Soil salinity shifts the community structure and diversity of seed bacterial endophytes of salt-sensitive and tolerant rice cultivars

  • Soil salinity due to accumulation of salts particularly sodium chloride affects agricultural lands and their vegetation. Generally, rice is a moderately sensitive plant with some cultivars with varying tolerance to salinity. Though there are physiological differences between salt-sensitive and salt-tolerant rice cultivars, both are still affected especially during high salinity and prolonged exposure. This also ultimately affects their indigenous bacterial endophytes particularly those that inhabit the rice seed endosphere. This study investigates the dynamic structure of seed bacterial endophytes of salt-sensitive and tolerant rice cultivars grown in different levels of soil salinity. Endophytic bacterial diversity was studied Terminal-Restriction Fragment Length Polymorphism (T-RFLP) analysis. Results revealed a very interesting pattern of diversity and shifts in community structure of bacterial endophytes in the rice seeds. There is a general decrease in diversity for the salt-sensitive rice cultivar, IR29 as soil salinity increases. For the salt-tolerant cultivars, IC32 and IC37, diversity interestingly increased at moderate salinity then decreased at high soil salinity. The patterns of community structure is also strikingly different for the salt-sensitive and salt-tolerant rice cultivars. IR29 has a more even distribution of abundance, but under soil salinity, the community shifted where Curtobacterium, Pantoea, Flavobacterium and Microbacterium become the more dominant bacterial communities. For IC32 and IC37, the dominant bacterial groups under normal stress conditions were also the dominant bacterial groups during salt stress conditions. Their seed bacterial community is dominated by endophytes belonging to Microbacterium, Flavobacterium, Pantoea, Kosakonia and Enterobacter. Stenotrophomonas and Xanthomonas have not changed in terms of abundance under different salinity stress level in the salt-sensitive and salt-tolerant rice cultivars. This study showed that soil salinity greatly influenced the seed bacterial communities of rice seeds irrespective of their physiological tolerance to salinity.

Varietal Differences in Quality Characteristics of Yukwa(Fried Rice Cookie) made from Fourteen Glutinous Rice Cultivars (14품종 찹쌀의 유과 가공성 비교)

  • Varietal difference in physicochemical characteristics of glutinous rice grain and interrelationships between these properties and the quality characteristics of Yukwa(fried rice cookie) were investigated on fourteen rice varieties, to obtained the basic informations for diversifying the utility of glutinous rice grain and for developing various glutinous rice cultivars adaptable to glutinous rice food processing. Among physicochemical properties of glutinous rice grain, the content of released reducing sugar during soaking treatment was the most positive correlation between the adaptability to Yukwa processing quality. CB243 and Sandong 71 were the most adaptable glutinous rice cultivars to make the Yukwa, because of its tested score in expansion volume, crispiness and sensory preference was higher than other glutinous rice cultivars.

Impact of Heat Stress on Pollen Fertility Rate at the Flowering Stage in Korean Rice (Oryza sativa L.) Cultivars

  • Rice is very sensitive to high temperatures during the reproductive stage, particularly during the flowering and anthesis periods. To determine how high temperatures result in sterility during the flowering period in Korean rice cultivars, groups of 11 cultivars were subjected to different temperature regimes (24, 28, 30, and 33℃) during the flowering stage using sunlit phytotrons. At an average of 33℃, all 11 rice cultivars reached anthesis earlier than at the other temperatures. Microscopy analyses revealed significant differences in pollen germination and pollen viability in cultivars grown at 33℃ compared to those cultured at lower temperatures. At 33℃, the cultivars had significantly lower fertility rates (47% reduction) than cultivars grown at 24℃. These findings are important as rice pollination and fertility depend on the pollen viability and germination. The present study shows that rice fertility is negatively affected by excessively high temperatures.

Effects of Temperature and Day-Length on Heading Habit of Recently Developed Korean Rice Cultivars

  • To understand the heading habit of recently developed 20 Korean rice cultivars, rice plants in a phytotron were exposed to different temperature: $22.5^{\circ}C(day\;27^{\circ}C/night\;18^{\circ}C),\;27.5^{\circ}C(day\;32^{\circ}C/night\;23^{\circ}C)$, and day-length conditions: 10, 12, 13, 14, 15 hours. Four rice cultivars (Sobibyeo, Juanbyeo, Ilpumbyeo and Shindongjinbyeo) showed relatively short Basic Vegetative Phase (BVP) of 17 to 18 days, while Dasanbyeo showed the longest (35 days) BVP, compared to other remaining 15 tested cultivars which exhibited 24 to 31 days of BVP. In this experiment, it was tried out to separate the eliminable vegetative phase into photosensitivity and thermo-sensitivity with two different pathways. Many tested cultivars, however, exhibited quite different responses under low temperature and / or long day-length conditions. Especially, Surabyeo and Juanbyeo were the most difficult cultivars to separate into photo- or thermo-sensitivity in that the eliminable vegetative phase of these two cultivars increased greatly only under low temperature $(22.5^{\circ}C)$ and long day-length (15 hr.) conditions. Regarding the heading response to temperatures, tested cultivars could be categorized into 2 groups. In 1st group of rice cultivars, the eliminable vegetative phase decreased almost equally as the temperature changed from $20.0^{\circ}C\rightarrow22.5^{\circ}C\rightarrow25.0^{\circ}C\rightarrow27.5^{\circ}C$. In contrast, the 2nd group of rice cultivars exhibited eliminable vegetative phase slowly decreasing when the temperature changed from $22.5^{\circ}C\rightarrow25.0^{\circ}C\rightarrow27.5^{\circ}C$, but rapidly decreasing when the mean temperature changed from $20.0^{\circ}C\;to\;22.5^{\circ}C$. All the cultivars belonged to 2nd group, the heading date would be very delayed if cool summer comes.

Pedigree Analysis of 17 High Quality Korean Rice Cultivars Using Web Database Systems

  • It is important to understand pedigree of rice cultivars commonly used for breeding. In this paper, pedigree tables for tracking the pedigree of 17 representative rice cultivars recommended by Rural development Adminstration (RDA) were completed and analyzed using two kinds of web database system; 'IRIS' and 'RRDB'. Seven cultivars, namely, 'Sangmibyeo', 'Ilpumbyeo', 'Saegewhabyeo', 'Surabyeo', 'Shindongjinbyeo', 'Ilmibyeo' and 'Jungwhabyeo' had 'Koshihikari' on the pedigree of their ancestor. Besides 'Koshihikari', the most feguently used ancestral germplasms among the high quality rice cultivars were 'Fujisaka 5', 'Kameno o' and 'Asahi', 'Fujisaka 5' was used as ancestral parent in 12 out of 17 cultivars. Interestingly, 'Kameno o' was used in pedigree of 16 out of 17 high quality varieties and 'Asahi' was used in the ancestral pedigree of all 17 varieties. 'Hwayeongbyeo' was used as one of parent in the breeding of 'Dongjin 1', 'Hwabongbyeo', 'Saegewhabyeo' and 'Junambyeo'. 'Ilpumbyeo' was used in the breeding pathway of 'Junambyeo' and 'Saegewhabyeo', 'Mangeumbyeo' itself was not enlisted as one of high quality rice cultivars, but was used as a breeding parent of three high quality varieties, namely, 'Saegewhabyeo', 'Hwabongbyeo' and 'Nampyeongbyeo'. Incorporated with evaluation data, pedigree will provide a valuable chance to genealogical tracking of agronomic traits such as disease resistance, grain quality and etc.

Diurnal Variation in Endogenous Gibberellin Levels of Rice Shoots

  • Diurnal changes in levels of endogenous gibberellins(GAs) were investigated in three rice cultivars i.e. Sangjubyeo, Shingeumobyeo(photo-neutral) and Chucheongbyeo(photosensitive). The rice cultivars were grown under a 12-hr photoperiod and endogenous GA levels were assayed by gas chromatography-mass spectrometry(GC-MS-SIM) every 3 h for 24 h. The endogenous bioactive $GA_1$ and its immediate precursor $GA_{20}$ contents were significantly different in both photosensitive and photo-neutral rice cultivars, though less pronounced differences were observed for endogenous $GA_{12},\;GA_{53},\;GA_{19}$, and $GA_8$ levels with in the three rice cultivars. The levels of bioactive $GA_1$ and its immediate precursor $GA_{20}$ were significantly higher in Chucheongbyeo than in the other two cultivars. In Chucheongbyeo, the $GA_1$ contents increased significantly from 11.00 to 17.00 o'clock, thus indicating a correlation with light. In Shingeumobyeo, $GA_1$ contents slightly increased during morning hours, while a similar hike in $GA_1$ contents was observed for Sangjubyeo during evening hours. $GA_{19}$ was found to be the most abundant GA form in the three rice cultivars. Our results suggested that GA production in rice depends upon the response potential of rice cultivars and that light positively correlated to GA production in photosensitive rice cultivar.

Study on rice double cropping in Southern Korea paddy field

  • Recently, the rice growing time was extended by the global warming. This study was conducted to investigate the possibility of rice double cropping system in southern Korea. The first transplanting was with six cultivars ('Kilala397', 'Baekilmi', 'Joun', 'Hanseol' and 'Jungmo1031') on April 15. All cultivars could be harvested at the end of July. Adaptable cultivars for the first cultivation were 'Kilala397', Baekilmi' and 'Joun'. The rice yields at the first cultivation was about 95% of local average yield. Although the yield was slightly less, the first cultivation was considered to have economic benefits, because of the high market price of rice. In the second transplanting was with five cultivars ('Manjong', 'Joun', 'Deabo', 'Jinok' and 'Kilala397') on April 15. All cultivars could be harvested in early November. Adaptable cultivars for the second cultivation were 'Jinok' and 'Kilala397'. The rice yields at the second cultivation was about below the 60% of local average yield, because there was the less growth than normal season cultivation. Consequently, southern Korea' annual rice double cropping system is considered to have no economic value yet. However, the research should be continued considering the temperature rise of global warming.

Incidence of Rice Blast on New Rice Cultivars Released in 1998 and Some Elite Line Observed at Different Locations and in Different Years in Korea (1998년도 벼 신품종 및 유망계통에 대한 연도 및 지역별 도열병 발생정도)

  • Incidence of rice blast on new rice cultivars and elite lines was observed from 1995 to 1997 in Icheon, Chuncheon, Jecheon and Naju areas. The observation was made in the nurseries and fields. In the nurseries, three cultivars Grubyeo, Sangjuchalbyeo and Suwon 414 showed moderate levels of resistance to leaf blast, with the disease index 0 to 5. From the field observation, it was found that cultivars Heugjinjubyeo and Unbong 18 were highly resistant to leaf blast, but susceptible to neck blast. In general, there was a great variation yearly and regionally in the incidence of neck blast within the same cultivar, ranging from 0 to 100%. However, the range of neck blast infection was relatively narrower in the cultivars Yunghaebyeo (0∼2.4%) and Suwon 414 (0∼2.2%) during observation period. Races of rice blast fungus were variable at different areas. Among them dominant races were KI-409, KJ-201 and KJ-301.

Indicative Responses of Rice Plant to Atmospheric Ozone

  • Differences in physiological and biochemical responses between sensitive and tolerant rice cultivars to ozone were investigated to develop reliable indications of early ozone damage. Three Korean local rice cultivars -sen-sitive cultivar Dongjin (DJ), moderately tolerant cultivar Hwayeong (HY) and tolerant cultivar Ilmee (IM) were exposed to ozone at the concentrations of 100 nl $\textrm{l}^{-1}$ or 200 nl $\textrm{l}^{-1}$ , 8 h per day for 10 days in a controlled-environment fumigation chamber. The rice cultivars seemed to be endurable to ozone stress at the concentration of 100 nl $\textrm{l}^{-1}$ which is frequently monitored during the growing season in summer. However, severe damage was induced and differential sensitivity was clearly noted among the rice cultivars at the higher ozone concentration. Activation of the glutathion (GR) -ascorbate peroxidase (APX) cycle was likely to be responsible for protection of rice plants against ozone exposure, relating difference in sensitivity of rice cultivars to ozone. Photosynthetic activity appeared to be one of sensitive responses, for which chlorophyll fluorescence and leaf greenness can together provide a very reliable index, a degree of photosynthetic damages by ozone. Formation of malondialdehyde (MDA) was also considered as an indication that can differentiate cultivars sensitivity to ozone. However, the changes in polyamines and total phenolics were not consistent with exposed ozone concentrations and/or ozone sensitivity of the cultivars. The behavior of polyamines and phenolics in the damaged plants at high ozone levels could be interpreted as an indication of ozone injury rather than activation of additional protection mechanisms scavenging active oxygen species formed by ozone. Several responses triggered by ozone could explain the differential sensitivity of the rice cultivars and be used as reliable indications of relative ozone damage to rice plant.

EFFECTS OF UV-B RADIATION ON GROWTH AND DEVELOPMENT OF RICE CULTIVARS (ORYZA SATIVA L.).

  • Elevated near-UV radiation, containing a large amount of UV-B and a small amount of UV-C, inhibited the development of leaves and tillers, the increase in biomass production, the elongation of plant height, the photosynthetic rate and the chlorophyll content in rice plants in a phytotron. Elevated UV-B radiation filtered through cellulose diacetate film or UV-31 cut filter (transmission down to 290 nm) similarly suppressed each growth component above. Near-UV radiation-caused injuries were alleviated either by elevated CO$_2$ atmosphere or by exposure to high irradiance-visible radiation. On the basis of these findings, we examined cultivar differences in the resistance to UV radiation-caused injuries among 198 rice cultivars belonging to 5 Asian rice ecotypes ( aus, aman, boro, bulu and tjeleh) from the Bengal region and Indonesia and to Japanese lowland and upland rice groups. It was shown that .various cultivars having different sensitivities to the effects of near-UV radiation were involved in the same ecotype and the same group, and that the Japanese lowland rice group and the boro ecotype were more resistant. Among Japanese lowland rice cultivars, Sasanishiki (one of the leading varieties in Japan) exhibited more resistance to near-UV radiation, while Norin 1 showed less resistance, although these two cultivars are closely related. It was thus indicated that the resistance to the inhibitory effects of near-UV radiation of rice cultivars is not simply due to the difference in the geographical situation where rice cultivars are cultivated. From the genetic analysis of resistance to the inhibitory effects of UV radiation on growth of rice using F$_2$ plants generated by reciprocally crossing Sasanishiki and Norin 1 and F$_3$ lines generated by self-fertilizing F$_2$ plants, it was evident that the resistance to the inhibitory effects of elevated near-UV radiation in these rice plants was controlled by recessive polygenes.
