대황 추출물의 항위염 작용과 유효성분에 관한 연구 (A Study on the Antigastritic Effects of Rheum Species Extracts and Their Active Components)

  • 현재 대황은 지표성분의 규격이 한국, 일본과 중국이 다르고, 유통시장에서 대황과 종대황이 혼재되어 있으며 다양한 기원식물이 혼재되어 수입되고 있다. 따라서 대황의 효능을 현대 약리학적으로 해석하는 과정을 통해 약재의 표준화, 과학화 및 위염에 대한 지표성분 검색에 기여할 수 있을 것으로 사료되어 Rheum tanguticum(당고특대황)과 그 비교생약으로 Rumex cripus(양제근), Rheum officinale(약용대황), Rheum palmatum(장엽대황) 및 Rheum undulatum(종대황)의 물 및 70% 에탄올 추출물을 비교하였다. 그 결과, 흰 쥐를 이용한 위염 모델에서 양제근 70% 에탄올 추출물(73.2%)와 종대황 물 추출물(71%)이 각각 70% 이상으로 우수한 위손상 억제활성을 보여주었지만, 당고특대황의 70% 에탄올 추출물 투여시 91.8%로 실험군 중 가장 우수한 위염 억제 효과를 확인할 수 있었다. 공격인자로써의 산을 중화시킬 수 있는 능력을 알아보기 위한 제산력 시험법으로는 당고특대황 물 추출물과 70% 에탄올 추출물이 각각 약 14.2%, 약 15.8%로 억제함을 알 수 있었다. 이 결과로 보아 양성대조군으로 사용한 hydrotalcite(49.9%)보다는 제산작용이 약하였지만 과량 혹은 장기간 복용하면 효과가 있을 것으로 생각된다. 소화성궤양, 위암 및 변연부 B세포 림프종 발생에 있어 중요한 원인 인자로 밝혀졌고, 기능성 소화불량증, 위의 과형성 용종, 철결핍성 빈혈 등 기타 질환과의 연관성들이 제시되고 있는 H. pylori에 대한 colonization 억제 활성을 확인한 결과 당고특대황의 70% 에탄올 추출물 $100{\mu}g/mL$에서 H. pylori에 대한 항균작용(+)을 확인할 수 있었다. DPPH 용액을 이용하여 free radical의 소거능을 $IC_{50}$으로 나타낸 결과, 당고특대황은 물 및 70% 에탄올 추출물에서 $IC_{50}$$25.9{\mu}g/mL$, $5.8{\mu}g/mL$로 양성대조군으로 사용한 L-ascorbic acid ($IC_{50}=6.5{\mu}g/mL$)와 유사한 free radical 소거능을 나타내었다. 이 결과로 당고특대황의 70% 에탄올 추출물이 휜 쥐 위염모델의 위손상을 가장 우수하게 억제하였으며 항산화 활성 또한 가장 우수한 것으로 확인되었다. 당고특대황의 성분인 chysophanol, chrysophanol-8-O-glc, desoxyrhaponticin desoxyrhapontigenin, emodin, isorhaponticin, 2-methoxy-4-hydroxyanthraquinone-5-O-glc, physcion, piracetannol-3'-O-glc, resveratrol, rhaponticin 및 rhapontigenin을 이용하여 실험한 결과로는 효성분들 중 physcion $100{\mu}M$과 rhaponticin $50{\mu}M$$100{\mu}M$에서는 실험에 사용한 성분 들 중 비교적 우수한 colonization 억제작용(+)을 나타내었고, DPPH radical 소거 활성은 isorhaponticin, piracetannol-3'-O-glc, resveratrol 및 rhaponticin에서 $IC_{50}$이 각각 $129.5{\mu}M$, $94.1{\mu}M$ $80.1{\mu}M$$104.6{\mu}M$로 확인되었다. 실험결과를 토대로 항위염 효과가 있는 우수한 약용식물로써 천연물 의약품개발 가능성을 확인하였고 나아가 한약재의 효능을 과학적으로 검증하고 한약재 표준화에 기여할 수 있을 것으로 기대된다.

식품위해성 미생물에 대한 대황(Rheum tanguticum) 메탄올 추출물의 항균활성 및 성분분석 (Antimicrobial Activity of Methanol Extract from Rheum tanguticum against Food Hazardous Microorganisms and the Composition of the Extract)

  • 식품 위해성 세균과 곰팡이에 대하여 대황 메탄을 추출물의 항균활성을 paper disc법으로 조사하였고 최소생육저해농도를 결정하였다. 또한 GC/MS를 이용하여 대황 메탄을 추출물에서 분리된 각 peak의 성분과 함량을 분석하였다. 대황 메탄을 추출물은 p. aeruginosa에 대해 강한 항균활성을 보였으며 S. aureus와 S. typhimurium에 대해서도 상당한 항균활성을 나타내었다. 대황 메탄을 추출물은 250mg/$m\ell$농도에서 S. aureus의 생육을 저지시켰으며 300mg/$m\ell$농도에서 p. aeruginosa와 S. typhimurium의 생육을 억제시켰다. 대황 메탄을 추출물을 GC/MS로 분석한 결과 전체 120가지의 성분이 분석되었으며 이 중 추출물에 함유된 ketones 화합물이 39%로 가장 많은 부분을 차지하고 있었다. 항균물질로 알려진 phenol성 화합물인 2-methoxy-phenol, 4-vinyl-2-methoxy-phenol 등이 대황 메탄을 추출물에서도 검출되었다.

Polysaccharide Extracted from Rheum Tanguticum Prevents Irradiation-induced Immune Damage in Mice

  • Aim: To investigate the protective effect of purified fraction 1 polysaccharide extracted from Rheum tanguticum RTP1 on irradiation-induced immune damage in mice. Methods: Kunming mice were randomly divided into five groups: normal group (NC), irradiation control group (IC), RTP1 low dose (200 mg/kg), middle dose (400 mg/kg) and high dose (800 mg/kg) groups. RTP1 was adminstered by the gastric route for 14 d, mice in the NC and IC groups being given by 0.9% sodium chloride solution in the same way. The mice in all groups except NC group were irradiated with 2.0 Gy $^{60}Co{\gamma}$-ray on the fourteenth day. Immune indives of non-specific immune function, cellular immunity and humoral immunity were assessed at the 24th hour after radiation. Results: Compared with the IC group, the spleen index, thymus index, rate of carbon clearance, phagocytic function of macrophages, lymphocyte proliferation, hemolysin value of blood serum and NK activity were increased markedly (P < 0.05 or P < 0.05). Conclusion: RTP1 has an obvious protective effects on damage in ${\gamma}$-ray radiated mice.

마우스 Raw 264.7 세포에 대한 4종 대황류 추출물의 항염증 활성 비교 (Comparative Study of Extracts from Rhubarb on Inflammatory Activity in Raw 264.7 Cells)

  • Rhubarb is the well-known and frequently used herbal medicine for the treatment of constipation, inflammation, and cancer. As described in the Korea and Chinese Pharmacopoeia, rhubarb consists of the roots and rhizomes of Rheum palmatum, R. officinale, and R. tanguticum. However, the pharmacological differences among rhubarb have not been scientifically established. In the present study, we investigated and compared the inhibitory effects of 70% ethanolic extracts from R. palmatum (RPE), R. officinale (ROE), R. nobile (RNE), and R. franzenbachii (RFE) on the production of inflammatory mediators, nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), interlukin-1beta ($IL-1{\beta}$), and tumor necrosis factor-alpha ($TNF-{\alpha}$) in RAW 264.7 macrophage cells. ROE, RNE, and RFE significantly inhibited the release of NO, $PGE_2$, $IL-1{\beta}$, and $TNF-{\alpha}$. RPE significantly reduced the release of $IL-1{\beta}$, but not NO, $PGE_2$, and $TNF-{\alpha}$. Overall, RFE was found to inhibit the release of $PGE_2$ and $IL-1{\beta}$, to a far greater degree than RPE, ROE, and RNE. Our results indicate that RFE possess the strongest anti-inflammatory activity among 4 tested rhubarb.

RAPD 분석을 통한 대황(大黃)과 종대황(種大黃) 감별용 SCAR 유전자 마커 개발 (Development of SCAR Markers for the Discrimination of Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma based on the RAPD)

  • Objectives : Due to the morphological similarity and frequent occurrence of intermediate forms as well as morphological variations of aerial part, the correct identification between Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma is very difficult. To develop a reliable method for correct identification and improving the quality standards of Rhei Radix et Rhizoma and Rhei Undulatai Rhizoma, we analyzed RAPD and developed SCAR marker. Methods : To amplify target DNA at the genomic level, 32 Operon 10-mer random primers were applied with four Rheum species, R. officinale, R. palmatum, R. tanguticum and R. undulatum. The nucleotide sequences were determined and species-specific primers were prepared depending on the species-specific RAPD amplicons after subcloned into the pGEM-Teasy vector. To develop the SCAR markers, species-specific PCR amplification and multiplex-PCR were carried out using the single species-specific primer pairs and combinations of them, respectively. Results : We used RAPD analysis of four Rheum plant species to obtain several species-specific RAPD amplicons. From nucleotide sequences of these RAPD amplicons, we developed two SCAR markers that amplified 314 bp and 390 bp DNA fragments in only R. undulatum but not in R. officinale, R. palmatum, R. tanguticum and R. undulatum, for distinguishing Rhei Undulatai Rhizoma and Rhei Radix et Rhizoma. Furthermore, we established SCAR markers for the simultaneous discrimination of the three species within a single reaction by using multiplex-PCR. Conclusions : These genetic markers can be used for the efficient discrimination of plants species and commercial herbal medicines between Rhei Undulatai Rhizoma and Rhei Radix et Rhizoma, to ultimately prevent indiscriminate distribution and prescription of these herbal medicines.

유통 대황의 Sennoside A 및 Anthraquinone 함량분석을 통한 품질관리 기준에 관한 연구 (Quantitative Analysis of Sennoside A and Anthraquinones for the Guideline for the Quality Control of Rhubarbs)

  • Rhubarb (Rhei Rhizoma) has been used for the various clinical purposes such as purgative, stomach protective and pain relief for a long time. However, rhubarb in current market has a problem of quality control under which many of rhubarb fail to meet the standard specified in Korean Pharmacopoeia. This study was carried out to validate the method for the evaluation of the quality of five rhubarbs and Rumex species; Rheum palmatum, R. officinale, R. tanguticum, R. franzenbachii, R. undulatum, and Rumex species. The content of sennoside A with five anthraquinones (aloe-emodin, rhein, emodin, chrysophanol and physcion) in five rhubarbs and one Rumex has been performed by using HPLC quantitation analysis. In results, only four samples in Palmata sect. were qualified with sennoside A and those samples were R. officinale and R. tanguticum. Samples of R. palmatum did not meet the standard contents of sennoside A. The contents of anthraquinones in Palmata sect. were two times larger than those in Rhapontica sect. Moreover the content variations of anthraquinones were smaller than those of sennoside A. Thus, anthraquinones can be the key characterizing molecules to control quality of rhubarb.

패턴분석법에 의한 대황의 비교 연구 (Comparative Study of the Rhei Rhizoma by Pattern Analysis)

  • Three species, such as Rheum palmatum L., R. tanguticum Maxim. and R. officinale Baillon are recognized as the source plants of Rhei Rhizoma in Korean Pharmacopeia. However, other herbal sources such as R. undulatum L. and Rumex crispus L. have been often misused as Rhei Rhizoma. A pattern analysis method to discriminate Rhei Rhizoma in Korean Pharmacopeia from other herbal plants using HPLC and TLC chromatograms was developed. The multivariate peak data of the chromatograms of methanol extracts of Rhei Rhizoma were used for hierarchical clustering analysis, principal components analysis and similarity calculation. Besides of the statistic analysis, TLC patterns of samples could be used as criteria of the discrimination. The developed pattern analysis method was specific and could be readily utilized for comprehensive evaluation of Rhei Rhizoma.
