Isolation and Enzyme Production of a Xylanase-producing Strain, Bacillus sp. AMX-4. (Xylanase를 생산하는 Bacillus sp. AMX-4 균주의 분리와 효소 생산성)

  • A bacterium producing the extracellular xylanase was isolated from soil and has been identified as a Bacillus sp. strain. The isolate, named Bacillus sp. AMX-4, was shown to be similar to B. subtilis strain on the basis of its chemical compositions. The xylanase of culture supernatant was most active at 50℃ and pH 6.0. The additional carbon sources including monosaccharides, disaccharides, wheat bran, and rice straw increased the enzyme productivity. Especially, the maximum xylanase productivity was reached 29.2 units/ml in LB medium supplemented with 1.5% (w/v) xylose, which was 16-folds more than that in LB medium. As the results of investigating the effects of xylose on cell growth and xylanase productivity of Bacillus sp. AMX-4, increase of xylanase production was owing to the induction of xylanase biosynthesis. It was also found that the enzyme production was in association with the growth of Bacillus sp. AMX-4.

Optimization of Laccase Production from Bacillus sp. PK4 through Statistical Design of Experiments

  • Statistical design of experiments was employed to optimize the media composition for the production of laccase from Bacillus sp. PK4. In order to find the key ingredients for the best yield of enzyme production from the selected eleven variables viz yeast extract, glucose, zinc sulphate, copper sulphate, potassium chloride, magnesium sulphate, calcium chloride, ferrous sulphate, sodium chloride, potassium dihydrogen phosphate ($KH_2PO_4$) and dipotassium hydrogen phosphate ($K_2HPO_4$), Plackett-Burman design was applied. The $MgSO_4$, $FeSO_4$, and $CuSO_4$ showed positive estimate, and their concentration optimized further. The steepest ascent method and Box-Behnken method revealed that 1.5 mM $MgSO_4$, 0.33 g/l $FeSO_4$ and 1.41 mM $CuSO_4$ were optimal for the laccase production by Bacillus sp. PK4. This optimization strategy leads to enhancement of laccase production from 2.13 U/ml to 40.79 U/ml. Agro-wastes residues replace the carbon source glucose in the optimized media namely sugarcane bagasse, wheat bran, rice husk, and groundnut shell, among these groundnut shells (117 U/ml) was found to enhance the laccase production significantly. The laccase produced by Bacillus sp. PK4 was found to have the potential to degrade persistent organic pollutant benzo[a]pyrene.

  • Pholiota sp. were collected from the forest of Kangwon province (Mt. Samak, Mt. Odae and Kangwon National University Forest Experimentation) from the early of August to the end of October 1997. Among these fungi, Poliota adiposa was selected for culture experiment because it is suitable for edible mushroom. The Optimal temperature for the mycelial growth of Pholiota adiposa was in the range of $28^{\circ}C$ while that of fruit body formation was $15{\pm}1^{\circ}C$. Brown sugar as carbon source and soybean flour as nitrogen source were good for mycelial growth in commercial liquid culture. The fruit bodies of Pholiota adiposa were artifitially produced on plastic bottle including poplar sawdust and rice bran (4:1).

Optimization of Culture Media for Solid-state Culture of Pleurotus ferulae

  • In order to elucidate the possibility of artificial production of p. ferulae by solid-state culture, the optimization of culture conditions was carried out. When $NH_4H_2PO_4$ and $CaCO_3$ were used in the cultures using test tube with 30 g of Populus sawdust at $25^{\circ}C{\pm}1$ in the dark, the favored mycelial growth was observed with $1\%$ of $NH_4H_2PO_4$ and the production of polysaccharide was 7.85 mg/100 mg of mycelium with $1\%$ of $CaCO_3$. The mixtures of $80\%$ of Populus Sawdust and $20\%$ of rice bran at $60\%$ of water content were determined to be optimal for the production of fruiting bodies in the sawdust culture. When three treatments containing various ratios of garlic powder were conducted, yields of fruiting bodies were drasti[ally higher than those of Synthetic mixture without garlic powder The highest yield (143 g/bag) was obtained with $7\%$ garlic powder. The yield of synthetic mixture containing $7\%$ of garlic powder was $83\%$ higher than that of Sawdust culture. The reason why garlic powder did support growth was not clear but it is possible that garlic powder might contain effective components for the formation of fruiting body. The optimal synthetic mixture composition consisted of cotton seed $77\%$, lime $6.4\%,\;K_2HPO_4\;0.2\%,\;KH_2PO_4\;0.2\%,\;CaHPO_4\;0.2\%$, corn flour $4\%$, wheat flour $5\%$, and garlic pow-der $7\%$.

Thermal Oxidative Stability of Various Vegetable Oils used for the Preparation of the Seasoned Laver Pyropia spp.

  • Seasoned laver Pyropia spp. is a traditional Korean seafood that has gained popularity worldwide because of its unique taste, texture, and health benefits. It is prepared by roasting a sheet of dried laver, to which vegetable oils have been applied, at an ultra-high temperature (UHT) of $300^{\circ}C$. Therefore, the oxidative stability of the oils is the most important factor in determining the shelf life of seasoned laver products. In this study, we investigated changes in the thermal oxidative stability of six major vegetable oils (sesame, perilla, sunflower, rice bran, canola, and olive) during the seasoned laver processing. The oxidation induction time of each oil from the seasoned laver products was decreased compared with the fresh oil. These results indicate that the UHT treatment ($300^{\circ}C$, 10 s) induced thermal oxidation of the oils. Among the six seasoned laver oils, the induction times of olive (OL, 8.02 h) and sesame (SE, 5.31 h) oils were significantly higher than the other oils. The acid values (AVs) of OL and SE oils from the seasoned laver were 0.49 and 0.79, respectively. On the other hand, perilla oil had the overall worst thermal oxidative properties (induction time: 0.35 h, AV: 2.82). Our results provide useful information about seasoned laver products for researchers or manufactures.

Effect of microbial product on microorganisms in soil and growth of cabbage and tomato (미생물제재 처리에 의한 토양 미생물상의 변화 및 배추와 토마토의 생장에 미치는 영향)

  • Effect of the microbial product, which consisted of Bacillus coagulans DL-1 and rice bran, on the microorganisms in soil and growth of cabbage and tomato was investigated. Bacillus congulans DL-1 was isolated form the soil and identified in this study. Total number of microorganisms in the soil treated with the microbial product was higher than the untreated soil. The growth of cabbage and tomato on the soil treated with microbial product was faster than that on the untreated soil. The treatment of microbial product in the soil resulted in the increase of useful microorganisms, which seemed to enhance the growth of cabbage and tomato. It seemed that microbial product can increase the number of certain microorganisms and change the ratio of different species of microorganisms.

The Foliage of Flemingia (Flemingia macrophylla) or Jackfruit (Artocarpus heterophyllus) as a Substitute for a Rice Bran - Soya Bean Concentrate in the Diet of Lactating Goats

  • Ninety lactating goats (Bachthao, Barbary, Beetal and Jamnapary breeds) were used in an experiment to investigate the replacement value of the tree fodders Flemingia (Flemingia macrophylla) and Jackfruit (Artocarpus heterophyllus). The foliages were used to replace the concentrate in diets based on chopped whole sugar cane (Sacharatum sp.), Para grass (Brachiaria mutica) and dried cassava root (Manihot esculanta). The concentrate was replaced by foliage of Jackfruit or Flemingia at 0%, 20%, 40%, 60% and 80% based on the crude protein (CP) content in the concentrate and foliages, respectively. Average milk yield was 1,617 g/day for goats fed Jackfruit compared to 1,532 g/day for those fed Flemingia. Increasing amounts of Flemingia foliage resulted in reduced dry matter intake and decreased milk yield but milk composition (CP, casein and fat content) was similar up to 60% replacement. Flemingia showed a poor potential as a supplement for lactating goats and replacement levels should not exceed 20% of the protein in the concentrate or 7.5% of the dry matter in the diet. With respect to the combination of milk production and net return over the control a CP replacement rate of 20% was the most promising. For Jackfruit there was similar feed intake and milk yield at a replacement level of 20% of CP in the concentrate (9.2% of DM intake) compared to the control diet. Milk yield at the level of 40% replacement (15% of the DM) in the diet was slightly reduced. Up to a level of CP replacement rate of 60% (21% DM in the diet) can be suggested for on-farm testing as a higher net return over the control was obtained on station.

Response of Growing and Laying Japanese Quails (Coturnix japonica) to Dietary Sunflower Seed Meal

  • High fibre (CF, 242 g/kg) sunflower seed meal (SSM) was incorporated in standard grower and layer quail diets at 0, 50, 75 or 100 g/kg replacing part of the soybean meal (SBM) and deoiled rice bran (DORB). Each grower diet was offered to 30 quails housed in battery cages from day one to 5 weeks of age. At the end of the growing period, Twelve female quails from each of the four respective dietary groups were transferred to the individual laying cages and their laying performance from 7 to 20 weeks of age were evaluated on diets containing SSM at similar levels as that during the growing period. The results of growth bioassay revealed that the live weight gain, feed intake, nutrient retention and carcass characteristics of quails in different dietary groups did not vary significantly (p>0.05), although the quails fed SSM based diets utilized the feed less (p<0.05) efficiently than those fed the control diet. During the laying period, the egg production and egg quality traits remained unaffected due to dietary treatments but feed required per unit egg production increased (p<0.05) as the level of SSM in diet was increased to 100 g/kg. Although, the cost of feed per kg decreased due to the incorporation of SSM in diet but the same was not reflected in feed cost per unit gain, meat or egg production as such parameters were comparable on all the diets. It could thus be concluded that the SSM can be incorporated in quail diets upto 100 g/kg for part of SBM, an item usually expensive and also in short supply in developing countries including India.

Atomization Characteristics Experiment of Pintle Type Nozzle by the PDPA (PDPA에 의한 Pintle형 노즐의 미립화 특성실험 -식물유를 중심으로-)

  • A simplified experiment was performed to figure out the atomization characteristics of highly viscous liquid of rice-bran oil by applying ultrasonic energy to improve the atomization of spray droplets. A spray system, an ultrasonic system, and three kinds of pintle-type nozzles(pin-edge angle: 5 , 10 , 15 ) were manufactured. To investigate the effects of ultrasonic energy on the atomization of a highly viscous liquid, a phase doppler particle analyzer was used for the measurement and calculation of spray droplets data. Nozzle opening pressures were chosen of 3 levels, i.e, 10, 13, 16 MPa. As a result, it could be concluded that the ultrasonic energy was effective to improve the spray atomization when applied to the fuel by means of 3 different nozzles because of the effects of the liquid fuel cavitation and relaxation between molecules caused by ultrasonic energy. The improvement rate of the spray atomization by the ultrasonic spray atomization by the ultrasonic spray compared with the conventional spray was about 10% increase in the case of pintle type nozzles. With the increase of pin-edge angles the distribution lines by nozzle opening pressures are declined for both conventional and ultrasonic sprays. This means that the increase of the pin-edge angle improves the atomization of sprays.

Impact of Hunger Experiences in Childhood or Adolescence on Diabetes among Korean Elders (한국 노인의 당뇨병과 소년기 굶주림 경험)

  • Objectives: To investigate the relationship between experience of hunger in childhood or adolescence and diabetes mellitus in old age. Methods: Stratified national samples of 421 men and 554 women aged 60-89 in South Korea were analyzed. Diabetes diagnosed by a doctor was used as the dependent variable. Experience of hunger in childhood was assessed with the question, "In your childhood, have you ever gone hungry with skipping a meal?" and "In your childhood, have you ever eaten porridge or some bran cake as main dish instead of rice because of lack of food?" Odds ratios (ORs) and 95% confidence interval (CI) were calculated by multiple logistic regressions. Results: ORs of diabetes by experience of hunger in childhood were 1.64(95% CI 0.95-2.83) for men and 1.58(95% CI 1.01-2.47) for women. Significant association of experience of hunger in childhood with prevalence of diabetes persisted in men after adjustment for adulthood and old age socioeconomic position indicators and other risk factors(OR 1.88(95% CI 1.04-3.41)). The effect of experience of hunger in 10-19 years was more prominent than that in less than 10 years in both gender. Conclusion: Experience of hunger in childhood or adolescence may increase the risk of diabetes in Korean elders.
