Construction of Various Copy Number Plasmid Vectors and Their Utility for Genome Sequencing

  • We developed various plasmid cloning vectors that are useful in the construction of genomic and shotgun libraries. Two medium copy vectors, pCUGlblu21 (pCb21) and pAGlblu21 (pAb21), which are resistant to kanamycin ($Km^R$) and chloramphenicol ($Cam^R$), respectively, are useful for cloning DNA inserts ranging from 5kb to 15kb. Two high copy vectors, pCUGlblu31 (pCb31) and pAGlblu31 (pAb31), containing $Km^R$ and $Cam^R$, respectively, are useful for DNA inserts less than 5kb. These vectors are well adapted for large-scale genome sequencing projects by providing choice of copy number and selectable marker. The small vector size is another advantage of these vectors. All vectors contain lacZa including multicloning sites that originated from pBluscriptllsk- for easy cloning and sequencing. Two medium copy vectors contain unique and rare cutting Swal (ATTTAAAT) restriction enzyme sites for easy determination of insert size. We developed two combined vectors, pC21A31 and pC31A21, which are combinations of (pCb21 + pAb31) and (pCb31 + pAb21), respectively. These two vectors provide four choices of vectors such as $Km^R$ and medium, $Cam^R$ and high, $Cam^R$ and medium, and $Km^R$ and high copy vectors by restriction enzyme cutting, dephosphorylation, and gel purification. These vectors were successfully applied to high throughput shotgun sequencing of rice, tomato, and brassica BAC clones. With an example of extremely biased hydro sheared 3 kb shotgun library of a tomato BAC clone, which is originated from cytogenetically defined peri-centromeric region, we suggest the utility of an additional 10 kb library for sequence assembly of the difficult-to-assemble BAC clone.

Analysis of Food Habits and Nutrients Intake of Nursery School Children Living in Anyang City, based on Z-score of Weight for Height (보육시설 아동의 신장별 체중 Z-score에 따른 식습관과 식이섭취 분석 -안양시 동안구 소재 보육시설 대상-)

  • To determine food preferences and nutrients intake of nursery school children, 199 children, ages 1 to 6 years, were assessed with a structured questionnaire completed by their mothers. The questionnaire included questions related to dietary habits, mother's interest in the child's health, family history of diseases, etc. The nutrients intake was measured by 24-hour recall method. Children's weight and height were measured by trained investigators on the same day. For analysis purposes, the subjects were grouped into underweight, normal and overweight groups based on the z-score of weight for height. Only 58.3% of the subjects were grouped into the normal weight group. On the other hand, 19.1% were in the underweight group and 22.6% were in the overweight group. Forty four percent of them showed unbalanced dietary habits such as preference for picky foods, 40.0% of them disliked vegetables, and 46.7% disliked minor grains other than rice. Children in the underweight group showed a higher score of inappropriate dietary habits compared to those of normal or obese groups. The level of nutrients intake of the subjects was higher than the Korean RDA value in general, except for Ca, calories, and Zn. However, 87.4% of the children showed a higher intake level than 125% of KRDA in protein, and 18.2% and 41.4% of them showed a lower intake level than 75% of KRDA in Fe and Ca, respectively. Children in the underweight group showed a higher percentage of eating less than 75% KRDA in niacin compared to those of the normal or overweight group (p<0.05) and we found a similar tendency in Fe, Zn and Vitamin $B_1$ intake.

Hygienic Study of Traditional Foodstuffs Subjected to the Mycotoxin (Mycotoxin을 중심으로 한 전통식품의 위생학적 연구)

  • Certain Fungi Including Aspergillus flavus produce low molecular secondary metabolite that is toxic to human and animals, which have been termed mycotoxin. Given the proper humidity and temperature like summer in Korea, are capable of growing of those hazard fungi and elaborating mycotoxin on almost any organic substrate such as traditional foodstuffs and their raw materials including rice, barley, corn, meju, doenjang and gochujang etc. Until now, some people have examined to isolate various fungi such as Aspergillus sp., Penicillium sp. and Fusarium sp. from traditional foodstuffs and raw materials, and have screened various mycotoxin producing strains. Some mycotoxin contamination such as aflatoxin, ochratoxin, deoxynivalenol (DON) and zearalenone etc. also have been confirmed from similar above samples. But these data are different each other and inconsistent in experimental conditions and methods. Especially, almost experiments have been finished for one time. So more consistent experimental method and data are necessary to evaluate objectiely the safety of traditional foodstuffs subjected to the mycotoxin. For this purpose, we have to apply a new advanced technology to develop more simple and rapid methods for determination of mycotoxin and also have to concentrate our efforts on activation of research and accumulation of technology nth sustaining investment of financial support and enlargement of research installation. With those harmonious efforts, it should be possible to examine continuously nd systematically the mycotoxin contamination in our traditional foodstuffs and to assure the safety of them. Then we can maintain and develop the better traditional foodstuffs suited to internationalization.

Movements and Home-range of Mallards by GPS-Mobile based Telementary (WT-200) in Korea (야생동물위치추적기(WT-200)를 이용한 청둥오리의 이동거리 및 행동권 연구)

  • Mallard (Anas platyrhynchos) is the abundant winter visitor in South Korea. Mallard migrates long distances between Russian Siberia and Korea. This species prefers a rice paddy area as their winter habitat. We captured birds using cannon-net, and attached the GPS-Mobile phone based Telemetry(WT-200) on Seven Mallards in the winter of 2011~2013. We were monitored wintering home-range and movement distance. We analyzed the tracking location data using ArcGIS 9.0 and calculated Kernel Density Estimation (KDE) and Minimum Convex Polygon (MCP). The average home-range in the wintering ground by MCP was $118.8km^2$(SD=70.1, n=7)and the maximum home-rang was $221.8km^2$ and the minimum was $27.7km^2$. Extents of home-range by KDE were $60.0km^2$(KDE 90%), $23.0km^2$(KDE 70%) and $11.6km^2$(KDE 50%). Mallard moved an average of 19.4 km from start site(attach to WT-200 site), maximum moved was 33.2 km and minimum moved was 9.4 km. The average distance of 0.8 km between GPS fixed point(range 0.2~1.6 km), maximum moved was 19.7 km. Mallard moved a very short distance in wintering season and showed a very high water-dependent trends in wintering site.

Species Composition of Orius spp. (Hemiptera: Anthocoridae) and Their Seasonal Occurrence on Several Plants in Korea (애꽃노린재류(Orius spp.)의 종류와 몇가지 식물에서 발생소장)

  • Field survey for Orius spp., predacious natural enemies of thrips pests, on several plants by beating or sweeping method revealed that O. sauteri, O. strigicollis, O. minutus, and O. nagaii habituated in open fields or in greenhouses of Korea. Among them, O. sauteri was the most dominant species at all areas (83.0~92.5%), followed by O. strigicollis and O. minutus. Especially, O. sauteri exclusively dominated on the economically important crops such as soybean, red pepper, and watermelon. On the other hand, O. strigicallis and O. nagaii occurred mainly on ornamental plants including chrysanthemum and rose and on rice, respectively, during summer-autumn season. The seasonal occurrence varied depending on the plant. Initial population of Orius spp. was first observed on white clover in late May, followed by on soybean, red pepper, an buck wheat in early June, indcating that movement of Orius spp. relates to the flowering time of the plant. Peak of Orius spp. abundance was found first in July and secondly in August or September. On white clover and buck wheat the first peak of abundance was one week earlier than on soybean and red pepper. Therefore, we suppose that wild plants such as white clover can serve as a reservoir for Orius spp. after overwintering.

Fungal Development, Respiration and Activity of Oxidative Enzymes in Rice Plants Inoculated with Pyricularia oryzae in Both Compatible and Incompatible Combinations (벼 도열병균에 감염된 친화 및 비친화 조합 벼에서의 균생장, 호흡 및 산화효소 활성)

  • Appressorial formation of Pyricularia oryzae on leaves showed no marked difference between highly resistant Tongil and susceptible Norm No. 6. Race N-2 of the blast fungus penetrated directly into motor cells of susceptible cultivar Norm No. 6, later extensively spreading hyphae were developed, while in the cultivar Tongil, after penetration, no further hyphal extension resulted. In discoloration of infected tissues, the highly resistant cultivar Tongil not only discolored rapidly, but also the percentage of discolored cells was higher than the susceptible cultivars, Jinheung and Norm No. 6. The respiratory rate, was generally higher in infected tissue than in healthy tissue. No significant difference in the respiration rate of resistant Suwon No. 180 was not found between the infected and healthy leaf tissue, whereas, in susceptible Jinheung, a marked increase in respiratory rate was caused by blast infection. The respiratory rate increased at the appearance of the first visible symptom in all cultivars resistant or susceptible. Higher peroxidase activity was found in infected tissues as compared with healthy tissue. Peroxidase activity increased in resistant and susceptible reactions. Particularly, in resistant reaction, the increase of the activity was more pronounced. In highly resistant reaction, there was no difference in peroxidase activity between healthy and infected tissues. Ascorbic acid oxidase, hydroquinone oxidase and catechol oxidase had the same trend as the peroxidase. In contrast, activity of catalase rather decreased in leaf tissues infected with compatible races of the fungus.

Occurrence of Major Diseases in Vegetable Growing under the Furnihsed Condition in Southern Part of Korea (남부 시설원예지대의 주요병 발생생태에 관한 연구)

  • The study was conducted to obtain the basic information on the diseases epidemics of vegetable crops grown in Namji, Jinju, Gimhae and Suncheon under the extremely varied and specified conditions, plastic film house. The disease survey was conducted from the end of April, 1951 to April 1982. Leaf mold and late blight were serious foliar disease in tomato during the seedling stage, especially when the infected seeds were sown. The diseases increased rapidly 35 days after seeding. In both continuous cultivation of cucumber and rotation with upland crops, incidence of Fusarium wilt was severe while incidence of the disease was negligible in cultivations after paddy rice or grafting on pumpkin. Downy mildew of cucumber was severe in Jinju and Suncheon area, however, it was not so serious in Namji area where the growing season of cucumber was unfavorable for the maximum disease incidence. Cucumber mosaic virus disease was prevalent in the areas surveyed and the disease incidence was increased rapidly after June. Powdery mildew prevailed at the early stage of cucumber growth after transplanting in Namji area. Root rot and wilting caused by Phytophthora capsici was as the most destructive disease in pepper grown under the vinyl house, especially in Namji and Jinju area where the pepper has been cultivated intensively. The Phytophthora attacked most parts of young plants during the winter time and then induced crown rot on the adults plants. Cultivation of pepper in vinyl house was almost impossible because of the Phytophora disease by the end of June. Virus diseases to tomato plants were prevalent throughout the surveyed area and the damage was also severe. In Jinju and Gimhae area leaf mold and late blight showed high infection rate in tomato during the harvest time.

Over-wintering of the Apple Snail, Pomacea canaliculata (Gastropoda: Ampullariidae) and Its Over-wintering Habitat in the Yeongnam District (영남지역의 왕우렁이 월동 및 월동처)

  • The effect of environmental factors on the over-wintering success of the apple snail, Pomacea canaliculata, which is considered as unable to hibernate during winter climate in Korea. This study was examined to determine the location of over-wintering habitats in the agricultural water system related with weed control using the apple snail in rice paddy field in the Yeongnam district from 2003 to 2006. Over-wintering success of the apple snail was observed at Hadong, Hapchen, Sanchung, Kimhae, Milyang, Changwon, Ulsan, Busan, Gyengju, Pohang, Chilgok, Seongju, Gumi, Sangju and Youngju in Yeongnam area. And the over-wintering locations of apple snail going north year by year. Accordingly, the successful over-wintering sites observed were deep wells, water running canals and brooks not dried conditions. A special feature of this sites was that the water of this sites was not frozen during winter season.

Attractiveness of Stink Bugs to Color, Height and Location of Aggregation Pheromone Trap (집합페로몬 트랩의 색깔, 설치높이 및 장소에 따른 노린재류의 유인효과)

  • Attractiveness of stink bugs to various colors, heights and locations of fish-net traps incorporated with aggregation pheromones was determined. Bean bug, Riptortus pedestris Fabricius, was most attracted to yellow color trap, followed by white, black, green, blue and brown. R. pedestris and one-banded stink bug, Piezodorus hybneri Gmelin, were most attracted to fish-net traps placed 80 cm above the ground regardless of sexes of two species of stink bugs. Between the sexes, however, R. pedestris females were more attracted to 40 and 80 cm above the ground than the males at those heights although the attraction was not significantly different at 120 and 160 cm heights. P. hybneri females tended to be attracted to 40 and 80 cm height traps but the attraction was the opposite at 120 and 160 cm heights. The highest attraction of R. pedestris was observed in a perilla field with no significant difference in the soybean field and border area in which no crop were cultivated between the perilla and soybean fields. R. pedestris was also more attracted to the sweet persimmon orchard than the soybean and medicinal crops fields, which was a significant difference between the two fields.

The optimum media size and composition for sawdust culture of Lentinula edodes (Strain: JNM3022) (표고버섯(JNM3022) 톱밥재배에 적합한 배지조성과 배지크기)

  • These experiments were conducted to search for the optimum media size and composition for culture of Lentinula edodes (JNM3022). Considering the incubation period and the successful culturing rate, the optimum media size was length 25 $cm{\times}diameter$ 14 cm (weight 2.5 kg). The saturation moisture amount of Oak sawdust was 251 ml/100 g. It is a half of pine sawdust 495 ml/100 g. Considering the hypha growing speed and hypha density, the optimum media composition was Oak sawdust 80%+ wheat bran 20% and Oak sawdust 80%+ rice bran 20% In 10 treatments of media composition. The optimum moisture amount of media for growing Lentinula edodes (JNM3022) was 55%. Injection pipe with 20 holes in media was reduced the incubation periods to 11 days when 20cm length media and 32 days when 30cm length media compare to control. The best method for browning media surface was change temperature condition above $10^{\circ}C$ and punching the vinyl bag. This treat could shortened the browning periods to 54 days.
