Purification and Characterization of Trypsin Inhibitor from Alismatis Rhizoma and its Binding Protein, 10-Formyltetrahydrofolate Dehydrogenase (택사(Alismatis Rhizoma)로부터 트립신 저해제의 정제와 특성 규명 및 이와 결합하는 단백질, 10-Formyltetrahydrofolate Dehydrogenase에 관한 연구)

  • Alismatis Rhizoma has been used as diuretics and antiphlogistics in the Chinese oriental medicine. A trypsin inhibitor was isolated from Alismatis Rhizoma using DEAE ion exchange column, trypsin affinity column, and FPLC chromatography, and its activity and characteristics were studied. The purifed Alismatis Rhizoma trypsin inhibitor (ARTI) was estimated to be about 22 kDa. The sequence determination on N-terminal amino acid residues and 84 amino acid residues has been completed, yet no homology has been found with trypsin inhibitors reported at NCBI. ARTI did not show inhibitory activities on chymotrypsin and elastase, however it exhibited a significant inhibitory activity on bovine trypsin, and formed a complex with rat liver 10-formyltetrahydrofolate dehydrogenase.

FPTase Inhibition Effect of Protostanes from Alismatis Rhizoma and Derivatives from Alisol B 23-acetate (택사 (Alismatis Rhizoma)에서 분리한 Protostane계 화합물과 그 유도체의 FPTase 억제활성)

  • The purpose of this research is to study of inhibitory activity of protostane type triterpens against farnesyl-protein transferase (FPTase). The ingredients of Alismatis Rhizoma, alisol B 23-acetate, C 23-acetate, alisols B and A 24-acetate, and thirteen synthetic analogues from alisol B 23-acetate exhibited inhibition activity against FPTase by scintillation proximity assay method. As a result, alisol C 23-acetate, one of the constituents of Alismatis Rhizoma, the synthetic analogues carboxylated and hydroxylated on branch chain of protostane exhibited a significant inhibitory activity. However, the compounds significantly lowered the inhibitory activity, when there is no 3 position keto on protostane skeletone.

Effects of Rhizoma Alismatis on Lipid Composition and TBARS Concentration in Rat Fed High Fat Diet (택사농축액이 고지방식이를 급여한 흰쥐의 지질구성 및 TBARS량에 미치는 영향)

  • Objectives : Effects of Rhizoma Alismatis extract on lipid composition and TBARS (thiobarbituric acid reactive substances) formation were investigated in rat fed high fat diet. Methods : Seventy male Sprague-Dawley rats were used in these experiments and housed with food and water ad libitum and Twenty rats were selected healthy over 400g body weight and fed high fat diet for 8 weeks. Rhizoma Alismatis extract was administerd at dose 100mg/kg/day, 200mg/kg/day and 300mg/kg/day p.o. for 4 weeks. Then Triglyceride, Total cholesterol, HDL-cholesterol, LDL-cholesterol, TBARS concentration and ALT(Alanine Aminotransferase), AST(aspatate transaminase) activities were determined. Results : Plasma total LDL-cholesterol showed a tendancy to decrease in all treated group(P<0.05), however plasma HDL-cholesterol, total cholesterol, triglyceride showed no significantly different. Rhizoma Alismatis extract feedings resulted in a decrease in plasma TBARS(p<0.05) concentration and AST, ALT activites but contents of TBARS in liver showed no significantly different among all the treatments. Conclusions : Reviewing these experimental results, it appears that Rhizoma Alismatis extract was effective for inhibiting lipid peroxidation and decreasing LDL-cholesterol concentration.

Effects of the Extracts from Hoelen alba, Alismatis Rhizoma and Atractylodes Rhizoma on Proliferation and Differentiation of 3T3-L1 Cells (3T3-L1 세포(細胞)의 증식(增殖) 및 분화(分化)에 미치는 백복령(白茯笭), 택사(澤瀉) 및 창출(蒼朮)의 영향(影響))

  • These studies were conducted to investigate the effects of the extracts from Hoelen alba, Alismatis Rhizoma and Atractylodes Rhizoma on the proliferation and differentiation of 3T3-L1 cells. The results were summerized as follows: Hoelen alba and Alismatis Rhizoma extracts inhibited the proliferation of preadipose 3T3-L1 cells. In inductive differentiation, all three extracts inhibited the adipose conversion in 2 days of initial-culture, Atractylodes Rhizoma extract inhibited the adipose conversion in 5 days of final-culture and Hoelen alba and Alismatis Rhizoma inhibited adipose conversion in treatment of whole term of culture. In spontaneous differentiation, Atractylodes Rhizoma extract increased the adipose conversion in 2 days of initial-culture, Hoelen alba and Alismatis Rhizoma increased the adipose conversion in 5 days of final-culture, all three extracts increased adipose conversion in treatment of whole term of culture. The 10% serum of mice treated with each sample did not affect, but the 5% serum of them inhibited the proliferation of 3T3-L1 cells. In inductive differentiation, the 10% serum of them inhibited the adipose conversion in treatment of whole term of culture.

The Literature Study of Research Trend of Alismatis Rhizoma and Relationship Between the Herbology and KCD (택사(澤瀉)의 국내·외 연구동향과 『본초학』, 한국표준질병사인분류의 상관관계에 대한 연구)

  • Objectives : This study was aimed to analyze the correlation between the Herbology and contemporary research results, KCD and terms of Alismatis Rhizoma. The study will present information that can be used to find the direction of further researches and be applied to the education of Herbology.Methods : Papers were searched in Riss4U and PubMed, and categorized. Medicine or pharmacy articles about Alismatis Rhizoma were matched with the Herbology treatment and KCD. KCD-codes and terms were arranged by the Herbology treatment. The Degree of Herbology research (HDR) was calculated by numbers of papers, study method, citation rates.Results : There were 392 articles about Alismatis Rhizoma. Among these there were 141 medicine and pharmacy articles about Alismatis Rhizoma, and 51 articles were matched to the Herbology treatment. Studies about Phlegm-Fluid Retention and Dizziness, Edema and Distention and Fullness was more active than others. Analysis of other articles showed that studies about anti-cancer, anti-biotic effect, anti-diabetes mellitus, anti-oxidant effect, anti-inflammation, hepatoprotective effect were active, too. In HDR, Phlegm-Fluid Retention and Dizziness was scored by 181, Edema- Distention and Fullness by 173, Hyperlipidemia by 83, Dysuria 64, Heat strangury pain by 18, Diarrhea and decreased urine volum by 0.Conclusions : 141 articles about Alismatis Rhizoma were analyzed and 51 articles were matched to the Herbology treatment. Studies about Phlegm-Fluid Retention and Dizziness, Edema and Distention and Fullness were more active than others. And studies about anti-cancer, anti-biotic effect, anti-oxidant effect, anti-inflammation, Osteoporosis should be more studied and be more discussed among Herbology researchers.

Repeated Dose Oral Toxicity Study of Alismatis Rhizoma in SD Rats (택사(Alismatis Rhizoma) 추출물의 반복 경구투여 독성 연구)

  • Objectives : This study was carried out to evaluate the repeated dose oral toxicity of Alismatis Rhizoma in Sprague-Dawley(SD) rats. Methods : Male and female rats were administered orally with Alismatis Rhizoma water extract of 500 mg/kg(low dosage group), 1,000 mg/kg(middle dosage group) and 2,000 mg/kg(high dosage group). We daily observed number of deaths, clinical signs and gross findings for 14 days(twice a day). After 14 days, we measured body and organs weight. Also we analyzed hematological changes. Results : No dead SD rats and no clinical signs were found during the experiment period. Also other specific changes were not found between control and treated groups in hematology and serum biochemistry. In addition no significant changes of gross body and individual organs weight. Conclusions : These results suggest that water soluble extract of Alismatis Rhizoma has not repeated dose oral toxicity and oral LD50 value was over 2,000 mg/kg in SD rats. As a result, we can determine Alismatis Rhizoma is a relatively safe substance.

Antimicrobial and Antioxidative Effects of Alismatis Rhizoma Extract (택사 추출물의 항균 및 항산화 효과)

  • Antimicrobial and antioxidative effects of Alismatis Rhizoma which has been used as a galenic for diuretic and antiphlogistic were investigated. In a paper disc test, water extract of Alismatis Rhizoma showed antimicrobial activities against almost all of the microbial species tested, while their activities of methanol and ethanol extract was low than the activity of water extract, and the antimicrobial activity of water extract was most prominent against Vibrio parahaemolyticus. Antioxidative activity against linoleic acid was not observed in methanol extract. In contrast to methanol and ethnol and ethanol extract, water extract showed a antioxidative activity against linoleic acid. However, the antioxidative of water extract was lower than that of 0.1% BHT.

Determination of Alisol B 23-acetate and Alisol C 23-acetate in Alismatis Rhizoma by HPLC-ESI-MS

  • An HPLC-ESI-MS method has been developed to identify and quantify two main tetracyclic triterpenes, alisol B 23-acetate and alisol C 23-acetate in the Alismatis Rhizoma (Taeg-Sa). The relative distribution of the two triterpenes in the methanolic extract of commercially available Alismatis Rhizoma was established by selective ion monitoring (SIM) mode via electrospray ionization (ESI) source. Regression equations revealed good linear relationship, and the correlation coefficients were 0.999 and 0.998 for alisol B 23-acetate and alisol C 23-acetate, respectively, between the peak areas of the components and their concentration in a range of $0.06-2.0{\mu}g/mL$. It was found that there were significant differences in the amount of alisol B 23-acetate and alisol C 23-acetate between Korean and Chinese origins. The results showed that this method could be used to identify the two components in Alismatis Rhizoma with high sensitivity and selectivity.

  • The quality of Alismatis Rhizoma was evaluated by reversed-phase high performance liquid chromatographic method. Alisol B 23-acetate was used as a standard marker for evaluation. This component was fully separated from the other components in the plant extracts on a ODS column. Identifcation of alisol B 23-acetate was carried out by comparing the LC/MS spectrum of separated peak from the extract with that of standard. Alisol B 23-acetate contents in Alismatis Rhizoma obtained from several herbal markets were varied from $0. 15\%$ to $0.56\%$.

  • The protective and therapeutic effects of Alismatis Rhizoma extracts on rat liver injury induced by carcinogenic polycyclic hydrocarbon, benzo(a)pyrene were investigated in rats. Serum and liver triglyceride, total-cholesterol level and serum HDL-cholesterol content were measured. Alismatis Rhizoma extracts inhibited the enhanced production of lipid in chemically induced liver injury. Among 4 fractions, CHCh fraction revealed the most powerful liver-protective effect. Our results strongly suggest that Alismatis Rhizoma be a promising liver-protective agent.
