Development of Leaf Mutant Cultivars of Cymbidium goeringii by Ethyl-methane-sulfonate (EMS) Treatment. (Ethyl-methane-sulfonate(EMS) 처리에 의한 춘란 잎 돌연변이 품종의 개발)

  • This study was for developing leaf chlorophyll mutant cultivars of Cymbidium goeringii by ethyl-methanesulfonate(EMS) treatment. Chlorophyll mutant rhizomes were easily produced by 0.2% EMS treatment in this genus. Among the mutants, they became dark brown about 50% of the rhizomes. When the dark-brown rhizomes were cultured in a solidified MS medium, new rhizomes were formed from part of the old ones. Chlorophyll mutant rhizomes were obtained from subcultured meristem tissues of newly-formed rhizomes. The rhizomes were cut and subcultured for a year and then became mutant plants. As the results, they produced 4 kinds of leaf mutant cultivars; zigzag-striped, comb-striped, net-striped, and dwarf types, indicating that the EMS treatment in the rhizome could produce versatile leaf chlorophyll regulating mutants. These results suggest that our method is useful for developing leaf mutant cultivars of this planta which they are estimated as higher commercial values.

Effects of Heat Pretreatment on the Quality under Storage of Fresh Ginger Rhizomes (저장 전 열처리가 생강의 이화학적 및 관능적 특성에 미치는 영향)

  • Fresh ginger rhizomes were heat-treated for 10 min, 30 min, or 60 min using hot air ($40^{\circ}C$, $50^{\circ}C$, or $60^{\circ}C$), and stored in low-density polyethylene (0.04 mm thickness) bags for 2 months at $12{\pm}1^{\circ}C$. We studied the effects of heat pretreatment on changes in gas levels after packaging, and quality characteristics of the rhizomes. Oxygen and carbon dioxide levels progressively fell and rose, respectively, as the temperature of heat treatment rose and the duration of such treatment was extended. The sprouting rate of ginger rhizomes treated at $40^{\circ}C$ was higher than that of other samples. Rotting, softening, and increasing pH of rhizomes were accelerated by treatment at higher temperature for a longer time. Weight loss and soluble solid levels were not affected by heat treatment. Sensory qualities such as appearance, odor, and overall acceptability of rhizomes fell with treatment at a higher temperature for a longer time. These results suggest that heat pretreatment has a detrimental effect on the quality of fresh ginger rhizomes.

Effects of Rhizome Length and Node Numbers on the Proliferation of Menyanthes trifoliata Cuttings (조름나물 삽목 시 지하경의 길이와 마디 수에 따른 증식효율)

  • To enhance the proliferation efficiency of Menyanthes trifoliata L. endangered species in Korea, various rhizome cuttings were tested based on cutting length and node numbers. Growth length and increased volume of rhizomes were highest on long cuttings of 10-18cm length. But relative growth length of rhizomes were higher in short cuttings of 2-4cm and medium cutting of 4-10cm length than in long cuttings. Relative increased volume of rhizomes was highest in medium cuttings. The node numbers were not contributed to the efficiency of relative growth of rhizomes. This study suggests that medium length cutting is the best practice for the proliferation of M. trifoliata.

Antioxidant Activity and Total Volatile Oil Content of Cassumunar Ginger (Zingiber montanum Roxb.) at Various Rhizome Ages

  • Cassumunar ginger (Zingiber montanum Roxb.) was grown in the experimental field at the Department of Horticulture, Kasetsart University, Thailand. The antioxidant activity and volatile oil content of rhizomes of varying age were measured. Antioxidant activity as determined using the DPPH (diphenyl-2-picrylhydrazyl) method differed significantly between samples of different ages. Antioxidant activity and rhizome age were positively correlated, with 22-month old rhizomes showing the highest radical scavenging activity (79.19%). Volatile oil was obtained by steam distillation of fresh rhizomes. The extraction yield of volatile oil was highest in l6-month old rhizomes (13.02 mL/kg). GC-FID data indicated the presence of three major compounds, sabinene, terpinen-4-ol and (E)-1-(3',4'-dimethylphenyl) butadiene (DMPBD), however none of the major components were correlated with the age of rhizome.

  • Korean folk medicine 'Sin Seong Cho' (神聖草)has been used to cure common cold, neuralgia and cancer of the stomach in Korea. The botanical origin of the crude drug has been no pharmacognostical confirmation on it. To clarify the botanical origin of 'Sin Seong Cho', the morphological and anatomical characteristics of the rhizomes of Davallia mariesii Moore and Neocheiropteris ensata (Thunb.) Ching growing in Korea were examined. As the result, it was made clear that 'Sin Seong Cho' from Korea was derived from the rhizomes of Davallia mariesii, rarely from the rhizomes of Neocheiropteris ensata.

  • One sesquiterpenoid was isolated from the methanol extract of Atractylodes rhizomes which has been used as traditional medicine for diuresis and the compound was established as selina-4(14),7(11)-dien-8-one by the spectroscopic methods. The compound$(IC_{50}<10\;ppm)$ has inhibitory effects on melanogenesis in B16 mouse melanoma.

Chemical Constituents of the Rhizomes of Sparganium stoloniferum (흑삼릉 근경의 성분)

  • The present study was carried out to evaluate biologically active components of the rhizomes of Sparganium stoloniferum and to supply the preliminary data for the chemotaxonomy and the medicinal application. Extraction and systematic fractionation of the rhizomes by column chromatography led to the isolation of six compounds from ethylacetate and n-butanol soluble fractions. Elucidation of the chemical structures of these compounds by physicochemical and apectral analysis demonstrated that compound I,II ,III,IV,V and Ⅵ were $\beta$-sitosterol, $\beta$-sitosterol-3-$\beta$-D-glucuronopyranoside, 3- (4-hydroxyphenyl)-2-propenoic acid, sorbose, 1-O-$\beta$-D-glucopyranosyl-(2S, 3R, 4E, 8Z)-2-[(2(R)-hydroxyeicosanoyl)amido]-4,8-octadecadiene-1,3-diol, and $\beta$-sitosterol-3-O-$\beta$-D-glucopyranoside, respectively.

  • In continued studies on cultivated Korean rhubarb rhizomes, three additional stilbene compounds were isolated from water extract by column chromatographic separation using Sepha dex LH-20, ODS-gel. Compound I, desoxyrhapontigenin(3,5-dihydroxy-4`-methoxystilbene), Compound II, rhapontigenin(3,3`,5-trihydroxy-4`-methoxystilbene), and compound III, piceatannol(3,3`,4`,5-tetrahydroxystilbene) were elucidated based on physico-chemical and spectroscopic evidences(UV, IR, $^1H$-NMR, $^13C$-NMR and Mass).

Rhizome Growth and Flowering of White Lotus (Nelumbo nucifera) at Muan Hoesan-Lake (무안 회산지(回山池) 백련꽃의 근경생장과 개화특성)

  • This study was conducted to examine characteristics of rhizome growth and flowering of white lotus (Nelumbo nucifera) plant in Muan Hoesan-lake for collecting the basic data on the cultivation and the flowering which is growing in Muan Hoesan-lake of Jeonnam province in Korea. We have planted from the seeds and the enlarged rhizomes of the white lotus which were planted in the plastic containers on April 20 and May 4, 2008, and cultivated under rain shelter condition. Rhizome growth pattern and the flowering characteristics of the white lotus were examined on September 4, 2008. Main rhizomes of the white lotus propagated by the seeds and the enlarged rhizomes were branched by 10% and 15%, respectively. Numbers of nodes in the bloomed and non-bloomed rhizomes of the white lotus were over 14 and below 11, respectively. Flower was not observed in the white lotus when propagated with seeds, however, flowers came out up to 80% by the enlarged rhizomes. First flowers were bloomed from the late July and to the early August, and the number of flowers were 4.0~4.4. The first flower stalks without branching of the main rhizome were emerged in 8.8th node, however, that with branching were 8.0th node. Time to flowering from the emergence of flower stalk on the above-ground parts were required 16 days. White flowers from the lotus were 15.3 cm in height and 28.2 cm in width, their longevity was 3.3 days and the averaged flower fresh weight was 29.2 g. The results conclude that optimum number of nodes on the main rhizomes for blooming the white lotus flowers should be over 8 nodes before the rhizome enlargement.

  • The anti-microbial and anti-inflammatory activities of the constituents from Atractylodes rhizomes were evaluated. Atracylone showed anti-microbial activity. Atractylone and attractylenolide I possessed considerable anti-inflammatory activity utilizing rat cotton pellet granuloma bioassay.
