Gamma Irradiation Up-regulates Expression of B Cell Differentiation Molecule CD23 by NF-κB Activation

  • Gamma irradiation ($\gamma$-IR) is reported to have diverse effects on immune cell apoptosis, survival and differentiation. In the present study, the immunomodulatory effect of a low dose $\gamma$-IR (5~10 Gy) was investigated, focusing on the role of NF-${\kappa}B$ in the induction of the B cell differentiation molecule, CD23/FceRII. In the human B cell line Ramos, $\gamma$-IR not only induced CD23 expression, but also augmented the IL-4-induced surface CD23 levels. While $\gamma$-IR did not cause STAT6 activation in these cells, it did induce both DNA binding and the transcriptional activity of NF-${\kappa}B$ in the $I{\kappa}B$ degradation-dependent manner. It was subsequently found that different NF-${\kappa}B$ regulating signals modulated the $\gamma$-IR-or IL-4-induced CD23 expression. Inhibitors of NF-${\kappa}B$ activation, such as PDTC and MG132, suppressed the $\gamma$-IR-mediated CD23 expression. In contrast, Ras, which potentiates $\gamma$-IR-induced NF-${\kappa}B$ activity in these cells, further augmented the $\gamma$-IR- or IL-4-induced CD23 levels, The induction of NF-${\kappa}B$ activation and the subsequent up-regulation of CD23 expression by $\gamma$-IR were also observed in monocytic cells. These results suggest that $\gamma$-IR, at specific dosages, can modulate immune cell differentiation through the activation of NF-${\kappa}B$, and this potentially affects the immune inflammatory response that is mediated by cytokines.

고강도 폴리머 콘크리트 휨부재의 응력블럭 (Stress Block of High Strength Polymer Concrete Flexural Members)

  • 본 연구에서는 압축강도 1400 kgf/$\textrm{cm}^2$ 를 갖는 불포화 폴리에스터 폴리머 콘크리트를 이용하여 제작한 C형 시험체에 대한 편심압축 시험을 통하여 폴리머 콘크리트 휨부재의 응력-변형률 관계를 구명하고 등가직사각형 응력블럭의 파라미터 $\alpha$, $\beta$1, ${\gamma}$를 산출하였다. 아울러 인장철근비를 달리하는 5종의 전단스팬비 4.0인 철근 보강 폴리머 콘크리트 보에 대한 휨시험에서 얻어진 극한모멘트 시험값과 C형 시험체 응력-변형률 곡선에서 산출되는 파라미터 $\alpha$=0.61, $\beta$1=0.73 를 이용하여 계산된 이론적인 극한모멘트 값을 비교하여 보았던 바, 인장철근비가 0.50 $\rho$b 이상일때 시험값과 이론값이 비교적 잘 부합됨을 확인 할 수 있었다. 그러나 폴리머 콘크리트 휨 부재에 대한 보다 정확한 설계기준의 마련을 위해서는 압축강도와 크기를 변수로 하는 C형 시험체에 대한 보다 많은 시험을 통한 파라미터의 체계적인 연구가 필요하다.

원심식(遠心式) 분무건조장치(噴霧乾燥裝置)의 작동조건(作動條件)에 따른 분무입자(噴霧粒子)의 입도예측(粒度豫測) 모델 (A Prediction Model of Droplet Size of Rotary Spray Dryer at Various Operating Conditions)

  • In an effort to localize the spray-dryer which is markedly used for drying food materials, a experiment was carried out with a wheel type atomizer locally designed and manufactured to evaluate the effect of rotational speed, feed rate and physical properties of liquid food material on the droplet size, and to develop a model to predict the droplet size sprayed at various operational conditions. The result are summarized as follows. 1. The frequency of droplet size sprayed from the atomizer at every treatment were similar to normal distributions. 2. Under the test conditions adopted in this study, that is, rotational speed of the atomizer ranging from 15,000 to 20,000 rpm (55.0 m/sec - 73.3 m/sec), feed rate from 14 to 37 kg/hr and viscosity of the material from 1.14 to 350 cP, the mean volume-surface dia. of droplets was decreased as increase in rotational speed and was not affected significantly by the feed rate and viscosity. 3. Through the dimensional analysis, a prediction model was developed as follows : $$\frac{Dvs}{r}=K[\frac{Q}{{\mu}r}]^a[\frac{rN^2}{g}]^b[\frac{{\rho}^2r^3g}{{\mu}^2}]^c[\frac{L}{r}]^d$$ and it was proved that the above model was better in degree of fitness than other models reported. 4. A prediction equation for the droplet size sprayed from the atomizer under the test was expressed as follows : $$\frac{Dvs}{r}=0.0215[\frac{Q}{{\mu}r}]^{0.06}[\frac{rN^2}{g}]^{0.3314}[\frac{{\mu}^2}{{\rho}^2r^3g}]^{0.0158}$$.

MicroRNA-27a Inhibits Cell Migration and Invasion of Fibroblast-Like Synoviocytes by Targeting Follistatin-Like Protein 1 in Rheumatoid Arthritis

  • Fibroblast-like synoviocytes (FLS) with aberrant expression of microRNA (miRNA) are critical pathogenic regulators in rheumatoid arthritis (RA). Previous studies have found that overexpression or silencing of miRNA can contribute to the development of miRNA-based therapeutics in arthritis models. In this study, we explored the effects of miR-27a on cell migration and invasion in cultured FLS from RA patients. We found that miR-27a was markedly downregulated in the serum, synovial tissue, and FLS of RA patients. Meanwhile, the expression of follistatin-like protein 1 (FSTL1) was upregulated, which suggests that FSTL1 plays a key role in RA development. The results of a Transwell assay showed that miR-27a inhibited FLS migration and invasion. However, miR-27a inhibition promoted the migration and invasion of FLS. In addition, the down-regulated expression of matrix metalloproteinases (MMP2, MMP9, and MMP13) and Rho family proteins (Rac1, Cdc42, and RhoA) was detected after treatment with miR-27a in RA-FLS by quantitative reverse transcription-PCR and western blot analysis. Then, a luciferase reporter assay validated that miR-27a targeted the 3-untranslated region (3'-UTR) of FSTL1. Moreover, miR-27a caused a significant decrease of FSTL1. In addition, the expression of TLR4 and $NF{\kappa}B$ was inhibited by miR-27a but increased by FSTL1 overexpression. In conclusion, we found that miR-27a inhibited cell migration and invasion of RA-FLS by targeting FSTL1 and restraining the $TLR4/NF{\kappa}B$ pathway.

제조합 균주 Escherochia coli가 생산하는 Bacillus stearothermophilus Acetyl Xylan Esterase의 정제 및 특성 (Purification and Characterization of Acetyl Xylan Esterase from Escherichia coli Cells Harboring the Recombinant Plasmid pKMG6)

  • Acetyl xylan esterase was produced by E. coli HB101 harboring a recombinant plasmid pKMG6 which contained the estI gene of Bacillus stearothermophilus. The maximum production was observed when the E. coli strain was grown at 37$\circC for 12 hours in the medium containing 0.5% acetyl xylan, 1.0% tryptons, 1.0% sodium chloride, and 0.5% yeast extract. The esterase produced was purified to homogeneity using a combination of ammonium sulfate fractionation, DEAE Sepharose CL-6B ion exchange chromatography and Sephacryl S-200 gel filtration. The native enzyme had an apparent molecular mass of 60 kd and was composed of two identical subunits of 29 kd. The N-terminal amino acid sequence of the polypeptide was Ala-X-Leu-Gln- Ile-Gln-Phe-X-X-Gln. The acetyl esterase displayed a pH optimum of 6.5 and a temperature opti- mum of 45$\circC. The heavy metal ions such as Ag$^{++}$, Hg$^{++}$ and Cu$^{++}$ inhibited nearly completely the activity of the esterase, and no specific metal ion was found to be required for the enzyme activity. The enzyme readily cleaved MAS, $\beta$-D-glucose pentaacetate, $\alpha$-naphthyl acetate, $\rho$-nitrophenyl acetate as well as acetyl xylan, but had no activity on $\rho$-nitrophenyl propionate, $\beta$-nitrophenyl butyrate or $\beta$-nitrophenyl valerate. The Km and Vmax values for MAS were 2.87 mM and 11.55 $\mu$mole/min, respectively. Synergistic behavior was demonstrated with a combination of xylanase and esterase from B. stearothermophilus in hydrolyzing acetyl xylan.

  • 보리차 추출액(抽出液)의 분무건조(噴霧乾燥)를 위(爲)한 분무(噴霧) 장치(裝置)로 paint용(用) spray gun을 사용(使用)할 경우 분무(噴霧)에 중요한 영향(影響) 요소(要素)로 작용하는 보리차 추출액(抽出液)의 물성(物性)을 조사하였으며, 각(各) 농도(濃度) 및 공기(空氣) 송입(送入) 압력(壓力)에서 공기일액체(空氣一液體) 혼(混) 합비(合比)를 산출(算出)할 수 있는 식(式)을 유도하였고 paint용(用) spray gun에 의해 생성(生成)되는 미세액적(微細液滴)의 범위(範圍)를 조사하였다. 1. 보리차 추출액(抽出液)의 고형분(固形分) 함량(含量)(C, %)과 비중(比重)(${\rho}$) 및 점도(粘度)$({\mu},\;cP)$ 와의 관계(關係)는 각각 C=245($C=245({\rho}-1),\;{\mu}=e^{0.17c}$과 같았다. 2. Atomizer를 통과(通過)하는 공기(空基)의 압력(壓力)(P, mmHg)과 공기(空氣)와 액체(液體)의 무게 혼합비(混合比)$(Ma/M{\ell}$)=aP+b의 관계(關係)가 성립(成立)하였다. 이 식(式)에서 a, b는 보리차 추출액(抽出液)의 농도(濃度)에 따라 정(定)해지는 계수이다. 3. 페인트용(用) spray gun의 분무(噴霧)에 의하여 형성(形成)되는 미세액적(微細液滴)은 보리차 추출액(抽出液)의 농도(濃度)가 $3.1%{\sim}25.2%$, 공기 송입(送入) 압력(壓力)이 $164{\sim}564mmHg$인 조건하(條件下)에서 $13{\mu}m{\sim}270{\mu}m$ 크기의 범위(範圍)에 있었다.

  • Ni$(C_6H_{4-n}F_n)(CO)_2$ ($C_6H_4$=benzyne, n=1-4) 착물의 전자구조 및 성질을 혼성 밀도 함수 B3LYP 이론을 이용하여 조사하였다. Benzyne 고리에 대한 방향족 성질과 벤젠핵과 무관한 화학적이동(NICS)을 분석하였다. 일-, 이- 및 삼-플루오르화 착물 중에서 3-F, 3, 6-F, 및 4-H는 각기 가장 안정한 이성질체였다. 고리 중심 상단의 여러 점에서 계산한 NICS 수치는 이들 착물의 상대 에너지에 의해 구한 값들과 일치하였다. 분자-내-원자(AIM) 분석에서 얻어진 Ni-C 결합거리는 모든 화학종에 대한 고리 임계점의 전자밀도(${\rho}_{rcp}$)와 좋은 상관성을 보였다.

토양의 용적밀도에 따른 포화수리전도도 및 음이온의 용출양상 (Effects of Soil Bulk Density on Saturated Hydraulic Conductivity and Solute Elution Patterns)

  • 시설 재배지내 염류집적의 원인의 하나인 토양의 용적밀도 변화가 염류집적에 미치는 영향을 조사하기 위해, 충남 예산군 오가면의 시설재배 10여년 연작지 토양의 Ap층을 이용하여 용적밀도 변화에 따른 수리전도도 변화와 전기전도도 및 음이온의 용출특성 변화를 토주(土柱)실험을 통해 조사하였다. 수리전도도(Y, cm/day)와 용적별도(X, $Mg/m^3$)간에는 부의 회귀관계 ($Y=-19.2X^2+6X+15.5$, (r=0.985)) 가 성립하여, 낮은 용적밀도에서는 큰 폭의 수리전도도 감소가 있으며, $1.4Mg/m^3$ 이상의 높은 용적밀도 조건에서는 용적밀도 변화에 따른 수리전도도 변화가 매우 미미 하였다. 위의 5 수준의 용적밀도 조건하에서 시간변화에 따른 용출액 중 용존 물질의 용출속도는 용적밀도 증가에 따라 크게 지연되었다. 용출액의 전기전도도가 최고점에 도달하는 시간을 비교한결과 용적밀도 $1.3Mg/m^3$ 이하에서는 토양단면 10cm를 통과하는데 약 36시간 이내의 시간이 요구되었으며, 용적밀도 $1.4Mg/m^3$ 이상에는 120시간 이상이 요구되어 두 조건사이 약 84시간의 사이가 있었다. 토양내 이행성이 높은 주요 3가지 음이온의 용출속도도 용적밀도 증가에 따라 크게 지연되었으며, 용적밀도 $1.2Mg/m^3$ 일 때 Chloride, Nitrate와 Sulphate가 90%이상 용출 되는데 각각 17, 15, 35시간이 소요된 반면, 용적밀도 $1.5Mg/m^3$ 일 때 각각은 90%이상 용출 되는데 약 130, 125, 213시간이 각각 소요되었다. 따라서 경작층 전후의 큰 용적밀도 차이를 보이고 있는 충남 예산 지역의 시설재배지내 염류집적의 원인들 중 경작층 이하인 지표면 아래 20~30cm 부근에 형성된 용적밀도(容積密度) $1.5Mg/m^3$ 이상인 경반층(硬盤層)에 의한 토양수 흐름과 각종 무기염류의 이동 지연으로 인해 크게 악화되고, 지표면에 염류집적이 가속화된 것으로 판단된다.

  • Magnetic and superconducting properties in a series of intermetallic compounds Y$_{1-x}Tb_xNi_2B_2C$ were investigated by measuring the temperature dependent magnetization, M (T), and resitivity, ${\rho}$ (T). As Tb concentration, x, is increased, the superconducting transition temperature, T$_c$, decreases and eventually disappears in the vicinity of x = 0.5 while Neel temperature, T$_N$, appears abruptly near x = 0.4 and increases linearly. Of particular interest is the collision of superconductivity and antiferromagnetism around x = 0.4. The linear decrease of T$_c$ for dilute Tb concentration seems to follow the Abrikosov-Gor'kov behavior, while the decay of T$_c$ below T$_c$ is expected to originate from the effective magnetic field on the conduction electrons. The Ginzburg-Landau theory was phenomenologically constructed to explain this competition of superconducting order parameter and antiferromagnetic order parameter with the multi-band model.

  • Novel anthracycline analogues 2-9 as potential anticancer agents were synthesized from daunomycin (1a) and doxorubicin (1b). Compounds 2, 6, and 7 were prepared by the nucleophilic displacement type esterification of a 14-bromodaunomycin (1c) with a sodium lactate, and stearic acid, respectively. Compounds 3-5 and 7-9 were prepared by the reaction of either daunomycin (1a) or doxorubicin (1b) with L-lactic and stearic acids in the presence of EDCI/PP reagents.
