The molecular mass was estimated to be about 52 kDa on SDS-PAGE and 37.4 kDa on Sephacryl S-300 gel filtration. The enzyme catalyzed the hydrolysis of both p-nitrophenyl-N-acetyl-$\beta$-D-hexosaminide (pNP-GlcNAc) and p-nitrophenyl-N-acetyl-$\beta$-D-hexosaminide (pNP-GalNAc) as substrates, which are typical properties of $\beta$-HexNAc'ase. The ratio of the pNP-GlcNAc'ase activity to the pNP-GalNAc'ase activity was 4.0. However, it could not hydrolyze chitin, chitosan, pNP-$\beta$-glucopyranoside, or pNP-$\beta$-glucopyranoside. The enzyme showed $K_m$, $V_{max}$ and $K_{cat}$ for pNP-GlcNAc of 1.65 mM, $79.49\;mM\;min^{-1}$, and $4.79{\times}10^6\;min^{-1}$, respectively. The comparison of kinetic values for pNP-GlcNAc and pNP-GalNAc revealed that the two enzyme activities are associated with a single binding site. The purified enzyme exhibited optimum pH and temperature for pNP-GlcNAc of 5.0 and $50^{\circ}C$, respectively. The enzyme activity for pNP-GlcNAc was stable at pH 5.0-5.5 and $20-40^{\circ}C$. The enzyme activity was completely inhibited at a concentration of 0.1 mM $HgCl_$ and $AgNO_3$, suggesting that the intact thiol group is essential for activity. Chloramine T completely inhibited the activity, indicating the possible involvement of methionines in the mechanism of the enzyme.

Eco-Friendly Control of Water Foxtail (Alopecurus aequalis) on Cultivation Fields of Forage Barley Sowed before Rice Harvest (벼 수확전 청보리 파종 재배시 친환경적 뚝새풀 관리)

  • This study was conducted to develop an eco-friendly control method for water foxtail in the field sowing barley seeds before rice harvesting. When sea water was applied pre-emergent, 1 and 2 leaves, little injury was observed on barley. Percent of water foxtail control with sea water (100%) was more than 50% at 1 leaf application timing. When sodium chloride was applied 400, 800 and $1,600kg\;ha^{-1}$, little injury was observed on barley. Percent of water foxtail control with sodium chloride was 36-44% at 30 days after sowing. When ferrous sulfate was applied at 400, 800 and $1,600kg\;ha^{-1}$, emergent injury was observed 35-50% on barley. Percent of water foxtail control with ferrous sulfate was 48-79% at 30 days after sowing. When barley was sowed at 200, 300 and $400kg\;ha^{-1}$, seeding rate standing plants have many of the more crops, water foxtail occurrence was low. And, Forage yield of 300 kg ha-1 seeding in the highest. Based on the results, it is possible to control water foxtail in the field sowing barley seeds before rice harvesting more than 50%.

Minimum Raising Duration of Infant Rice Seedling for Machine Transplanting (벼 기계이앙용 어린모 최소 육묘기간)

  • The duration of raising seedling of infant rice seedling(IRS) in machine transplanting is 8 to 10 days. This experiment was conducted to investigate the minimum duration of IRS's raising seedling and its method by the combination of several treatments such as plant growth regulator, root-break-seat and temperature. The rice seeds of Hwaseongbyeo was soaked in the solution of metalaxyl for 24 hours at room temperature. Metalaxyl (25% wettable powder), a fungicide, was used in 1, 000 times dilution as a promting substance on the root-mat formation of IRS. The application of fungicide in the nursery soil for the controlling of damping-off and physiologyical seedling rot was omitted due to the metalaxyl seed-soaking treatment. Seeding rate was 220g per seed tray (30$\times$60$\times$3cm). To promote the root-mat formation of IRS, the sheets of polyethylene vinyl and absorptive paper were placed bottom the seed tray. The root-mat formation of IRS was promoted at higher temperature and longer duration of raising seedling. The metalaxyl-treated seeds markedly increased the root length and rooting activity of IRS as compared with the control, thus the root-mat formation was excellent. The absorptive paper with polyethylene vinyl as root-break-seat showed a better root-mat formation compared with control, polyethylene vinyl, alone. The minimum duration of raising seedling of IRS was 5 days after sowing based on the root-mat formation and seedling height under the condition of metalaxyl seed treatment, absorptive paper with polyethylene vinyl as a root-break-seat and the raising seedling temperature 30/2$0^{\circ}C$ (day /night).

Influence of Rainfall During the Ripening Stage on Pre-Harvest Sprouting, Seed Quality, and Longevity of Rice (Oryza sativa L.)

  • The influence of rainfall during the ripening stage on pre-harvest sprouting, seed viability, and seed quality was investigated in two Korean rice cultivars, Shindongjin and Hopum. When the rainfall was artificially treated in a greenhouse, HP started to pre-harvest sprouting at three days of rainfall treatment (DRT), but Shindongjin did not show pre-harvest sprouting at 40 DAH treatment and just 0.3~0.8% at 50 DAH, which was much lower than 15.3~25.8% of Hopum in the same treatment. After harvest, the seed germination of Hopum decreased about 10~25% compared to non-treated seeds, but that of Shindongjin decreased much little rate than that of Hopum. The seed longevity tested by accelerated aging decreased with prolonged rainfall period in both cultivars, but the varietal difference was clear; Shindongjin could withstand longer accelerated aging than Hopum. Shindongjin maintained its germination (>50%) ability after 15 days of accelerated aging regardless of the rainfall treatment period and time, but Hopum dropped below 50% germination ability after only 5 days of accelerated aging. In conclusion, rainfall during the ripening stage induced not only pre-harvest sprouting, but also reduced seed quality and longevity during storage, which varied between two cultivars.

Food Preference Test of the Korean Elderly Menu Development (노인식 제공을 위한 일부 노인의 음식 기호도 조사)

  • This study was performed to test food preferences of the Korean elderly for Korean elderly menu development. The elderly subjects preferred steamed rice cooked with 1.2 times water than that with 1.5 times water (p < 0.001) , whole rice grain gruel than the gruel with ground grain (p < 0.05) . Green tea powder gruels developed as a snack, there were no significant differences in the preference of the elderly between gruel with different contents of green tea powder (2, 3, $4\%$). The preferences of the salt concentration of the soup were tested by using soybean sprout soup and soybean paste soup. The elderly preferred 0.7 - $0.8\%$ salted bean sprout soup and $7\%$ soybean paste soup. The elderly preferred Kimchi cut in widths of 1 - 2 cm than that in 0.5 cm. The elderly preferred sweeter grape jam ($67\%$ vs. $50\%$ or $37\%$ sugar content) and were not concerned about chewing seeds. This result could be useful to develop menus for the elderly. Studies on Practical Methods to Control Seed Vigour in Several Food Crops (주요식량작물 종자세 개선에 관한 연구)

  • To improve seed vigour in rice, barley and soybean, several methods of presowing treatment, using chemicals and priming in polyethylene glycol solution, were investigated. Gibberellic acid($GA_3$) slightly improved germination of rice, but other chemical treatments showed no beneficial effect on seed vigour. Aged seeds were primed in polyethylene glycol solution then rinsed and germinated with drying back. In general mean germination time increased and percentage germination decreased with in- creasing water potential of the priming solution, but there were no significant effects on spread of germination times. Priming also showed no marked improvement in germination under cold, wet, or osmotic conditions. None of the treatments used was successful in practically improving the seed germination and vigour of the tested crops. However, seed treated with GA$_3$ gave the best overall germination response.

Classification and Expression Profiling of Putative R2R3 MYB Genes in Rice

  • MYB genes, comprising group of related genes found in animal, plant, and fungal genomes, encode common DNA-binding domains composed of one to four repeat motifs. MYB genes containing two repeats (R2R3) constitute largest MYB gene family in plants. R2R3 MYB genes play important roles in regulation of secondary metabolism, control of cell shape, disease resistance, and hormone response. Eight-four R2R3 MYB genes were retrieved from rice genome for functional characterization of MYB genes. Analysis of MYB domains revealed each MYB domain contains three ${\alpha}$-helices with regularly spaced tryptophan residues. R2R3 MYB genes were divided into four subfamilies based on phylogenic analysis result. Real-time PCR analysis of 34 MYB genes revealed 12 MYB genes were highly expressed in seeds than in leaves, whereas 4 genes were highly expressed in leaves.

Incidence of Altermaria Species in Red Pepper and Sesame from Korea and Their Ability to Produce Mycotoxins (한국산 고추와 참깨에 발생하는 Alternaria의 종류와 이들의 진균독소 생성능력)

  • Alternaria alternata and A. solani were identified from 130 Alternaria isolates obtained from red pepper fruits, and three species including A. alternata, A. sesami and A. sesamicola were detected from 150 isolates from sesame seeds. Among the 4 Alternaria species, A. alternata was the predominant fungus from both plants, having incidence of 95.4% in red pepper and 56.0% in sesame. Of the total 280 isolates, cultures on autoclaved rice of 75 isolates were tested for toxicity to 21-day-old virgin female rats. Out of 50 isolates of A. alternata, 17 were lethal to rats, inducing congestion and hemorrhage of stomach and intestine and kidney enlargement, and 8 caused lack of weight gain or weight loss. The other 25 isolates of A. alternat and all the isolates of A. sesami, A. sesamicola and A. solani, showed no significant indication of toxicity. Production of mycotoxins in the rice cultures of the above 75 isolates belonging to 4 species was analyzed. Alternaria cultures were extracted with methanol and purified by using solvent partition, thin-layer chromatography, and high performance liquid chromatography. Of the four species tested, all produced alternariol (AOH) and alternariol monomethyl ether (AME), three (A. alternata, A. sesami and A. sesamicola) produced alternuene (ALT) and altertoxin-I (ATX-I), and only A. alternata produced tenuazonic acid (TA). TA was produced by all of the highly toxic (lethal to rats) isolates of A. alternata, but not by any nontoxic isolates.

  • This study was conducted to investigate the incidence of bacterial leaf blight (BLB) in Milyang 30 which was previously considered to be resistant to Xanthomonas campestris pv. oryzae Seeds of Milyang 30 were collected from Suweon, Milyang and Haenam. When rice plants from different source were inoculated at the maximum tillering stage and flag leaf stage, reactions of Milyang 30 were consistant. The susceptibility of Milyang 30 was found to be due to infection with tome isolates in the pathotype II of X. campestris pv. oryzae.
