Comparative Nutrition of Traditional Korean Diet (전통 한국 식이의 비교 영양학)

  • Rice is the primary main dish of Traditional Korean diet. Although there have been changes in food consumption and nutrient intake among Koreans, traditional dietary pattern is stil dominant among Koreans. Traditional Korean diet has emphasized breakfast, which is the most frequently missed meals in Korea today but important for daily work performance and health. Compared to diets of the U.S. and Greece, Korean diet is high in carbohydrate and low in fat and cholesterol due to low intake of meat. Koreans also consume large amount of plant food, which makes fiber content of diet to be high. However fruit and milk consumption tends to be low in Korea. Koreans use fermented food, including kimchi, very frequently as well as foods cooked and consumed at high temperature and over direct fire. Traditional cooking methods are time consuming which limits the usage among modern city dwellers with working women. Despite the strengths of traditional Korean diets in reducing risk factors of chronic diseases, preservation of the tradition in modern Korean society requires special attention and efforts to make them more adaptable to contemporary life styles.

  • Antioxidative noudle products were developed with mixed material and antioxidative liquid extracted from natural products. Matrrials were mixed with wheat flour, rice flour, starch and green tea. 3% of green tea was suitable mixing ratio from the mastication data of texture meter, and proper water contents were 33% to prepare these noodles. The suitable mixing ratio of materials were wheat flour(85%), starch( 12%) and greentea(3%) from the physical data of texture meter. Since AUC value of antioxidative liquid was more higher than that of tap water, antioxidative noodle products were expected for good health and preservation of some diseases.

  • ;Bacterial blight caused by Xanthomonas oryzae pv. Olyzae is one of the most important diseases of rice. Host resistance, which relies on single, dominant resistance genes, is the only reliable method to control the disease at present. Pathogenic variation of the bacteria has been shown to follow the deployment of resistance genes in commercial cultivars. Information on the factors and the mechanisms for genetic variation of this pathogen is limited. Further, we have no clear evidence of whether population variability is due to sexual recombination or to variation introduced by mutations or intragenic recombination in a clonally maintained population.(omitted)itted)

Biological Control of Plant Diseases With Bacillus Species

  • Biocontrol is playing a more and more important role in plant disease management. Evidences show that there are optimum prospects for people to apply biocontrol approach to control plant disease or to study the mechanism of antagonism.“The study of Antagonistic Protein of Bacillus spp.to Xanthomonas oryzae pv. oryzae”has been worked in our laboratory since 1986. One hundred and thirty antagonistics bacteria were screened out, most of them belonged to Bacillus spp., and showed very strong inhibitive effect to various plant pathogens. Nine antagonistic proteins (peptides) were purified (P11-I, P11-II, B8, B826-I, B826-II, A30-I, A30-II, G35). Two antagonistic protein related DNA fragments (B826-I, A30-II) were cloned and sequenced. B826-I DNA fragment composed by 905 bp, and it contained two ORF encoding 95, and 54 amino acids, respectively. By using Rifr and Kamr as the selective markers, we found the bacteria could colonize on rice leaf for at least 40 days. In greenhouse the antagonistic bacteria showed certain degree of control efficiency.

Effect of Tricho-compost against Seedling Blight Disease of Wheat Caused by Sclerotium rolfsii

  • The efficacy of formulated Trichoderma harzianum-based Tricho-compost, seed treatment with Tricho-inocula, and chemical fungicide Provax 200 WP against foot and root rot diseases of wheat caused by Sclerotium rolfsii was tested in the pot house and in the research field of Plant Pathology Division, Bangladesh Agricultural Research Institute, Gazipur, Bangladesh. Tricho-compost was prepared with a mixed substrate of cow dung, rice bran, and poultry refuse colonized by T. harzianum. Seedling mortality of wheat was significantly reduced by the Tricho-compost, Tricho-inocula, and Provax 200 WP both in the pot house as well as in the field experiments. The yield of wheat was sharply increased over the control due to the T. harzianum formulations and Provax 200 WP. Among the treatments, soil application of Tricho-compost was more efficient in reducing seedling mortality and accelerating plant growth with an increased yield of wheat with S. rolfsii-inoculated pot cultures and field experiments.

  • Isotianil is a novel fungicide which induces systemic acquired resistance in plants. It has excellent preventive effects as low dosages against rice blast which is one of the most serious diseases in rice. The Maximum Residue Limit (MRL) of Isotianil in republic of korea was set to 0.1 mg/kg in rice, so it is necessary to determine levels of Isotianil residues in agricultural commodities for controlling food safety. Therefore, The purpose of this study was to develop analytical method for the determination of isotiical residues in agricultural commodities using GC-NPD/MSD. Isotianil was extracted with acetonitrile from apple, chinese cabbage, hulled rice, mandarin, pepper, and soybean. The extract was diluted with saline water, and then dichloromethane partition was followed to recover this fungcide from the aqueous phase. A solid phase extraction with Florisil cartridge was additionally employed for final clean up. Isotianil was analyzed and quantitated by GC-NPD and confirmed by GC-MSD. Average recovery of Isotianil ranged from 70.0 to 103.9% in six representative agricultural commodities with relative standard deviations less than 10%, and limit of quantification (LOQ) was 0.05 mg/kg.

A Survey of Viral Diseases of Proso Millet (Panicum miliaceum L.) and Sorghum (Sorghum bicolor L.) in South Korea (국내 기장 및 수수의 바이러스병 발생 조사)

  • Throughout year 2015 to 2016, 101 proso millet and 200 sorghum samples were collected from five provinces in South Korea. The samples were subjected to paired-end RNA sequencing and further analyzed by RT-PCR. The results indicated that Rice black-streaked dwarf virus (RBSDV) was detected from sorghum collected in Gyeongsang province. The other four viruses, including RBSDV, Rice stripe virus (RSV), Barley virus G (BVG), and Cereal yellow dwarf virus (CYDV), were detected from proso millet. Among four viruses, both RSV and RBSDV were identified high frequency from proso millet collected from Gyeongsang province. Otherwise, BVG was nearly equally identified from five provinces, suggesting that the virus was supposedly widespread nationwide. RBSDV was first identified from both proso millet and sorghum in South Korea. The other virus annotated CYDV identified proso millet was shown to have relatively low identities compared to CYDV previously reported, suggesting that the virus might be new member of Polerovirus.

Supplementary Effects of Black Rice (Oryza sativa L.) Aleurone Layer Extract on Body Fat, Serum Lipid, and Serum Hormone Levels in Ovariectomized Rats (흑미 호분층 추출물의 급여가 난소절제 흰쥐의 체내 지방, 혈중 지질 및 관련 호르몬 수준에 미치는 영향)

  • This study was conducted to evaluate suppressive effects of black rice aleurone layer extract (BRE) on body fat, serum lipid, and hormone levels in ovariectomized rats. The rats were divided into five groups (n=8/group): sham operated group (S); ovariectomized control group (OVX); ovariectomized group treated with isoflavone 10 mg/kg B.W. (IF); ovariectomized group treated with BRE 30 or 90mg/kg B.W. (BRE-30 or -90). High fat diet (45% calories from fat) was fed to all rats for 12 weeks. Body weight gain, body fat weight, and adiposity index increased in the OVX group, but they significantly decreased in IF or BRE supplementation. Serum triacylglycerol and leptin levels decreased in BRE groups while serum adiponectin level significantly increased compared to that of the OVX group. Hepatic total lipid, triglyceride, and cholesterol levels increased in the OVX group, but they decreased in BRE supplementation (p>0.05). These data suggest that black rice aleurone layer may be a useful food source to decrease obesity and its related diseases by modulating lipid metabolism in estrogen-deficiency model.

Effect of Osmotic Pressure on hCTLA-lg Production in Transgenic Rice Cell Suspension Cultures (형질전환된 벼세포 배양에 있어서 삼투압 조절에 따른 hCTLA4-lg 생산성 변화)

  • An immunosuppressive agent, human cytotoxic T lymphocyte antigen 4 (hCTLA4), is used for the prevention of graft rejection and treatment of autoimmune diseases. hCTLA4-Ig, a CTLA4-immunoglobulin fusion protein, was produced and secreted from transgenic rice cell suspension cultures using rice a-amylase (RAmy3D) expression system. In this system, hCTLA4-Ig expression was regulated metabolically by sugar starvation. For the purpose of improving hCTLA4-Ig production, the effects of osmotic pressure was investigated in suspension cultures of transgenic rice cells. The highest production level was achieved at 40 mM sorbitol $(140\;mOsm{\cdot}kg^{-1}\;H_2O)$. Using the medium with 8 mM glucose, the level of hCTLA4-Ig in the medium reached 45.3 mg/L. By adjusting the osmotic pressure of induction medium, it was found that the hCTLA4-Ig production could be increased up to 2.1-fold compared with that in batch culture.

Survey of Major Disease Incidence of Rice in Korea during 1999-2008 (1999-2008년도 벼 주요 병해 발생 개황)

  • During the period from 1999 to 2008, we surveyed the major diseases incidence of rice in Korea. Leaf blast showed 0.85% of the diseased leaf area in 2000 and then consistently decreased to 0.02% by 2008. However, panicle blast irregularly appeared by the years depending on weather conditions for disease development from heading to milk stage of rice. Diseased lesion height of sheath blight had the lowest (10.1%) incidence in 2001 and the highest (21.4%) incidence in 2007. A negative corelation ($r=-0.88^*$) was measured between diseased lesion height of sheat blight and sunshine hours from mid- to late August. In Chunbuk, Gyeongbuk, and Gyeongnam provinces, brown spot occurrence declined since 2001, however, in Chungnam, Jeonbuk, and Jeonnam provinces, it increased since 2005. False smut drastically decreased after showing the highest incidence of 7.59% in panicles in 2000. The correlation coefficient between diseased panicles of false smut and rainfall during the late August was calculated as $0.94^{**}$. Bacterial leaf blight severely increased in the chronically infested fields in Jeonnam, Jeonbuk, Gyeongnam, and Chungnam province since 2003.
