Comparisons of inorganic amounts in paddy fields, rice straw and seed with varying severity of brown spot caused by Cochliobolus miyabeanus

  • In order to elucidate influence of nutritional status on rice brown spot caused by Cochliobolus miyabeanus, rice cultivation soils and rice straws were collected from paddy fields where ice brown spot occurred severely, moderately, a little and none respectively. Rice plant materials were analyzed to measure inorganic nutrients in rice straws and rice seeds. Analysis of chemical properties of rice paddy soil showed that EC and contents of available phosphate, cation and silicic acid in soil with severe infections were lower than those in healthy soil. This result suggests that amount and holding capacity of nutrient contents in soils collected from paddy field with infection of C. miyabeanus are relatively low compared to those in soils collected from healthy paddy field. Analysis of inorganic nutrients in rice straws showed that amount of macronutrient elements such as silicic acids, available phosphate and total nitrogen, and micronutrients such as copper, iron and zinc in rice straws from paddy field with infection were lower than those in healthy soil. Especially amount of iron and silicic acid were very low in rice straws from paddy field soils with infection Amount of inorganic nutrients such as iron and zinc in rice seeds was the same trend as those of rice straws. These results showed that one of major factors affecting rice brown spot was amount of nutrient contents in soil and rice straw.

  • In order to study a relationship between soil nutrients and rice brown spot occurrence, paddy field soils, rice grains and straws collected from different paddy fields with different disease degrees of brown spots were analyzed for inorganic nutrients. Brown spot was prevalent in the rice grown in nutrient-deficient soils, which is especially low in macronutrient elements (phosphoric acid, potassium, silicic acids) and micronurients (calcium, magnesium). The soil, however, was high in sodium while organic nutrients and pH level were similar to others. The rice straws with severe brown spot were low in inorganics such as ferrous, copper, T-N, and $P_{2}O_{5}$ while the rice grains with brown spot were low in ferrous, MgO, Zn, and Mn. In the analysis of field type and nitrogen level, the highest disease severity was found in sandy-type field soil, followed by salty-type field soil and disease severity decreased as application level of nitrogen fertilizer increased. As a summary, the most important factor for effective brown spot control in rice is maintenance of proper nutrients in sandy-type field and control of sodium level in salty-type field soil.

Discrimination Method of Brown Planthopper, Nilaparvata lugens(Stal) Nymphs by the Fluorescent Spots between Compound Eyes in Rice Paddies (벼논에서 벼멸구(Nilaparvata lugens Stal)약충 두부의 형광성 반점에 의한 식별법)

  • Several species of planthoppers such as brown planthopper, N. lugens (Stal) (BPH), smaller brown planthopper, Laodelphax striatellus (Fall n) (SBPH), and white-backed planthopper, Sogatella furcifera (Horv th) (WBPH) are distributed in Korean rice paddies but not easy to discriminate them in situ. Accurate discrimination of them is an indispensable process in the forecasting for their outbreak and control. Especially, innovative discrimination method for BPH was required because BPH was one of the most important insect pest of rice. Nymphs and adults of BPHs, SBPHS, and WBPHS, thus, were examined their morphological characteristics in the paddies and laboratory. The nymphs of BPH had different characters from those of SBPH and WBPH. The nymphs of BPH had white fluorescent spot between bottom of compound eye and antenna, while there was no that spot the other two species. The white spot was the brightest at the nymphs just after hatching and getting weaker as the nymph developed. At last the white spot was totally disappeared at the adult stage. This white spot was innovative criterium to discriminate nymphs of BPH, SBPH, and WBPH in rice paddies.

Overwintering of the Pathogen and Factors Affecting Disease Develpoment of Rice Brown Spot Caused by Cochliobolus miyabeanus (벼깨씨무늬병균의 월동과 발병조건에 관한 연구)

  • This study was carried out to investigate factors affecting disease development of rice brown spot caused by Cochliobolus miyabeanus in the field and under the greenhouse conditions. In normal paddy fields, the pathogen was more frequently isolated from dried rice straws than wet straws after overwintering. The pathogen was also readily isolated from wet rice straws of salty paddy fields although infection rate of the fields was low. The disease symptom was first observed at 2 to 3days after inoculation when the seedlings were kept for 6 hours in dew chamber. The number of lesions peaked at 14 hr incubation in cultivars Chucheongbyeo and 24 hr incubation in cultivar Daesanbyeo and Tetep. Sixty three-d-old seedlings were more susceptible than 21 to 49-d-old seedlings regardless of cultivar. There was some difference in number of lesions observed, but the lesions were evenly spread regardless of leaf position. Light on the rice seedlings at 3,000 lux or 6,000 lux showed more severe symptoms than those in the dark.

Morphological Characters and Seed Transmission of Bipolaris panici-miliacei Causing Leaf Spot of Common Millet (기장 점무늬병균 (Bipolaris panici-miliacei)의 형태적 특징과 종자전염)

  • Bipolaris panici-miliacei, Cercospora fusimaculans, Fusarium moniliforme and Rhizoctonia solani were pathogenic fungi detected from 5 seed samples of common millet (Panicum miliaceum). Morphological characters of B. panici-miliacei were as follows. Conidiophores were dark olivaceous brown, simple, cylindrical, geniculate, and septate. Conidia were fusoid, dark olivaceous brown, tapering gradually toward the ends, straight to slightly curved, 3~13 distoseptate, and 29.4~155.4$\times$10~26 ${\mu}{\textrm}{m}$ in size with dark hilum included within the contour of the basal cell. Seed infection with B. panici-miliacei caused seed rotting, coleoptile spot, and seedling blight of common millet plants. According to the inoculation experiments, B. panici-miliacei showed strong virulence on the young seedlings of common millet, but very weak virulence on the young seedlings of rice (Oryza sativa) and foxtail millet (Setaria italica).

Screening of Antagonistic Bacteria Having Antifungal Activity against Brown Spot and Sheath Rot of Rice (벼 깨씨무늬병 및 잎집썩음병에 항진균 활성을 갖는 길항 미생물의 탐색)

  • Brown spot and sheath rot of rice are caused by fungal pathogens such as Curvularia lunata, Cochliobolus miyabeanus, and Sarocladium oryzae, and cause losses such as reduced rice yield and quality, which is an enormous problem with serious long-term effects. To search biological control agents of phytopathogenic fungi, five kinds of useful Bacillus-like isolates which are excellent in extracellular enzyme activity and produce siderophore were selected from paddy soil of Sunchang in Korea. The selected isolates were tested for excellent antifungal activity against three of the phytopathogenic fungi that frequently occur in rice, and JSRB 177 strain had the most excellent antifungal activity. Based on the experimental results, JSRB 177 is finally selected as a candidate for biological control and identified to Bacillus subtilis through 16S rRNA sequence analysis. In addition, physiological characteristics of JSRB 177 confirmed by analysis of carbohydrate fermentation patterns and enzyme production ability. Based on the above results, JSRB 177 is expected to be used as a biological control agent for the rice pathogenic fungi. In the future, further studies related to industrialization such as port test and establishment of mass production process are needed.

Constituents of Antimutagenic Factor from Brown Rice (현미의 항돌연변이 활성물질의 구성성분)

  • To investigate the constituents of antimutagenic factor from brown rice, methanol extracts were fractionated into ether, ethylacetate, buthanol and aqueous fractions. The ether fractions showed distinct antimutagenic effect and active spot were selected by silica gel chromatography. The specific activity of active spot decreased with isolation of the active components from the methanol extract. Qualitative analyses of the active spot by using various spraying reagents revealed that ninhydrin and orcinol did not develop colored reactions. But, ferric chloride, 2,7-dichlorofluorescein, antimony pentachloride, phosphomolybdic acid, bromothymol blue and rhodamine 6G led to colored reactions. These results suggested that the consitituents of active material were neither polar nor nitrogen-containing compounds and that they may contain phenolic compounds and fatty acid derivatives. Main compounds of the active spot were analyzed to be o-hydroxy benzyl alcohol(saligenin), octanoic acid(caprylic acid), 9,12-cis-octadecadienoic acid(linoleic acid), 11-cis-octadecenoic acid(oleic acid), hexadecanoic acid(palmitic acid), 1H-indole-2-carboxylic acid and 1,2-benzenedicarboxylic acid(phthalate) in GC/Mass spectrum, and antimutagenicity of these active compounds using standard regeant was reconfirmed in S. typhimurium reversion assay.

Bacillus vallismortis EXTN-1-Mediated Growth Promotion and Disease Suppression in Rice

  • Bacillus vallismortis EXTN-1, a biocontrol agent in cucumber, tomato and potato was tested in rice pathosystem against rice fungal pathogens viz. Magnaporthe grisea, Rhizoctonia solani and Cochliobolus miyabeanus. Apart from increasing the yield in the bacterized plants (11.6-12.6% over control), the study showed that EXTN1 is effective in bringing about disease suppression against all the tested fungal pathogens. EXTN-l treatment resulted in 52.11% reduction in rice blast, 83.02% reduction in sheath blight and 11.54% decrease in brown spot symptoms. As the strain is proven as an inducer for systemic resistance based on PR gene expression in Arabidopsis and tobacco models, it is supposed that a similar mechanism works in rice, bringing about disease suppression. The strain could be used as a potent biocontrol and growth-promoting agent in rice cropping system.

Seedling Establishment and Yield of Direct Subsurface Seeded Rice as Influenced by $CaO_2$ Coating and Seeding Rates (과산화석회 분의와 파종량이 침수 토중직파재배 벼의 입모와 수량에 미치는 영향)

  • To establish the direct seeding cultivation method in rice (Oryza sativa L.). direct subsurface seeding with different oxygen suppliers and seeding rates was conducted from May to Oct. 1989. at the experimental field of Suncheon National University. In direct subsurface seeding. calcium peroxide enhanced the ratio of seedling establishment. However. as number of plants per unit area increased. the occurrence of field lodging became progressively severe. so that the enhanced seedling establishment did not increase the yield. The optimal seeding rate and the number of sowing seeds per equally spaced spot were estimated to be 3.3Kg seeds/l0a and 3 seeds per spot with CaO$_2$-coated seeds. and 5.6Kg/10a and 5 seeds per spot with uncoated seeds. respectively, According to the results from the path coefficient analysis. percent of filled spikelets was identified to be the most influential variables on the variability in brown rice yield. so that improving percent of filled spikelets by inhibition of field lodging was the most important matter to increase the yield.
