Kwanok, Fujisaka #5, Paldal, and Suwon #82 as japonica type and IR-262 and CP-slo as indica type of rice seeds were selected for this experiment among varieties grown in Korea. Activities of crude enzymes extracted from germinating seeds of these varieties on malathion and p-nitrophenyl acetate as substrates, esterase zymograms with 1-naphthyl acetate as substrate, and some observations are summarized as follows: 1. Activities per unit volume of crude enzyme preparations on malathion were in the order of Kwanok>IR-262>Fujisaka #5>CP-slo>Paldal>Suwon #82. 2. Esterase zymograms on agar-gel electrophoretograms exhibited three to four bands two electrodes with little difference among varieties, nevertheless showing a wide and strongly-colored band toward cathode. Suwon #82 has a somewhat different pattern from others. 3. Enzyme activities per milligram protein with p-nitrophenyl acetate as substrate were in the order of CP-slo>IR-262>Paldal>Kwanok>Suwon #82>Fujisaka #5, indicating that activities of indica type are much stronger than those of japonica type, but not in agreement with results with malathion. 4. Malathion did not much inhibit the esterase activity at the concentration of 0.2PPM on electrophoretograms. 5. It is supposed that there is a complex esterase system hydrolyzing malathion and p-nitrophenyl acetate in germinating rice seeds.

This study was carried out to clarify the effect of sprout length, burried depth of seed and CaO$_2$ seed coating on emergence, seedling stand and early growth of rice in puddled-soil drill seeding at Junbuk in 1995. The cultivar tested was Dongjinbyeo(Japonica type). Although the longer sprouted seeds were caused the better seedling stand, 4mm sprout length of seeds appeared to be appropriate for good seedling stand and seeding uniformity, while 6mm sprouted seed resulted in ununiform seedling establishment. Rapid emergence and increased seedling stand were obtained by CaO$_2$ seed coating, which had effect more on soaked seed than on sprouted seed, but the seedling stand of sprouted seeds significantly reduced at deeper than 1cm seeding depth and also emergence rate of sprouted seeds significantly reduced at deeper than 2cm seeding depth.
Agrobacterium-mediated gene transfer has recently been developed to improve rice transformation. In this study, 3 different transformation methods were tested including soaking, co-cultivation, and vacuum infiltration. Agrobacterium tumefaciens GV3101 harboring the binary vector pGreen:: LeGSNOR was used in this experiment. This study aimed to identify the most appropriate method for transferring LeGSNOR into rice. Vacuum infiltration of the embryonic calli for 5 min in Ilpum resulted in high transformation efficiency based on confirmation by PCR, RT-PCR, and qRT-PCR analyses. In conclusion, we described the development of an efficient transformation protocol for the stable integration of foreign genes into rice; furthermore, the study results confirmed that PCR is suitable for efficient detection of the integrated gene. The vacuum infiltration system is a potentially useful tool for future studies focusing on transferring important genes into rice seed calli, and may help reduce time and effort.

Bacterial grain rot by Burkholderia gluae cause severe damage in seedling and grain of rice after heading season. This seed-borne pathogen play a role as first infection agent that could be cause disease following cropping season. Until now the direct isolation of the bacteria has some trouble by interference of other bacteria existed inside seed. This study established convenient identification method as simple isolation with KB medium from seed showing symptom and using PCR identification. By this isolation method, B. glumae was isolated from 40 to 50% in brown rice and inner hull, however, there were saprophytic bacteria and fungi outer hull. In PCR identification with Ogf4 and Ogr3 primer to these 25 isolates, the amplified products were presented in all of the collections but not in 10 saprophytic germs. The isolation rate was constant to 3 months stored seeds. This result provide a rapid and convenient isolation and identification of B. glumae.
Somatic embryos derived from cell suspension cultures of rice were singly alginate-encapsulated to be used as artificial seeds. When placed on half strength MS solid medium,73% of the encapsulated somatic embryos were capable of germination Encapsulation per se did not affect the germination frequency of embryos. When incubated by wrapping with moistured non-sterile filter paper, 60% of the encapsulated somatic embryos germinated. However encapsulated zygotic embryos without endosperm showed a high germination frequency regardless of the sterility of the incubation conditions. The results suggest that a greater susceptibility of somatic embryos to contaminants is attributed to lower germination frequency of encapsulated somatic embryos in non-sterile conditions.
This experiment was conducted on rice (cv. 2005 Thaoi) seeds to study whether priming with deep sea water (DSW) results in enhancement of seed emergence and seedling growth and to identify the optimum concentration of Deep Sea Water (DSW) for priming. Two experiments were conducted subsequently. In experiment 1, four concentrations of the DSW (10%, 20%, 30% and 40%), and in experiment 2, five concentrations of DSW (10%, 15%, 20%, 25% and 30%) were prepared and seeds were primed for 24 hours duration at $25^{\circ}C$. Beside this, hydro priming with plain water was also included as a control. Experiments were laid out in Completely Randomized Design (CRD) with three replications. Result showed that 20% DSW seed priming treatment had improved the emergence, seedling height, number of roots and root length as compare to other with DSW or without DSW treatments. Beyond 20% DSW priming (i.e. 25%, 30% and 40%) were not suitable for priming the seed. On the basis of seedlings growth parameters; emergence, seedling height, root number and length, and shoot root ratio, 20% DSW priming was the best priming treatment.
For the purpose of finding out the effect of heat treatment on biological response and mutation rate, rice seeds were heat treated before and after gamma irradiation. 1. At a dose of 20 KR, pre-irradiation heat treatment showed reduced biological damage and increased mutation rate as compared with non-heat treatment. 2. Mutation frequency was increased in post-treatment of heat shock than in pre-irradiation heat treatment and non-heat treatment. 3. Pre-irradiation heat treatment at 6$0^{\circ}C$ for 30 minutes markedly reduced the biological damage and increased the mutation rate. 4. Mutation spectrum in heat treatment was different from non-treatment.
Analytical method of carbofuran (2,3-dihydro-2, 2-dimethyl-7-benzofuranyl methyl carbamate) residues and its persistence in rice seeds, rice seedlings, rice plants and soils were studied by gas-liquid chromatographic analysis using electron capture detector. 1. The effective column material for clean-up is Florisil (5% $H_2O$)+Alumina (4% $H_2O$)+absorbent mixture with rinsing the first 300l of eluants to remove impurities in the column materials. 2. The method of applying an gelatin encapsulated carbofuran to the root zone of rice plant is the longest persistence in its residues. 3. By seed treatment, no carbofuran residues were detected in rice seeds and seedlings. 4. The amounts of carbofuran residues in rice seedlings is in proportion to the soaking time of rice seedlings in carbofuran solution rather than the concentration of the chemical. 5. Applying carbofuran by root zone has the higher and the loger residual effect than broadcast. 6. Persistence of carbofuran in the high clay content soil is longer than in the low clay content soil. 7. No carbofuran residue was detected in rough rice at havesting time.

Transposon-mediated insertional mutagenesis provides one of the most powerful tools for functional studies of genes in higher plants. This project has been performed to develop a large population of insertional mutations, and to construct databases of molecular information on Ds insertion sites in rice. Ultimate goals are to supply genetic materials and information to analyze gene function and to identify and utilize agronomically important genes for breeding purpose. Two strategies have been employed to generate the large scale of transposon population in a Japonica type rice, Dongjin Byeo; 1) genetic crosses between Ac and Ds lines and 2) plant regeneration from seeds carrying Ac and Ds. Our study showed that over 70% of regenerated plants generally carried independent Ds elements and high activity of transposition was detected only during regeneration period. Ds-flanking DNA amplified from leaf tissues of F2 and T1 (or T2) plants have been amplified via TAIL-PCR and directly sequenced. So far, over 65,000 Ds lines have been generated and over 9,500 Ds loci have been mapped on chromosomes by sequence analysis. Database of molecular information on Ds insertion sites has been constructed, and has been opened to the public and will be updated soon at http://www.niab.go.kr. Detailed functional analysis of more than 30 rice mutants has been performed. Several Ds-tagged rice genes that have been selected for functional analysis will be briefly introduced. We expect that a great deal of information and genetic resources of Ds lines would be obtained during the course of this project, which will be shared with domestic and international rice researchers. In addition to the Japonica rice, we have established the tagging system in an rice line of indica genetic background, MGRI079. MGRI079 (Indica/Japonica) was transformed with Agrobacteria carrying Ac and Ds T-DNA vectors. Among transgenic lines, we successfully identified single-copy Ds and Ac lines in MGR1079. These lines were served as ‘starter lines’ to mutagenize Indica genetic background. To achieve rapid, large scale generation of Ds transposant lines, MGR1079 transformants carrying homozygous Ac were crossed with ones with homozygous Ds, and $F_2$seeds were used for plant regeneration. In this year, over 2,000 regeneration plants were grown in the field. We are able to evaluate the tagging efficiency in the Indica genetic background in the fall.
To investigate the presence of brassinosteroids in rice bran and pㅐlished rice, they were extracted with MeOH. The extracts were purified through sequential procedure of solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 column chromatography and charcoal adsorption chromatography. The activity of brassinosteroids was monitored by the rice inclination test and its presence was confirmed in each purification step. The purified active components were further separated by TLC and HPLC. Brassinosteroids in active fractions of rice bran were identified as castasterone and teasterone by HPLC. We acknowledge that our work is probably the first report of brassinosteroids in mature seeds of rice The more amount of brassinosteroids was confirmed in rice bran than polished rice. The contents of castasterone and teasterone which were identified in rice bran were 0.15 ng/g and 0.37 ng/g, respectively.
