Vasodilation Effect of the Water Extract of Alcohol Steamed Rheum undulatum L. in Rat Thoracic Aorta (종대황의 주습 수치 방법에 따른 백서의 흉부대동맥 혈관이완에 미치는 영향)

  • We have examined the relaxative response to the water extract of Rheum undulatum L. (ERU) and water extract of alcohol steamed Rheum undulatum L. (SRU) with isolated thoracic aorta from sprague dawley (SD) rat. Rat thoracic aorta was investigated in vessel segments suspended for isometric tension recording by polygraph. Responses to ERU and SRU were investigated in vessels precontracted with 5-hydroxytryptamine(5-HT). We found that the thoracic aorta segments responded to ERU and SRU with a dose-dependent vasorelaxation : The thoracic aorta segments responded to ERU and SRU with a dose-dependent vasodilation. The amounts of emodin were 0.063%, 0.076% and 0.145% in ERU and SRU, respectable. The 5-HT induced contraction at 10-4M were inhibited by 85.2±4.76% and 84.0±2.91% after addition of the 0.1mg/mL water extract of ERU and SRU. The 5-HT induced contraction at 10/sup -3/M were inhibited by 100% after 10/sup -3/M emodin. In conclusion, vasodilation effect of the water extract of Rheum undulatum L. in rat thoracic aorta was not decreased according to the processing of alcohol steamed Rheum undulatum L.

The Effect of Rheum palmatum L. and Rheum undulatum L. on Rat Thoracic Aorta and Abdominal Aorta

  • Objectives: To examine the relaxational response to the water extract of Rheum palmatum L. and Rheum undulatum L. on rat thoracic aorta and abdominal aorta. Methods: Segments of thoracic aorta and abdominal aorta obtained from rats immediately after delivery were mounted in organ baths superfused on a polygraph. Results : We found that the thoracic aorta segments responded to the water extract of genus Rheum with a dose-dependent vasorelaxation. At $10^{-4}$ M 5-HT, the maximal contraction force was 93.5% of the maximum KCI-response. The 5-HT induced contractions at $10^{-4}$ M were inhibited by 86.4% and 62.1 % after addition of the high concentrations of R. palmatum root (RPR) and leaf (RPL) and R. undulatum root (RUR) and leaf (RUL). At 10 mg/ml RPR and RUR, the relaxational response at thoracic aorta and abdominal aorta with and without endothelium were 86.4%, 83.2%, 85.8%, and 62.1% of the maximum 5-HT induced contraction. Conclusion: Our result showed that RPL and RUL induced dose-dependent vasorelaxation on rat thoracic aorta and abdominal aorta, and that RPL and RUL roots have more potent effects than the leaves.

  • Oxidative stressand glia-associated chronic inflammation have been linked to the pathophysiology of Alzheimer's disease. Rhei rhizoma has been commonly used as a purgative and a haemostatic agent in traditional oriental medicine. Recently, the methanol extract from a dried root of Rheum undulatumhas been shown to have anti-allergic and anti-inflammatory effects. Anti-Apoptotic Effect of Rheum undulatum Water Extract in Pancreatic ${\beta}-cell$ Line, HIT-T15

  • Sopungsungi-won has been used as a traditional medicine for diabetes and it has been proved to be a potential remedy for type 2 diabetes mellitus. We previously reported that water extract of Sopungsungi-won exhibits anti-diabetic effects both in vivo and in vitro experiments. In the present study, we have chosen to examined anti-apoptotic effect of Rheum undulatum, which is the main component of Sopungsungi-won, on pancreatic ${\beta}-cells$, HIT-T15, against hydrogen peroxide $(H_2O_2)$. oxidative stress. To investigate the anti-apoptotic effect of Rheum undulatum water extract (RUWE) against $H_2O_2-induced$ apoptosis in pancreatic ${\beta}-cell$ line of hamster, HIT-T15, MTT assay, DAPI staining, TUNEL assay, RT-PCR and caspase-3 enzyme assay were performed. The morphological analysis demonstrated that cells treated with $H_2O_2$ exhibited classical apoptotic features, while such changes was reduced in cells pre-treated with RUWE. In addition, RUWE pre-treated cells prior to $H_2O_2$ treatment induced increase of levels of bcl-2 expression and decrease of caspase-3 enzyme activity compared to cells treated with $H_2O_2$ only. These results provide the possibility of usage of RU in patients with progressively deteriorated diabetes.

Screening and Identification of Fungicidal Compounds Derived from Medicinal Plants against Cucumber Powdery Mildew (약용식물로부터 오이흰가루병에 대한 항균성물질 탐색 및 동정)

  • This experiment was conducted to test the fungicidal activity of extracts from 50 medicinal plants to powdery mildew (Sphaerotheca fulinginea) and identify the bioactive substances. Among the medicinal plants tested, the water extract of Rheum undulatum was the most effective in spore germination inhibition, which inhibited by 100% at 200-fold dilution. Also, 50-fold dilution of water extract, 100-fold dilution of alcohol extract, 500-fold dilution of crude extract from Rheum undulatum and even 1000-fold dilution of reference chemical inhibited powdery mildew of cucumber more than 60%. 500-fold dilution of crude extract inhibited powdery mildew of cucumber 100% by twice spray treatment. There was phytotoxcity at the 100-fold dilution, but was not recognized this injury at the 500-fold dilution of crude extract. From our research to identify bioactive substance using HPLC, GLC and Mass spectrum analysis, it indicated that Rheum undulatum extract contained tentatively 1,8-dihydroxy-3-methyl-9,10-anthracenedione and 1,8-dihydroxy-3-methoxy-6-methyl-9,10-anthracenedione.

Effects of Rheum Plants on Blood Platelet Aggregation (대황류생약의 혈소판응집억제작용)

  • In order to clarify the anti-Ohyul activity of rhubarb, we investigated the effects of water extract from rhizomes of four different rhubarb on blood platelet aggregation induced by arachidonic acid, ADP, collagen and PAF in vitro. The cultivated Korean rhubarb rhizomes (Rheum undulatum) exhibited the most potent inhibitory action on the aggregation induced by arachidonic acid and also among the four fractions, stilbene components containing part showed strong inhibitory action. These inhibitory effect may partially contributed to anti-Ohyul activity of rhubarb.

Both in vivo and in vitro experiments with water extract of Sopungsungi-won have been reported to exhibit anti-diabetic effects in our previous studies. Cytotoxicity of Crude Extracts of Rheum uudulatum L. with Human Kidney Epithelial Cell A498 (신장 상피세포주 A498을 이용한 대황(Rheum undulatum L.)추출물의 세포독성)

  • We have evaluated cytotoxic effects of four crude extracts of methylene chloride, ethyl acetate, butanol, water layer isolated Rheum undulatum in A498 cell line, human kidney epithelial cells. The cytotoxic evalutation was measured by colorimetric assay using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide(MTT) , neutral red(NR) and sulforhodamine protein B(SRB). These results obtained are as follows : MTT, NR and SRB quantities were significantly decreased in cultured A498 cells treated four crude extracts by increased concentrations. The cell cytotoxic effect of crude extracts of butanol layer was more stronger than others layer. The values of MTT$\sub$50/, NR$\sub$50/, SRB$\sub$50/ of crude extract of butanol layer and were measured both 0.63 mg/ml, 0.65 mg/ml, and 0.68 mg/ml, respectively and the values of water layer were 0.84 mg/ml, 0.82 mg/ml. and 0.80 mg/ml. respectively in cultured A498 cell line.

Screening of Medicinal Plants with Antifungal Activity on Major Seedborne Disease (주요종자전염병 억제를 위한 항균성 약용식물탐색)

  • Antifungal activity on major seedborne diasease of crops was screened by the treatment of the extracts from 50 medicinal plants in vitro and in vivo. The extracts of garlic and taxus, Rheum undulatum, Achiranthes japonica, Glycyrrhiza uralensis, Oenothera lamar kiana treated with the blotting filter paper and water agar methods inhibited the growth of Pyricularia oryzae, Alternaria sesamicola, Colletotrichum gloeosporioides, and Alternaria brassicicola among the tested plants. Antifungal activities on infected seeds by soaking methods were shown even at the dilution of the extracts by 10 times. The activity was the highest in soaking seeds at $25^{\circ}C$ for 24 hours. The effect of plant extract on seed germination was not significant as compared with untreated seed. However, early growth of seedling was increased by the treatment of extracts. The extract of taxus slightly inhibited the seed germination of radish and chinese cabbage but those of Achirunthes japonica, Glycyrrhiza uralensis and Oenthfera lamarkiana showed severe damage on the seed germination and early growth of seedling.
